The Oxidative Stress by Hair Dyeing Changes the Antioxidant Defense Systems and Strongly Relates to the Plasma Vitamin E Concentration

  • Reactive oxygen species can be generated in the skin by hair dyeing. The aim of this study was to find out the effects of the oxidative-type hair dye application in young women on the antioxidant systems. We investigated the lipid peroxide levels, glutathione (GSH) levels, and the antioxidant enzyme activities including superoxide dismutase (SOD), glutathione peroxidase (GSHPx) in plasma and erythrocytes and catalase (CAT) in erythrocytes, and DNA damages in lymphocytes. Also, plasma concentrations of antioxidant vitamins, vitamin A and E, were measured and the correlations between various antioxidant parameters and oxidative damages were evaluated The antioxidant enzyme activities in plasma (GSHPx) and in erythrocytes (SOD and CAT) were decreased significantly after hair dyeing. 1be lipid peroxide and GSH levels were not affected in both plasma and erythrocytes. No significant difference was found in the concentrations of both vitamin A and E between before and after hair dyeing. However, DNA damages expressed as the tail extent moment (TEM) and tail length (TL) were significantly (p<0.001) increased. The plasma vitamin E concentration was correlated with DNA damages (TEM: r=-0.590, p<0.01 and TL: r=-0.533. p<0.01) and RBC SOD activity (r=0.570, p<0.05). In turn, RBC SOD activity was significantly correlated with both plasma MDA levels (r=-0.412, p<0.05) and DNA damages (TM: r=-0.546, p<0.01, TL: r=-0.493, p<0.01). Our results demonstrated that the exposure to hair dyeing produced lymphocyte DNA damage and modification of the antioxidant enzyme activities. Also, there were very strong associations between plasma vitamin E concentration, RBC SOD activity and DNA damage induced by hair dyeing. It suggests that the antioxidant status of a subject is likely to be related to the extent of the harmful effects caused by hair dyeing.

Comparative Genomics Study of Candidatus Carsonella Ruddii; an Endosymbiont of Economically Important Psyllids

  • Candidatus Carsonella ruddii is an endosymbiont that resides in specialized cells within the body cavity of plant sap-feeding insects called psyllids. The establishment of symbiotic associations is considered one of the key factors for the evolutionary success of psyllids, as it may have helped them adapt to imbalanced food resources like plant sap. Although C. ruddii is defined as a psyllid primary symbiont, the genes for some essential amino acid pathways are absent. Complete genome sequences of several C. ruddii strains have been published. However, in-depth intra-species comparison of C. ruddii strains has not yet been done. This study therefore aimed to perform a comparative genome analysis of six C. ruddii strains, allowing the interrogation of phylogenetic group, functional category of genes, and biosynthetic pathway analysis. Accordingly, overall genome size, number of genes, and GC content of C. ruddii strains were reduced. Phylogenetic analysis based on the whole genome proteomes of 30 related bacterial strains revealed that the six C. ruddii strains form a cluster in same clade. Biosynthetic pathway analysis showed that complete sets of genes for biosynthesis of essential amino acids, except tryptophan, are absent in six C. ruddii strains. All genes for tryptophan biosynthesis are present in three C. ruddii strains (BC, BT, and YCCR). It is likely that the host may depend on a secondary symbiont to complement its deficient diet. Overall, it is therefore possible that C. ruddii is being driven to extinction and replacement by new symbionts.

Expression of Cytoplasmic 8-oxo-Gsn and MTH1 Correlates with Pathological Grading in Human Gastric Cancer

  • Background: Cancers have dysfunctional redox regulation resulting in production of reactive oxygen species (ROS), damaging DNA, RNA and free NTPs, and causing the accumulation of oxidative nucleic acids in cytoplasm. The major types are 8-oxo-7,8-dihydroguanine(8-oxoGsn) in RNA and 8-oxo-7,8-dihydro-2' deoxyguanosine(8-oxodGsn) in Mt-DNA. The MTH1 protein sanitizes oxidized nucleotide pools from NTPs to monophosphates, preventing the occurrence of transversion mutations. This study concerned cytoplasmic 8-oxodGsn/Gsn and MTH1 expression in gastric cancer and para-cancer tissues and elucidated roles of nucleic-acid oxidation and anti-oxidation. Materials and Methods: A polymer HRP detection system was used to detect 8-oxo-Gsn/dGsn and MTH1 expression in 51 gastric cancer and para-cancer tissue samples. Analyses of patient clinical and pathological data were also performed. Results: The expression of MTH1 and the 8-oxo-dGsn/Gsn ratio were significantly higher in cancer tissues than para-cancer tissues (P<0.05). Cytoplasmic 8-oxo-Gsn and MTH1 were both found to positively correlate (P<0.05) with tumor differentiation, while no significant associations were found with gender, age, invasion depth, lymph node metastasis and clinical stage (P>0.05). Conclusions: We found 8-oxo-dGsn/Gsn and MTH1 are both highly expressed in gastric cancer tissues, especially in well differentiated lesions. In addition, oxidated mtDNA is prevalently expressed in gastric cancers, while 8-oxo-Gsn expression in cytoplasmic RNA is a bit lower, but more selectively.

Functional Prediction of Imprinted Genes in Chicken Based on a Mammalian Comparative Expression Network

  • Little evidence supports the existence of imprinted genes in chicken. Imprinted genes are thought to be intimately connected with the acquisition of parental resources in mammals; thus, the predicted lack of this type of gene in chicken is not surprising, given that they leave their offspring to their own heritance after conception. In this study, we identified several imprinted genes and their orthologs in human, mouse, and zebrafish, including 30 previously identified human and mouse imprinted genes. Next, using the HomoloGene database, we identified six orthologous genes in human, mouse, and chicken; however, no orthologs were identified for SLC22A18, and mouse Ppp1r9a was not included in the HomoloGene database. Thus, from our analysis, four candidate chicken imprinted genes (IGF2, UBE3A, PHLDA2, and GRB10) were identified. To expand our analysis, zebrafish was included, but no probe ID for UBE3A exists in this species. Thus, ultimately, three candidate imprinted genes (IGF2, PHLDA2, and GRB10) in chicken were identified. GRB10 was not significant in chicken and zebrafish based on the Wilcoxon-Mann-Whitney test, whereas a weak correlation between PHLDA2 in chicken and human was identified from the Spearman's rank correlation coefficient. Significant associations between human, mouse, chicken, and zebrafish were found for IGF2 and GRB10 using the Friedman's test. Based on our results, IGF2, PHLDA2, and GRB10 are candidate imprinted genes in chicken. Importantly, the strongest candidate was PHLDA2.

Therapeutic Potential of an Anti-diabetic Drug, Metformin: Alteration of miRNA expression in Prostate Cancer Cells

  • Background and Aims: Prostate cancer is the most commonly diagnosed cancer in males in many populations. Metformin is the most widely used anti-diabetic drug in the world, and there is increasing evidence of a potential efficacy of this agent as an anti-cancer drug. Metformin inhibits the proliferation of a range of cancer cells including prostate, colon, breast, ovarian, and glioma lines. MicroRNAs (miRNAs) are a class of small, non-coding, single-stranded RNAs that downregulate gene expression. We aimed to evaluate the effects of metformin treatment on changes in miRNA expression in PC-3 cells, and possible associations with biological behaviour. Materials and Methods: Average cell viability and cytotoxic effects of metformin were investigated at 24 hour intervals for three days using the xCELLigence system. The $IC_{50}$ dose of metformin in the PC-3 cells was found to be 5 mM. RNA samples were used for analysis using custom multi-species microarrays containing 1209 probes covering 1221 human mature microRNAs present in miRBase 16.0 database. Results: Among the human miRNAs investigated by the arrays, 10 miRNAs were up-regulated and 12 miRNAs were down-regulated in the metformin-treated group as compared to the control group. In conclusion, expression changes in miRNAs of miR-146a, miR-100, miR-425, miR-193a-3p and, miR-106b in metformin-treated cells may be important. This study may emphasize a new role of metformin on the regulation of miRNAs in prostate cancer.

  • Heterostyly continues to fascinate evolutionary biologists interested in heredity, evolution, breeding, and adaptive function. Polymorphism demonstrates how simply inherited developmental changes in the location of plant sexual associations can have important consequences for population pollination and mating biology. In contrast to homozygous self incompatibility, only a small number of mating phenotypes can be maintained in the population because insect pollinators have limitations in achieving multiple segregation sites for pollen deposition. Field studies of pollen tube growth have shown that reciprocal style-stamen polymorphisms function to increase the capacity of insect-mediated cross-pollination. The genetic pattern of style morphs is well established in various taxa, but despite recent advances, the identity, number, and structure of the genes controlling the heteromorphic syndrome have been poorly elucidated. The phenomenon of heterostyly in buckwheat has been controlled by gene complex concentrate to S-locus. Homomorphic autogamous buckwheat strains were established by the interspecific hybridization. Backcrossing of this line to the common buckwheat (pin) and selecting homostylar progenies made it possible to introduce the self-compatible gene into common buckwheat. In the result, we obtained the BC9F2 generation, and defined the strong linkage between flower type and self-incompatibility by microscopic observation of pollen tube growth. This finding suggests that self-incompatibility character is not controlled by one gene. Moreover, we defined the strong linkage between flower type and self-incompatibility. It strongly supports the S supergene theory. Therefore, we have plan to elucidate the heterostyly self-incompatibility by using molecular genetics, proteome analysis and apply to exploitation of buckwheat improvement. In near future, the expression of heterozygous syndromes in genus Fagopyrum with single isolated heterozygous species may provide clues to early stages of polymorphic assembly and shed light on evolutionary models of heterozygous strains.

Mineralogical Studies on Sulfide Ore Species of the Tong Myeong Tungsten Deposits (동명중석광산산(東明重石鑛山産) 유화광물(硫化鑛物)의 광물학적(鑛物學的) 연구(硏究))

  • The skarn type tungsten deposits in Jechon area are developed in the contact aureole of Jurassic granodiorite and lower Paleozoic limestone beds. The Tong Myeong mine contains scheelitebearing skarns found at and near the contacts between crystalline limestone and hornfels. Although the skarns are heterogeneous, there are clear patterns in the preferred associations and nonassociations of minerals on all scales. The skarn show a zonal arrangement from limestone to hydrothermal vein as follow: wollastonite skarn, clinopyroxene skarn, clinopyroxene-garnet skarn, garnet skarn, and vesuvianite skarn. Scheelite, abundant in all skarn units except wollastonite skarn and also in quartz veins near orebodies, is everywhere strongly correlated with pyrrhotite. It is implied that it was a stable phase throughout the evolution of the zoned skarns, at least in pyrrhotite.forming environments. Deposition of scheelite was probably widely caused by increasing $a_{Ca^{2+}}$ in the fluid, resulting from associated and interrelated reactions: $FeCl_2\;aq+H_2S\;aq{\rightarrow}FeS+2H^{+}+2Cl^-$; and $CaCO_3+2H^+{\rightarrow}Ca^{+2}+H_2CO_3$. The spectral reflection powers of nine sulfide species were studied, for three mineralization stage. The shapes and characteristics of the spectral reflectivity profiles are significant in their control of other optical properties. The characteristics of the Vickers microhardness and the optical symmetry for the minerals studied are discussed. Broad radicle groupings of the sulfides can be made with regard to the reflectivity-microhardness values.

Allelopathic Potentials of Larix leptolepis on Germination of Several Forest Tree Species (몇가지 삼림수종(森林樹種) 종자발아(種子發芽)에 대(對)한 낙엽송엽(落葉松葉) 추출물(抽出物)의 Allelopathic 효과(效果))

  • Inhibitory effect of L. leptolepis fallen leaf extracts on the germination of Pinus densiflora, ${\times}$ Pinus rigitaeda, Pinus rigida, Pinus thunbergii and Larix leptolepis was investigated. Germination of those seeds in culture room and in pot at field showed the highly significant inhibitory effects by the aqueous extracts from L. leptolepis fallen leaves. Among them L. leptolepis was the most severely inhibited by the aqueous extracts used. To identify allelopathic substances, thin-layer chromatography was employed. Gallic, ferulic, t-cinnamic and vanillic acids were identified from fallen leaves of L. leptolepis. From the results, it is assumed that autotoxicity of L. leptolepis may act as inhibitory factors on germination of the species in natural stands.

  • 1. The plant communities on Mt. Sulak, a typical alpine mountain in central Korea was researched from the standpoint of ecological survey. 2. The plant on this mountain can largely be classified into 2 categories, the warm zone (below 1100m) and the frigid zone. The former is again classified into a broad-leaved tree formation and a needle-leved tree formation, and the latter is calssified into a broad-leaved tree formation and needle-leaved tree formation, and shrub formation. 3. The broad-leaved tree formation in the warm zone can largely be classified into a Carpinus laxiflora-syringa Palibiniana var. kanibayashi association(1), a Carpinus erosa-Acer Pseudo-sieboldianun association(2), and a Quercus mongolica- Tripterygium Regelii association(3). 4. The needle-leaved tree formation in the warm zone can also largely ben classified into a Pinus densiflora- Miscanthus sinensis association(4), Abies holophylla- Tripterygium regelii association(5), Pinus kordiensis- Rhododendron schlippenbachii association(6) In the association(4), the canopy of the Pinus densiflora is gradually being occupied by the Quercus mongolica which is one kind of broad-leaved trees. 5. We can fully see a DryoPteris type of the flour layer plant type in the association (3) and also see a Sasamorpha type in the association (5). 6. The broad-leaved tree formation in the frigid zone can largely be classified into Quercus mongolica- Rhododendron schlippenbachii association (8). The constituents of tree and subtree layers in (7) and (8) associations are similar to those association (3), but the constituents of the shrub layer are different from those of associtation (3) due to the difference in height. 7. The needle-leaved formation in the frigid zone can largely be classified into Thuja koraiensisRodgersia podophylla var. viritis association (9) and Abies nephrolepis-Acer tschonoskii var. rubripes asscciation (10). The species of alpine plants or subalpine plants are gradually increased in this formation. 8. The shrub formation can also largely be classified into a Rhododendron mucronulatum- Patrinia saniculaefolia association (11) and Pinus pumila associatio (12). Association (12) has largely developed on the windy place. Association (12) was burned due to the bombing during the Korean war, but now we recoginized there occurred an invasion of Tripterygium Regelii in such a place. 9. The herb layer species which constitute the shrub formation are mostly alpine or subalpine plants of small size, and their kinds are also very few. 10. The growth of the Moss layer is especially good because of the varied conditions of the habitat. The kinds which can easily be seen are the genus of Thunidium, Haplocladium, Brachythecium, Macromitrium, Holonitrium, Atrichum, Schwstchkeopsis, Grimmia, Hedwigia, Rhynchostegium and Mnium. 11. The genus of the Sphagnum densly grows like a mat at the acid and moisturous place above 1100m. The authors should express their thanks to father. Competition Relation and Forage Yields by Different Sowing Rate in Grass and Legume Mixtures (몇가지 화본과목초(禾本科牧草)와 콩과목초(科牧草)를 혼파(混播)할 경우 파종비율(播種比率)에 따른 목초(牧草)의 생산성(生産性)과 경합관계(競合關係))

  • This experiment was carried out to investigate the competition in mono-and binary-associations of four forage species; ochardgrass (Dactylis glomerata L. cv. Potomac), tallfescue (Festuca arundinacea Scnreb, cv. Fawn), white clover (Trifolium repns L. cv. California), and red clover (Trifolium pratense L. cv. Kenland) as affected by different sowing rate. Each species was grown in interspecific stands in field. And samplings of shoot were taken at first cliping stages. The interspecific competition was evaluated by calculating traditional methods: aggressivity, relative yield total (RYT), relative crowding coefficient (RCC), and compensation index (CI) with shoot dry matter production and calories of nutrient ingredient of each species in mono-and binary-mixture. Compared mixture with monoculture. RYT based on dry weight was lucrative in mixture except for orchardgrass-redcover (1:0.1,1:1). RYT of calculated calories with produced nutrients was in inlucrative in mixture of orchardgrass-redclover's 1:0.1 mixture by RYT<1.0. Generally, RYT was the highest in mixture of 1:0.5 sowing rate. Aggressivity of grass was poor competition tendency that raised up legume's sowing rate, particularly, with whiteclover. In grasses, tallfescue was stronger competitor than orchardgrass with legume in mixture, RCC of grasses against legume's growth was depressed in higher sowing rate with legume. Grass's growth depression was severer with whiteclover than redclover in mixture.
