Degradation oof Triphenylmrthane Dyes by Citobacter sp. (Citrobacter sp.에 의한 Triphenylmethane계 색소의 분해)

  • The Optimal condition for degradation of crystal violet and other triphenylmethane dyes by Citrobacter sp. SK-3 isolated from the activated sludge of dye manufacturing factory was investigated. The optimal culture medium for the degradation of triphenylmethane dye was composed of minimum inorganic salt medium supplemented with 0.5% galactose, 0.1% beef extract, with the initial pH of 8.0 to 9.0. Under this condition, Citrobacter sp. SK-3 degraded 200 ppm of crystal violet completely within 24 hours. Citrobactre sp. SK-3 also degraded efficiently malachite green, pararosaniline, brilliant green, methyl violet, basic fuchsin and methyl red. Analysis of the degradation products of crystal violet through this layer chromatography and high performance liquid chromatography indicated that the methyl groups bound to crystal violet backborn were gradually demethylated to pentamethyl-, tetramethyl- and trimethylpararosaniline.

The Effect of Endotoxin on Gene Expression and Total Amount of Surfactant Protein A (내독소가 Surfactant Protein A의 유전자 발현과 총단백량에 미치는 영향에 관한 실험적 연구)

  • Background : Surfactant protein A (SP-A) is important in the regulation of surfactant secretion, synthesis and recycling. SP-A has important roles in regulating surfactant metabolism as well as in determining surfactant's physical properties. Since systemic sepsis is one of the common causes of acute respiratory distress syndrome (ARDS) and abnormalities in surfactant function have been described in ARDS, the authors investigated the effects of endotoxemia on the accumulation of mRNA encoding SP-A and SP-A protein content. Methods : Adult rats were given various doses of intraperitoneal endotoxin from Salmonella enteritidis and sacrificed at different times. SP-A mRNA was measured by filter hybridization method. Lung SP-A protein content was determined by double sandwich ELISA assay using a polyclonal antiserum raised in rabbits against purified rat SP-A. Results : 1) The accumulation of SP-A mRNA in the endotoxin treated group 24 hours after 2mg/kg and 5mg/kg endotoxin treatments was significantly increased 50.9% and 27.3%, respectively, compared to the control group (P<0.001, P<0.025). 2) The accumulation of SP-A mRNA 24 hours in the 5mg/kg endotoxin treated group was significantly increased by 26.5% compared to the control group (P<0.01). 3) Total amount of lung SP-A was not altered at 24 hours by various doses of treatment. Total lung SP-A content 144 hours after endotoxin administration was significantly decreased by 51.4% compared to the control group (P<0.01). Conclusions : The specific regulation of SP-A by various time course in vivo is evident. The late decline in SP-A protein content was unexpected and suggests that SP-A may be differentially regulated during lung inflammation. The functional significance of these alterations remains to be clarified.

  • Cryposporidium species have a wide host range. These coccidian parasites are found in close association with epithelial cells of many species of animals including mm. The gastrointestinal tract is most commonly affected in young ruminants and this parasite is thought to be considerable importance in calf diarrhea complex. Major outbreaks of cryptosporidiosis have been reported in calves, lambs, pigs and others including avian species. Cryptosporidiosis is transmitted by oocysts of Cryptosporidium species. Because cryptosporidiosis is common infection among animals, early literature considered it a zoonosis. Human infections contracted from calves, cats, and horse feces. However, the resrvoir host is longer considered the major source of infection. Mild cases of disease have been reported in farm workers. Immunosuppressed, very young and very old persons should avoid contact with this parasite because it may cause severe diarrhea. In order to detect of Crytosporidium sp infection from feces of cattle and pigs at Chonbuk Iksan and Kunsan area, sedimentation and modified acid fast stain were applied. The positive rate of Cryptosporidium sp infection from 1, 176 of cattle and 267 of pigs were 0.5 % and 16.8%, respectively. According to area in Iksan and Kunsan, the positive rates were 0.4% and 0.9% from cattle, and 18.9% and 12.1% from pigs, respectively. In cattle, positive detection rate was 0.6% in milking cows but not in Korean cattle.

  • We cloned the 4.8 kb BglII fragment containing genes downstream pHENX7 from Pseudomonas sp. strain DJ77. The restriction map of the resultant clone, recombinant plasmid pYCS500, was determined. Sequencing analysis of the 465 bp HindIII-ClaI fragment revealed an open reading frame of 282 bp that was then designated phnM. The deduced polypeptide is 93 amino acid residues long with a $M_r$ of 10,008. The PhnM has 37.3-53.9% identity with plant-type ferredoxin proteins such as NahT, XylT, DmpQ, AtdS, PhlG, PhhQ and TbuW and contains the motif similar to well-conserved functional domains of those proteins.

Analysis of the Inhibitory Effect of two Bacterial Strains on Metarhizium anisopliae Induced Fatality Rates in Protaetia Brevitarsis

  • Bacterial species, Bacillus amyloliquefaciens and Lactobacillus species (L. sp.5-1), are known to inhibit the growth of pathogenic bacteria and fungi. Metarhizium anisopliae is a pathogenic fungal species which causes fatal damage to P. brevitarsis populations. Therefore, we investigated the inhibitory effect of B. amyloliquefaciens and L. sp. 5-1 on M. anisopliae induced fatality rates in P. brevitarsis. Samples of M. anisopliae-infected sawdust were treated with strain B. amyloliquefaciens KACC10116, strain L. sp. 5-1 KACC19351, and a combination of the two. P. brevitarsis were fed treated sawdust samples, and their subsequent fatality rate was monitored. The fatality rate fell below 1.5% after 10 days and decreased by approximately 40% after 15 days. On average, the fatality rate decreased by 20%, compared to the control. The difference in the decrease in fatality rate between B. amyloliquefaciens treatment and L. sp. 5-1 treatment was not significant. Results indicate that both strains exhibit high anti-fungal activity, which may be useful in environmental purification efforts. These strains may be used for effective prevention of fungal infection in P. brevitarsis.

Studies on Biodegradation of Synthetic Detergents (합성세제의 성분해에 관한 조사연구)

  • ABS, Alkyl benzene sulfonate, persists for long periods in stream because of its resistance to biologic degradation. Its bio-degradation is very varied in the environments. This investigation was therefore undertaken in order to know the biodegradation of synthetic detergents which are comprising soft and hard forms from February 1976 to September 1976. The biodegradations by spontaneuos settling and aeration were determined. The reduction rates of ABS, DBS, and LAS were 9.8%, 13.7% and 10.4% by the settlings for 5 days at $25^{\circ}$ and 63.3%, 27.2%, and 43.9% by aeration respectively. It was not contributed effectively to biodegradate the synthetic detergents by the simulaneous incubation with micro-organisms isolated from sewage such as Enterobacter sp., E. coli, Bacillus sp., Flavobacterium sp., Pseudomonas sp., Staphylococcus sp., and etc. Tricking filter could also eliminate some amount of detergent. By the application of this investigation, it could be designed that detergents would be regulated in abuse in context with water pollution and be treated at a proper process in the sewage treatment plant to be installed.

  • Alkalophilic sp. YC-335 isolated from soil was capable of producing large amount of cyclodextrin glycosyltransferase (CGTase) in culture broth. This enzyme was successively purified 52.9 folds with 17.8 yield by ethanol precipitation, DEAE-Toyopearl column chromatography and Sephadex G-100 column chromatography. The purified enzyme have a molecular weight of approximately 75,000 estimated by SDS polyaerylamide gel electrophoresis. The optimum pH and temperature for the enzyme activity were 6.0 and 5$0^{\circ}C$, respectively. The enzyme stable between pH 6 and 10, and up to 5$0^{\circ}C$. The thermostability of the enzyme was increased up to 6$0^{\circ}C$ by the addition of 15mM CaCl$_2$.

  • Tremella fuciformis produces white jelly fruitbody which is used as a special food in the orient. Symbiotic relationship between T. fuciformis and Hypoxylon sp. is important for mass production of fruitbody in T. fuciformis. T. fuciformis showed the peak of 24mg/2mL on the 9th day, after that mycelial growth maintained a gentle curve. Protein content increased into 0.69㎍/mL in rapid, T. fuciformis fruiting body maintained high galactose, mycelia of T. fuciformis showed 42.6% trehalose.

Purification and Characterization of an Alkaline Protease Produced by Alkalophilic Bacillus sp. DK1122 (호알칼리성 Bacillus sp. DK1122 균주가 생산하는 알칼리성 단백질 분해효소의 정제 및 특성)

  • An alkaline protease was purified and characterized from an alkalophilic microorganism, Bacillus sp. DK1122, isolated from soil in central Korea. The optimum temperature and pH for the growth of the producer strain were 40℃ and pH 9.0, respectively. The protease was produced aerobically at 40℃ after 24 h incubation in modified Horikoshi I medium (pH 9.0) containing 0.5% (w/v) glucose, 0.8% (w/v) yeast extract, 0.5% (w/v) polypeptone, 0.1% (w/v) K2HPO4, 0.02% (w/v) MgSO4·7H2O, 1% (w/v) Na2CO3, and 3% (w/v) NaCl. The alkaline protease was purified by 70% ammonium sulfate precipitation of the culture supernatant of Bacillus sp. DK1122, followed by CM-Sepharose chromatography. The molecular weight of the enzyme was estimated to be 27 kDa on the basis of SDS-PAGE. The optimum temperature and pH for the protease activity were 60℃ and pH 9.0, respectively. Addition of CaCl2 increased the thermal stability of the purified protease, where 90% of protease activity was retained at 60℃ for up to 3 h. Consequently, it is expected that the alkaline protease from this study, exhibiting stability at pH 7–9 and 60℃, may be promising for application in the food and detergent industries.

Distribution of Carabid Beetles(Coleoptera: Carabidae) in different forests of Central Kangwon-do (강원 중부지역 산림내의 딱정벌레과(科) 분포에 관한 연구)

  • This study deals with the structure of Carabid beetle community surveyed in different stands. Species richness and abundance of Carabid beetles were 1,162 individuals, 23 species, 10 genera in the Pinus densiflora stand, 418 individuals, 23 species, 11 genera in the Pinus koraiensis stand, 782 individuals, 25 species, 11 genera in the Larix leptolepis stand, 1,383 individuals 25 species, 12 genera in the Quercus variabilis stand. Dominant species were Synuchus cycloderus(19.8%), Synuchus nitidus(16.6%), Pterostichus pertirax(16.2%) in the Pinus densiflora stand. Pterostichus sp.1(27.9%), Synuchus nitidus(24.4%), Synuchus cycloderus(10.2%) in the Pinus koraiensis stand, Synuchus sp.1(40.0%), Synuchus nitidus(9.3), Pterostichus sp.1(7.4%) in the Larix leptoepis stand. Pterostichus sp.2(17.4%), Synuchus cycloderus(14.4%). Pterostichus pertirax(14.1%) in the Quercus variabili stand. Shannon species diversity index(H') in the Pinus densiflora, Pinus koraiensis, Larix leptolepis and Quercus variabilis stand were 2.11, 2.25, 2.23 and 2.32 respectively and Evenness index(J') were 0.64, 0.72, 0.69 and 0.72, respectively. Community similarity index showed the lowest value (0.66) between Quercus variabilis stand and Pinus koraiensis stand and the highest value (0.83) between Pinus densiflora stand and Larix leptolepis stand.
