Trichoderma disease of oyster mushroom has not been effectively detected in the field for testing its resistance against the disease with its varieties. In this study, we investigated the methods to detect its resistance in the laboratory by using media, which enables us to understand the relevant characteristics (e.g., lysis, toxin enzyme, mycelial growth rate). In coculturing with strains of Trichoderma and oyster mushroom, it is possible to observe the difference in the resistance of oyster mushroom against Trichoderma with the phenomena of barrage reaction, overgrowth and lysis. We also observed the inhibition of mycelial growth of oyster mushroom using the dilution method with 48-well plate, but could not observed the inhibition of mycelial growth using the filter paper method of cultural supernatant. In simultaneously culturing both Trichoderma and oyster mushroom, it was possible to detect the inhibition of the mycelial growth of oyster mushroom, but Trichoderma mycelium did not overgrow against oyster mushroom. We found that the pathogenicity was efficient in using solid medium with the phenomena of overgrowth and lysis by inoculating Trichoderma on top of mycelia of oyster mushroom. In conclusion, the methods (e.g., coculture method, dilution method with 48-well plate, post-inoculation method) are recommended to detect the resistance of oyster mushroom against Trichoderma disease.
Objectives : The objective of this study is to investigate the suppressing effect of the cervi pantotrichum cornu pharmacopuncture on the expression of iNOS mRNA and production of NO in synoviocytes from artificially arthritis-induced mice. Methods : In vitro test, synoviocytes extracted from a knee joint of a mouse were cultivated, and the herbal extract of cervi pantotrichum cornu($0.4mg/m{\ell}$, $0.6mg/m{\ell}$, $0.8mg/m{\ell}$, and $1.0mg/m{\ell}$) was added into the wells of synoviocytes to suppress the expression of iNOS mRNA and production of NO. In vivo test, each ten mice were allocated into three groups; Normal group, CIA-elicitated group(CIA), and group treated with cervi pantotrichum cornu pharmacopuncture after CIA elicitation(CCA). The extract of cervi pantotrichum cornu was injected into the acupoint of $SP_{10}$ to observe the changes of foot thickness in mice and the suppression of MIF, TNF-$\alpha$, NF-${\kappa}B$ p65, and iNOS. Results : In vitro test, the expression of iNOS mRNA and production of NO were dose-dependently decreased in the wells of synoviocytes treated with PMA. In vivo test, the suppression of MIF, TNF-$\alpha$, NF-${\kappa}B$ p65, and iNOS was clearly shown in the pieces of the synovial joint treated with the extract of cervi pantotrichum cornu. The foot thickness also decreased dose-dependently. Conclusions : It is speculated that the cervi pantotrichum cornu pharmacopuncture can be applicable to the therapy of rheumatoid arthritis by suppressing the expression of iNOS mRNA and production of NO.

Ten green leafy vegetables were cultivated from medicinal plant seed. Their nutritional content, taste, appearance and texture were evaluated. Moisture content was $91.23{\pm}0.00%$ in $Saururus$$chinensis$ (Sc), $90.39{\pm}0.05%$ in $Perilla$$frutescens$ (Pa) and $66.78{\pm}0.05%$ in $Agastache$$rugosa$ (Ar). The fat content was 8.89% in $Salvia$$plebeia$ (Sp) and 0.13% in $Peucedanum$$japonicum$ (Pj). The protein content was $11.1{\pm}0.00%$ in Ar and $0.54{\pm}0.03%$ in Pj. Total sugar content was $20.06{\pm}0.03%$ in $Geum$$japonicum$ (Gj), $12.73{\pm}0.03%$ in $Coixlacry$$majobi$ (Cm) and $1.04{\pm}0.03%$ in $Atractylodes$$japonica$ (Aj). Medicinal leafy vegetables had higher glucose, sucrose and fructose, but lower maltose. Ascorbic acid content was $33.67{\pm}1.40$ mg% in Sc, $27.91{\pm}2.91$ mg% in Pa and $15.68{\pm}1.09$ mg% in At. Chlorophyll content was higher in $Perilla$$frutescens$ var. $acuta$ Kudo than other leafy vegetables. The evaluation also showed that Cm, Pa and Aj tasted sweeter and had low acidity.

Coloriztion algorithms, which hide color information into gray images and find them to recover color images, have been developed recently. In these methods, it is important to minimize the loss of original information while the color components are embedded and extracted. In this paper, we propose a colorization method using a wavelet packet transform in order to embed color components with minimum loss of original information. In addition, the compensation processing of color saturation in the recovered color images is achieved. In the color-to-gray process, an input RGB image is converted into Y, Cb, and Cr images, and a wavelet packet transform is applied to the Y image. After analyzing the amounts of total energy for each sub-band, color components are embedded into two sub-bands including minimum amount of energy on the Y image. This makes it possible not only to hide color components in the Y image, but to recover the Y image with minimum loss of original information. In the gray-to-color process, the color saturation of the recovered color images is decreased by printing and scanning process. To increase color saturation, the characteristic curve between printer and scanner, which can estimate the change of pixel values before and after printing and scanning process, is used to compensate the pixel values of printed and scanned gray images. In addition, the scaling method of the Cb and Cr components is applied to the gray-to-color process. Through the experiments, it is shown that the proposed method improves both boundary details and color saturation in the recovered color images.

Biological control has many advantages as a disease control method, particularly when compared with pesticides. One of the most important benefits is that biological control is an environmental friendly method and does not introduce pollutants into the environment. Another great advantage of this method is its selectivity. Selectivity is the important factor regarding the balance of agricultural ecosystems because a great damage to non target species can lead to the restriction of natural enemies' populations. The objective of this research was to evaluate the effects of several different bacterial isolates on the efficacy of biological control of soil borne diseases. White rot caused by Sclerotium cepivorum was reported to be severe disease of garlic and chive. The antifungal bacteria Burkholderia pyrrocinia CAB08106-4 was tested in field bioassays for its ability to suppress white rot disease. In field tests, B. pyrrocinia CAB08106-4 isolates suppressed white rot in garlic and chive, with the average control efficacies of 69.6% and 58.9%, respectively. In addition, when a culture filtrate of B. pyrrocinia CAB08106-4 was sprayed onto wounded garlic bulbs after inoculation with a Penicillium hirstum spore suspension in a cold storage room ($-2^{\circ}C$), blue mold disease on garlic bulbs was suppressed, with a control efficacy of 79.2%. These results suggested that B. pyrrocinia CAB08106-4 isolates could be used as effective biological control agents against both soil-borne and post-harvest diseases of Liliaceae. Chinese cabbage clubroot caused by Plasmodiophora brassicae was found to be highly virulent in Chinese cabbage, turnips, and cabbage. In this study, the endophytic bacterium Flavobacterium hercynium EPB-C313, which was isolated from Chinese cabbage tissues, was investigated for its antimicrobial activity by inactivating resting spores and its control effects on clubroot disease using bioassays. The bacterial cells, culture solutions, and culture filtrates of F. hercynium EPB-C313 inactivated the resting spores of P. brassicae, with the control efficacies of 90.4%, 36.8%, and 26.0%, respectively. Complex treatments greatly enhanced the control efficacy by 63.7% in a field of 50% diseased plants by incorporating pellets containing organic matter and F. hercynium EPB-C313 in soil, drenching seedlings with a culture solution of F. hercynium EPB-C313, and drenching soil for 10 days after planting. Soft rot caused by Pectobacterium carotovorum subsp. carotovorum was reported to be severe disease to Chinese cabbage in spring seasons. The antifungal bacterium, Bacillus sp. CAB12243-2 suppresses the soft rot disease on Chinese cabbage with 73.0% control efficacy in greenhouse assay. This isolate will increase the utilization of rhizobacteria species as biocontrol agents against soft rot disease of vegetable crops. Sclerotinia rot caused by Sclerotinia sclerotiorum has been reported on lettuce during winter. An antifungal isolate of Pseudomonas corrugata CAB07024-3 was tested in field bioassays for its ability to suppress scleritinia rot. This antagonistic microorganism showed four-year average effects of 63.1% of the control in the same field. Furthermore, P. corrugata CAB07024-3 has a wide antifungal spectrum against plant pathogens, including Sclerotinia sclerotiorum, Sclerotium cepivorum, Botrytis cinerea, Colletotrichum gloeosporioides, Phytophotra capsici, and Pythium myriotylum.
The effect of metallothionein expression on the metal resistance and removal by recombinant Saccharomyces cerevisiae containing the plasmid pJW9 was investigated. The recombinant strain S. cerevisiae BZ-pJ was constructed by transforming the host strain S. cerevisiae BZ3l-1-7Ba with the gene coding for a metal-binding protein, metallothionein. Introduction of the MT gene yielded an increase in the minimum inhibitory concentration (MIC) of copper more than three times compared with the host strain. The minimum inhibitory concentrations of $Cr^{2+}, Znr^{2+} and Pb^{2+}, $ were not different for the two strains. The recombinant yeast grown in a medium containing 8mM CuSO4 was able to remove copper with a capacity of 18.9mg $Cu^{2+}$/g dry cell. In a mixture of copper and zinc, the presence of copper relieved the toxic effects caused by zinc, resulting in an enhancement of the final cell density and the specific growth rate of the recombinant yeast. The capability to remove copper by the recombinant yeast was linearly proportional to the copper concentrations in the medium. The efficiency of copper removal was rather constant regardless of the initial copper concentrations. The specific removal of zinc was dependent on the zinc concentrations in media, though, and such dependence was not so pronounced as the concentration of copper.

To compensate the problems of chemical assay in detoxification of recalcitrant and a practical approach in selection of bioremediation bacteria, a simple and rapid toxicity evaluation system was constructed based on Lumbricus rubellus. Long term-culture and specific equipment are not necessary, and semi-quantitative analysis of toxicity at sub-lethal concentration is possible by measuring of dose-dependent increased yellowish secreted compounds. When the toxicity of endosulfan, its metabolites and structurally related chemicals were measured for 24 h, the results were coincided with previous reports. Toxicity was found in endosulfan, endosulfan sulfate, aldrin, and dieldrin, respectively. Rapid and economic selection of endosulfan-detoxifying bacteria was possible using our system. Klebsiella pneumoniae KE-1, K. oxytoca KE-8 and Pseudomonas sp. KS-2P, reported endosulfan degrading bacteria, ameliorated the endosulfan toxicity, whereas E. coli, B. subtilis and other bacteria failed to protect the toxicity of endosulfan in L. rubellus. Our results suggest that the constructed system is useful to selection of microorganism as well as toxicity evaluation against toxic recalcitrants.

The treatment efficiency of rotating biological contactors (RBCs) for the high strength of dairy wastewater was investigated. Two different systems were conducted composing of a single RBC with tapered aeration reactors for the system A and a sequential RBCs following tapered aeration reactors for the system B. Experiments using dairy wastewater were conducted for 50 days period of time, in which hydraulic rates were maintained at the constant ratios of 346L per day and variable BOD concentrations were at the range from 1,358mg/L to 829mg/L, the $COD_{cr}$, concentration of the range were from 2,384mg/L to 1,329mg/L, the range of T-N concentrations was from 66mg/L to 38mg/L, and 50% of internal recycle and 50% of sludge return were performed. Results indicated that the removal efficiencies of the system B were higher than those of the system A. The removal efficiencies of system A for the BOD, $COD_{cr}$, T-N and T-P were 97.8%, 96.7%, 87.2% and 82%, respectively. The removal efficiencies of system B for the BOD, $COD_{cr}$, T-N and T-P were as of 98.5%, 98.5%, 91.3% and 89%.

Acupuncture has fairly good weight-reducing effect in treating simple obesity due to the neuroendocrine regulation. In this study, the antiobesity effects of herbal acupuncture(HA) with Sang-hwang(phellinus linteus) at Fuai(SP16) were investigated in the rat fed on high-fat(HF) diet. Sang-hwang mushroom has been proven to have anti-carcinogenic effects and Sang-hwang extracts are highly effective in treatment and preventive treatment of AIDS, diabetes and high blood-pressure. To determine whether the Sang-hwang herbal acupuncture may have the anti-obesity effect, male Sprague-Dawley(4-wk-old) rats were fed a HF diet for 5 wk, which produced significant weight gain compared to rats were fed a normal diet, and then herbal acupuncture were treated for 3 wk in HF diet group. The body weight, food consumption, food effeciency ratio(FER), body fat mass, plasma nitric oxide(NO) were investigated in rats fed on normal diet, HF diet, and HF diet with HA(HF-diet-HA) groups. NO has been proposed to be involved in the regulation of food intake. In addition, the expression of appetite peptides such as orexigenic peptide neuropeptide Y(NPY) and the anorectic peptide cholecystokinin(CCK) were observed in the hypothalamus. HF-HA group reduced body weight gain, FER, body fat contents and NO concentration compared to HF diet group. The expression of NPY was reduced in arcuate nucleus(ARC), and CCK was increased in the paraventricular nucleus(PVN) after treatment of HA. In conclusion, Sang-hwang HA reduced adipocity, plasma NO and hypothalamic NPY, but increased CCK expression in the HF dietinduced obesity rat, therefore HA may have anti-obesity action through regulating body weight and appetite peptide of the central nervous system.
Heavy reliance on organosynthetic chemical methods to control agricultural pest tends to deteriorate and lower fertility of soil. We studied annual occurrence of earthworm in environment-friendly agroecosystem and attempted to determine the degree of soil contamination. Candidate bioindicators of earthworm in agroecosystem were selected through analysis of earthworm diversities. Depending on the degree of pesticide use, the fields were devided into conventional, less-pesticide, pesticide free, and organic fields. A nearby field not in agricultural use was chosen as a control. This study was carried out in the greenhouses (lettuce and grape) in 2006 and in orchards (apple, pear, peach, citrus and grape) in 2007. Earthworm was collected from 50 $\times$ 50 $\times$ 10 cm$^3$ of respective soil. We found 11 species from 5 genera and 3 families, and 12 species from 4 genera and 3 families in the greenhouses and in orchards, respectively. The contents of organic matter in soil in the greenhouses were variable but quite less so in orchards. Amynthas heteropodus occurred most frequently in both sites suggesting that it could be adopted as a bioindicator in agroecosystem in Korea.
