Two Gram-negative, nonmotile, coccobacilli, SW-$3^T$ and SW-$100^T$, were isolated from sea water of the Yellow Sea in Korea. Strains SW-$3^T$ and SW-$100^T$ contained ubiquinone-9 (Q-9) as the predominant respiratory lipoquinone and $C_{18:1}\;{\omega}9c$ and $C_{16:0}$ as the major fatty acids. The DNA G+C contents of strains SW-$3^T$ and SW- $100^T$ were 44.1 mol% and 41.9 mol%, respectively. A neighbor-joining tree based on l6S rRNA gene sequences showed that the two isolates fell within the evolutionary radiation enclosed by the genus Acinetobacter. Strains SW-$3^T$ and SW-$100^T$ exhibited a l6S rRNA gene similarity value of 95.7% and a mean DNA-DNA relatedness level of 9.2%. Strain SW-$3^T$ exhibited l6S rRNA gene sequence similarity levels of 93.5-96.9% to the validly described Acinetobacter species and fifteen Acinetobacter genomic species. Strain SW-$100^T$ exhibited l6S rRNA gene sequence similarity levels of less than 97.0% to the other Acinetobacter species except Acinetobacter towneri DSM $14962^T$ (98.0% similarity). Strains SW-$3^T$ and SW-$100^T$ exhibited mean levels of DNA-DNA relatedness of 7.3-l6.7% to the type strains of some phylogenetically related Acinetobacter species. On the basis of phenotypic, phylogenetic, and genetic data, strains SW-$3^T$ and SW-$100^T$ were classified in the genus Acinetobacter as two distinct novel species, for which the names Acinetobacter marinus sp. novo (type strain SW-$3^T$=KCTC $12259^T$=DSM $16312^T$) and Acinetobacter seohaensis sp. novo (type strain SW-$100^T$=KCTC $12260^T$=DSM $16313^T$) are proposed, respectively.
This study was conducted to analyze the morphological characteristics such as flower color, flower type, flower diameter and flower stalk, and the main annual production yield, and genetic similarity of twenty four Gerbera species growing in Korea. Most of flower colors were pink. The numerical order of flower color was pink, orange, red, double-colored, and milk-white. Majority of flower types were sin81e or semidouble flowers. A few species were double flowers. flower diameters were from 7 ㎝ to 12 ㎝, showed significant differences compared to other characteristics. Flower stalks were ranged from 55 ㎝ to 65 ㎝. Only one species was the shortest as 55 ㎝. The others were similar size as about 65 ㎝. Main annual production yields were between 190 and 400 blossoms. Fifty seven reproducible polymorphic bands from eighty primers were used for analyses of genetic similarity. The genetic similarity of 24 collected Gerberas was largely classified into five groups. The average similarity coefficient was 0.72 ranged from 0.50 to 0.90. The highest similarity coefficient was shown between 'Sardana' with red/white flower color and double flower type, and 'Tamara' with orange flower color and double flower type as 0.90.
As a part of the research program 'Survey of freshwater organisms and specimen collection', freshwater samples were collected from Lakes Soyang and Chungju in 2016. Hundreds of bacterial strains were isolated from the samples and were identified based on 16S rRNA gene sequences. Among the bacterial isolates, strains showing higher than 98.7% sequence similarity with validly published bacterial species not reported in Korea were selected as unrecorded bacterial species. Based on 16S rRNA gene sequence similarity, 17 strains were identified as unrecorded bacterial species in Korea. The 17 bacterial strains were phylogenetically diverse and belonged to four phyla, seven classes, 13 orders, 14 families, and 16 genera. At generic level, the unreported species were affiliated with Caulobacter, Paracoccus, and Mesorhizobium of the class Alphaproteobacteria, Deefgea, Undibacterium, Chitinimonas, Inhella, and Sphaerotilus of the class Betaproteobacteria, Vibrio and Cellvibrio of the class Gammaproteobacteria, Sanguibacter and Clavibacter of the phylum Actinobacteria, Lactococcus of the phylum Firmicutes, Deinococcus of the class Deinococci, and Chryseobacterium and Flavobacterium of the phylum Bacteroidetes. The unreported species were further characterized by examining Gram reaction, colony and cell morphology, biochemical properties, and phylogenetic position. The detailed description of the 17 unreported species are also provided.
Introgression from genetically modified plants (GMPs) may be dependent on the genetic similarity to wild relative plants. In Korea, many wild plant species are botanically related to the cultivated plants that have a potential to be genetically transformed. The controversy for hazards of GMPs is continuing because the studies on gene flow or introgression are little. Based on the systematic criteria, we have surveyed Korean wild plant species that showed the similarity to cultivating crops. The consideration for feasibility of genetic pollution (introgression of transgene) is necessary for the successful accomplishment in the practical use of GMPs. Although the detrimental effects of GMPs on wild relatives have not been clearly verified, Korean wild plant species related to crop plant (potential GMP) have to be investigated with respect to the introgression. Korean flora consists of ca. 5,500 species. Among them, 1,448 species are classified as weed species (966 native, 325 naturalized, and 167 escaped ones), which is vulnerable to GMPs in term of introgression. We suggested the principal Korean wild plants related to major crops that might be affected by GMPs via introgression. The investigated species herein are selected based on the morphological and phenological relationship. It is necessary to verify the genetic relationship between cultivated plants and wild relatives sing more precise molecular techniques, which provide the information of likelihood for the introgression of transgene.
The molecular taxonomic relationship of nine species in the genus Moniliella Stolk & Dakin and Trichosporonoides Haskins & Spencer and six species of other yea나-like fungi was examined by sequencing analysis of large subunit rDNA D1/D2 variable domain. The fifteeen species fell into two major groups corresponding with their genetic relationships. The nine species of the genus Moniliella and Trichosporonoides were placed at the same cluster. similarity values based on the D1/D2 domain sequences were 45.4-100% among species of genus Moniliella, 45.2-84.4% among genus Trichosporonoides species, and 45.6-90.1% among species of genus Moniliella and Trichosporonoides. Identical sequence similarity was observed between M. suaveolens var. nigra and M. suaveolens. A colse relationship of M. mellis. and M. acetoabutens is observed. The result of this study provided and insight into the genetic origins of genus Moniliella and Trichosporonoides species as well as their genetic relationships. Genus Moniliella and Trichosporonoides are closely related to each other based on sequence analysis of the large subunit rDNA D1/D2 region and we suggest combination of the genus Moniliella and Trichosporonoides to single genus.
Mt. Yeonae is at Gijang-gun in Busan and is surrounded by farming lands on three sides. The search for the species composition and dynamics of local communities were studied at Mt. Yeonae of how spatial similarity decays with geographic distance. The index values of Z$\ddot{u}$rich-Montpellier School's phytosociology at the 12 plots was compared to a distribution of similarly using 20 m quadrates at 12 sites. The specific communities were five including Pinus densiflora - Quercus variabilis community. Six species were significant similarity between neighboring sites by using the spatial autocorrelation coefficient, Moran's I. If Mt. Yeonae was destroyed by an artificial action, some spatial correlated species such as P. densiflora and Q. variabilis will be collapsed because of no maintaining the effective population sizes.
Seagrasses are marine angiosperms of ecological importance in providing shelter and food to aquatic species as well as maintaining the carbon cycle on earth. Phyllospadix iwatensis is a seagrass of the family Zosteraceae and is distributed along the eastern coast of Korea. The nucleotide sequences of P. iwatensis nuclear genes encoding 18S ribosomal RNA (rRNA) and internal transcribed spacer-1 (ITS-1) were determined for molecular phylogenetic analysis. Genomic DNA was isolated from P. iwatensis and used for PCR amplification of 18S rRNA and ITS-1. Examination of the 18S rRNA sequence of P. iwatensis showed a close (99% similarity) relationship to Zostera noltii, another genus of Zosteraceae, but a distant (84% similarity) evolutionary relationship to other macroalgal Laminariales species. Further discrepancies found in ITS-1 nucleotide sequences between closely related species indicate that the sequence information could be used for species identification.
Habitat pattern and distribution of the water birds were analyzed along the Hyungsan River on nine sites during fifteen months from January 2001 to March 2002. Water birds including 16,186 individuals and 86 species in total were observed from all the surveyed sites at the downstream of the Hyungsan river. Site 5, which has been observed 4,669 individuals and 60 species of the birds, was the most favorite habitat for the water birds in the Hyungsan River. And the diversity of water birds from the Hyungsan river showed the fewest species and individual numbers of the water birds at the site 2 located at the estuary of the river and site 3, the small reservoir for filtering of the draining water from the industrial area. The similarity between habitats in the Hyungsan river was divided of three groups by the cluster analysis with the bird species and individual numbers such as coastal area, estuary sites, and rice paddy field areas.
The purpose of this study is to evaluate the vegetation recovery status of Mudeungsan National Park Jungmeorijae, Jeungsimsa district restoration site, and the Shimwon Valley ecological landscape restoration site in Jirisan National Park. Compared to the control plots, the Jungmeorijae restoration site was analyzed to have height growth of about 73.5%, the average species diversity index of about 75.2%. and the average similarity index was recovered to 7.75%. In the case of the restoration site in Jeungsimsa district, the height growth compared to the control plots was about 69.2%, the average species diversity index was about 55.0%. and the average similarity index was recovered to 25.65%. In the case of the Shimwon Valley ecological landscape restoration area, the height growth compared to the control plots was about 32.6%, the average species diversity index about 176.7%. and the average similarity index was recovered to 0.85%. The restoration site of the Jeungsimsa district was planted with relatively large trees during restoration work, and it took a relatively long time(20 years). Also, the site had less limiting factors due to the low elevation, allowing the degree of vegetation recovery to be higher than that of other sites.
DNA-DNA hybridization has been established as an important technology in bacterial species taxonomy and phylogenetic analysis. In this study, we analyzed how the efficiency with which the genomic DNA from one species hybridizes to the genomic DNA of another species (DNA-DNA hybridization) in microarray analysis relates to the similarity between two genomes. We found that the predicted DNA-DNA hybridization based on genome sequence similarity correlated well with the experimentally determined microarray hybridization. Between closely related strains, significant numbers of highly divergent genes (>55% identity) and/or the accumulation of mismatches between conserved genes lowered the DNA-DNA hybridization signal, and this reduced the hybridization signals to below 70% for even bacterial strains with over 97% 16S rRNA gene identity. In addition, our results also suggest that a DNA-DNA hybridization signal intensity of over 40% indicates that two genomes at least shared 30% conserved genes (>60% gene identity). This study may expand our knowledge of DNA-DNA hybridization based on genomic sequence similarity comparison and further provide insights for bacterial phylogeny analyses.
