Seasoning of Commercial Wood Using Solar Energy (태양에너지를 이용한 유용목재의 건조)

  • This study investigated the temperatures and relative humidities in the semi-greenhouse type solar dryer with a black rock-bed heat storage and without heat storage and outdoor temperature and relative humidity at 9 a.m. and 2 p.m.. A comparison was made of the drying rates, final moisture contents, moisture content distributions, casehardening stresses, drying defects, volumetric shrinkage of dried lumber for solar- and air-drying from the green condition of mixtures of Douglas-fir, lauan, taun, oak and sycamore 25mm- and 50 mm-thick lumber during the same period for four seasons, and heat efficiencies for solar dryer with and without the heat storage for saving of heat energy and the cost of lumber drying using the solar energy. The results from this study were summarized as follows: I. The mean weekly temperatures in the solar dryers were 3 to $6^{\circ}C$ at 9 a.m. and 9 to $13^{\circ}C$ at 2 p.m. higher than mean outdoor temperature during all the drying period. 2. The mean weekly relative humidities in the solar dryers were about 1 to 19% at 9 a.m. higher than the outdoor relative humidity. and the difference between indoor and outdoor relative humidity in the morning was greater than in the afternoon. 3. The temperatures and relative humidities in the solar dryer with and without the heat storage were nearly same. 4. The overall solar insolation during the spring months was highest and then was greater in the order of summer, atumm, and winter month. S. The initial rate of solar drying was more rapid than that of air drying. As moisture content decreased, solar drying rate became more rapid than that of air drying. The rates of solar drying with and without heat storage were nearly same. The drying rate of Douglas-fir was fastest and then faster in the order of sycamore, lauan, taun and oak. and the faster drying rate of species, the smaller differences of drying rates between thicknesses of lumber. The drying rates were fastest in the summer and slowest in the winter. The rates of solar drying during the spring were more slowly in the early stage and faster in the later stage than those during the autumn. 6. The final moisture contents were above 15% for 25mm-thick air dried and about 10% for solar dried lumber, but the mean final MCs for 50mm-thick lumber were much higher than those of thin lumber. The differences of final MC between upper and lower course of pile for solar drying were greater than those of pile for air drying. The differences of moisture content between the shell and the core of air dried lumbers were greater than those of solar dried lumber, smallest in the drying during summer and greatest in the drying during winter among seasons. 7. Casehardening stresses of 25mm- and 50mm-thick dried lumber were slight, casehardening stress of solar dried lumber was severer than that of air dried lumber and was similar between solar dried lumber with and without heat storage, Casehardening stresses of lumber dried during spring were slightest and then slighter in the order of summer, autumn, and winter. Casehardening stresses of Douglas -fir, sycamore and lauan were slight, comparing with those of taun and oak. 8. Maximum initial checks of 25mm-thick lumber occurred above and below fiber saturation point and those of 50mm-thick lumber occurred in the higher moisture content than thin lumber. As the moisture content decreased, most of checks were closed and didn't show distinct difference of the degree of checks among drying methods. The degree of checks were very slight in case of Douglas-fir and lauan, and severe in case of taun and oak. The degree of checks for 50mm-thick lumber were severer than those for 25mm-thick lumber. 9. The degree of warpage showed severe in case of oak and sycamore lumber, but no warping was found in case of Douglas-fir, lauan and taun. 10. The volumetric shrinkages of taun and oak were large and medium in case of Douglas-fir, lauan and sycamore. 11. Heat efficiencies of solar dryer with heat storage were 6.9% during spring, 7.7% during summer, 12.1% during autumn and 4.1% during winter season. Heat efficiency of solar dryer with heat storage was slightly greater than that of without heat storage. As moisture content of lumber decreased, heat efficiency decreased.

Establishment of PCR Conditions for the Identification of Stenotrophomonas maltophilia Isolated from Boar Semen and Antimicrobial Susceptibility Patterns of the Isolates (돼지 정액에서 분리된 Stenotrophomonas maltophilia 확인을 위한 PCR 기법 개발 및 분리 균주의 항생제 감수성 양상)

  • Bacteria are frequently contaminated during the collection and processing procedures of boar semen. Of the contaminants, Stenotrophomonas (S.) maltophilia is a Gram-negative bacterium that is widely distributed in a variety of habitats. Although PCR assays have been developed for the detection of S. maltophilia, they cross-react with some species of Xanthomonas. In this study, we designed a primer set for the detection of S. maltophilia in order to target the chiA (GenBank accession no. NC_010943) gene. The specific PCR products were amplified from S. maltophilia only, not from other tested strains that are frequently found in semen. The detection limit of the PCR was $1.5{\times}10^3$ CFU/ml with pure-cultured S. maltophilia and $1.5{\times}10^4$ CFU/ml with S. maltophilia spiked in semen. Twenty-six (5.9%) S. maltophilia were isolated from 440 semen samples. The PCR results exhibited 98.9% agreement with a comparison of S. maltophilia isolation. Also, the sensitivity and specificity of the PCR were 100% and 98.7%, respectively. In the antimicrobial susceptibility test, S. maltophilia isolates were highly susceptible to enrofloxacin and florfenicol, while the majority of them were resistant to amoxicillin/clavulanic acid, apramycin, ceftiofur, penicillin, and spectinomycin. These results indicated that the PCR using the chiA gene was proven to be reliable and effective for the detection of S. maltophilia with high levels of sensitivity and specificity.

Comparison of Antioxidant, Cytotoxicity and Flavonoid Content of Stachys sieboldii Miq. vs. Lycopus lucidus Turcz. Leaf Extracts (초석잠 및 택란 잎 추출물의 항산화 및 세포독성 활성과 총 플라보노이드 함량 비교)

  • In this study, the antioxidant and cytotoxic effects and the flavonoid contents of leaf extracts from Stachys sieboldii Miq. and Lycopus lucidus Turcz. were compared. The flavonoid contents of the acetone + methylene chloride (A+M) and methanol (MeOH) extracts of L. lucidus Turcz. leaves were 55.7 and 233.2 mg/g, respectively. In a DPPH assay, A+M and MeOH extracts from L. lucidus Turcz leaves had a greater scavenging effect than those of S. sieboldii Miq. leaves (p<0.05). In an ABTS assay, MeOH extracts from S. sieboldii Miq. and L. lucidus Turcz (0.5 mg/ml concentration) leaves had scavenging effects of 85% and 91%, respectively (p<0.05), suggesting that both of the MeOH extracts had greater scavenging effects than both A+M extracts. In a 120 min ROS production assay, all tested extracts decreased the cellular ROS production induced by H2O2 compared to that produced by exposure to the extract-free control. The MeOH extract from L. lucidus Turcz leaves had a greater inhibitory effect on cellular ROS production (p<0.05). Treatment with A+M and MeOH extracts from both S. sieboldii Miq. and L. lucidus Turcz. leaves showed a dose-dependent increased cytotoxicity against the growth of AGS, HT-29 cancer cells, and HT-1080 (p<0.05). Both A+M extracts had a greater inhibitory effect on the growth of all cancer cells than both MeOH extracts. These results suggest that the MeOH extract of L. lucidus Turcz. leaves is effective in scavenging free radicals and inhibiting cellular oxidation, while the A+M extract inhibits proliferation of three types of cancer cell.

Seed Set in Close Pollination and Backcross of Interspecific F1 of Lilium spp. (백합 근연수분 및 종간 교잡종 F1 여교잡시 종자형성)

  • We studies seed set in the interspecific F1 /backcross hybrids of Lilium species. In the interspecfic hybrid of L. longiflorum cv. Gelria with L.${\times}$ fomolongi cv. Raizan,93% fruit set was obtained by stigmatic pollination in comparison to 53% from cut-style pollination. Accordingly, the number of seed set resulting from stigmatic and cut-style pollination was 147 and 53, respectively. Pollination o( both stigmatic and cut-style pollination resulted in 47% fruit set in the hybrid of L. longiflorum cv. Lorina with L.${\times}$ fomolongi cv. Raizan. However, stigmatic pollination formed 413 seeds, whereas only 24 seeds were obtained by cut-style pollination in this cross. The hybrid of L.${\times}$ fomolongi cv. Raizan with L. longiflorum cv. Come set 40% fruit with a total of 43 seeds by stigmatic pollination. However, no fruit set was observed in cut-style pollination in this hybrid. Backcrossing the F1 hybrid by cut-style pollination of L. longiflorum cv. Lorina ${\times}$ Asiatic hybrid cv. Chicago with the latter parent led to 53% fruit set, and 109 embryos were obtained. Likewise, backcrossing following cut-style pollination of L. longiflorum cv. Lorina ${\times}$ Asiatic hybrid cv. Corsia with the latter parent formed 67% fruits and 107 embryos. However, in the remaining interspecific hybrids, cut-style pollination set no fruit.

Zooplankton Abundance in Korean Waters (한국근해 동물성 부유생물의 주요군의 양적 분포)

  • Plankton samples used for the present study were collected by the NORPAC net during the CSK cruises in the Korean waters in March and August, 1967. Regional and seasonal variations in the zooplankton biomass (wet weight, mg/㎥) were noticed in the Korean waters. In March the highest biomass, 130mg/㎥ on the average, occurred in the southern part of Japan Sea, but the lowest biomass of less than 50mg/㎥ occurred in the Yellow Sea and the western sea of Cheju Island Contrally, in August, the average biomass of 120mg/㎥ was measured in the Yellow Sea, the western sea of Cheju Island and the coastal waters of southern Korea, while the biomass of Japan Sea was the lowest of the regions surveyed. In comparison with the zooplankton biomass, total number of zooplankton per cubic meter of water strained also showed regional and seasonal fluctuations. In general, variations in the number of zooplankton specimens follows the same trend as in the biomass. The largest number, up to 800mg/㎥ on the average, occurred in the southern part of Japan Sea in March and the lowest number, less than 200mg/㎥ occurred in the Yellow Sea and the western sea of Cheju Island. In August, as shown by the biomass fluctuations, the largest number of zooplankton 850mg/㎥ on the average occurred in the Yellow Sea, the western sea of Cheju Island and the coastal region of southern Korea. But the lowest number of less than 500mg/㎥ was found in the Japan Sea. Among the various groups of zooplankton examined, the following were dominant components of the zooplankton population: Copepoda, Chaetognatha, Siphonophora, Euphausiacea, Cladocera, Appendicularia, and Amphipoda. The zooplankton conposition was significantly differed between the Japan Sea and Yellow Sea. Copepods which usually occupied over 66% in the Japan Sea and thd Korean Strait samples occupied only 42% of the catches in August, while cladocerans and chaetognaths were relatively abundant, i. e., 15 and 18% of the total organisms. The most dominant species of copepods and chaetognaths were Paracalanus parvus, Oithona similis, Acartia clausi, Calanus helgolandicus, Sagitta enflata, S. bedoti, S. elegans and S. crassa.

Morphological Comparison Hysterothylacium sp. and Anisakis simplex (Nematoda: Anisakidae) from Wild Black Rockfish, Sebastes schlegeli, and Histopathological Host Reaction (자연산 조피볼락, Sebastes schlegeli의 소화관에 기생하는 Hysterothylacium sp.와 Anisakis simplex (Nematoda: Anisakidae)의 형태학적 비교 및 숙주의 조직병리학적 반응)

  • Numerous anisakids were parasitic near the digestive tract of the black rockfish, Sebastes schlegeli and some anisakids observed on the liver of the host. Anisakids in the host were identified three species (Hysterothylacium sp., Anisakis simplex, A. pegreffii) and Hysterothylacium sp. was the high occurrence of anisakid worms in the host. Hysterothylacium sp. was shorter and thinner than A. simplex. Both of anisakids observed lip, mouth, nerve ring, excretory pore and excretory duct in the anterior portion. In the mid portion, anisakids had esophagus, ventriculus and intestine and especially, Hysterothylacium sp. had intestinal ceacum and ventricular appendage. There was conical nodulose apex at the end of the posterior portion in Hysterothylacium sp. and spine in A. simplex. SEM examination revealed that there was three lip near the mouth of Hysterothylacium sp. Dorsal lip was approximately 65 ${\mu}m$ and paried lateroventral lip were approximately 60 ${\mu}m$ in the width. All lips were found double papilla (approximately 8 ${\mu}m$ in the width). The body width of the Hysterothylacium sp. and A. simplex was approximately 480 ${\mu}m$ and 900 ${\mu}m$ respectively. The hight of the lateral alae was about 7 ${\mu}m$ and width of papilla on the cornical nodulose apex was about 3.3 ${\mu}m$ in Hysterothylacium sp. The hight of spine was approximately 20 ${\mu}m$ in A. simplex. There was mainly Hysterothylacium sp. in the intestinal lumen of the host. The nematod worms were parasitic near the mucosal fold and in the submucosal. In the mucosal epidermal layer, it was increased mucous cells by the infection of the parasites.

Application for Identification of Food Raw Materials by PCR using Universal Primer (일반 프라이머를 이용한 PCR의 식품원료 진위 판별에 적용)

  • In order to determine an authenticity of food ingredient, we used DNA barcode method by universal primers. For identification of animal food ingredients, LCO1490/HCO2198 and VF2/FISH R2 designed for amplifying cytochrome c oxidase subunit1 (CO1) region and L14724/H15915 for cytochrome b (cyt b) region on mitochondrial DNA were used. Livestock (cow, pig, goat, sheep, a horse and deer) was amplified by LCO1490/HCO 2198, VF2/FISH R2 and L14724/H15915 primers. Poultry (chicken, duck, turkey and ostrich) was amplified by LCO1490/HCO 2198 and VF2/FISH R2 primers. But, Fishes (walleye pollack, herring, codfish, blue codfish, trout, tuna and rockfish) were only amplified by VF2/FISH R2 primers. For plant food ingredients, 3 types of primers (trnH/psbA, rpoB 1F/4R and rbcL 1F/724R) have been used an intergenic spacer, a RNA polymerase beta subunit and a ribulose bisphosphate carboxylase region on plastid, respectively. Garlic, onion, radish, green tea and spinach were amplified by trnH/psbA, rpoB 1F/4R and rbcL 1F/724R. The PCR product sizes were same by rpoB 1F/4R and rbcL 1F/724R but, the PCR product size using trnH/psbA primer was different with others for plants each. We established PCR condition and universal primer selection for 17 item's raw materials for foods and determine base sequences aim to PCR products in this study. This study can apply to determine an authenticity of foods through making an comparison between databases and base sequences in gene bank. Therefore, DNA barcode method using universal primers can be a useful for species identification techniques not only raw materials but also processed foods that are difficult to analyze by chemical analysis.

Seedling Emergence and Growth Affected by Priming and $GA_3$ Treatments to Three Campanulan Plant Seeds (Priming과 $GA_3$ 처리에 따른 도라지, 더덕 및 만삼의 출아(出芽)와 생장(生長))

  • Seed germination test done in laboratory does not coincide with field emergence in general. The experiments were carried out to examine the effect of priming and $GA_3$, treatment to seeds of Platycodon grandiflorum; Codonopsis lanceolata and C. pilosula on lapsed time to first seedling emergence, seedling emergence, morphological characters and growth and the cause of poor emergence of C. pilosula. No-treatment as Control (water), priming or $GA_3$ treatment was done with only distilled water for 2 days, $CA(NO_3)_2$ 150 mM for 2 days or $GA_3$ 0.1 mM for 3 days, respectively. Seedling emergence rate was counted every 2 days but morphological characters and dry weight of shoot and root were measured on 38 days after sowing. Their internal seed structures were examined with Scanning Electron Microscope. C. pilosula had poorer seedling emergence rate than P grandiflorum and C. lanceolata showing nearly same rate: Compared to the other treatment (s) P. grandiflorum displayed higher rate in priming and $GA_3$, treatments but C. lanceolata or C. pilosula did the greatest rate in only $GA_3$ or priming treatment, respectively. $GA_3$ treatment to seeds of P. grandiflorum and C. lanceolata shortened the lapsed time to seedling emergence in comparison with Control, 2-days water imbibition before sowing. In all the species plant height and number of leaves per seedling became shorter and less in priming treatment than the other treatments except plant height of C. Pilosula while their hypocotyl length was nearly same in all treatments. Although priming treatment had nearly similar effect to morphological characters, $GA_3$ treatment forced greater shoot, root and aftermath total dry weight per seedling. Poor seedling emergence of C. pilosula was caused by its seed defect like cleavage or lack of embryo, poor development of embryo and endosperm or their separation.

Analysis of Molecular Epidemiological Properties of Staphylococcus aureus Isolates from Domestic Animals and Human Patients by PCR (Polymerase Chain Reaction을 활용한 국내 동물과 사람환자에서 분리한 Staphylococcus aureus 분리주의 분자역학적 특성분석)

  • This study was conducted to analyze the molecular epidemiological properties and to select the most efficient and reliable PCR method on 116 of Staphylococcus aureus (S. aureus) isolates from Korean cattle, black goat, pig, dog, chicken, mouse and also human clinical cases from hospital. The distribution patterns of SSG [species specific genes; coagulase (coa), protein A (spa), nuclease (nuc) and aroA (RsaI) gene] were analyzed by PCR method. Among the SSGs, the nuc-gene was found in all strains $(100\%)$ tested and followed by coa-gene $(87.9\%)$, spa-gene $(91.4\%)$ and aroA-gene $(26.7\%)$, in order. The genetic subtyping by RFLP method was performed on the coa [AluI] and aroA-gene [RsaI] PCR products. The mecA-gene PCR and PCR-RFLP techniques were chosen to detect and verify of MRSA strains. Only the human strains $(12.1\%)$ were detected the positive mecA-gene products (533 bp), which were divided into two specific bands [201 & 332 bp] by HhaI enzyme digestion. On coa-gene and spa-gene typing, coa-gene was typed with ten kinds of genotype and coa-3 type were determined as the most predominant genotype, while spa-gene was divided into eleven kinds of genotype and also spa-7 type were selected the most prevalent genotype based on their genetic variations. On the aroA and coa-gene subtyping by PCR-RFLP, aroA-gene products were discriminated with only seven types of genotype, while coa-gene products were further divided into an eleven genotype, respectively. In comparison of SID values of five PCR based typing methods, the coa-PCR-RFLP (SID0.894) was evaluated the most efficient and reliable tools and followed by coa-PCR (SID0.883) and aroA-PCR-RFLP (SID0.462), in order. In conclusion, we could determined that the coa-PCR-RFLP method was the most suitable genetic analysis tool for S. aureus and MRSA strains from domestic animals and humans.

Carrageenan Content and its Chemical Composition of Eucheuma cottonii from the Coast of Philippine (Philippine산(産) Eucheuma cottonii의 carrageenan 함량(含星)과 그 성상(性狀)에 대하여)

  • One species of Rhodophyceae namely Eucheuma cottonii from the coast of Philippine was analyzed with respect to the content of carrageenan and such chemical characteristics as the content of sulphate and 3,6-anhydro-D-galactose and its solubility in potassium chloride solution. In addition, the same chemical properties were tested in the fractions separated by the different concentrations of potassium chloride. In comparison of the results of carrageenan in Eucheuma cottonii samples from Philippine and Chondrus ocellatus samples from Korea, carrageenan content in Eucheuma cottonii was higher than that of Chondrus ocellatus. Both samples showed more than forty-five percent carrageenan content. The Eucheuma cottonii carrageenan showed a higher 3,6-anhydro-D-galactose than the Chondrus ocellatus carrageenan. The sulphate content was higher in Chondrus ocellatus than Eucheuma cottonii. In fractionation of carrageenan by the solubility methods using potassium chloride solution, the yield of Eucheuma cottonii was highest in fraction I, fraction III was next and fraction II was the lowest.
