Current status of pathogen infection in cultured eel Anguilla japonica between 2000 and 2010 (2000~2010년 우리나라 양식산 뱀장어, Anguilla japonica의 병원체 감염현황)

  • Disease survey was conducted to investigate the cause of high mortality in 23 farms of eel, Anguilla japonica, during the period from 2000 to 2010. Seven kinds of fish pathogens were confirmed in the tested fish, which included: Pseudodactylogyrus sp. (infection rate: 65.5%, 19/29 samples), aquabirnavirus (ABV, 62.5%, 15/24 samples), Edwardsiella sp. (44%, 11/25 samples), anguillid herpesvirus 1 (AngHV1, 16.7%, 2/12 samples), Heterosporis sp. (10.3%, 3/29 samples), Vibrio sp. (8%, 2/25 samples), Trichodina sp. (3.4%, 1/29 samples). The rate of single infection was 44.8% (13/29 samples), while 62.1% (18/29 samples) showed mixed type of infection with 2 to 5 different pathogens. The most predominant mixed infection were Pseudodactylogyrus sp.-Edwardsiella sp. (10.3%, 3/29 samples), Pseudodactylogyrus sp.-ABV (10.3%, 3/29 samples), Edwardsiella sp.-ABV (6.9%, 2/29 samples). Infection by the above infectious parasites, bacteria and virus was confirmed in tested eels of juvenile to adult sizes that were cultured at $25{\sim}30^{\circ}C$ in almost the examined farms.

Characterization and Purification of Anti-Complement Polysaccharide from Spirodela polyrhiza

  • We purified and characterized a crude polysaccharide from Spirodela polyrhiza with anti-complement activities. The crude polysaccharide fraction (SP-0) which had potential anti-complement activity was extracted in hot water for 4 hrs at 10$0^{\circ}C$. The ethanol-precipitate, the crude polysaccharide traction (SP-1), showed a potent anti-complement activity. Further purification of the crude polysaccharide (SP-1) was carried out by cetavlon, ion exchange chromatography and gel column chromatography. Among cetavlon fractions, SP-4 showed the most potent anti-complement activity. When 100 $\mu\textrm{g}$/mL of SP-4 was incubated with an equal volume of normal human serum (NHS), the TCH$_{50}$ was reduced by about 78%. When the SP-4 fraction was further purified by DEAE-Sepharose (Cl$^{[-10]}$ ), the SP-4IIa, SP-4IIb and SP-4IIc, absorbed fractions, were almost the same as the anti-complement activities of SP-4. SP-4IIc, having the greatest potential activation and the highest yield by ion exchange chromatography, was further purified by gel column chromatography on a Sepharose CL-6B column. Four polysaccharide fractions of SP-4IIc-1, SP-4IIc-2, SP-4IIc-3 and SP-4IIc-4 were obtained, consisting mainly of arabinose, rhamnose, galactose and glucose, with approximate molecular weights of about 305,000, 132,000, 64,000 and 12,000, respectively. Among these subfractions, SP-4IIc-1 had the most potent anti-complement activity. When the SP-4IIc-1 aggregate was applied to a gel column chromatography in 10 mM and 50 mM NaCl solution, the position of the peak fractions shifted to a low molecular weight region, and the molecular weight of SP-4IIc-1 decreased with increased NaCl concentration in the gel column chromatography. It was found that the self-aggregation formed spontaneously in void volume by gel column chromatography using Sepharose CL-6B in water and the self-aggregation significantly affected the anti-complement function.

  • Mesangial expansion caused by cell proliferation and glomerular extracellular matrix accumulation is one of the earliest renal abnormalties observed at the onset of hyperglycemia in diabetes mellitus. Transcription factor Sp1 is implicated in the transcriptional regulation of a wide range of genes participating in cell proliferation, and is assumed to play an essential role in mesangial expansion, transforming growth factor (TGF)-$\beta$1, plasminogen activator inhibitor (PAI)-1. We have generated a phosphorothioated double-stranded Sp1-decoy oligodeoxynucleotide that effectively blocks Sp1 binding to the promoter region for transcriptional regulation of TGF-$\beta$1 and PAI-1. The Sp1 decoy oligodeoxynucleotide suppressed transcription of these cytokines and proliferation of primary rat mesangial cells in response to serum stimulation. These results suggest that the Sp1 decoy oligodeoxynucleotide could bea powerful tool in preventing the pathogenesis of renal hypertrophy.

Improvement of FK506 Production in the High-Yielding Strain Streptomyces sp. RM7011 by Engineering the Supply of Allylmalonyl-CoA Through a Combination of Genetic and Chemical Approach

  • FK506, a widely used immunosuppressant, is a 23-membered polyketide macrolide that is produced by several Streptomyces species. FK506 high-yielding strain Streptomyces sp. RM7011 was developed from the discovered Streptomyces sp. KCCM 11116P by random mutagenesis in our previous study. The results of transcript expression analysis showed that the transcription levels of tcsA, B, C, and D were increased in Streptomyces sp. RM7011 by 2.1-, 3.1-, 3.3-, and 4.1-fold, respectively, compared with Streptomyces sp. KCCM 11116P. The overexpression of tcsABCD g enes in Streptomyces sp. RM7011 gave rise to approximately 2.5-fold (238.1 μg/ml) increase in the level of FK506 production compared with that of Streptomyces sp. RM7011. When vinyl pentanoate was added into the culture broth of Streptomyces sp. RM7011, the level of FK506 production was approximately 2.2-fold (207.7 μg/ml) higher than that of the unsupplemented fermentation. Furthermore, supplementing the culture broth of Streptomyces sp. RM7011 expressing tcsABCD genes with vinyl pentanoate resulted in an additional 1.7-fold improvement in the FK506 titer (498.1 μg/ml) compared with that observed under non-supplemented condition. Overall, the level of FK506 production was increased approximately 5.2-fold by engineering the supply of allylmalonyl-CoA in the high-yielding strain Streptomyces sp. RM7011, using a combination of overexpressing tcsABCD genes and adding vinyl pentanoate, as compared with Streptomyces sp. RM7011 (95.3 μg/ml). Moreover, among the three precursors analyzed, pentanoate was the most effective precursor, supporting the highest titer of FK506 in the FK506 high-yielding strain Streptomyces sp. RM7011.

Characterization of the molecular features and expression patterns of two serine proteases in Hermetia illucens (Diptera: Stratiomyidae) larvae

  • To investigate the molecular scavenging capabilities of the larvae of Hermetia illucens, two serine proteases (SPs) were cloned and characterized. Multiple sequence alignments and phylogenetic tree analysis of the deduced amino acid sequences of Hi-SP1 and Hi-SP2 were suggested that Hi-SP1 may be a chymotrypsin- and Hi-SP2 may be a trypsin-like protease. Hi-SP1 and Hi-SP2 3-D homology models revealed that a catalytic triad, three disulfide bonds, and a substrate-binding pocket were highly conserved, as would be expected of a SP. E. coli expressed Hi-SP1 and Hi-SP2 showed chymotrypsin or trypsin activities, respectively. Hi-SP2 mRNAs were consistently expressed during larval development. In contrast, the expression of Hi-SP1 mRNA fluctuated between feeding and molting stages and disappeared at the pupal stages. These expression pattern differences suggest that Hi-SP1 may be a larval specific chymotrypsin-like protease involved with food digestion, while Hi-SP2 may be a trypsin-like protease with diverse functions at different stages.

  • The abundance of earthworm in the agro-ecosystem was assessed on the basis of 1,846 individuals collected from May to October 2006. Material was found from litter layers in soils by hand sorting and digging. They encompassed 3 families, 5 genera, and 15 species. Species composition and abundance were as follows: Amynthas agrestis 20.9%, Amynthas koreanus 20.3%, Amynthas heteropodus 15.4%, Amynthas sp. 8.7%, Drawida japonica 8.0%, Eisenia fetida 7.2%, Amynthas hilgendorfi 7.0%, Bimastos parvus 3.2%, Amynthas hupeiensis 2.5%, Aporrectodea caliginosa 1.8%, Amynthas corticis 1.8%, Aporrectodea tuberculata 1.3%, Eisenia andrei 1.2%, Amynthas sp. 20.4%, and Amynthas sp. 10.3%. Of 1,046 individuals, the A. agrestis, A. koreanus, and A. heteropodus were the most numerous members with abundance of 56.7%. Disc-shaped male pore region is useful for classification of Amynthas, and in the survey, we collected two new species with male discs from lettuce cultivated region, Amynthas sp. 1 Amynthas sp. 2. The clitellate vs. aclitellate ratio of the earthworm was approximately 1.25:1.0.

  • From the rhizoplane and rhizosphere of pepper, tomato, lettuce, pasture, and grass, unsoluble inorganic phosphate solubilizing bacterial strains were isolated using plate base assay on Pikovskaya's medium. Two strains, CL-1 and CL-2, which produced largest halo on plates (indicative of phosphate solubilization)were selected for further studies. Based on these biochemical and 16S rRNA analysis strains CL-1, CL-2 were found to be as species of Pseudomonas sp. and Kluyvera sp., respectively. In broth assay Pseudomonas sp. CL-1 and Kluyvera sp. CL-2 solubilized insoluble phosphate by 193.4 mg and $493.6P\;mg\;L^{-1}$, respectively after $3^{rd}$ day inoculation. These effecient phosphate solubilizing bacteria have a potential to be developed as microbial based fertilizer in future.

  • Thirty new species consisting of 24 poecilostomatoid and six siphonostomatoid copepods are described as associates of marine invertebrates from the Moluccas. New taxa in the order Poecilostomatoida are Amarda curvus n. sp., Anchimolgus gracilipes n. sp., A. partenuipes n. sp., A. parangensis n. sp., A. hastatus n. sp., Andrianellus papillipes n. sp., Exodontomolgus communis n. gen. n. sp., Jamescookina moluccensis n. sp., Odontomolgus flammeus n. sp., O. parvus n. sp., O. pavonus n. sp., Paranchimolgus parallelus n. gen. n. sp., and Scyphuliger karangmiensis n. sp. in the family Anchimolgidae; Enalcyonium circulatum n. sp. and E. ceramensis n. sp. in the family Lamippidae; Parastericola rimosus n. gen. n. sp. in the family Lichomolgidae; Pseudanthessius truncus n. sp. and P. planus n. sp. in the family Pseudanthessiidae; Acanthomolgus gomumuensis n. sp., A. dispadactylus n. sp., A. bandaensis n. sp., A. ambonensis n. sp., Kombia avitus n. sp. and Pionomolgus moluccensis n. sp. in the family Rhynchomolgiae. New taxa in the order Siphonostomatoida are Cryptopontius acutus n. sp. in the family Artotrogidae; Asteropontius fungicola n. sp., A. gonioporae n. sp., Collocheres humesi n. sp. and C. amicus n. sp. in the family Asterocheridae; and Molucomes ovatus n. gen. n. sp. in the family Stellicomitidae. Species new to the Moluccas and new host records are also included. Lists of 263 species of associated copepods known from the Moluccas and their 135 species of invertebrate hosts are provided.

Four New Species of the Genus Hyattella (Dictyoceratida: Spongiidae) from Korea

  • Four new species of the genus Hyattella (Dictyoceratida: Spongiidae); H. sinchangensis n. sp., H. mara n. sp., H. chuja n. sp. and H. lendenfeldi n. sp., were collected from Jeollanam-do and Jeju-do, Korea. Hyattella sinchangensis n. sp., H. mara n. sp., H. chuja n. sp. and H. grobosa Lendenfeld, 1889 are very similar to each other in some characters. However, Hyattella sinchangensis n. sp. characterized by no special dermal lamella and no tertiary fibres, and more thin secondary fibres than H. globosa. Hyattella mara n. sp. is differs in having well developed dermal lamella, slightly fasciculated primary fibres, and thick tertiary fibres. Hyattella chuja n. sp. differs in surface characters with a slightly elevated part, skeleton with abundant spongine, and very complex meshes. Hyattella lendenfeldi n. sp. is similar to H. tenella (Lendenfeld, 1889) in skeletal structure, thickness of primary and secondary fibres, but differs in growth form.
