Current Status and Tasks of Contaminant Migration Experiment Using Underground Research Laboratory (지하연구시설을 이용한 오염물질 이동실험 현황 및 과제)

  • Research and development for disposal of contaminants including radioactive wastes in deep underground have been carried out from laboratory works. However, validation and reliability of the data from the laboratory are arguing issues because they are not obtained from real disposal situations. Underground research laboratory (URL) is not only a solution to overcome such limitations, but also a valuable facility for performance assessment as an engineering scale. However, it requires much budget, and environmental issues can give rise to social conflicts easily. Such considering points related to URL are discussed as well as current status of worldwide URLs are introduced. Furthermore study plans for solute transport in a small-scale underground research tunnel (KURT), which was authorized recently as an non-radioactive facility in Korea, also described.

Precipitation Process in Cu-0.2%Cr-0.05%Zr Alloy Studied by the Electrical Resistivity Measurements (전기저항 측정에 의한 Cu-0.2%Cr-0.05%Zr 합금의 시효석출 거동)

  • The precipitation process in Cu-0.2 wt.%Cr-0.05 wt.%Zr alloys has been studied by electrical electrical resistivity measurements. The kinetics of precipitation could be well described by Johnson-Mehl-Avrami equation, $f(t)=1-\exp(-kt^n)$. The values of n were found to be in the range of 0.36~0.42 at first stage and 1.3~1.6 at second stage. The activation energy was determined by cross-cut method and was 80~89 kJ/mol. The value is similar to the energy for the migration of either a vacancy or a vacancy-solute complex through the lattice.

  • In this study, numerical model for transport of radionuclide and colloid was developed. In order to solve reaction-migration governing equation for colloid and radionuclide, Strang-splitting Sequential Non-Iterative (SNI), which is one of Operator Splitting Method, was used for numerical method and this was coded by MATLAB. From the verification by comparing the simulation results with analytical solution considering only solute transport and rock diffusion, the Pearson's correlation coefficient was greater than 0.99 which demonstrates the accuracy of the model.

  • As Al-Li based alloys are to find widespread use in aerospace and other structural applications in which their low-density high specific stiffness properties be exploited, their mechanical properties must adequately match those of the which they intended to replace. In order to develop these purposed, the precipitation behaviors of the rapidly solidified Al-Li-Ti alloys aged at various temperature were investigated. ${\delta}'$ phase precipitated homogeneously in the matrix during not only melt quenching but also aging at the aging temperature of $160^{\circ}C\;and\;210^{\circ}C$. The addition of the Ti in Al-Li alloy promoted the formation of $\delta$' phase. The addition of Ti on the ${\delta}'$ solvus line had a little effect over the thermodynamics ${\delta}'$ solvus line. The reason for these behavior was that the ${\delta}'$ phase was suppressed to precipitate as much as supercooling by melt quenching. The discontinuous precipitation reaction occurred by the preferential growth of ${\delta}'$ phase due to the migration of grain boundary provided the driving force dependent of solute concentration fluctuations.

Insights from an OKMC simulation of dose rate effects on the irradiated microstructure of RPV model alloys

  • This work studies the defect features in a dilute FeMnNi alloy by an Object Kinetic Monte Carlo (OKMC) model based on the "grey-alloy" method. The dose rate effect is studied at 573 K in a wide range of dose rates from 10-8 to 10-4 displacement per atom (dpa)/s and demonstrates that the density of defect clusters rises while the average size of defect clusters decreases with increasing dose rate. However, the dose-rate effect decreases with increasing irradiation dose. The model considered two realistic mechanisms for producing <100>-type self-interstitial atom (SIA) loops and gave reasonable production ratios compared with experimental results. Our simulation shows that the proportion of <100>-type SIA loops could change obviously with the dose rate, influencing hardening prediction for various dose rates irradiation. We also investigated ways to compensate for the dose rate effect. The simulation results verified that about a 100 K temperature shift at a high dose rate of 1×10-4 dpa/s could produce similar irradiation microstructures to a lower dose rate of 1×10-7 dpa/s irradiation, including matrix defects and deduced solute migration events. The work brings new insight into the OKMC modeling and the dose rate effect of the Fe-based alloys.

Hydrogeological Properties of Geological Elements in Geological Model around KURT (KURT 지역에서 지질모델 요소에 대한 수리지질특성)

  • To develop site characterization technologies for a radioactive waste disposal research in KAERI, the geological and hydrogeological investigations have been carried out since 1997. In 2006, the KURT (KAERI Underground Research Tunnel) was constructed to study a solute migration, a microbiology and an engineered barrier system as well as deeply to understand geological environments in in-situ condition. This study is performed as one of the site characterization works around KURT. Several investigations such as a lineament analysis, a borehole/tunnel survey, a geophyscial survey and logging in borehole, were used to construct the geological model. As a result, the geological model is constructed, which includes the lithological model and geo-structural model in this study. Moreover, from the results of the in-situ hydraulic tests, the hydrogeological properties of elements in geological model were evaluated.

The Candidate Tumor Suppressor Gene SLC8A2 Inhibits Invasion, Angiogenesis and Growth of Glioblastoma

  • Glioblastoma is the most frequent and most aggressive brain tumor in adults. Solute carrier family 8 member 2 (SLC8A2) is only expressed in normal brain, but not present in other human normal tissues or in gliomas. Therefore, we hypothesized that SLC8A2 might be a glioma tumor suppressor gene and detected the role of SLC8A2 in glioblastoma and explored the underlying molecular mechanism. The glioblastoma U87MG cells stably transfected with the lentivirus plasmid containg SLC8A2 (U87MG-SLC8A2) and negative control (U87MG-NC) were constructed. In the present study, we found that the tumorigenicity of U87MG in nude mice was totally inhibited by SLC8A2. Overexpression of SLC8A2 had no effect on cell proliferation or cell cycle, but impaired the invasion and migration of U87MG cells, most likely through inactivating the extracellular signal-related kinases (ERK)1/2 signaling pathway, inhibiting the nuclear translocation and DNA binding activity of nuclear factor kappa B ($NF-{\kappa}B$), reducing the level of matrix metalloproteinases (MMPs) and urokinase-type plasminogen activator (uPA)-its receptor (uPAR) system (ERK1/2-$NF-{\kappa}B$-MMPs/uPA-uPAR), and altering the protein levels of epithelial to mesenchymal transitions (EMT)-associated proteins E-cardherin, vimentin and Snail. In addition, SLC8A2 inhibited the angiogenesis of U87MG cells, probably through combined inhibition of endothelium-dependent and endothelium-nondependent angiogenesis (vascular mimicry pattern). Totally, SLC8A2 serves as a tumor suppressor gene and inhibits invasion, angiogenesis and growth of glioblastoma.
