The aim of this study was to investigate the effects of production systems and milk collection periods on the somatic cell count (SCC), some microbiological properties, total aerobic mesophilic bacteria (TAMB), coliform, Staphylococcus aureus (S. aureus), yeast and mould) and antibiotic residue of milk; in Turkey. Milk samples were collected from 9 conventional farms and 9 organic farms during one year time, at six different months (December 2013 to October 2014), and all farms were selected from the same geographical locations. All organically managed farms had organic production certificates given by the Republic of Turkey Ministry of Food, Agriculture and Livestock. The count of TAMB, coliform, and coagulase positive S. aureus were affected by production systems at the level of p<0.01; yeast and mold, and somatic cell count (SCC) were affected at the level of p<0.05. But, differences according to months were statistically significant only on TAMB (p<0.01) and coliform (p<0.05) counts. The general means of TAMB, coliform and yeast and mould counts of the organic milk (OM) were significantly lower (p<0.05), while the general means of SCC and coagulase positive S. aureus count of the OM was significantly higher (p<0.05) compared to conventional milk (CM). Antibiotic residue was determined in one of the CM sample and in two of the OM samples. Our study is the first research that compared conventional and organic milk in Turkey. This study indicated that the microbiological quality of OM was the higher in terms of TAMB, coliform and yeast and mould, whereas was the lower in relation to SCC and coagulase positive S. aureus counts. But, the quality of both milk types should be improved.
The present study was conducted to develop a milk filtering system for decreasing somatic cell count (SCC) in bulk tank milk. The pore sizes of the filter were 0.1, 0.3, 0.4, 0.5 and $0.8{\mu}m$. The rate of SCC reduction of $1^{st}$ grade milk on $0.1{\mu}m$ filter was 76% and significantly higher than other treatments. The rates of SCC reduction for 0.3, 0.4, 0.5 and $0.8{\mu}m$ were 35, 32, 18 and 6.4%, respectively. The effects of the milk filtering system on bacterial count and milk fat content were minimal. The milk flow rates per minute between the filter sizes were similar. But discharge pressures were increased as the pore size of the filter decreased. In conclusion, Considering the rate of SCC reduction, discharge pressure and cost, $0.4{\mu}m$ filter could be recommended.
Mastitis is characterized by physical, chemical and bacteriological changes in the milk and pathological changes in the glandular tissue of the udder and affects the quality and quantity of milk. The bacterial contamination of milk from the affected cows render it unfit for human consumption and provides a mechanism of spread of diseases like tuberculosis, sore-throat, Q-fever, brucellosis, leptospirosis etc. and has zoonotic importance. Somatic cell count (SCC) is a useful predictor of intramammary infection (IMI) that includes leucocytes (75%) i.e. neutrophils, macrophages, lymphocytes, erythrocytes and epithelial cells (25%). Leucocytes increase in response to bacterial infection, tissue injury and stress. Somatic cells are protective for the animal body and fight infectious organisms. An elevated SCC in milk has a negative influence on the quality of raw milk. Subclinical mastitis is always related to low milk production, changes to milk consistency (density), reduced possibility of adequate milk processing, low protein and high risk for milk hygiene since it may even contain pathogenic organisms. This review collects and collates relevant publications on the subject.
The objective of this study was to develop models to predict SCC (somatic cell count) in unhomogenized milk by visible and near-infrared (NIR) spectroscopic technique. Total of 100 milk samples were collected from dairy farms and preserved to minimize propagation of bacteria cells during transportation. Reductive reagents such as methyl red, methylene blue, bromcresol purple, phenol red and resazurin were added to milk samples, and then colors of milk were changed based on SCC of milk. For optimal reductive reagents, reaction time was controlled at 3 level of reaction time. A spectrophotometer was used to measure reflectance spectra from milk samples. The partial least square (PLS) models were developed to predict SCC of unhomogenized milk. The PLS results showed that milk samples with reductive reagents had a good correlation between predicted and measured SCC at 5 minutes of reaction time in the visible range. The PLS models with resazurin reagent had the best performance in $400{\sim}600\;nm$. The prediction results of milk samples with resazurin had 0.86 of correlation coefficient and 14,184 cell/mL of SEP.
The study was undertaken to find out the normal mean and variations in somatic cell count (SCC) of milk in crossbred and indigenous cows as influenced by stage of lactation, parity and season. On day of milk sampling the udders were tested for mastitis by California Mastitis Test (CMT). Only those cows, which were found negative in the CMT, were taken in the study. Paritywise differences in SCC were not significant between the 1st to 6th lactation and above. Similarly, stage of lactation effect, when tested at 30 day intervals, did not differ significantly. However, the seasons significantly (p<0.05) affected SCC count of milk. The SCC was lower during cold ($1.10{\times}10^5cells/ml$) and hot-dry ($1.11{\times}10^5cells/ml$) season then during hot-humid season ($2.14{\times}10^5cells/ml$). On an average SCC recorded were 1.26, 1.31, 1.54 and $1.61{\times}10^5$ cells per ml respectively in Tharparkar, Sahiwal, Karan Swiss and Karan Fries cows irrespective of stage of lactation, parity and season. Further, crossbred Karan Swiss and Karan Fries cows behave similar to the indigenous Tharparkar and Sahiwal cows but are more vulnerable to hot-humid climate then indigenous ones. Significant correlation between the SCC and milk yield during different stages of lactation (1.38 to $1.74{\times}10^5cells/ml$) and parity (1.47 to $1.63{\times}10^5cells/ml$) suggested that the SCC/ml of milk was higher during the later stages of lactation.
The objective of this study was to determine the relationship between udder and teat characteristics and somatic cell count (SCC). A total of 749 (73.1%) milk samples from 1,024 quarters of 259 Holstein cows contained less than 200,000 somatic cells/ml, while 132 (12.9%) quarters contained more than 500,000 somatic cells/ml. Prior to data analysis, somatic cell counts were transformed to natural logarithm. The mean SCS {$log_e(SCS/10^3)$} of milk samples from the front quarters was lower than milk samples from the rear quarters. The highest SCS was observed from cows with the step-shaped udder and the pear-shaped teats, respectively. Increased SCS was observed from cows with large teat diameter, short teat length, short distance between the teat tip to floor (p<0.05) and with increase in parity, respectively.
The objective of the present study was to investigate the accuracy of mastitis diagnostic indicators for different dairy goat breeds. Biweekly milk samples were collected from individual half mammary gland of seven Saanen and seven Alpine dairy goats in the period of 40 to 120 days in milk. With threshold value set at 2.8 and 3.1 for Alpine and Saanen dairy goats, respectively, log (SPC) offered good sensitivity (0.89, 0.93), specificity (0.88, 0.95), positive predictive value (0.75, 0.85) and negative predictive value (0.95, 0.98) as a mastitis diagnostic tool. The correlations of log (SPC) with milk yield, log (SCC), ALP, LDH, $Na^{+}$, $K^{+}$ and EC were significant in Saanen dairy goats (p<0.05), with the highest correlation coefficient (0.653) existing between log (SPC) and log (SCC). The correlations of log (SPC) with milk yield, milk fat, milk protein, log (SCC), $Na^{+}$, $K^{+}$, EC were significant in Alpine dairy goats (p<0.05), with the highest correlation coefficient (0.416) existing between log (SPC) and log (SCC). There were different best-fit regression equations with different multiple diagnostic indicators for Saanen and Alpine dairy goats. In conclusion, different breeds of dairy goats may have to adapt different mastitis diagnostic parameters for a better diagnosis.
This study was initiated in an effort to determine the normal mean and variations of the somatic cell count (SCC) in milk of buffaloes as influenced by the milking time, stage of lactation, parity and season. The buffaloes were hand milked at 13 and 11 h. interval during evening and morning respectively. On the day of milk sampling the udders were tested for mastitis by California Mastitis Test (CMT). Only those buffaloes, which were found negative in the CMT, were included in the sampling plan. The mean values for morning and evening were 1.09 (range 0.39-1.76) and $0.97(range\;0.57-2.46){\times}10^5cells/ml$, respectively which did not differ significantly. When data of the morning and evening values was compared on the basis of total cell secretion in milk, even then there was no statistical difference between the morning and the evening values, thereby suggesting that no diurnal variation existed in SCC of milk. Paritywise differences were not significant between the 1st to 5th lactation and above. Similarly stage of lactation effect, when tested at 30 day intervals, did not differ significantly. Significant (p<0.05) correlation coefficients (r) between SCC and milk yield during different stages of lactation and parity suggested that SCC per ml of milk was higher during the later stages of lactation. SCC was higher in primiparous than in multiparous buffaloes. On an average the SCC recorded was $1.0{\times}10^5cells/ml$ of milk irrespective of time of milking, parity and stages of lactation. The SCC was low during cold and hot-dry season but were high during the hot-humid season (p<0.05), the respective values being 0.76, 1.08 and $1.35{\times}10^5cells/ml$. These values were lower than the SCC already reported in cows suggesting less stressful condition of the udder of buffaloes in this study.

Somatic cells count (SCC), milk yield (MY) and pedigree information of 2,791 first lactation cows that calved between 1990 and 2010 on 259 Thai farms were used to estimate genetic parameters and trends for SCC and its genetic association with MY. The SCC were log-transformed (lnSCC) to make them normally distributed. An average information-restricted maximum likelihood procedure was used to estimate variance components. A bivariate animal model that considered herd-yr-season, calving age, and regression additive genetic group as fixed effects, and animal and residual as random effects was used for genetic evaluation. Heritability estimates were 0.12 (SE = 0.19) for lnSCC, and 0.31 (SE = 0.06) for MY. The genetic correlation estimate between lnSCC and MY was 0.26 (SE = 0.59). Mean yearly estimated breeding values during the last 20 years increased for SCC (49.02 cells/ml/yr, SE = 26.81 cells/ml/yr; p = 0.08), but not for MY (0.37 kg/yr, SE = 0.87 kg/yr; p = 0.68). Sire average breeding values for SCC and MY were higher than those of cows and dams (p<0.01). Heritability estimates for lnSCC and MY and their low but positive genetic correlation suggested that selection for low SCC may be feasible in this population as it is in other populations of dairy cows. Thus, selection for high MY and low SCC should be encouraged in Thai dairy improvement programs to increase profitability by improving both cow health and milk yield.
