  • The properties of sea water in deep-sea are still quite poorly understood, even though those of the surface water have been sufficiently investigated. In this study, the specific conductance of sea water was measured under the circumstances : pressure of 1 to 2,000 bars and temperature 10 to $35^{\circ}C$ as well as over the concentration of 6.228 to 19.372 per mil chloronity. The specific conductance gave the linearity with the given pressure. The initial slope, ${\triangle}k/{\triangle}P$ of the specific conductance n versus the pressure P over the pressure 1 to 1,000 bars showed a large positive value, but the second slope over the pressure 1,000 to 2,000 bars was relatively less positive value. And our equations were comparable with the empirical formular obtained by Horns et al. (1963).

Main Factors affecting on Photosynthesis under shading condition of Codonopsis lanceolata Trautv. (차광조건에서 더덕의 광합성속도에 미치는 주요 요인)

  • This study was carried out in order to investigate the main traits related to photosynthesis by multiple regression analysis. The photosynthetic rate showed more significant increase in shaded leaves of plant than in non-shaded leaves of plant after shading treatment for 44 days. Negative correlation was found between net photosynthetic rate and SLA(specific leaf area). However, the values obtained from the linear equation were different and were highly significance. From the above results, net photosynthetic rate of shading treatment is higher than that of control in the same SLA. We also found that there were correlation between stomatal conductance and SPAD as well as net photosynthetic rate and stomatal conductance. By the multiple regression analysis, SPAD value, SLA, stomatal conductance and leaf water potential showed a higher correlation coefficient of $r=0.848^{**}$. From the results, out of 4 factors(SPAD value, SLA, stomatal conductance, leaf water potential) stomatal conductance was main factor in the view points of partial regressing.

  • Effect of conductance factors on performance of vacuum system was simulated for optimum design of vacuum system. In this investigation, the feasibility of modeling mechanism for VacSim$^{Multi}$ simulator was proposed. Application specific design of vacuum system is required to meet the particular process conditions for various industrial implementations of vacuum equipments. Geometry and length, diameter of exhaust pipeline were modeled as simulation modeling variables for conductance effects. Series vacuum system was modeled and simulated with varied dimensions and structures of exhaust pipeline. Variation of pipeline diameter showed the more significant effects on vacuum characteristics than that of pipeline length variations. It was also observed that the aperture structure of pipeline had the superior vacuum characteristics among the modeled systems.

Expression of $Ca^{2+}$-activated $K^+$ Channels and Their Role in Proliferation of Rat Cardiac Fibroblasts

  • Cardiac fibroblasts constitute one of the largest cell populations in the heart, and contribute to structural, biochemical, mechanical and electrical properties of the myocardium. Nonetheless, their cardiac functions, especially electrophysiological properties, have often been disregarded in studies. $Ca^{2+}$-activated $K^+\;(K_{Ca})$ channels can control $Ca^{2+}$ influx as well as a number of $Ca^{2+}$-dependent physiological processes. We, therefore, attempted to identify and characterize $K_{Ca}$ channels in rat Cardiac fibroblasts. First, we showed that the cells cultured from the rat ventricle were cardiac fibroblasts by immunostaining for discoidin domain receptor 2 (DDR-2), a specific fibroblast marker. Secondly, we detected the expression of various $K_{Ca}$ channels by reverse transcription polymerase chain reaction (RT-PCR), and found all three family members of $K_{Ca}$ channels, including large conductance $K_{Ca}$ (BK-${\alpha}1-\;and\;-{\beta}1{\sim}4$subunits), intermediate conductance $K_{Ca}$ (IK), and small conductance $K_{Ca}$ (SK$1{\sim}4$ subunits) channels. Thirdly, we recorded BK, IK, and SK channels by whole cell mode patch clamp technique using their specific blockers. Finally, we performed cell proliferation assay to evaluate the effects of the channels on cell proliferation, and found that the inhibition of IK channel increased the cell proliferation. These results showed the existence of BK, IK, and SK channels in rat ventricular fibroblasts and involvement of IK channel in cell proliferation.

Effects of Pharmacological Modulators of $Ca^{2+}-activated\;K^+$ Channels on Proliferation of Human Dermal Fibroblast

  • Employing electrophysiological and cell proliferation assay techniques, we studied the effects of $Ca^{2+}$ -activated $K^+$ channel modulators on the proliferation of human dermal fibroblasts, which is important in wound healing. Macroscopic voltage-dependent outward $K^+$ currents were found at about -40 mV stepped from a holding potential of -70 mV. The amplitude of $K^+$ current was increased by NS1619, a specific large-conductance $Ca^{2+}$-activated $K^+$ (BK) channel activator, but decreased by iberiotoxin (IBTX), a specific BK channel inhibitor. To investigate the presence of an intermediate-conductance $Ca^{2+}$-activated $K^+$ (IK) channels, we pretreated the fibroblasts with low dose of TEA to block BK currents, and added 1-EBIO (an IK activator). 1-EBIO recovered the currents inhibited by TEA. When various $Ca^{2+}$-activated $K^+$ channel modulators were added into culture media for 1∼3 days, NS1619 or 1-EBIO inhibited the cell proliferation. On the other hand, IBTX, clotrimazole or apamin, a small conductance $Ca^{2+}$-activated $K^+$ channel (SK) inhibitor, increased it. These results suggest that BK, IK, and SK channels might be involved in the proliferation of human dermal fibroblasts, which is inversely related to the channel activation.

Modulation of Large Conductance $Ca^{2+}-activated$ $K^+4$ Channel of Skin Fibroblast (CRL-1474) by Cyclic Nucleotides

  • Potassium channels in human skin fibroblast have been studied as a possible site of Alzheimer disease pathogenesis. Fibroblasts in Alzheimer disease show alterations in signal transduction pathway such as changes in $Ca^{2+}$ homeostasis and/or $Ca^{2+}-activated$ kinases, phosphatidylinositol cascade, protein kinase C activity, cAMP levels and absence of specific $K^+$ channel. However, little is known so far about electrophysiological and pharmacological characteristics of large-conductance $Ca^{2+}$-activated $K^+$ ($BK_{Ca}$) channel in human fibroblast (CRL-1474). In the present study, we found Iberiotoxin- and TEA-sensitive outward rectifying oscillatory current with whole-cell recordings. Single channel analysis showed large conductance $K^{+}$ channels (106 pS of chord conductance at +40 mV in physiological $K^+$ gradient). The 106 pS channels were activated by membrane potential and $[Ca^{2+}]_i$, consistent with the known properties of $BK_{Ca}$ channels. $BK_{Ca}$ channels in CRL-1474 were positively regulated by adenylate cyclase activator ($10{\mu}M$ forskolin), 8-Br-cyclic AMP ($300{\mu}M$) or 8-Br-cyclic GMP ($300{\mu}M$). These results suggest that human skin fibroblasts (CR-1474) have typical $BK_{Ca}$ channel and this channel could be modulated by c-AMP and c-GMP. The electrophysiological characteristics of fibroblasts might be used as the diagnostic clues for Alzheimer disease.

Effects of Ozone on Crops and Protective Effects of Ethylenediurea as an Anti-Oxidant

  • Phytotoxic effects of ozone and ethylenediurea (EDU) on soybean (Glycine max) and spinach (Spinacia oleracea) were observed by using open-top field chamber system (OTC). Gas exchange rates (photosynthesis. stomatal conductance and transpiration rates) of soybean plants were decreased by 20% to 30% by ambient ozone and resulted in 30% reduction of seed yields. In OTC. ambient ozone and 0.12 $\mu$l/l $O_3$ decreased gas exchange rates of spinach by 25% to 40% and by 50%. respectively. The protective effect of EDU against ozone induced injury was obtained at 100 mg/l on soybean. and at 250 mg/l on spinach, respectively. The excessive application of EDU. however. inhibited photosynthesis. transpiration. and stomatal conductance without any specific visible damage.

  • In the experiment carried out on 20 college students, recorded were frontal, temporal and occipital EEG, skin conductance response, skin conductance level, heart rate and respiration rate during listening to two music fragments with different affective valences and white noise administered immediately after negative visual stimulation. Analysis of physiological patterns observed during the experiment suggests that affective auditory stimulation with music is able to selectively modulate autonomic and cortical activity evoked by preceding aversive visual stimulation and to restore initial baseline levels. On other hand, physiological responses to white noise, which does not possess emotion-eliciting capabilities, evokes response typical for orienting reaction after the onset of a stimulus and is rapidly followed by habituation. Observed responses to white noise were similar to those specific to attention only and had no evidence for any emotion-related processes. Interpretation of the obtained data is considered in terms of the role of emotional and orienting significance of stimuli, dependence of effects on the background physiological activation level and time courses of attention and emotion processes. Physiological parameters are summarized with regard to their potential utility in differentiation of psychological processes induced by auditory stimuli.

  • We cloned the cDNA encoding the neuron-specific $\beta$-subunit ($\beta$4) of large-conductance calcium-activated potassium channels from rat brain and determined the DNA sequences of the entire coding region (GenBank accession; AY028605). The deduced amino acid sequences of r$\beta$4, 210 amino acids in length, are closely related to the $BK_{Ca}$ $\beta$4 subunits of other species but show only limited sequence homology to other $\beta$-subunits, $\beta$1-$\beta$3.(omitted)d)

  • We examined the psychophysiological effects of navigation in a virtual reality (VR). Subjects were exposed to the VR, and required to detect specific objects. Ten electrophysiological signals were recorded before, during, and after navigation in the VR. Six questionnaires on the VR experience were acquired from 45 healthy subjects. There were significant changes between the VR period and the pre-VR control period in several psychophysiological measurements. During the VR period, eye blink, skin conductance level, and alpha frequency of EEG were decreased but gamma wave were increased. Physiological changes associated with cybersickness included increased heart rate, eye blink, skin conductance response, and gamma wave and decreased photoplethysmogram and skin temperature. These results suggest an attentional change during VR navigation and activation of the autonomic nervous system for cybersickness. These findings would enhance our understanding for the psychophysiological changes during VR navigation and cybersickness.
