Isoflavones are reported to playa role in menopausal women as a phytoestrogen, which can replace estradiols in hormone replacement therapy (HRT). Recently; due to the risk of breast cancer by HRT, phytoestrogens (e.g. isoflavones) have been focused as an alternative therapy in menopause. In the study, we investigated whether isoflavones derived from Sophorae fructus (SISO) have more benefit than that of soybean isoflavones in estrogen deficient rats. We found that SISO effectively controled $H_2O_2$ comparing with the baseline (p<0.01 vs. post value of OVX-Cont), and the blood sugar and weight were also controlled with decreasing patterns. Additionally, in LDH assay for cytoprotective effect in Neuro-2a cell line, SISO protected cells from the damage by SNAP (p<0.05). In conclusion, SISO may have more beneficial effect in enhancing the menopausal signs than that of soybean isoflavones and the cytoprotective effect in neuron cells suggests that SISO can play a certain role in neuroprotection after menopause.

Considerable beneficial effects on osteoporosis from soy intake in postmenopausal women are being reported (Murkies et al.1998, Arjmandi et al. 1996). Isoflavones(IF), one of phytoestrogenic substances in the soybean, have been suggested for the effect. Our preliminary study showed that even the young chicks fed IF-rich diet tended to have higher bone ash content than those fed IF-poor diet. IF have been reported to decrease bone ash content than those fed IF-poor diet, IF have been reported to decrease bone loss by reducing bone resorption. Soybean meal has been one of the most important protein sources in poultry diets. We assume that the IF intake through dietary soybean meal could give meaningful influences on the birds. This study was carried out to determine effects of dietary IF from soybean sources on bone development in young chicks. Soy protein concentrate (SPC, IF-poor)and an IF concentrate(Phyto-Nutramin) were formulated together with purified-type ingredients to provide three different levels(25, 240 and 480 mg/kg) of total IF. Control diet(240 mg/kg) was prepared with isolated soyprotein(ISP, IF-rich). The diets were fed to 96, 3-day old, layer-type, male chicks, divided into 4 treatments with 3 replications for 3 weeks. Chicks fed the ISP diet had better gain/feed ratios than fed the SPC diets. Birds fed the diets with higher levels of IF tended to show higher values in serum total and ionized Ca% and tibial bone density, length % ash, stiffness and strain. This trend however, appeared less significant at the end of third week. No noticeable differences in sizes of comb and testicle and serum alkaline phosphatase activities were observed among the dietary groups. These results suggest that dietary isoflavones from soybean sources could be associated with chemical composition and physical properties of bone in sizes of comb and testicle and serum alkaline phosphatase activities were observed among the dietary groups. Isoflavones found in soybeans are present as glycosides and aglycones, which differ according to their chemical structure. The absorption rate and bioavailability of aglycones are greater than those of glycosides. It is known that aglycone isoflavones in soybean was converted from glycoside isoflavones by activating of endogenous β-glucosidase under drying (40~60℃) conditions after soaking. In this study, we compared and analyzed the proximate composition and isoflavone contents of soybean powders prepared under dried after soaking were analyzed. In the comparison of the proximate compositions, the moisture contents of white soybean powder dried after soaking (WSPDS) and black soybean powder dried after soaking (BSPDS) were decreased as compared with those of the control white soybean powder (CWSP) and control black soybean powder (CBSP). Whereas the contents of other proximate compositions were increased. The aglycone isoflavone contents were higher in WSPDS than in CWSP. Whereas the aglycone isoflavone contents of BSPDS had a similar content those of CBSP because of the component characteristics of black soybean. In conclusion, we believe that it is appropriate to identify and utilize the component characteristics of soybean varieties in order to induce an increase in the functional component content of soybeans under drying conditions after soaking.

Phytoestrogens have been used as a food supplement to prevent osteoporosis. The isoflavones in the phytoestrogens are daidzein, genistein and formononetin which are present in various herbs. This study examined the quantity of isoflavones in medicinal herbs, which can be used as a phytoestrogen supplement; soybean. These isoflavones were quantified using high performance liquid chromatography (HPLC) with a UV/VIS detector. The concentration of daidzein in Puerariae Radix was $10,436.16{\pm}2,143.83\;mg/kg$ of the dried herb, which was much higher than that extracted from soybeans, $341.47{\pm}18.96\;mg/kg$. The amount of genistein in Sophorae flavescentis Radix ($336.09{\pm}50.89mg/kg$) was approximately 11 times higher than that extracted from soybean ($30.03{\pm}7.17mg/kg$). The level of formononetin in Dalbergiae odoriferae Lignum, $2,189.14{\pm}136.46mg/kg$, was the highest among the herbs tested. The total isoflavone content of Puerariae Radix was approximately 30 times higher than that extracted from soybean. Therefore, plants from the family Leguminosae, particularly Puerariae Radix, can be a good source of phytoestrogens.

A simple HPLC quantification method was developed for genistein, genistin, daidzein, and daidzin in soybeans and soybean products. The procedure used a $4.6{\times}100\;mm$$Chromolith^{(R)}$ RP-18e column with a mobile phase of 1% HOAc in 20% MeOH to 1% HOAc in 80% MeOH for 10 min. The injection volume was $2\;{\mu}L$ at a flow rate of 2 mL/min. Detection was carried out under UV at 254 nm. Under these conditions, the major isoflavones daidzein, daidzin, genistein, and genistin in soybean and soybean pastes were eluted within 7 min with baseline separation. Optimal extraction of the above four major isoflavones was achieved when 40 g of soybean or soybean paste was refluxed in 100 mL of 95% ethanol for 2 hr. Ten different soybean cultivars and nine commercial soybean pastes were analyzed by this method. The total isoflavone content was highest in the cultivar Somyung ($2,497\;{\mu}g/g$ dry weight). The isoflavone content in soybean pastes varied widely from manufacturer to manufacturer (an almost five-fold difference between the highest and lowest values). Such variations were presumably due to differences in fermentation conditions, type of soybeans used, and levels of such additives as starch and salt.
The purpose of this study was to examine whether soybean protein isolate with rich isoflavones which was the same amount as used for earlier study prevents bone induced by ovarian hormone deficiency. Forty female Sprague-Dawley rats (body weight 210 $\pm$ 5 g) were divided into two groups, ovariectomy (OVX) and sham surgery groups, which were each randomly divided into two subgroups that were fed casein and soybean protein isolate with rich isoflavones (isoflavones 3_4 mg/g protein). The diets were fed for 9 weeks after operation. The results of this study indicate that body weight gain and food intake were higher in OVX groups than in SHAM groups regardless of diets. Weight gain were higher in soy isolate groups than in casein groups. Serum Ca concentration was lower in OVX rats than in SHAM rats when fed casein diet. Within the OVX groups, serum Ca concentration was higher in rats fed soy isolate diet than in rats fed casein diet. Serum ALP were higher in OVX groups than in SHAM groups. Within the OVX, soy isolate groups, indices of serum osteocalcin, were a trend for greater than those of the OVX casein group. Crosslink value were increased in ovariectomy groups. Spine BMD and femur BMD of ovariectomy groups were significantly lower than SHAM groups. However, soy isolate with rich isoflavones group in ovariectomy groups, spine BMC and femur BMC were significantly increased after 9 weeks. In conclusion, the soy protein with rich isoflavones was beneficial for bone in ovareiectomized rats and the results were similar to the findings of our previous study which showed a beneficial effect on bone in growing rats.

In order to establish the quality standard of commercial soybean paste, pH, color, moisture, crude protein, isoflavone, free amino acid and organic acid contents of the fermented soybean food in both traditional and commercial products were analyzed. The crude protein contents were from 5.0 to 11.9% in commercial soybean pasted of Samjang and traditional soybean paste of Chungugjang, and the latter showed higher value than those of the former. The contents of isoflavones in commercial soybean paste were less than those of traditional soybean paste. Total contents of free amino acids were distributed from 301 to 4,602mg%, respectively, in which Chungugjang showed the smallest value of 301mg% and the traditional soybean paste showed the most abundant one of 4,602mg%. The ratio of glutamic acid to total free amino acids were 15.9% and 30.1% in traditional soybean paste and commercial soybean paste, respectively. The major organic acid both commercial soybean paste and Samjang was citric acid containing 109.9~196.5mg%. On the other hand, that of traditional soybean paste and Chungugjang was oxalic acid containing 82.8~130.1mg%. This results might be caused by the differences of brewed periods, microflora, processing methods and used raw materials.
Phytoestrogens, especially Yak-kong or soybean-derived isoflavones have been traditionally used as a supplement of estrogen for preventing postmemopausal osteoporosis in oriental folk medicine. In our previous study, the treatment of Yak-kong and soybean increased estrogen receptor-a (ERa) expression and proliferation of MG-63 osteoblastic cells. In contrast, the increase of estrogen receptor-$\beta$ (ER$\beta$) expression in proliferating MG-63 cells with Yak-kong and soybean treatment was less pronounced, which suggested that ER$\beta$ may play a role rather in the regulation of bone cell differentiation To determine the role of ER$\beta$ in Yak-kong or soybean mediated regulation of bone cell differentiation, we established MG-63 cell lines stably expressing either ER$\beta$ or antisense ER$\beta$ RNAs. Increased expression of ER$\beta$ did not affect ERa expression and proliferation of MG-63 cells. However, increased expression of ER$\beta$ in MG-63 cells (ER$\beta$-MG63 cells) selectively enhanced Yak-kong or soybean induced expression of osteoprotegerin (OPG), a novel soluble glycoprotein which is secreted from osteoblasts and mediates the signal for osteoclast differentiation. Inhibition of ER$\beta$ expression by antisense ER$\beta$ RNAs (As-ER$\beta$-MG63) caused these cells to insensitize Yak-kong or soybean induced expression of OPG but increased MG-63 cell proliferation. Furthermore, the comparable effects between Yak-kong and the combined treatment of genistein and daidzein at $0.5{\times}l0^{-8}$ M, which is a concentration of these two isoflavones similar to Yak-kong at 0.001 mg/mL, on OPG expression in ER$\beta$-MG63 cell demonstrate that the enhanced expression of OPG with Yak-kong treatment is mediated by the synergistic effect of low leveled isoflavones in the extracts. Together, coupled with low level of ER expression in osteoclasts, our data demonstrate that ER$\beta$ in osteoblasts plays an important role in Yak-kong and soybean mediated inhibition of osteoclast differentiation indirectly by enhancing the expression of OPG.
This study was performed to investigate the possibility of using three commercial bifidobacteria as a starter for soybean paste fermentation. In order to determine susceptibility to inhibition by high concentrations of salt in soybean paste, cell growth of three strains in sterilized soybean paste was analyzed. Bifidobacterium breve KCTC 5081 was the most resistant to salt, whereas Bifidobacterium bifidum KCTC 5082 showed low cell viability. Conversion efficiencies from glycoside isoflavone to aglycon isoflavone in soybean paste ranged from 11.3~28.6%, with Bifidobacterium longum KCTC 5734 the best strain. Therefore, B. longum KCTC 5734 may be used as a starter for Cheonggukjang fermentation, which is low-salt fermented soybean paste.
