Differences and Variations among Anguilla japonica, Muraenesox cinereus and Conger myriaster from the Yellow Sea

  • Genomic DNAs were extracted from the muscle of twenty-one specimens of three eel species collected in Anguilla japonica (AJ), Muraenesox cinereus (MC) and Conger myriaster (CM) from the Yellow Sea, respectively. In the present study, 7 oligonucleotides primers generated 191 specific loci in the AJ species, 226 in the (MC) species and 181 in the CM species, respectively. The primer BION-02 generated the most loci (a total of 83), with an average of 11.86 in the AJ species. The specific loci generated by oligonucleotides primers exhibited inter-individual-specific characteristics, thus revealing DNA polymorphisms. With regard to average bandsharing value (BS) results, individuals from Conger myriaster species (0.808) exhibited higher bandsharing values than did individuals from Muraenesox cinereus species (0.729) (P<0.05). The longest genetic distance (0.430) displaying significant molecular difference was also between individual no. 01 within Anguilla japonica eel species and individual no. 04 within Anguilla japonica species. In this study, the dendrogram resulted from reliable seven oligonucleotides primers, indicating three genetic clusters composed of group I (ANGUILLA 01~ANGUILLA 07), group II (MURAENESOX 08~MURAENESOX 14) and group III (CONGER 15~CONGER 21). The existence of species differentiation and DNA polymorphisms among three eel species were detected by PCR analysis. As mentioned above, a dendrogram revealed close relationships between individual identities within three eel species. High levels of a significant genetic distance among three eel species showed this PCR approach is one of the most suitable tools for individuals and/or species biological DNA studies.

Comparison of Cenangium Dieback Fungus Isolated from Three Different Species of Pine

  • Dieback of pine branches or twigs with brown needles occurs most commonly on Pinus species after severe winter in Korea. In this study, Cenangium ferruginosum was isolated from infected stems, branches, and twigs of Pinus koraiensis (C1), P. densiflora (C2), and P. thunbergii (C3). Morphological and cultural characteristics of the isolates were than compared. There were no significant differences in the morphological characteristics of conidia and ascospores produced by the three isolates. However, cultural differences were observed among the isolates. Optimum temperatures for mycelial growth of C1, C2, and C3 were 15, 20, and $20^{\circ}$, respectively. C1 produced a few conidia and no ascospores, while C2 and C3 produced abundant ascospores and conidia. While optimum temperatures for mycelial growth ranged from 15 to $20^{\circ}$, mycelial growth was also relatively good at lower temperatures of 5-$10^{\circ}$. Conidiomata and conidia were produced on MSA (malt extract soya peptone agar) after 25-30 days of incubation in the dark at $15^{\circ}$. Apothecia were produced by altering culture condition from 15 to $20^{\circ}$, and incubating for 35-60 more days. Optimum temperature for ascospore and conidium germination was $20^{\circ}$. RAPD analysis revealed that there was high similarity of 0.78 between C2 and C3, and low similarity of 0.31 between C2 or C3 and C1.

Riboprint and Virulence Gene Patterns for Bacillus cereus and Related Species

  • A total of 72 Bacillus cereus strains and 5 Bacillus thuringiensis strains were analyzed for their EcoRI ribogroup by ribotyping and for the presence or absence of seven virulence-associated genes. From these 77 strains, 42 distinctive ribogroup were identified using EcoRI, but the two species could not be discriminated by their EcoRI ribogroup. The 77 strains were also examined by PCR for the presence of seven virulence-associated genes, cerAB, pi-plc, entFM, bceT, hblA, hblC, and hblD. All five Bacillus thuringiensis strains were positive for these genes. Although differences in the patterns of virulence genes were observed among the different B. cereus strains, within any given ribogroup the patterns of the seven virulence genes was the same. Pulsed-field gel electrophoresis (PFGE) analysis in combination with available chromosomal maps for a selected group of B. cereus strains revealed significant differences in their chromosome size and the placement of virulence genes. Evidence for significant rearrangements within the B. cereus chromosome suggests the mechanism through which the pattern of virulence-associated genes varies. The results suggest linkage between ribogroups and virulence gene patterns as well as no apparent containment of the latter within any particular species boundary.

한국 산딸기 집단에서 잎 형태 특성에 나타난 표현형 변이 (Phenetic Variability in Leaf Morphological Characteristics of the Korean Rubus crataegifolius Populations)

  • Rubus crataegifolius is a shrub species and is primarily distributed throughout Asia and Europe. We collected 550 representative samples of the eleven populations in Korea to estimate leaf variation within species. The 35 morphological characteristics allowed us to resolve well-supported fixed characteristics and variable characteristics. Sixteen of twenty-three quantitative characteristics and five of twelve qualitative characteristics showed significant differences among populations. We argued that the population differentiation can accounted for the variation in phenetic characteristics such as spine in this species, but were less successful in accounting for variation in other traits. Within a polystatistic leaf structure, their morphological differences could be modulated by ecological pressure such as temperature, altitude, duration of sunshine, and solar radiation. The phenogram showed two distinct clades. One was a group in central Korea and the other was a group in the southern regions of Korea. If morphological characteristics in R. crataegifolius populations reflect their ecological niche, morphology should be regarded with caution as the main criterion for population studies in R. crataegifolius.

한국산 참서대과 Cynoglossidae 어류 6종의 상두개골계(epicranical bony system) (Epicranial Bony System of Six Species in Family Cynoglossidae (Pisces, Pleuronectiformes) from Korea)

  • 1992년부터 1994년까지 우리나라의 연안에서 채집된 참서대과 어류 6종의 상두개골계(epicranical bony system) 에 대해 비교 검토하였다. 상두개골 가운데 erisma 와 frontal element 의 발달정도에 따라 1. Paraplagusia japonica, 2.Cynogossus joyneri 와 C.robustus, 3.c. semilaveis와 C. abbreviatus, 4.C. interruptus 의 4 group 으로 구분되엇고, erisma를 지지하는 proximal radials 의 수에서도 P.japonica와 C. robustus 가 10-11개, C. joyneri 10-12 . C. semilaevis 9-11. C. abbreviatus 9-10 , C. interruptus 7-8개로 종간에 많은 차이를 나타냈다. 이러한 골격의 형태적 차이는 종간의 유연관계를 나타내는데 중요한 형질로 사료된다.

한국산 다슬기류 ( Semisulcospira spp. ) 두 종의 세포학적 연구 (A Cytological Study on Two Species of Genus Semisulcospira ( Gastropoda: Pleuroceridae ) in Korea)

  • The melaniid snails belonging to genus Semisulcospira were collected in the Kangwha and Yonchon areas of Korea in 1986 through 1987 in order to carry out a cytotaxonomic study, The snails were first narcotized with menthol and fixed with 70% ethyl alcohol for morphological identification. The gonads of adult snails were used for chromosome analyses by the technique of Imai et al. (1977) with minor modification. Slide preparations were observed under high power fields using a Leitz light miscroscope. The results obtained in the present stuedy are summarized as follows: 1)The sanils collected from Kangwha and Yonchon areas were identified as Semisulcospira forticosta(Martens, 1886)and S. gottschei (Martens, 1886) respectively.2)No specific differences were obwerved in details of the chromosome cycle between S. forticosta and S. gottschei.3) Diploid chromosome numbers observed at mitotic metaphase were 36. There was no difference in chromosome numbers between S. forticosta and S. gottschei.4) There were morphological differences in the karyotypes of the two species. The spermatogonial metaphase karyotype of S. forticosta consists of six pairs of metacentric, eleven pairs of submetacentric, and one pair of acrocentric chromosomes. The spermatogonial metaphase karyotype of S. gottschei consists of five pairs of metacentric, tselve pairs of submetacentric, and one pair of acrocentric chromosomes. Summarizing the aboxe results, the two species of Semisulcospira employed in this study have same chromosome numbers(2n=36)with different karyotypes.

도시 내 가로수의 광선투과량에 따른 온도저감 효과 - 서울시 서초구를 중심으로 - (Temperature Reduction Effect According to Light Transmittance of Urban Street Trees - Focused on Seocho-gu in Seoul -)

  • With rapid urbanization and reckless urban development in the 21st century, the urban environment has gradually gotten worse, and urban heat island effect has been dramatically intensified. Thus, the importance of street greenery that can mitigate the urban heat island effect has further been highlighted. In this regard, this study was aimed at selecting suitable plant species for street greenery to reduce the urban heat island effect. Towards this end, five roads located in Seocho-gu, Seoul were selected as study sites, and plant species composition and difference of surface temperature were compared and analyzed in relation to the light transmittance. The street with the greatest temperature difference is Bangbae-ro(Platanus occidentalis). On the other hand, the road with the lowest temperature difference is Nambusunhwan-doro(Metasequoia Glyptostroboides). The effect of temperature reduction was found to be associated with light transmittance.Bangbae-ro(Platanus occidentalis) with the lowest light transmittance showed the highest temperature difference and Nambusunhwan-doro(Metasequoia Glyptostroboides) with the highest light transmittance showed the lowest temperature difference. It is analyzed that there are most differences in temperature when the amount of lights coming in between the crown is small. The temperature reduction effect can be obtained by planting deciduous broad-leaved trees. Also species with dense crown and broad width of crown will be able to maximize the effect of temperature reduction. In future studies, it will be necessary to expand the other species of trees in the street, and analyze the germicidal trees and shrubs as well as the differences in the packaging materials.

수 종 목본식물의 화력학적 연구

  • 온대낙엽수림의 수 종 목본식물에서 생육초기의 기온과 개엽시기와 관계를 규명하기 위하여 남한산성과 지역에서 1991, 1992 및 1993년 대관령 지역에서 1992년, 3월부터 5월까지 겨울눈의 파열 시기, 개엽이 완료된 시기, 개화 및 낙화의 시기를 관찰 조사하고 이것을 기상자료와 관련시켜 분석하였다. 남한산성 지역에서 개엽이 이른 종은 신갈나무, 진달래, 개벚나무, 노린재나무 등으로 4월 초순이었으며, 늦은 종은 굴참나무, 떡갈나무, 다릅나무 등으로 4월 하순이었다. 그리고 가장 빠른종인 진달래 (4월 8일)와 가장 늦은 종인 굴참나무, 다릅나무 및 떡갈나무 (5월 4일)의 차이는 27일 이었다. 두 지여간 동일 수종의 개엽시기를 비교하면, 남한산성의 것은 대관령 지역의 것보다 8~24일 빨랐다. 한편, 두 지역간 개화시기의 차이는 0~22일로 개엽시기의것보다 적었다. 겨울눈의 파열시기는 일적사온량지수(YDI)와 관계가 있지만 개엽이 완료되는 시기나 개화시기를 예측하는 데는 YDI보다 Nuttonoson의 온량지수(Tn)가 더욱 유효한 것으로 나타났다.

서부 열대 태평양의 눈다랑어와 황다랑어의 먹이에 관한 연구 (Diets of Bigeye and Yellowfin Tunas in the Western Tropical Pacific)

  • 눈다랑어와 황다랑어의 먹이의 종류와 섭이 습성을 비교하기 위하여 서부 열대 태평양에서 연승어업으로 어획된 이들 다랑어의 위내용물을 조사하였다. 눈다랑어와 황다랑어의 먹이는 주로 어류, 두족류, 갑각류이었다. 이들 두 종의 다랑어에서 동정된 먹이생물의 총 종수는 어류가 15종, 갑각류 6종 그리고 두족류가 1종이었으며 그 중 가장 중요한 먹이는 샛비늘치류인 Myctophum sp였다. 황다랑어와 눈다랑어의 먹이를 분석해본 결과 먹이의 종류에 있어서는 두 종간에 큰 차이가 없었으며 이것으로 보아 같은 해역에 서식하는 다랑어류는 먹이 습성이 유사함을 알 수 있었다. 그러나, 먹이 가운데 몇몇 종 (Myctophidae, Alepisauridae, Oplophoridae, Gammaridae, and Onychoteuthidae)의 경우 그 중요도에 있어서 두 다랑어간에 현저한 차이를 보였다.

Morphological and Molecular Characterization of Heterodera schachtii and the Newly Recorded Cyst Nematode, H. trifolii Associated with Chinese Cabbage in Korea

  • The sugar beet cyst nematode, Heterodera schachtii is a well known pathogen on Chinese cabbage in the highland fields of Korea. However, a race of cyst forming nematode with close morphological resemblance to H. trifolii was recently isolated from the same Chinese cabbage fields. Morphological species differentiation between the two cyst nematodes is challenging, with only minor differences between them. Thus, this study described the newly intercepted H. trifolii population, and reviewed morphological and molecular characteristics conceivably essential in differentiating the two nematode species. A comparison of morphometric measurements of both infective juveniles and vulval cones of cysts showed significant differences between the two cyst nematodes. When total RNA and genomic DNA were extracted from a mixed field population, COI genes and ITS regions were clearly amplified with primers of the two Heterodera species, suggesting that Heterodera population collected from the Chinese cabbage field consisted of a mixture of two species. COI and ITS of H. trifolii were predominantly amplified from nucleotides prepared from H. trifolii monoxenic population whereas those of H. schachtii were strongly detected in H. schachtii monoxenic cultures. Thus, this study confirms the coexistence of the two species in some Chinese cabbage fields; and the presence of H. trifolii in Korea is reported here for the first time.
