A New Report on Three Species of Sordariomycetes Class Isolated from Soil in Korea

  • Three fungal strains belonging to the class Sordariomycetes, namely BH-06, 17-039 and BE12-1, were isolated from soils in Korea and identified as Chaunopycnis alba, Myrothecium cinctum and Humicola olivacea, respectively. These species were confirmed according to their morphological characteristics and phylogenetic relationships determined based on internal transcribed spacer regions, as well as large subunit, small subunit, ${\beta}$-Tubulin and RNA polymerase II largest subunit sequences of ribosomal DNA. These three species are the first members of the Sordariomycetes reported in Korea.

  • In an ongoing survey of Korean indigenous fungi, three fungal strains belonging to the Sordariomycetes were isolated from soil samples. These strains were designated KNU16-001, KNU16-002, and KNU16-009, and identified as Ambrosiella grosmanniae, Acremonium sclerotigenum, and Trichocladium asperum, respectively, based on morphological characterization and phylogenetic analysis using internal transcribed spacer region sequences of ribosomal DNA. This is the first report of these species in Korea.

  • During a survey of fungal diversity in the class Sordariomycetes, 3 fungal strains, CNUFC-KMHY6-1, CNUFC-MSW24-2-11, and CNUFC-GW2S-4 were isolated from soil and freshwater samples, respectively in Korea. The strains were analyzed both morphologically and phylogenetically on the basis of internal transcribed spacer and RNA polymerase II second largest subunit gene sequences. On the basis of their morphology and phylogeny, CNUFC-KMHY6-1, CNUFC-MSW24-2-11, and CNUFC-GW2S-4 isolates were identified as Arcopilus aureus, Memnoniella echinata, and Stachybotrys sansevieriae, respectively. To the best of our knowledge, Ar. aureus and M. echinata have not been previously recorded in Korea, and this is the first report of S. sansevieriae from freshwater niche.

First report of six Sordariomycetes fungi isolated from plant litter in freshwater ecosystems of Korea

  • Freshwater ecosystems provide a complex environment for microorganisms. In this study, we isolated diverse fungal strains from plant litter in freshwaters. These strains were identified using molecular phylogenetic analyses of rDNA and/or other gene sequences (TUB, GAPDH, and EF1). In addition, we examined their morphological characteristics by microscopy and cultural characteristics on several media. We identified six previously unrecorded Sordariomycetes species in Korea, i.e., Colletotrichum godetiae, Discosia rubi, Robillarda sessilis, Monochaetia dimorphospora, Idriella lunata, and Phialemoniopsis endophytica. Of these, D. rubi and M. dimorphospora exhibited high extracellular amylase, lipase, and protease activities, suggesting that these fungal isolates might play an important role as decomposers in freshwater ecosystems. Plant litter could thus be a good source for isolating and investigating previously undocumented fungal species in freshwater environments.

Molecular and Morphological Characterization of Three New Fungal Species of the Class Sordariomycetes from Korea

  • Three fungal strains belonging to the class Sordariomycetes were isolated from soil collected on Jeju Island and Gyeongsangbuk-do, Korea. They were identified as Diaporthe endophytica (KNU-JJ-1809), Faurelina indica (KNU-JJ-1830), and Trichoderma ivoriense (KNU-4-KH1). KNU-JJ-1809 produced beta conidia that were straight, curved, hyaline, smooth-walled, with a diameter of 16.5-25.0×0.6-1.7 ㎛. The conidia of strain KNU-JJ-1830 were hyaline to light green, thin, clavate, round, truncate base, had guttules at both ends, with a diameter of 2.5-5.2×1.7-3.8 ㎛. The conidia of strain KNU-4-KH1 were oblong or ellipsoidal, smooth-walled, greenish, with a diameter of 2.2-4.4×2.2-3.6 ㎛. Internal transcribed spacer regions, partial large subunit, translation elongation factor 1-alpha, β-tubulin, and calmodulin genes were used to confirm the strains, and their cultural and morphological characteristics. To our knowledge, this is the first report on D. endophytica, F. indica, and T. ivoriense in Korea.

Pezizomycotina (Ascomycota) Fungi Isolated from Freshwater Environments of Korea: Cladorrhinum australe, Curvularia muehlenbeckiae, Curvularia pseudobrachyspora, and Diaporthe longicolla

  • Fungi are ubiquitous and indispensable components of nearly all ecosystems on earth, including freshwater environments. A survey of fungal diversity in freshwater environments of Korea led to the identification of four unrecorded Pezizomycotina (Ascomycota) species in 2016 and 2017, based on morphology and molecular phylogeny; these included Cladorrhinum australe, Curvularia muehlenbeckiae, Curvularia pseudobrachyspora (Dothideomycetes), and Diaporthe longicolla (Sordariomycetes).

  • Sordariomycetes is the second largest class of Ascomycota, distributed throughout various habitat including terrestrial and aquatic environments and also existing as endophytes. We isolated endophytic fungal strains in Korea, identifying them based on their morphological characteristics and molecular analyses, using eight specific DNA regions for accurate genus identification. We identified five previously unrecorded endophytic fungal species in Korea: Chaetomium subaffine, Colletotrichum jiangxiense, Colletotrichum sydowii, Diaporthe vacuae and Neurospora tetraspora. In this study, we describe the morphological characteristics and present our phylogenetic analyses of these five fungal species.

  • 본 연구에서는 제주 습지 환경에서 채집한 수생식물에서 내생균류를 분리하였다. 분리된 균을 형태학적 특징과 internal transcribed spacer 영역의 염기서열 분석을 통하여 동정한 결과, NNIBRFG2982, NNIBRFG2984, NNIBRFG3040 균주는 각각 Acremonium tubakii, Colletotrichum cliviae, Cylindrocarpon pauciseptatum으로 확인되었다. 이들 3종의 균류는 Ascomycota문의 Sordariomycetes강에 속하는 균류로 국내 미기록종으로 보고하는 바이다.

A New Species of Thelonectria and a New Record of Cephalotrichum hinnuleum from Gunwi and Ulleungdo in Korea

  • Three fungal strains belonging to the class Sordariomycetes were isolated from soils collected from Gyeongsangbuk-do in Korea. They were identified as Cephalotrichum hinnuleum (UD CT 1-3-3 and KNU-19GWF1) and Thelonectria chlamydospora sp. nov. (UD ST 1-2-1). T. chlamydospora sp. nov. was morphologically identical to T. truncata, but its specific macroconidial dimensions, lower number of septations, and chlamydospore diameter render it distinct from the strains of the genus Thelonectria. The strains UD CT 1-3-3 and KNU-19GWF1 were developed flat, velvety to felty, and golden gray to brown-gray after 14 days of incubation at 25 ℃ on PDA. These strains were produced polyblastic conidiogenous cells and conidia were pale brown to brown, smooth, thin-walled, subglobose to ellipsoidal, arranged in chains, and the diameters of 6.7-9.0 × 3.7-5.1 ㎛. The strains were also confirmed by using the multi-locus genes using internal transcribed spacer (ITS) regions, partial large subunit (LSU), translation elongation factor 1α (TEF1-α), β-tubulin (TUB2), and actin (ACT) genes. This is the discovery of T. chlamydospora sp. nov. and Cephalotrichum hinnuleum, a new record from Korea.

Isolation and Identification of Two Unreported Sordariomycetes Fungi in Korea: Pestalotiopsis clavata and Botryotrichum iranicum

  • Two fungal strains, designated KNUF-21-006 and KNUF-21-028, were isolated from soil samples collected from Gyeongbuk Province, Korea. The strain KNUF-21-006 was similar to other Pestalotiopsis species in terms of morphological characteristics, including whitish to pale brown mycelium, conidial shape, and size. The isolate had aerial hyphae that produced black fruiting bodies on the mycelium. The conidia were fusoid to ellipsoid, four-septate, and appendage-bearing. Phylogenetic analysis using the internal transcribed spacer (ITS) region, translation elongation factor 1-alpha (TEF), and β-tubulin (TUB) gene sequences confirmed that the closest relationship of the isolate at the species level was with Pestalotiopsis clavata. The strain KNUF-21-028 exhibits similar morphological characteristics to other Botryotrichum species, including white aerial mycelium with sulcate and irregular margins, conidial shape, and size. The conidia were globose, single, and hyaline. Upon molecular analysis-using the ITS region, large subunit (LSU) rRNA gene, and TUB gene sequences-the fungus was identified as Botryotrichum iranicum. This is the first record of these fungal species in Korea.
