Immobilization of the Antarctic Bacillus sp. LX-1 α-Galactosidase on Eudragit L-100 for the Production of a Functional Feed Additive

  • Partially purified ${\alpha}$-galactosidase from Bacillus sp. LX-1 was non-covalently immobilized on a reversibly soluble-insoluble polymer, Eudragit L-100, and an immobilization efficiency of 0.93 was obtained. The optimum pH of the free and immobilized enzyme was 6.5 to 7.0 and 7.0, respectively, while there was no change in optimum temperature between the free and immobilized ${\alpha}$-galactosidase. The immobilized ${\alpha}$-galactosidase was reutilized six times without significant loss in activity. The immobilized enzyme showed good storage stability at $37^{\circ}C$, retaining about 50% of its initial activity even after 18 d at this temperature, while the free enzyme was completely inactivated. The immobilization of ${\alpha}$-galactosidase from Bacillus sp. LX-1 on Eudragit L-100 may be a promising strategy for removal of ${\alpha}$-galacto-oligosaccharides such as raffinose and stachyose from soybean meal and other legume in feed industry.

  • The optimal culture condition of Bacillus sp. P16 was investigated for production of an extracellular endo-splitting chitosanase. The best carbon and nitrogen sources for the chitosanase production were chitosan and tryptone, respectively. The best condition for the maximum activity was at $37^{\circ}C$ in a medium containing 0.5% powdered chitosan, 1% tryptone, and 1% NaCl(at initial pH 7.0) in a rotary shaker(200 rpm). In a jar fermenter, the culture duration shortened to $6{\sim}12$ hr for maximum activity and the enzyme activity increased about 100% compared with that of flask culture.

Isolation and Characterization of Pseudomonas sp. KM10, a Cadmium- and Mercury-resistant, and Phenol-degrading Bacterium

  • A bacterium which is resistant to both mercury and cadmium, and also capable of utilizing phenol as a carbon and energy source, was isolated from the Kumho River sediments near Kangchang Bridge, Taegu, Korea. The isolate was labeled Pseudomonas sp. KM10 and characterized. The bacteria grew in 4 mM $CdCl_2$and in $70{\mu}M$ $HgCl_2$. The bacteria efficiently removed over 90% of 1 g/l phenol within 30 h. In the presence of 1.250 g/l phenol, the growth of the microorganism was slightly retarded and the microorganism could not tolerate 1.5 g/l phenol. Curing of plasmid from the bacteria was carried out to generate a plasmidless strain. Subsequent experiments localized the genes for phenol degradation in plasmid and the genes for mercury resistance and cadmium resistance on the chromosome. Dot hybridization and Southern hybridization under low stringent conditions were performed to identify the DNA homology. These results showed significant homologies between the some sequence of the chromosome of Pseudomonas sp. KM10 and merR of Shigella flexneri R 100, and between the some sequence of the chromosome of Pseudomonas sp. KM10 and cadA of Staphylococcus aureus pI258. The mechanism of cadmium resistance was efflux, similar to that of S. aureus pI258 cadA, and the mechanism of mercury resistance was volatilization, similar to that of S. flexneri R100 mer.

Isolation and Characteristics of Brachybacterium sp. SY -97 Killing the Harmful Dinoflagellate Cochlodinium polykrikoides (유해적조생물 Cochlodinum polykrikoides를 살멸하는 Brachybacterium sp. SY-97의 분리 및 특성)

  • A bacterial strain SY-97 that showed algicidal activity against Cochlodinium polykrikoides was isolated from coastal water of Uljin (eastern coast of Korea) in August, 2005. The isolated strain was identified as Brachybacterium sp. by morphological and biological tests, and analysis of 16S rDNA sequence. The optimal culture conditions for the growth of strain SY-97 were $30^{\circ}C$, initial pH 7.0, and salinity 2.0%. From the result of cell culture insert experiment, Brachybacterium sp. SY-97 is assumed to produce secondary metabolites which have algicidal activity. When 10% culture filtrate of this strain was applied to C. polykrikoides ($1.2{\times}10^4\;cells/m{\ell}$) cultures, 100% of C. polykrikoides cells was destroyed within 15 hours. The released algicides were heat-tolerant to $100^{\circ}C$ and stable in pH $6.0{\sim}10.0$. These results suggest that Brachybacterium sp. SY-97 is potentially useful for controlling outbreaks of C. polykrikoides.

Purification and Characterization of Cyclodextrin Glycosyltransferase from Alkalophilic Bacillus sp (호알카리성 Bacillus sp.의 cyclodextrin glycosyltransferase의 정제와 특성)

  • Alkalophilic sp. YC-335 isolated from soil was capable of producing large amount of cyclodextrin glycosyltransferase (CGTase) in culture broth. This enzyme was successively purified 52.9 folds with 17.8 yield by ethanol precipitation, DEAE-Toyopearl column chromatography and Sephadex G-100 column chromatography. The purified enzyme have a molecular weight of approximately 75,000 estimated by SDS polyaerylamide gel electrophoresis. The optimum pH and temperature for the enzyme activity were 6.0 and 5$0^{\circ}C$, respectively. The enzyme stable between pH 6 and 10, and up to 5$0^{\circ}C$. The thermostability of the enzyme was increased up to 6$0^{\circ}C$ by the addition of 15mM CaCl$_2$.

The Inhibitory Effects of Intestine-oriented Lactobacillus sp. KP-3 on the Accumulation of Heavy Metals in Sprague Dawley rats (Sprague Dawley 쥐에서 장내 유래 Lactobacillus sp. KP-3의 중금속 축적 저해 효과)

  • To investigate the effect of lactic acid bacteria on the heavy metal adsorption from internal organs and blood, lactic acid bacteria were isolated from human feces. Some strains resistant to heavy metals were selected by incubation in agar media containing each of chrome and cadmium salts. Among them, a strain named KP-3 was ultimately chosen due to its higher growth rate in selective broth medium containing the heavy metals at the concentration of 0.01%. The strain was identified as Lactobacillus sp. based on its morphological, cultural and physiological characteristics. For evaluating the ability to prevent accumulation of heavy metals by selected Lactobacillus sp. strain in vivo, Sprague Dawley rats were fed with heavy metal salts (cadmium, chrome and lead) with or without cultured whole cells for 7 days. The amounts of heavy metals accumulated in liver, kidney and blood were analyzed. As a result, chrome was accumulated to kidney mostly, and lead was frequently found in liver and kidney. Experimental group (rats fed with lactic acid bacteria) showed less accumulation of heavy metal than control group (rats fed with saline solution). The inhibition rates of heavy metal accumulation were calculated to 41.8% (Cd), 33.4% (Cr) and 44.2% (Pb). Especially, feeding lactic acid bacteria strongly reduced accumulation of cadmium in blood. The results showed that feeding Lactobacillus sp. KP-3 could prevent the bioaccumulation of heavy metals in the living body.

  • Various physico-chemical and biological methods have been used to remove. cyanobacteria which causes blooms and releases toxin. The purpose of the following experiment is aimed finding out which cyanobacteria are affected by $_L-lysine $ and what concentration of$_L-lysine $ inhibits cyanobacteria. The 20 samples of Microcystis sp. have been tested. To prove the growth inhibition on Microcystis sp., double-layered agar method and microplate method have been used. When the concentration of $_L-lysine $ is as heavy as 100 ${\mu}g\; ml^{-1}$~300 ${\mu}g\; ml^{-1}$, some Microcystis sp. have made halo zone. Some Microcystis sp. have shown so high activity as to be inhibited in their growth by the $_{L}$-lysine of concentration 10 ${\mu}g\; ml^{-1}$ with microplate method. These activities are various in accordance with every species. In additions, the microplate method has been proven to be an easy way which examine the lytic activity on the species of algae.e.

Physiological Properties of Extracts and the Chemical Composition of Tetraselmis sp. JK-46 Cultured with Deep Seawater (해양심층수로 배양한 해양미세조류 Tetraselmis sp. JK-46의 성분 조성 및 생리활성)

  • This study examined Tetraselmis sp. JK-46 isolated from seawater from the East Sea. Deep seawater (DSW) had a greater effect on the growth of Tetraselmis sp. JK-46 than surface seawater (SSW). The crude protein, lipid, carbohydrate and ash contents of Tetraselmis sp. JK-46 cultured with DSW were 27.2, 37.1, 13.2 and 26.3 %, respectively, and these values were similar to the results for samples cultured with SSW. The contents of Mg, Ca, Fe and K in the DSW cultured samples were 7080.3, 1009.6, 251.2, and 2749.7 mg/100 g, respectively. The fatty acid compositions of Tetraselmis sp. JK-46 cultured with DSW and SSW were 53.7 and 49.0 % polyunsaturated fatty acids (PUFA) and 25.7 and 30.7 % saturated fatty acids (SFA), respectively. The total amino acid contents of the samples cultured with DSW and SSW were 7392.6 and 6376.0 mg/100 g respectively. The 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity of Tetraselmis sp. JK-46 extracts increased with the concentration of the chloroform and ethyl acetate fractions. The half maximal inhibitiory concentrations ($IC_{50}$) of the chloroform and ethyl acetate fractions of DSW and SSW cultured samples were 1.2 and 2.6 mg/mL, and 3.1 and 3.3 mg/mL, respectively. The ethyl acetate fractions of DSW and SSW cultured samples has anticoagulant activity and the activated partial thromboplastin times (APTT) were 93.4 and 89.3 sec., respectively. The chloroform and ethyl acetate fractions showed antimicrobial activity against Bacillus subtilis, Escherichia coli and Candida albicans.

  • An organic solvent-tolerant lipase of Pseudomonas sp. BCNU 154 that was isolated from wastewater in the industrial complex region had optimal activity at $37^{\circ}C$ and pH 8. This crude extracellular lipase from BCNU 154 exhibited maximum stability in toluene, retaining about 6.01 U/ml (117.53%) activity for 2 h. $Ca^{2+}$, $Mg^{2+}$, $NH_4{^+}$, and $Na^+$ ions and triton X-100 activated the enzymes, whereas $Ba^{2+}$, $Hg^{2+}$, and $Zn^{2+}$ ions inhibited their activity. Pseudomonas sp. BCNU 154 lipase revealed stable activity comparable to that of the commercial immobilized Novozym 435. Thus, this organic solvent-tolerant lipase could have potential as a whole cell biocatalyst in industrial chemical processes without the use of immobilization.

Effect of Salt Concentration and Turbidity on the Inactivation of Artemia sp. in Electrolysis UV, Electrolysis+UV Processes (해수의 염 농도와 탁도가 전기, UV 및 전기+UV 공정의 Artemia sp. 불활성화에 미치는 영향)

  • This study was conducted to investigate the effect of salt concentration and turbidity on the inactivation of Artemia sp. by electrolysis, UV photolysis, electrolysis+UV process to treat ballast water in the presence of brackish water or muddy water caused by rainfall. The inactivation at different salt concentrations (30 g/L and 3 g/L) and turbidity levels (0, 156, 779 NTU) was compared. A decrease in salt concentration reduced RNO (OH radical generation index) degradation and TRO (Total Residual Oxidant) production, indicating that a longer electrolysis time is required to achieve a 100% inactivation rate in electrolysis process. In the UV process, the higher turbidity results in lower UV transmittance and lower inactivation efficiency of Artemia sp. Higher the turbidity resulted in lower ultraviolet transmittance in the UV process and lower inactivation efficiency of Artemia sp. A UV exposure time of over 30 seconds was required for 100% inactivation. Factors affecting inactivation efficiency of Artemia sp. in low salt concentration are in the order: electrolysis+UV > electrolysis > UV process. In the case of electrolysis+UV process, TRO is lower than the electrolysis process, but RNO is more decomposed, indicating that the OH radical has a greater effect on the inactivation effect. In low salt concentrations and high turbidity conditions, factors affecting Artemia sp. inactivation were in the order electrolysis > electrolysis+UV > UV process. When the salt concentration is low and the turbidity is high, the electrolysis process is affected by the salt concentration and the UV process is affected by turbidity. Therefore, the synergy due to the combination of the electrolysis process and the UV process was small, and the inactivation was lower than that of the single electrolysis process only affected by the salt concentration.
