In vitro Evaluation of Different Feeds for Their Potential to Generate Methane and Change Methanogen Diversity

  • Optimization of the dietary formulation is the most effective way to reduce methane. Nineteen feed ingredients (brans, vegetable proteins, and grains) were evaluated for their potential to generate methane and change methanogen diversity using an in vitro ruminal fermentation technique. Feed formulations categorized into high, medium and low production based on methane production of each ingredient were then subjected to in vitro fermentation to determine the real methane production and their effects on digestibility. Methanogen diversity among low, medium and high-methane producing groups was analyzed by PCR-DGGE. The highest methane production was observed in Korean wheat bran, soybean and perilla meals, and wheat and maize of brans, vegetable protein and cereal groups, respectively. On the other hand, corn bran, cotton seed meal and barley led to the lowest production in the same groups. Nine bacteria and 18 methanogen 16s rDNA PCR-DGGE dominant bands were identified with 83% to 99% and 92% to 100% similarity, respectively. Overall, the results of this study showed that methane emissions from ruminants can be mitigated through proper selection of feed ingredients to be used in the formulation of diets.

Effect of Ethanol Extract of Quercus mongolica Leaf as Natural Food Preservative (신갈나무 잎 에탄올 추출물의 식품보존제 효과)

  • This study was investigated to determine the antimicrobial effect of the ethanol extract of Quercus mongolica leaf on microbial growth. The ethanol extract at the concentration of $250\;\mu\textrm{g}/mL\;and\;500\;\mu\textrm{g}/mL$ inhibited the growth of gram positive and gram negative food-borne disease bacteria for 40 hours in tryptic soy broth, respectively. Antimicrobial activity of the ethanol extract from Quercus mongolica leaf was not affected by pH and heat treatment. The comparision between ethanol extract and commercially available preservatives on antimicrobial activity in food system was conducted. When the 0.1% ethanol extract of Quercus mongolica leaf was added to pine needle drink and carrot juice, antimicrobial activity was similar to those of containing 0.05% benzoic acid and 0.5% grapefruit seed extract. Also addition of 2~3% ethanol extract to the soybean paste inhibited the microbial growth up to 7 week, comparable to the inhibition of 2% ethanol. Thus, this results indicate that the ethanol extract of Quercus mongolica leaf may be useful as natural antimicrobial agents.

Determination of the Adulteration of Sesame Oils Sold in Markets by Gas Chromatography (시중 참기름에 혼입된 이종기름에 관하여)

  • This study primarily attempted to establish the method for the determination of the adulteration in the sesame oil. First of all, extensive experiment was conducted to determine the composition of genuine sesame oil prepared from Korean, Japanese, Taiwanese and Chinese sesame seed. Sesamin and sterols in unsaponfiable matter were examined along with fatty acid in saponifiable fraction by Gc. There was no significant difference in the composition of sesamin and sterols in sesame oils prepared from Korean and foreign seeds. The ranges of sesamin and ${\beta}-sitosterol$ against campesterol were 3.32~5.46 and 2.39~2.99 respectively in all samples. Similiar composition of fatty acids was showed in all pure sesame oils, in which the contents were 8.37~lO.09% palmitic acid, 4.61~5.50% stearic acid, 35.24~39.97% oleic acid, 43.04~49.76% linoleic acid, O.21~O.31% linolenic acid and 0.40~O.69% arachidic acid. Among the commercial sesame oils sold in Markets, three sesame oils from Japan revealed low sesamin, high linoleic acid and linolenic acid, and low oleic acid and stearic acid, suggesting the adulteration with soybean oil.

Physico-chemical Characteristics and Utilization of Raw Materials for Mushroom Substrates (버섯 병재배 배지재료의 이화학성과 활용)

  • To provide a basic information on the chemical concentrations of different raw materials used in mushroom cultivation, the raw materials were collected from 13 Flammulina velutipes, 14 Pleurotus eryngii, and 8 P. ostreatus farms and analyzed to calculate moisture contents, pH, total carbon(T-C), total nitrate(T-N), total phosphate(T-P) and 13 different cations. In our results, the C : N ratios of cotton seed meal, beet pulp, and corn-cob were 6~17, 12~29 and 56~127, respectively. Depending on the companies which process these materials, the range of C:N ratio of soybean curd residue was approximately either 8~9 or 14~17 with wider range of C : N ratio of the raw materials imported from other countries without the detection of heavy metals. In this study, the formula was provided to calculate the composition of mixed media for mushroom cultivation based on the ingredient chart of different raw materials.

A Study on Trans Fatty Acids Contents of Vegetable Oils Added to Can (통조림에 주입된 유지중 Trans 지방산 함량에 관한 연구)

  • Trans fatty acids are formed during refining process of vegetable oils (deodorization), hydrogenation, the high temperature treatment of oils and rancidity. Trans fatty acids contents were measured in vegetable oils added to tuna, oyster and mussel Can by Glass Capillary Gas Chromatography. Also Acid Values, Peroxide Values, iodine Values and Saponification Values of vegetable oils added to Can were determined. The results were as follows; 1. Among vegetable oils added to Can, trans fatty acids isomer of cotton seed oil were mostly t,c-18:2 and t,c,c-18:3. 2. Total average contents of trans fatty acids of soybean oil added to tuna Can (Ab) were shown the highest values among tuna (Aa, Ab), oyster, mussel (Cb) Cans. 3. All of oils added to Can had been keeping safe quality during 1 year or more with low Acid and Peroxide Values.

  • This paper is a study on the development and consumer preference of the soup stew HMR new products, and the main contents are as follows. A total of nine products were studied, including three developed products and six market products. For this study, the demographic characteristics, the awareness and attitude of HMR products were investigated. In addition, the consumer panel evaluated overall liking, appearance, color, flavor, and taste using the nine-point recertification scale, while the strength assessed viscosity, sweetness, saltiness, sourness, and Umami. Familiarity, health degree, Purchase intention and Recommendation intention were investigated and the reasons for preference and non preference were analyzed by multi-response method. The consumer preference analysis indicates that the product is competitive among existing products and that it would be desirable to improve after identifying the causes of non-preferred factors.

  • 콩의 원산지는 만주를 중심으로한 동북아시아를 원산지로 추정하고 있으며, 동아시아인 한국, 중국, 일본의 주요 식재료로 다양하게 이용되어왔다. 콩은 단백질과 지방의 공급원으로서 뿐 아니라 최근에는 콩의 생리활성 물질이 항암효과, 골다공증, 신장질환과 같은 질병예방에도 효과가 있는 것으로 알려지면서 활용가능성이 높아지고 있다. 본 연구는 유전자원센터에 보존 중인 콩 수집단에서 농업형질, 기초정보 등을 토대로 핵심집단을 작성하고 그 중 한국, 중국, 일본 수집단 각 425, 93, 42자원의 개화일수, 생육일수, 주당협수, 100립중, 주당수량과 같은 양적형질을 비교하였다. 개화일수와 생육일수는 중국수집 자원에서 평균38일, 107일로 가장 빠르며, 한국수집 자원은 평균 53일, 127일로 가장 늦었다. 협당립수는 중국수집 자원에서 협당 평균3립으로 가장 많았으며, 한국과 일본 수집자원은 각 2.3과 2.4립으로 유사하였다. 주당협수는 한국 수집자원에서 주당 평균 327.4개로 가장 많고 일본이 248.0개 중국이 224.4순으로 조사되었다. 100립중은 일본 수집 자원이 평균 33.6g으로 대립이 많고 한국 26.5g , 중국 21.1g 이었다. 주당수량은 한국 수집자원이 평균 119.8g, 일본 84.2g, 중국 76.8g순이었다. 이들 5개 양적형질을 토대로 군집분석한 결과 한국 자원은 대다수가 C3 그룹에 속하고, C1 그룹에 일부가 속하였으며, 일본 자원은 C3 그룹과 C2 그룹에, 중국 자원은 C2 그룹에 속하여 한국과 일본자원이 유사하고 중국자원과는 차이를 보이는 것으로 나타났다. 이러한 특성조사 결과는 자원의 수집, 도입시 다양성 확보를 위한 기초정보로 이용할 수 있다.

Effects of a Mungbean Cultivar, Jangannogdu on Nymphal Development, Adult Longevity and Oviposition of Soybean Stink Bugs (장안녹두가 노린재류의 약충발육, 성충수명 및 산란에 미치는 영향)

  • Nymphal development, adult longevity, and oviposition of six different species of stink bugs were compared on seeds of a soybean cultivar, Taekwangkong, and two mung bean cultivars, Kyungseonnogdu and Jangannogdu in the insect rearing room ($28{\pm}2^{\circ}C$, 16L:8D). Nymphs of six species of stink bugs normally developed on Taekwangkong seed. While nymphs of Riptortus clavatus, Halyomorpha halys, and Dolycoris baccarum developed on seeds of Kyungseonnogdu and Jangannogdu normally, all nymphs of Plautia stali, Piezodorus hybneri and Nezara antennata died after 4th instars on Kyungseonnogdu and after 3rd instars on Jangannogdu. Each instar period tended to be increased as nymphal stages were progressed. Nymphal period was shortest on Taekwangkong seed, followed by Kyungseonnogdu and Jangannogdu, irrelevant to stink bugs species. Emergence rate, adult longevity, and preoviposition period of 6 stink bugs were different depending on the reared seeds. Of 6 different species examined, Riptortus clavatus, Halyomorpha halys, and Dolycoris baccarum which fed on Jangannodu did not emerge, Plautia stali, Piezodorus hybneri, and Nezara antennata which fed on Jangannodu did not oviposit.

Antioxidant and Cytoprotective Effects of Socheongja and Socheong 2, Korean Black Seed Coat Soybean Varieties, against Hydrogen Peroxide-induced Oxidative Damage in HaCaT Human Skin Keratinocytes (HaCaT 인간 피부 각질세포에서 과산화수소 유도 산화 손상에 대한 소청자 및 소총2호의 항산화 및 세포보호 효능)

  • Black soybeans are used as food sources as well as for traditional medicines because they contain an abundance of natural phenolic compounds. In this study, total phenolic contents (TPCs) of Korean black seed coat soybean varieties Socheongja (SCJ), Socheong 2 (SC2) and Cheongja 2 (CJ2) as well as their antioxidant capacities were investigated. Among them, TPCs were abundantly present in the order of CJ2$H_2O_2$-stimulated HaCaT human keratinocytes. Our results revealed that treatment with SCJ and SC2 prior to $H_2O_2$ exposure significantly increases the viability of HaCaT cells, indicating that the exposure of HaCaT cells to SCJ and SC2 conferred a protective effect against oxidative stress. SCJ and SC2 also effectively inhibited $H_2O_2$-induced apoptotic cell death through the blocking of mitochondrial dysfunction. SCJ and SC2 also attenuated the phosphorylation of Histone H2AX. Furthermore, they effectively induced the levels of thioredoxin reductase (TrxR) 1, a potent antioxidant enzyme, which is associated with the induction of nuclear transcription factor erythroid-2-like factor 2 (Nrf2); however, the protective effects of SCJ and SC2 were significantly reversed by Auranofin, a TrxR inhibitor. These results indicate that they have protective activity through the blocking of cellular damage related to oxidative stress via the Nrf2 signaling pathway. In conclusion, our study indicated that SCJ and SC2 might potentially serve as novel agents for the treatment and prevention of skin disorders caused by oxidative stress.

A Study on the Formation, Contents of Foods, and Antioxidative Effect of Conjugated Linoleic Acid (Conjugated Linoleic Acid의 형성과 식품중의 함량 및 항산화효과에 관한 연구)

  • A research was carried out to determine the formation, contents in foods, and antioxidative effects of conjugated linoleic acid (CLA). CLA was known as a mixture of positional isomer of linoleic acid (LA), that was included in milk, meat, and fish. The formation of CLA from methyl linoleate and soybean oil (SBO) storecd at 20${\pm}$1$^{\circ}C$ was higher than at 40${\pm}$1$^{\circ}C$, and CLA formation from methyl linoleate stored at 20${\pm}$1$^{\circ}C$ was over 13 times higher than early amounts(188 ppm) and was higher than that from SBO. In edible vegetable oils, the content of CLA were the highest in canola oil (CAO, 348 ppm) but were decreased during storage at 40${\pm}$1$^{\circ}C$, while the content of CLA in cotton seed oil (CSO) were 292 ppm, which increased dramatically (1322 ppm) during 28 days of storage at 40${\pm}$1$^{\circ}C$. Because the peroxide value (POV) of CSO at that time was very low (10.05 meq/kg $.$ oil), CLA occurrence of CSO was shown to be very available during storage at temperature. CLA content of milk from a market ranged 293∼2148 ppm, which depended on the manufacturing, companies. In meat, the CLA content was very high in pork (2379 ppm), and among fishes, that of spanish mackerel was the highest (1040 ppm, almost same as beef, which increased greatly (2039 ppm) during boiling with seasoning. Antioxidative effect of CLA on SBO was almost same as that of BHT until 7 days of storage at 40${\pm}$1$^{\circ}C$, but decreased greatly after that period. In case of com oil (CNO), antioxidative effects of CLA were higher than those or BHN and tocopherol, suggesting that the effect was different depending on the kinds of oils used as substrates. During heating at 180${\pm}$1$^{\circ}C$, antioxidative effect of CLA on SBO appeared almost same as those or BHT and tocopherol, and it was also shown greater effects in heating at high temperature (180${\pm}$1$^{\circ}C$) than at low temperature(40${\pm}$1$^{\circ}C$).
