청국장 분말을 첨가한 롤빵의 물성 및 관능적 특성 (Rheological Properties and Sensory Characteristics of Roll Bread with Chungkukjang Powder)

  • The purpose of this study was to investigate quality characteristics of roll bread with the addition of Chungkukjang powder(0, 5, 10, 15, 20%). The antioxidative activity of antioxidative substances produced from several bacterial strains isolated from fermented foods were tested by DPPH $(\alpha,\alpha’-diphenyl-\beta-picrylhydrazyl)$ free radical scavenging activity. The most optimal medium condition for the production of antioxidative substance from Bacillus sp. FF-7 was 2% galactose as carbon source and 1% tryptone as nitrogen source. In color values, with increasing Chungkukjang powder content, the "lightness" (p<0.05) decreased in the crumb bread, while the redness(p<0.05) and yellowness(p<0.05) increased. The volume and specific volume of roll bread with 5.0% Chungkukjang powder were the highest(p<0.05). In the rheology measurements for the roll bread, the hardness (p<0.05) decreased with the addition of 5.0% Chungkukjang powder, but then increased with further additions of Chungkukjang up to 10.0%. Highest sensory scores for color, flavor, taste(p<0.05), texture(p<0.05) and overall acceptance(p<0.05) were obtained from roll bread with 5.0% Chungkukjang powder and control. From the results of this study, it was assumed that the development of food products using Chungkukjang is prospective in response to health-oriented consumers.

  • 썩은 나무 잎에서 세포외 생물고분자 응집제를 생 산하는 흰 곰팡이를 분리하여 Pestalotiopsis sp. M01로 동정하였다. 이 균주가 생산하는 생물고분자 응집제의 응집활성에 대한 배양 조건을 무기염 배지인 Czapek-Dox 배지를 기초로 하여 탄소원, 질소 원, 배지의 pH멸, 그리고 옹도별로 조사하였다. 그 결과로서 응집활성 조건은 3 % sucrose, 0.3 % $KN0_3$, pH 7, 그리고 $25^{\circ}C$에서 최대 응집활성을 나타내었다. 반면에 이 균주의 생육 조건은 3% su-crose, 0.3 % $KN0_3$, pH 5, 그리고 $25^{\circ}C$에서 최대 생육을 나타내었다.

  • An Actinomycetes strain JB isolated from Mt. Hanla had a strong antimicrobial activity against gram positive bacteria and tumor cell growth inhibition. Especially, it couldn't degrade starch and casein as organic compounds. It was resist on lincomycin and rifampicin. The spore mass of strain JB which was arethrospore was white. DAP of the cell wall was L, L-DAP. Antimicrobial material was heat stable, dissolved in ethyl acetate, and not dissolved in butanol. In the pressnce of 0.1% phenol and 4% sodium chloride, strain JB could grow, but it didn't growth at below $10^{\circ}C$. Strain JB didn't use dextran, sodium acetate and sodium citrate as sole carbon source and L-cystein and L-thereonine as nitrogen source. The filtered broth of strain JB had the antimicrobial activity against gram positive bacteria, especially Staphylococcus aureus (ATCC 65389) and the growth inhibition of tumor cell line.

재조합 Bacillus subtilis로부터 endoxylanase의 과발현 및 분비생산 (Overexpression and Secretory Production of Endoxylanase from Recombinant Bacillus subtilis)

  • To overproduce endoxylanase from a recombinant Bacillus subtilis harboring the pJHKJ4 plasmid, the effects of carbon and nitrogen sources on the cell growth and expression level of endoxylanase were investigated in the flask cultures. Among the various carbon and nitrogen sources tested, glucose and maltose as carbon source and yeast extract as nitrogen source were found to be the most effective for the cell growth and the endoxylanase expression. When the concentration of glucose was increased from 0.5% to 5%, the highest activity of extracellular endoxylanse, 166 unit/$m\ell$, was observed at 2% glucose. In case of maltose, the endoxylanase was stably produced at the level of 180 unit/$m\ell$, regardless of the concentration of maltose. The higher the concentration of yeast extract, the greater cell growth and endoxylanase expression were obtained. However, the highest endoxylanase activity per unit cell mass was observed with 1% yeast extract. With the optimized medium (2% glucose, 1% yeast extract, etc), about 630 unit/$m\ell$ of endoxylanse was expressed through the batch fermentation in a fermentor, which expression level corresponded to about 0.7 g-endoxylanase protein /$\ell$. It was also found that the plasmid was stably maintained above 70% level, and more than 90% of endoxylanase activity was detected in the extracellular medium.

색소에 접합된 $\beta$-glucan을 이용한 $\beta$-glucan 분해효소 생산 균주의 분리 및 동정 (Isolation and identification of $\beta$-glucan degrading enzyme producing bacterium using coloured $\beta$-glucan)

  • A bacterium K-4-3, producing $\beta$-glucan hydrolyzing enzyme, was isolated from soil and identified to be Bacillus subtilis by its morpholohical and physiological characteristics. $\beta$-glucan was coloured using cibacron blue 3G-A and cross linded by the addition of 1, 4-butanedioldiglycidyl ether. This substrate was used for the isolation of $\beta$-glucanase producing microorganism. The $\beta$-glucan hydrolyzing enzyme actibity from isolated K-4-3 strain was also measured using the modified substrate. Bacillus subtilis K-4-3 produced the highest extracellular $\beta$-glucan hydrolyzing activity in the basal medium containing $\beta$-glucan as a carbon source, peptone and tryptone as a nitrogen source, and magnesium sulfate as an inorganic salt. The optimum temperature and initial pH for $\beta$-glucanase production by Bacillus subtilis K-4-3 were $37^{\circ}C$ and pH6. The highest enzyme activity was obtained at the culture age of 54 hrs with rotary shaking at $37^{\circ}C$. The crude enzyme showed the highest activity at pH 7.5-8.0 and $65^{\circ}C$.

Effect of Multiple Freeze-Thaw Cycles on Lipid Degradation and Lipid Oxidation of Grass Carp Surimi Containing Different Amounts of Pork Back Fat

  • Fresh grass carp was used to produce surimi samples that were supplemented with 50 g/kg, 100 g/kg, or 150 g/kg pork back fat. The lipid composition, lipase activity, lipid oxidation index, and lipoxygenase activity of samples subjected to repeated freezethaw process were determined to assess the effects of the added fat on lipolysis and lipid oxidation of grass carp surimi. Freeze-thaw treatment increased free fatty acid content, mainly due to the decomposition of phospholipids and some neutral lipids by lipase. With repeated freeze-thaw treatment, the levels of free fatty acids and phospholipids were correlated with the lipid oxidation indexes and lipoxygenase activity, indicating that lipid degradation can promote lipid oxidation. In the same freeze-thaw cycle, surimi products with high fat content are more vulnerable to oxidative damage, neutral lipids are the main source of free fatty acids in the early stage of freeze-thaw, and phospholipids are the main source of free fatty acids in the late stage.

수학적 인식에서 '활동'이 갖는 의미에 대한 고찰 (On the Meaning of 'Activity' in Mathematical Cognition)

  • In this study, I consider the meaning of activity as the source of mathematical knowledge. Mind-body dualism of Descartes which understands that knowledge precedes activity is somewhat overcomed by Ryle who understands that knowledge and activity are two sides of the same coin. But his discussion cannot offer the explanation about the foundation of rightness or the development of rules which can be expressed propriety of activity or rationality. Contrary to these views, Piaget solve this problem by the reasonability of 'the whole system of activity'. The theory of Dewey can be evaluated as an origin of activism of Piaget. Piaget considers knowledge as the system of activity itself, whereas Dewey considers knowledge as 'the result of activity'. This view of Dewey is related to the view of pragmatism which considers 'practice' is more important than 'theory'. The nature of 'activity' in this study has to be understanded as the interaction or the relation between the subject and the object. If we understand activity like this, we can explain that the whole structure of activity has the 'wholeness' that cannot be simply restored to the sum total of 'parts' and the new structure is a self-regulative transformation system which includes former structure continuously.

오존 및 광선의 처리가 맥아 효소활성에 미치는 영향 (Effects of Ozone and Light Illumination on the Enzymation Activity of Malt)

  • In order to increase the enzymatic activity of malt used as a source of traditional processing foods, the enzymatic activities of various barley were examined and the effects of ozone and light illumination treatment on the enzymatic activities of amylase, amylase, and glucanase in malt during man ufacture were also examined. Barley didn't show amylase activity prior to soaking, but the activity of barley increased quickly after soaking. Glutinous barley showed the highest amylase activity among Duru barley, Ol barley, two rowed barley and naked barley. Naked barley showed the lowest activity. The amylase activity was the highest in Duru barley and decreased in the order of in glutinous barley, naked barley and two rowed barley. It was showed that the enzymatic activity of malt was higher than that of control when malt was soaked for 24hr at the concentration of 0.3ppm of ozone. The enzymatic activity of malt treated with light illumination was higher than that of control. The bud and root of light illuminated malt was much stronger than that of control. The root of light illuminated malt was much shorter than that of control. In addition, light illuminated malt showed a little green color which matches the demand of consumer. These studies demonstrated that both ozone and light illumination treatment increased the enzymatic activity of malt to result in high quality of malt manufacture.

사회연결망 분석과 자료포락분석 기법을 이용한 소프트웨어 함수 우선순위 분석 연구 (Priority Analysis for Software Functions Using Social Network Analysis and DEA(Data Envelopment Analysis))

  • To remove software defects and improve performance of software, many developers perform code inspections and use static analysis tools. A code inspection is an activity that is performed manually to detect software defects in the developed source. However, there is no clear criterion which source codes are inspected. A static analysis tool can automatically detect software defects by analyzing the source codes without running the source codes. However, it has disadvantage that analyzes only the codes in the functions without analyzing the relations among source functions. The functions in the source codes are interconnected and formed a social network. Functions that occupy critical locations in a network can be important enough to affect the overall quality. Whereas, a static analysis tool merely suggests which functions were called several times. In this study, the core functions will be elicited by using social network analysis and DEA (Data Envelopment Analysis) for CUBRID open database sources. In addition, we will suggest clear criteria for selecting the target sources for code inspection and will suggest ways to find core functions to minimize defects and improve performance.

  • 단일 쌍극자 모델을 source localization 문제에 적용시키는 것은 초보적이기도 하지만 필수적이기도 하다. 시abf레이션을 이용하여 단일 쌍극자를 추적함으로써 얻은 결과는 실제 인간의 뇌에 관한 EEG 임상 실험에 여러가지 정보를 제공해줄 수 있기 때문이다. 이번 논문에서는 EEG실험에서의 전극 배치가 S/N(signal to noise ratio)과 추정 오차 사이에 어떤 영향을 미치는 가를 Monte Carlo 시뮬레이션으로 조사하였다. 머리모델은 3중 구각 모델을 사용하였고 이를 이용하여 forward problem을 계산하였다. 쌍극자 파라미터를 minimization하는 문제는 simplex method를 이용하여 계산하였다. 컴퓨터 시뮬레이션 결과, 특이한 점은 전극의 밀도와 입체각에 의해 쌍극자 파라미터 오차가 변화했다는 사실이다. 이것은 곧바로 전극 배치와 연관이 된다. 실제 EEG 실험에서 전극배치를 어떻게 했는가에 따라 그에 따른 오차가 변화한다.
