  • DNA markers have been studied and used intensively to identify plant species based on molecular approaches. The genus Medicago belongs to the family Fabaceae and contains 87 species distributed from the Mediterranean to central Asia. Five species of Medicago are known to be distributed in South Korea; however, their morphological characteristics alone cannot distinguish the species. In this study, we analyzed the phylogenetic relationships using collected five species of Medicago from South Korea and 44 taxa nucleotide information from NCBI. The constructed phylogenetic tree using gibberellin 3-oxidase 1 and tRNALys (UUU) to maturase K gene sequences showed the monophyly of the genus Medicago, with five species each forming a single clade. These results suggest that there are five species of Medicago distributed in South Korea. In addition, we designed polymerase chain reaction primers for species-specific detection of Medicago by comparing the plastid sequences. The accuracy of the designed primer pairs was confirmed for each Medicago species. The findings of this study provide efficient and novel species identification methods for Medicago, which will assist in the identification of wild plants for the management of alien species and living modified organisms.

  • Phytoextraction, one type of phytoremediation processes, has been widely used in the removal of heavy metals from polluted soil. This paper reviewed literature on metal uptake by plants and characterized the metal uptake by types of metals (Zn, Cu, Pb, Cd, and As), plant species, initial metal concentrations in soil and the distribution of metals in different parts of plants. The potential of metal accumulation and transport by plants was closely related to plants species, types of metals, and initial metal concentrations in soil. The plants belonging to Brassicaceae, Solanaceae, Poaceae, and Convolvulaceae families have shown the high potential capacity of Cd accumulation. The Gentianaceae, Euphorbiaceae, and Polygonaceae families have exhibited relatively high Pb uptake potential while the Pteridaceae and Cyperaceae families have shown relatively high Zn uptake potential. The Pteridaceae family could uptake a remarkably high amount of As compared with other plant families. The potential metal accumulation per plant biomass has increased with increasing initial metal concentration in soil up to a certain level and then decreased for Cd and Zn. For As, only Pteris vittata had a linear relationship between initial concentration in soil and potential of metal uptake. However, a meaningful relationship for Pb was not found in this study. Generally, the plants having high metal uptake potential for Cd or Pb mainly accumulated the metal in their roots. However, the Euphorbiaceae family has accumulated more than 80% of Pb in shoot. Zn has evenly accumulated in roots and stems except for the plants belonging to the Polygonaceae and Rosaceae families which accumulated Zn in their leaves. The Pteridaceae family has accumulated a higher amount of As in leaves than roots. The types of metals, plant species, and initial metal concentration in soil influence the metal uptake by plants. It is important to select site-specific plant species for effective removal of metals in soil. Therefore, this study may provide useful and beneficial information on metal accumulation by plants for the in situ phytoremediation.

  • The purpose of this study was to conduct phenology monitoring of forest plant species in Mt. JuWang National Park, thereby establish long-term prediction and management system for species susceptible to climate change, and utilize the result as basic materials necessary for conservation of plant genetic resources in accordance with changes in their growth environment. Global Positioning System coordinates were marked on each indicator species and a specific number ticket was provided to each plant. Changes in their blooming time, time of blossoms falling, time of leaves bursting into life, and time of leaves turning, and time of leaves falling were recorded. Investigation was made once per week from April 10 in 2010 to November 30 in 2011 except for the time period between July and August when investigation was made biweekly. The investigated plants concerned 12 kinds-nine species of trees and three kinds of herbs. According to the result of the penology monitoring of Mt. JuWang National Park, their time of leaves bursting into life, time of leaves turning, and time of leaves falling were largely earlier in 2011 than in 2010. However, it is hard to say that it is due to the factor of climate change. Long-term collection of climate data and continuous monitoring of plant phenology are considered necessary in order to examine correlation between climate change and seasonal change patterns of plants.

  • Sweet persimmon (Diospyros kaki Thunb.) is widely cultivated in the southern part of Korea and its cultivation is increasing. However, anthracnose disease caused by Colletotricuhum species is one of the major hinderances to the cultivation and production of sweet persimmon. Therefore, in the current study, PCR was used to specifically detect Colletotrichum spp., based on the sequences of the ITS II regions in the rDNA. Using the sequence data, CO-1 was designated to detect Colletotrichum together the with ITS 4 primer. The result showed that a single segment of ca. 500 bp was observed only in Colletotrichum, but not in any other fungal and bacterial isolates. The annealing temperatures and template DNA quantites were also investigated to identify optimal conditions for detection. Using these species-specific primers, a unique band was obtained at annealing temperatures ranging from $55^{\circ}C\;and\;61^{\circ}C$ and template DNA levels from 10 pg- $10{\mu}g$.
