Sesame has been consumed for centuries as flavoring ingredient in eastern Asian countries, especially Korea. Sesame seeds have been used as health food for traditional medicine to prevent disease in Asian countries for several thousand years. Sesame seed has higher oil content (around 50%) than most of the known oilseeds. Sesame oil is rich in monounsaturated and polyunsaturated fatty acids. Extraction of sesame has developed significantly over the years. The mechanical method was an early means of separation which was physical pressure to squeeze the oil out. Nowadays, solvent extraction becomes the commonly used commercial technique to recover oil from oilseeds. In this study, we investigated extraction efficiency and quality of oil affected by cultivars and extraction methods of sesame seed. Different variables were investigated; roasting temperature ($170{\sim}220^{\circ}C$), extraction methods (solvent and physical pressure), forced ventilation system and cultivars. The Contents of B(a)P in sesame oil after roasting at $170{\sim}220^{\circ}C$ were 0.30~2.53 ppm. When we introduced forced ventilation system during roasting, B(a)P Contents were decreased up to 36%. The Oil extraction efficiency on sesame seed was statistically depending on the cultivars and extraction methods. The oil extraction yields of solvent and physical pressure extraction were 56.3% and 44.6%, respectively. Many of sesame cultivars and genetic resources are linolenic acid content of less than 0.5%. The results supported that we have developed a safe and high quality sesame oil processing methods for small and medium-sized companies.
The effect of water activity on degradation of pigments in dried lavers, Porphyra tenera Kjellm. was examined when stored at room temperature for fifty days. Chlorophyll pigment was extracted with methanol-petroleum ether mixture solvent(2:1 v/v), partitioned in ether, and analysed spectrophotometrically at 662 nm as chlorophyll a. The degradation products of chlorophyll were isolated on sugar-starch column(85:15 w/w) with n-propanol-petroleum ether solution(1:200 v/v) as a developing solvent. The isolated green colored zones were analysed individually at the wavelengths of 650, 662, and 667 nm as allomerized product, chlorophyll a retained, and pheophytin formed respectively. Carotenoida were also extracted with the methanol mixture solvent, partitioned in ether, and finally redissolved in acetone after the evaporation of ether in a rotary vacuum evaporator. The total carotenoid content was measured as lutein at 450 nm. From the results, it is noted that the rate of chlorophyll degradation reached a minimum at 0.11 to 0.33 water activity while progressively increased at higher moisture levels resulting in rapid conversion of chlorophyll to pheophytin. At lower activity, autocatalysed oxidizing reaction like allomerization seemed prevailing the acid catalysed conversion reaction. The loss of carotenoid pigment was also greatly reduced at the range of 0.22 to 0.34 water activity with much faster oxidative degradation at both higher and extremely lower moisture levels. These two moisture levels indicated above at which the both pigments exhibited maximum stability are considerably higher than the BET monolayer moisture which appeared 7.91 percent on dry basis at Aw=0.10 calculated from the adsorption isothermal data of the sample at $20^{\circ}C$. The rate of pigment loss in heat treated samples at 60 and $100^{\circ}C$ for 2 hours prior to storage somewhat decreased, particularly at higher moisture levels although the final pigment retention was not much stabilized.
Elimination reactions of $(E)-2,4-(NO_2)_2C_6H_2CH=NOC_6H_3-2-X-4-NO_2$ (1a-e) promoted by $R_3N/R_3NH^+$ in 70 mol % MeCN(aq) have been studied kinetically. The reactions are second-order and exhibit Br$\ddot{o}$nsted ${\beta}$ = 0.80-0.84 and ${\mid}{\beta}_{lg}{\mid}$ = 0.39-0.42, respectively. For all leaving groups and bases employed in this study, the ${\beta}$ and ${\mid}{\beta}_{lg}{\mid}$ values remained almost the same. The results can be described by a negligible $p_{xy}$ interaction coefficient, $p_{xy}={\partial}{\beta}/pK_{lg}={\partial}{\beta}_{lg}/pK_{BH}{\approx}0$, which describes the interaction between the base catalyst and the leaving group. The negligible pxy interaction coefficient is consistent with the $(E1cb)_{irr}$ mechanism. Change of the base-solvent system from $R_3N$/MeCN to $R_3N/R_3NH^+$-70 mol % MeCN(aq) changed the reaction mechanism from E2 to $(E1cb)_{irr}$. Noteworthy was the relative insensitivity of the transition state structure to the reaction mechanism change.
