A report of 29 unrecorded bacterial species belonging to the phylum Bacteroidetes in Korea

  • Within a comprehensive, widescale investigation of indigenous prokaryotic species in Korea, 29 bacterial strains in the phylum Bacteroidetes were isolated from diverse environmental habitats that included soil, plant roots, natural caves, tidal flats, freshwater from lakes, and seawater. Based on their high 16S rRNA gene sequence similarities (>99.1%) and the formation of robust phylogenetic clades with the closest type species, each strain likely belonged to an independent and predefined bacterial species. There are no publications or official reports of the isolation of these 29 species in Korea. Our study provides strong evidence that seven species in three genera in the order Cytophagales, 15 species in 13 genera in the order Flavobacteriales and seven species in five genera in the order Sphingobacteriales, all within the phylum Bacteriodetes, are new reports of bacterial species in Korea. Gram reaction, colony and cell morphology, basic biochemical characteristics, isolation source, and strain IDs are described in the species description section.

광양만 및 섬진강 하구에서 채집된 어류의 공간적 군집 특성 (Spatial Characteristics of Fish Assemblage in Seomjin River Estuary and Gwangyang Bay)

  • 섬진강 하구와 광양만은 국내에서 유일하게 대규모 둑이 없는 자연상태를 유지하고 있어 생태학 및 경제학적으로 굉장히 중요한 위치에 있다. 또한, 다양한 어류의 주요 서식처로 알려져 있는 해역이다. 어류 군집에 대한 정보는 환경상태를 평가할 수 있는 적합한 지표군이기 때문에 지속적인 모니터링이 필요하다. 본 연구에서는 2020년~2021년 5월과 9월에 총 8지점 섬진강 하구 (ES1~ES3)와 광양만 (ES4~ES8)에 출현하는 어종을 파악하고 어류 군집구조의 시공간적 변화를 파악하기 위해 조사를 진행하였다. 채집된 어류는 총 2강 14목 42과 68속 78종, 5,111개체가 채집되었다. 조사기간 내 채집된 우점종으로는 보구치가 2,370개체가 채집되었으며, 상대 풍부도 (Relative abundance)는 46.4%로 나타났다. 아우점종은 주둥치가 1,643개체가 채집되어 상대 풍부도 32.1%가 나타났다. 어류 군집은 Bray-Curtis 유사도에 따라 총 3개의 그룹으로 나타났으며, 각각 기수역, 연안의 내측과 외측으로 나누어졌다. 1그룹은 주로 담수역 지점과 기수역에서 서식하는 어류가 주로 채집되었으며, 3그룹에서는 연안에 서식하는 어류들만 채집되었다. ANOSIM (Analysis of similarity) 분석을 진행한 결과, Group 2와 Group 3 사이에서 가장 큰 유의한 차이 (ANOSIM R=0.783, p=0.001)가 나타났고, 두 그룹에 기여하는 종은 보구치와 갯장어였다. 본 연구는 섬진강 하구부터 연안 외측까지 공간에 따른 어류 군집 변화를 나타내었다. 이러한 결과는 공간에 따라 변화하는 주요종에 대한 장기적인 모니터링을 제시하며, 경제성이 높은 어종의 보호와 관리를 위한 중요한 기초 자료로 활용될 것으로 사료된다.

A report on 24 unrecorded bacterial species of Korea isolated in 2016, belonging to the orders Rhizobiales and Sphingomonadales in the class Alphaproteobacteria

  • In 2016, as a part of the research program 'Survey of Korean Indigenous Species', diverse environmental samples were collected from various sources of freshwater, seawater, soil, wetland, reclaimed land, sand, pine forest, plant root, ginseng field, solar saltern, and caves. Thousands of bacterial strains were isolated from the diverse samples and identified based on 16S rRNA gene sequence analyses. The present study, as a phylogenetic subset of the primary research program, reports 24 unrecorded bacterial species in Korea that belong to the orders Rhizobiales and Sphingomonadales in the class Alphaproteobacteria. Based on the high 16S rRNA gene sequence similarities (>98.8%) and formation of a robust phylogenetic clade with the closest type species, it was determined that each strain belonged to each independent and predefined bacterial species. There is no official report that these 24 bacterial species have been described in Korea; therefore, 10 species of nine genera in the order Rhizobiales and 14 species of seven genera in the order Sphingomonadales are described for unreported alphaproteobacterial species in Korea. Gram reaction, colony and cell morphology, biochemical properties, and isolation sources are also provided in the species description section.

  • A total of 38 bacterial strains within the classes Bacilli and Deinococci were isolated from various sources in Korea. Samples were collected from animal intestine, urine, soil, tidal flat mud, and kimchi. In the sequence comparison and phylogenetic analysis of 16S rRNA sequences, the 38 isolates were assigned to the classes Bacilli and Deinococci with sequence similarities more than 98.7%. Twenty-four strains and 13 strains were classified the order Bacillales and Lactobacillales in the class Bacilli, respectively. In the order Bacillales, there were nine species in the genus Bacillus, seven species in the genus Paenibacillus, and the remaining eight species in the genera Domibacillus, Halobacillus, Virgibacillus, Lysinibacillus, Paenisporosarcina, Planococcus, Savagea, and Staphylococcus. In the order Lactobacillales, there were four species in the genus Lactobacillus, three species in the genus Leuconostoc, three species in the genus Lactococcus, and the remaining three species in the genera Aerococcus, Enterococcus, and Streptococcus. One species was related to the genus Deinococcus of the order Deinococcales. Most of the isolated strains were Gram-stain-positive, but some were Gram-stain-variable or Gram-stain-negative. Cells were rod or cocci-shaped. Based on the results of 16S rRNA analysis, we report 38 strains as previously unrecorded species to Korea, and the basic characteristics of strains are described herein.

열린 하구인 섬진강 하구 및 광양만 연안 어류 군집의 시공간적 변화 (Spatio-temporal Variation of Fish Communities in Open Estuary, Seomjin River Estuary and Gwangyang Bay Coast)

  • 강-하구-해양 연속체는 지구상에서 가장 생산적이고 역동적인 수중 생태계 중 하나이다. 어류는 생활사, 섭식 습관, 생태적 지위가 서로 다르고, 광범위하기 때문에 이를 통해 다양한 시공간적 규모에서 환경 상태를 평가할 수 있는 적합한 지표 생물이다. 2019년 3월부터 10월까지 3회에 걸쳐 섬진강-섬진강 하구-광양만 연안 연속체 내에서 조사된 어류는 멸종위기종 2종을 포함하여 31과 49종으로 나타났다. Bray-Curtis 유사도에 따라 2개의 그룹(1, 2그룹)과 두 그룹 내 속하지 않는 2개의 지점(3, 4그룹)으로 구분되었는데, 1그룹은 담수역 지점과 기수역 지점들이 주로 구성되었으며, 2그룹은 광양만 연안역 지점들인 ES4~8로만 구성되었다. 공간적인 변수와 시간적인 변수를 이용하여 ANOSIM 분석을 진행한 결과, 섬진강-광양만 연속체의 어류 군집은 계절적인 요인(ANOSIM R=0.273, P=0.002)의 차이보다 공간적인 요인(ANOSIM R=0.398, P=0.001)의 차이가 상대적으로 더 중요하였다. 구분된 그룹들 중에서도 특히 1그룹과 2그룹 간에 유의한 차이가 존재하였으며(ANOSIM R=0.556, P=0.001), 보구치(9.4%), 날개망둑(6.9%), 전어(5.9%)가 이 두 그룹의 차이에 크게 기여하는 분류군으로 나타났다. 섬진강-광양만 생태계 내 분포하는 어류상은 공간적으로 크게 구분되었으며, 종수와 개체수는 계절적인 차이를 보이는 것으로 나타났다. 본 연구는 강-하구-해양 연속체의 연속적인 환경 내에서 분포하는 어류 군집의 변화, 주요어종의 보호 및 관리를 위한 기초자료로 활용될 수 있을 것이다.

  • A new species of marine Halacaridae, Copidognathus laevisetosus is recorded from Yeongdeok coast, Korea. In morphological aspects, the present new spe챠es is closely related to C. koreanus Chatterjee and Chang. Morphological similarities and dissimilarities between them are discussed. Copidognathus cerberoideus Bartsch is reported here for the first time from Korea and away from its type locality.

제주산 짠물응애류 (응애목: 짠물응애과)의 1신종 1미기록종 (A New Species and A New Record of Halacarid Mites (Acari: Halacaridae) from Jejudo Island, Korea)

  • 제주 해안의 조간대 해조틈에서 채집한 짠물응애과의 1신종과 1 한국미기록종을 기록한다. 신종인 제주뽀족입짠물응애 (Copidognathus jejuensis n. sp.)는 C. dentatus. C. eblingi, C. dentipes 등과 밀접한 형태적 유사성을 보여주고 있다. 이들 종과 신종간의 형태적 유사점과 상이점들을 고찰하였다. 한국미기록종인 곰보뽀족입짠물응애 (C. polyporus)는 모식산지 외의 지역에서는 최초로 보고된다. 지금까지 한국에서 기록된 뽀족입짠물응애속의 5종에 대한 검색표를 작성하였다.

김치로부터 Lactobacillus sakei 생육저해 Leuconostoc 및 Weissella 속 균주의 분리 (Isolation of Leuconostoc and Weissella Species Inhibiting the Growth of Lactobacillus sakei from Kimchi)

  • Kimchi is a group of traditional fermented vegetable foods in Korea and known to be the product of a natural mixed-fermentation process carried out principally by lactic acid bacteria (LAB). According to microbial results based on conventional identification, Leuconostoc mesenteroides and Lactobacillus plantarum were considered to be responsible for the good taste and over-ripening of kimchi, respectively. However, with the application of phylogenetic identification, based on 16S ribosomal RNA gene similarities, a variety of Leuconostoc and Lactobacillus species not detected in the previous studies have been isolated, together with a species in the genus Weissella. Additionally, Lactobacillus sakei has been accepted as the most populous LAB in over-ripened kimchi. In this study, Leuconostoc and Weissella species inhibiting the growth of Lb. sakei were isolated from kimchi for future applications to do with kimchi fermentation. From 25 kimchi samples, 378 strains in the genera Leuconostoc and Weissella were isolated and 68 strains identified as Lc. mesenteroides, Lc. citreum, Lc. lactis, W. cibaria, W. confusa, and W. paramesenteroides exhibited growth inhibition against Lb. sakei. Most of the strains also had antagonistic activities against Lb. brevis, Lb. curvatus, Lb. paraplantarum, Lb. pentosus, and Lb. plantarum. Their antagonistic activities against Lb. sakei were more remarkable at lower temperatures of incubation.

A Benthic Polychaete Assemblage off the Korean South Coast(Gwangyang Bay and Yeosu Sound)

  • We investigated the benthic polychaete assemblage in Gwangyang Bay and Yeosu Sound in February 1997. The sediment was an almost entirely muddy facies. The benthic macrofauna comprised 295 species occurring at a mean density of 875 $indiv./m^2$. Polychaetes were the major faunal component; there were 94 species at mean density 765 $indiv./m^2$. The highest abundance and species richness occurred in the Myodo south and north channels, in the mouth of Gwangyang Bay, and in the Noryang channel mouth. The most abundant polychaete was Tharyx sp. (47.9%), followed in rank order by Heteromastus filiformis (9.6%), Melinna cristata (9.3%), and Lumbrineris longifolia (7.3%). Cluster analysis divided the study area into four station groups based on station similarities in benthic polychaete assemblages: the Glycinde-Prionospio cluster in the western inner bay, the H. filiformis cluster in the middle inner bay, the Melinna-Lumbrineris cluster in the Myodo south-north channel, and the Tharyx cluster in the eastern main channel region. The sediment type of Gwangyang Bay has changed gradually from sandy to muddy. Dominant species have also changed from Chone teres and Lagis bocki to Tharyx sp., which is a potential organic pollution indicator.

Cochleicola gelatinilyticus gen. nov., sp. nov., Isolated from a Marine Gastropod, Reichia luteostoma

  • A yellow, rod-shaped, non-motile, gram-negative, and strictly aerobic bacterial strain, designated LPB0005T, was isolated from a marine gastropod, Reichia luteostoma. Here the genome sequence was determined, which comprised 3,395,737 bp with 2,962 protein-coding genes. The DNA G+C content was 36.3 mol%. The 16S rRNA gene sequence analysis indicated that the isolate represents a novel genus and species in the family Flavobacteriaceae, with relatively low sequence similarities to other closely related genera. The isolate showed chemotaxonomic properties within the range reported for the family Flavobacteriaceae, but possesses many physiological and biochemical characteristics that distinguished it from species in the closely related genera Ulvibacter, Jejudonia, and Aureitalea. Based on phylogenetic, phenotypic, and genomic analyses, strain LPB0005T represents a novel genus and species, for which the name Cochleicola gelatinilyticus gen. nov., sp. nov. is proposed. The type strain is LPB0005T (= KACC 18693T = JCM 31218T).
