Identification of a Pathogen-Induced Glycine max Transcription Factor GmWRKY1

  • On screening pathogen-resistant soybean, we identified a WRKY type transcription factor named a Glycine max WRKY1 (GmWRKY1). Expression of GmWRKY1 gene was induced in the soybean sprout by Pseudomonas infection. The GmWRKY1 was expressed in all of the tissues with high levels in stems, leaves and developing seeds. The protein Gm WRKY1 contains highly conserved two WRKY DNA-binding domains having two $C_2-H_2$ zinc-finger motif ($C-X_{4-5}-C-X_{22-23}-H-X-H$) in its N-terminal and C-terminal amino acid sequences. In electrophoresis mobility shift assay, the GmWRKY1 protein bound specifically to W-box elements in the promoters of defense related genes. These results demonstrated that GmWRKY1 is one of the soybean WRKY family genes and the plant-specific transcription factors for defense processes.

Antifungal Properties of Streptomyces bacillaris S8 for Biological Control Applications

  • Soybean (Glycine max), a crucial global crop, experiences yearly yield reduction due to diseases such as anthracnose (Colletotrichum truncatum) and root rot (Fusarium spp.). The use of fungicides, which have traditionally been employed to control these phytopathogens, is now facing challenges due to the emergence of fungicide-resistant strains. Streptomyces bacillaris S8 strain S8 is previously known to produce valinomycin t through a nonribosomal peptide synthetase (NRPS) pathway. The objective of this study was to evaluate the antifungal activity of S. bacillaris S8 against C. truncatum and Fusarium sp., assessing its efficacy against soybean pathogens. The results indicate that strain S8 effectively controlled both above-ground and underground soybean diseases, using the NRPS and NRPS-related compound, suggesting its potential as a biological control in plant-microbe interactions. These findings underscore the pivotal role of the stain S8 in fostering healthy soybean microbial communities and emphasize the significance of microbiota structure studies in unveiling potent biocontrol agents.

Heterodera glycines-Induced Syncytium Structures Related to the Nematode Growth and Reproduction in Susceptible Soybean Cultivars

  • The production of soybean cyst nematode (SCN), Heterodera glycines, including female formation and fecundity was much higher in SCN race 14 (R14) than in race 3 (R3) in susceptible soybean cultivars Bragg (intolerant), Lee74 (moderately tolerant), and PI 97100 (highly tolerant). The nematode body was also significantly larger in R14 than in R3 at 20 days after inoculation, but the further nematode growth appeared to be slower in R14 than in R3, resulting in no significant difference between the two races at 30 days after inoculation. Within each race, no significant difference was observed in the growth and reproduction among the soybean cultivars tested. Syncytial areas near the nematode lip regions (infection sites) were measured for each soybean cultivar-SCN race combination. R14 induced significantly larger syncytia than R3. Bragg had relatively larger syncytia than Lee74 and PI 97100, but the difference among the soybean cultivars was minimal or not significantly different. Syncytium occupation in the stelar region differed only between PI 97100 and the other two cultivars, which may be somewhat, but not exactly, related to tolerance levels. Syncytial cytomplasm was degenerated more with R14 and in Bragg than with R3 and in Lee74 and PI 97100, respectively. In light microscopy, degenerated syncytia were characterized by depleted and loose cytoplasm with less plastids than normal-looking (intact) syncytia which had dense syncytial cytoplasm. Electron microscopy revealed that degenerated syncytia contained highly vacuolated cytoplasm with degenerated plastids. The above results suggest that structural characteristics of syncytia may match the nematode growth and reproduction.

Differential Subcellular Responses in Resistance Soybeans Infected with Soybean Cyst Nematode Races

  • Early nematode development and subcellular responses in resistant soybean lines PI 88788 and PI 437654 infected with races 3 (R3) and 14 (R14) of soybean cyst nematode (SCN), Heterodera glycines Ichinohe, were compared. SCN R14 nematodes penetrated and developed significantly more than R3 at 5-6 days after inoculation. Both races also penetrated and developed more in PI 88788 than in PI 437654. Syncytia, characterized by cell wall dissolution and cellular hypertrophy, were developed more in PI 88788 than in PI 437654 and more by R14 than R3, for which less necrotic responses occurred in the former than the latter. This suggests that the latter two may be more resistant and less virulent than the former two, respectively. A common structural feature found in each of PI 437654 and PI 88788 in relation to SCN-resistance was the formation of prominent cell wall appositions and nuclear degeneration prior to cytoplasmic degradation in syncytial cells, respectively. Necrosis and cell wall apposition are types of hypersensitive responses occurring at early stages of the nematode infection so that these structural modifications indicate the inhibition of initial syncytial development related to the early nematode development. As soybean cultivars and lines with identical or similar genotypes have the same types of structural features related to SCN-resistance, the structural modifications induced by SCN infection may result from the expression of inheritable resistance genes, of which the information can be used for breeding soybean cultivars and lines specifically resistant to SCN races.

분얼형(分蘖型) 옥수수 교잡종(交雜種)과 대두(大豆)의 교호작(交互作)에 대한 수량(收量) 비교(比較) 시험(試驗) (Comparison of Yield in Aiternating Crop System Mixed of Tillering Hybrid Corn and Soybean)

  • 대두(大豆)와 분얼형(分蘖型) 옥수수의 상호간(相互間)에 생육의 안정성(安定性)을 도모하고 동시에 최대수량을 얻고자 파종비율을 달리한 교호작(交互作)으로 하여 생체(生體), 건물(乾物) 및 종실중(種實重)의 상호(相互) 비교(比較)를 통해 적정(適定) 파종비율(播種比率)을 구명하고자 실시한 결과는 다음과 같다. 1. 콩의 간장(桿長)은 단작(單作)의 경우 59.5cm 였는데 콩의 파종비율를 낮추고 옥수수의 비율을 증가 시킨 처리에서 크게 도장(徒長)되었으나 상대적으로 콩의 비율이 증가된 2:1, 3:1, 3:2, 3:3 처리에서는 비교적 콩의 초형(草型)이 단작형(單作型)으로 안정(安定)되었다. 2. 대부분 위와같이 도장된 처리구에서는 분지수(分枝數)와 경태(徑太)가 크게 감소되었고 도대율(倒代率)의 증가가 뚜렸한 현상을 보였다. 3. 10a당 생체중(生體重)은 콩의 비율이 옥수수에 비해 상대적으로 낮은 1:1, 1:2, 1:3 처리구가 단작에 비해 크게 감소 되었으나 콩의 비율을 증가 시킨 2:1, 2:3, 3:1, 3:2, 3:3 등에서는 오히려 단작(單作)보다 크게 증가 되었다. 4. 10a당 건물중(乾物重) 역시 같은 경향이었으나 10a당 종실수량의 경우에는 단작이 276kg으로 가장 높았고 2:1, 3:3처리구 역시 비교적 높게 나타났다. 5. 옥수수의 생육 특성을 보면 평균 간장(桿長)이 208.9cm, 주당(株當) 펑균 분얼수(分蘖數)는 2.1개, 주당(株當) 평균 이삭수(數)는 4.0개로 콩의 파종비율의 증감(增減)에서 다소의 변이(變異)를 보였으나 유의차(有意差)는 인정 되지 않았다. 6. 각 처리간 10a당 생체중, 건물중, 종실중은 다소의 유의차를 보였는데 생체중과 건물중은 3:1, 2:1처리구에서 단작보다 1000kg와 100kg정도가 각각 높았고 종실중은 모든 처리구에서 단작의 438kg보다 높았는데 특히 3:2, 2:1처리에서 530kg 정도로 높게 나타났다. 7. 두 공시 작물을 파종 비율에 따라 10a 당으로 환산(換算)한 총량(總量)을 보면 생체중의 경우 3:1, 3:2, 2:1처리에서 콩 단작의 5721kg이나 옥수수 단작의 4358kg 보다 높았고 건물중의 합은 옥수수 단작, 2:1, 3:1순으로 높았는데 종실중은 2:1, 3:1처리구에서 430kg 이상이었다.

인공안개처리에 따른 콩의 잡초경합 양상 및 제초제반응 특이성 (Specificity in Weed Competition and Herbicide Response of Soybean under Foggy Condition)

  • 안개처리에 따르는 콩의 생육ㆍ잡초와의 경합 양상 및 제초제반응을 검토하여 안개상습지에서 작물피해 최소화 및 잡초방제체계 설정을 위한 기초자료를 제공코자 온실내 포트 시험으로 수행하였다. 안개조건에서 콩의 초장 및 엽영은 전 생장기간에 걸쳐서 비안개조건에 비해 증가되었으며 파종 후 100일째 최종 콩의 수량은 초장이 14% 증가한데 반해 수당협수, 협당 입수 및 100입중이 각각 오히려 16, 24 및 23% 감소하였다. 주요발생초종은 바랭이, 피, 미국개기장, 흰명아주 및 한련초 등이었으며, 비안개조건보다 안개조건에서 그 발생량이 훨씬 컸다. 안개조건에서 경합기간별 콩의 초장변화는 경합기간이 길수록 감소 정도가 컸으나 비안개처리와의 차이는 경미하였다. 안개조건에서 경합기간별 콩의 초장은 40일 경합기간까지에서는 비안개조건에 비해 증가하였으나 60일 이상의 경합기간에서부터 감소되는 경향이었다. 그러나 안개조건에서 수당협수는 경합기간처리와 무관하게 감소하였다. 제초제별 초장을 근거로 한 약해는 다소 인정되나 안개로 인해서 Qu-izalpfop를 제외한 제초제들에서는 무처리와 유사하게 증가한 초장을 각각 보였다. 수당협수 및 100립종은 무처리와 유사하게 나타났으나 안개조건하에서는 비안개조건에 비해 유사하거나 감소되는 경향을 보였다. 처리 후 40일제에 생체중을 근거로 한 잡초방제가로 볼 때 토양처리된 Ethalfluralin 및 Trifluralin과 경엽처리된 Sethoxydim 및 Quizalofop는 안개유무에 관계없이 모두 97%이상의 방제가를 나타었다.

열대환경하에 있어서 대두재식밀도가 각종형질에 미치는 영향 (The effect of plant spacing on several agronomic traits of a soybean variety under the tropical environment)

  • This experiment was conducted to determine the optimum row width and plant spacing within row under the tropical environment for a leading soybean variety Palmetto and it was carried out at Eakmat Experiment Station in Vietnam. The experiments were arranged in a split plot design with four replications and the test was repeated twice in two years. Variations for seed yield due to the distance between rows were significant at 1 per cent level in 1967 test(dry season growing) and at 5 per cent level in 1968 test. Significant differences for plant height, lodging, maturity, number of branches per plant, and number of pods per plant due to the row width were not found in both tests, while significant differences in difference spacing within row was found in all traits studied. Interaction between width of row and spacing within row for seed yield and plant height were found at five percent level in 1967 test. These results indicate that close planting may increase in seed yield and plant height in both seasons, and decrease in loadging. From these studies, one could be understood that the plant population, particularly in dry season, plays decisive roles on seed yield in soybean culture, and the maximum plant height and minimum value of lodging index were also observed in closest spacing plots. The highest soybean yield in late planting would be expected by a combinations of 30 cm(between rows)${\times}$5cm(between hills) plot, while 40cm ${\times}$ 5cm planting method would be suggested for rainy season growing. Highest seed yield was obtained at closet spacing in both dry and rainy season, and these results led to drilling method in seeding where about 20 seeds per meter of row could be recommended. Besides the seed yield, the close planting may produce some advantages, such as increase plant height and decrease lodging, weeds and erosion of surface soil.

결명재배가 토양선충밀도와 콩의 생육에 미치는 영향 (Effect of Pre-cultivation of Cassia tora on Soil Nematode Population and Soybean Growth)

  • 콩을 연작해온 밭토양에 경명을 재배하므로써 토양선충밀도를 줄일 수 있으며 이것이 콩생육에 어떤 영향을 미치는가를 알기 위하여 콩단작구, 결명단작구, 콩-결명혼작구 및 간작구를 만들어 1년간 재배한 후 그 자리에 3년동안 콩을 재배하면서 선충밀도와 콩의 생육을 조사하엿다 1. 콩단작구에 비하여 결명혼작구 및 간작구에서 토양선충밀도는 크게 낮았고 결명단작구에서는 결명의 생육이 왕성해지면서 토양선충이 완전히 없어 졌다. 2. 결명단작구에 이듬해부터 콩을 계속 재배했어도 토양선충의 밀도는 아주 낮았고 결명혼작고 또는 간작구에 콩을 계속 재배하여도 3년차까지는 콩연작구보다 선충밀도가 훨씬 낮았다. 3. 결명의 뿌리에는 뿌리혹이 착생하지 않는것이 특징인데 콩과 결명을 혼작 또는 간작하여도 콩뿌리에 형성된 뿌리혹의 수는 콩단작구와 차이가 없었다. 4. 콩 성열기의 주당입수, 주당입수 및 주당입중은 결명과 혼작 또는 간작하여도 콩단작구와 차이가 없었다. 결명단작구, 혼작구 및 간작구의 뒷자리에 이듬해부터 콩을 재배한 결과 2년차까지는 콩연작구보다 생육이 더 좋았다.

Topping Effect on Growth and Yield of Soybean Growth in Paddy Field

  • This study was conducted to determine the effects of two plant populations (28 and 14 plants per $m^2$) and two toppings in conventional plant population (28 plants per $m^2$) on soybean (Glycine max L. cv. Pungsannamulkong) cultivated in the paddy field. The two topping time were taken at 6$^{th}$ to 7$^{th}$ and 8$^{th}$ to 9$^{th}$ trifoliolate leaf stages in the conventional plant population. Experimental design for growth data was a randomized complete block with three replications, and samples were taken at R1 (July 31), R3 (August 19), R5 (September 2) and R7 (September 23) growth stages. The branch number of soybean was relatively higher in the low plant population (14 plants per $m^2$) and with the topping at the 6$^{th}$ to 7$^{th}$ leaf stage, in the conventional plant population (28 plants per $m^2$), and with topping at the 8$^{th}$ to 9$^{th}$ trifoliolate leaf stage in descending order. The highest average branch length of soybean was observed in the low population and the longest branch length was observed from the soybean with topping at the 6$^{th}$ to 7$^{th}$ leaf stage. The leaf number per plant was decreased in order of in the low population, with the topping at 6$^{th}$ to 7$^{th}$ trifoliolate leaf stage, with the topping at 8$^{th}$ to 9$^{th}$ trifoliolate leaf stage, and in the conventional population. The leaf area was high in the low population and with topping at 6$^{th}$ to 7$^{th}$ trifoliolate leaf stage and was relatively low in the conventional population and with the topping at 8$^{th}$ to 9$^{th}$ trifoliolate leaf stage in soybean. The dry weight of leaves and branches was high in the low population and with the topping at 6$^{th}$ to 7$^{th}$ trifoliolate leaf stage and was relatively low in the conventional population and with topping at 8$^{th}$ to 9$^{th}$ trifoliolate leaf stage. The leaf number per plant was high in the low population and with topping at 6$^{th}$ to 7$^{th}$ trifoliolate leaf stage and was relatively low in the conventional population and with topping at 8$^{th}$ to 9$^{th}$ trifoliolate leaf stage.
