Inflorescence architecture mainly contributes to final grain yield in crops. Sorghum inflorescence is basically composed of one fertile sessile spikelet (SS) and two infertile pedicellate spikelets (PS). To identify regulatory factors involved in the inflorescence architecture, we screened an EMS mutagenesis population from the pedigreed sorghum mutant library. We found inflorescent architecture mutants, named as multi-seed mutants, msd, with gained fertile ability in PS and also an increased number of floral branches. In natural sorghum populations, it is not common that are fertile. A detailed dissection of developmental stages of wild type and msd1 mutant described that the PS in wild type do not have floral organs, including ovary, stigma, filament and anther, while the msd1 mutants generate intact floral organ in the sessile spikelet. We found MSD1 encoded a TCP transcription factor using bulk segregant analysis (BSA) of F2 population, and was a strongly enriched expression during inflorescence developmental stages. We proposed that MSD1 functions to suppress floral organ maintenance at PS during inflorescence development in Sorghum. To explore the regulatory network associated with PS fertility, whole genome expression profiling was performed at 4 different developmental stages in 6 various tissue types between wild type and msd1. Taken together, we demonstrated that MSD1 was involved in the plant hormone and maybe influenced program cell death in PS via the activation of plant hormonal pathway.
The aim of this study is to select the superior resources of high yield, high content of functional material optimal to mechanical harvesting by the evaluation of crop growth and yield characteristics in sorghum germplasm. One hundred accessions of sorghum germplasm were used in this experiment. Days from seeding to heading date showed the range from 68 to 94 days with the highest frequency proportion was the group from 80 to 85 days, which occupied 34% (34 plant resources) of 100 germplasm. Ear types of 100 sorghum germplasm could be classified as 7 types of broom-tillering, half broom-tillering, extreme open-loose type, open-loose type, intermediate type, compact type, extreme-compact type of which intermediate type was the highest ratio of 28% (28 plant resources) of 100 germplasm. Yield showed the range from 106 to 365 ㎏/10a with the highest frequency proportion of it was the group from 150 to 200 ㎏/10a, which occupied 44% (44 plant resources) of 100 germplasm. Among 100 sorghum germplasm, 18 ideal resources of high yield and short plant height appropriate for mechanical harvesting were selected. In order to evaluate high content of functional substance, selected 18 resources were analyzed for total polyphenol content, DPPH radical scavenging activity and total anthocyanin content. Finally, we selected 5 resources of short plant height, high yield, high content of total polyphenol and high DPPH radical scavenging activity among 18 genetic resources.
To obtain basic information on forage sorghum F$_1$ hybrid breeding a total of 16 lines were selected from 311 introduced sorghum germplasm accessions, assessed and classified by the taxonomic distance and principal component analysis. The lines of which plant height and morphological characters were diverse and the 50% flowering date was similar to each other, were selected for parental lines in sorghum $\times$ sweet sorghum and sorghum $\times$ sudangrass crossing groups. Three varietal groups were classified by the average linkage cluster analysis based on the D$^2$ computed in eleven characters. Group I, II and III included 6 lines of sudangrass, 4 lines of sweet sorghum and 6 lines of grain sorghum, respectively. In the result of principal component analysis for eleven characters, about 82% of total variation could be appreciated by the first four principal components, the first principal component was highly loaded with head compactness and shape, l00-seed weight, plant color and grain covering, the second principal component with flowering date, plant height and awnness.
Kjeldahl method used in many materials from various plant parts to determine protein contents, is laborious and time-consuming and utilizes hazardous chemicals. Near-infrared (NIR) reflectance spectroscopy, a rapid and environmentally benign technique, was investigated as a potential method for the prediction of protein content. Near-infrared reflectance spectra(1100-2400 nm) of coarse cereal grains(n=100 for each germplasm) were obtained using a dispersive spectrometer as both of grain itself and flour ground, and total protein contents determined according to Kjeldahl method. Using multivariate analysis, a modified partial least-squares model was developed for prediction of protein contents. The model had a multiple coefficient of determination of 0.99, 0.99, 0.99, 0.96 and 0.99 for foxtail millet, sorghum, millet, adzuki bean and mung bean germplasm, respectively. The model was tested with independent validation samples (n=10 for each germplasm). All samples were predicted with the coefficient of determination of 0.99, 0.99, 0.99, 0.91 and 0.99 for foxtail millet, sorghum, millet, adzuki bean and mung bean germplasm, respectively. The results indicate that NIR reflectance spectroscopy is an accurate and efficient tool for determining protein content of diverse coarse cereal germplasm for nutrition labeling of nutritional value. On the other hands appropriate condition of cereal material to predict protein using NIR was flour condition of grains.
This study was conducted to obtain fundamental information on forage sorghum breeding in forage crop field of Livestock Experiment Station at Suwon from 1986 to 1991. The charcterization of sorghum germplasm was performed through 1986 to 1987, and after parental lines were selected from diverse sorghum germplasm on the basis of flowering date, plant height and several morphological characters for forage sorghum Fl hybrids. The range of variation of 50% flowering date and plant height were greater in order of forage sorghum sudangrass and male sterile line of grain sorghum. The average flowering date was earlier in sudangrass and male sterile line of grain sorghum than forage sorghum lines from the tested sorghum germplasms. And the average plant height was tall in order of forage sorghum, sudangrass and male sterile lines of grain sorghum. There were remarkable morphological variations between sudangrass lines and male sterile lines of grain sorghum such as plant color, leaf midrib color, glume color, seed coat color, head compactness and shape, awns, grain covering and 100 seed weight.
Crop characteristics of 179 sorghum (Sorghum bicolor L. Moench) resources collected in Korea were investigated in order to establish basic data for the improvement of crop breeding. Spike types of 179 sorghum resources were classified as 5 types of open-loose type, broom-tillering type, half broom-tillering type, extreme-compact type and compact type, of which broom-tillering type was the highest ratio of 38.0% (68 plant resources) of 179 germplasm. In the existence and nonexistence of spike awn in 179 sorghum resources, 28.5% (51 plant resources) showed existence of spike awn, whereas the rest of 71.5% (128 plant resources) had no spike awn. Seed type was classified as 5 types, of which circle-shape showed the highest ratio of 43.0% (77 plant resources) and the lowest was inclined-circle shape by 7.3% (13 plant resources). Seed color was classified as 4 colors of brown, white, whitish brown, and yellowish brown, of which yellowish brown was the highest ratio of 84.4% (151 plant resources) among them. Days from seeding to heading date showed the range from 67 to 88 days with the average of 77.4 days and the highest frequency proportion of it was the group from 76 to 80 days, which occupied 37.4% (67 plant resources) of 179 germplasm. 179 sorghum resources showed high variation in the range of culm length with the average of 253.8 cm and group from 92 cm to 360 cm of culm length showed the highest frequency proportion of 20.6% (37 plant resources). Spike length showed the range from 15 to 49 cm with the average of 28.8 cm and the highest frequency distribution of it was the group from 31 to 35 cm with the proportion of 26.3% (47 plant resources), whereas the lowest was below 15 cm with the proportion of 0.6%. Days from seeding to physiological maturity showed the range from 110 to 146 days with the average of 125.5 days and the highest frequency proportion (39.7%) of it was the group from 111 to 115 days, while there were also occupied with 2 plant resources below 110 days and 23 plant resources over 141 days among 179 germplasm. Number of grains per spike showed the range from 163 to 4,532 grains with the average of 2,068.6 grains and group from 1,601 to 2,000 grains per spike was the highest frequency distribution with the proportion of 25.7% (46 plant resources). 1000 grains weight showed the range from 10.6 to 38.1g with the average of 25.6g and group from 26.0 to 30.0g was the highest frequency distribution with the proportion of 44.1% (79 plant resources).

Sorghum bicolor is considered as copper-tolerant species. The present study was conducted to understand the copper tolerance mechanism in Sorghum seedling roots. Morphological and effects of Cu on other interacting ions were observed prominently in the roots when the plants were subjected to different concentrations (0, 50, and $100{\mu}M$) of $CuSO_4$. However, the morphological characteristics were reduced by Cu stress, and the most significant growth inhibition was observed in plants treated with the highest concentration of $Cu^{2+}$ ions ($100{\mu}M$). In the proteome analysis, high-throughput two-dimensional polyacrylamide gel electrophoresis coupled with MALDI-TOF-TOF mass spectrometry was performed to explore the molecular responses of Cu-induced sorghum seedling roots. In two-dimensional silver-stained gels, a total of 422 differentially expressed proteins (${\geq}1.5-fold$) were identified using Progenesis SameSpot software. A total of 21 protein spots (${\geq}1.5-fold$) from Cu-induced sorghum roots were analyzed by mass spectrometry. Of the 21 differentially expressed protein spots from Cu-induced sorghum roots, a total of 10 proteins were up-regulated, and 11 proteins were down-regulated. The abundance of the most identified protein species from the roots that function in stress response and metabolism was significantly enhanced, while protein species involved in transcription and regulation were severely reduced. The results obtained from the present study may provide insights into the tolerance mechanism of seedling roots in Sorghum.
Variation of major characteristics of 140 sweet sorghum germplasm which had been collected from domestic and foreign countries were investigated to study the possibility of bioethanol production in reclamed soil of Korea. Averages of culm length, ear length, the number of nodes, stem diameter, fresh weight yield, and growth duration from seeding to heading were 175 cm, 26 cm, 9, 11.6 mm, 12.1 t/ha, and 96 days, respectively. Sugar content, which has a great effect on bioethanol, ranged from 5.7 to 23.5 Brix (average 14.1 Brix). Sugar contents of selected two varieties were more than 20 Brix. Fresh weight yield ranged from 20 to 50 t/ha (average 12.1 t/ha). Fresh weight yield of selected three varieties were more than 30 t/ha. Culm length showed highly significant positive correlation with the ear length, the number of nodes, stem diameter, fresh weight yield, and growth duration from seeding to heading.
