Three Korean native steers ($779{\pm}24$ kg) fitted with duodenal cannulas were used in a $3{\times}3$ Latin square design to investigate the influence of oral administration of soluble proteins, intact casein (IC) and acid hydrolyzed casein (AHC), on gastro-intestinal hormone (GIH) secretion in the blood and pancreatic ${\alpha}$-amylase activity in the duodenum. Oral treatment consisted of a basic diet (control), IC (C+100% protein), or AHC (C+80% amino acid, 20% peptide) for 21 d. Blood and duodenum samples were collected for measurement of serum GI hormones, and pancreatic ${\alpha}$-amylase activity was determined at 900, 1030, 1330, 1630, and 1930 h after feeding on d 21 of treatment. The levels of serum cholecystokinin (CCK) and secretin in the IC treatment group were higher compared to the other treatment groups (p<0.05). In addition to the changes in CCK and secretin levels upon IC treatment, the pancreatic ${\alpha}$-amylase activity in the duodenum was higher in the IC group compared to the control diet group (p<0.05). The response of serum ghrelin to IC and AHC treatment was in accordance with the response of serum secretin. The level of peptide fragments flowing in the duodenum was higher in the IC treatment group than the other treatment groups (p<0.05). In conclusion, this study demonstrated that an increase in duodenal CCK and secretin upon IC oral administration increased pancreatic ${\alpha}$-amylase secretion. In addition, ghrelin may be associated with GI hormone secretion in Korean native steers.

Colostrum deprived, newborn pigs (N=12, $1.64{\pm}0.05kg$) were used to study the renal threshold of carnitine, and effects of emulsified medium-chain triglyceride (MCT, tri-8:0) feeding on kinetics of plasma carnitine and urinary carnitine excretion. An arterial catheter was inserted through an umbilical artery, and a bladder catheter was inserted via the urachus. Piglets were oro-gastrically gavaged with one of six carnitine levels (0, 60, 120, 180, 240, $480{\mu}mol/kg\;W^{0.75}$) with (+MCT) or without medium-chain triglycerides (-MCT) in 0.9% NaCl solution. Blood was sampled into heparinized tubes at 0, 1, 2, 4, 6, 8, 14, and 20 h after gavage, and urine was collected and pooled into 1 h or 2 h composite samples to determine free- and short-chain carnitine concentrations. Plasma from the 12 newborn piglets before gavage contained $10.6{\pm}1.2{\mu}mol/L$ free carnitine and $7.2{\pm}0.6{\mu}mol/L$ acid-soluble acyl carnitine. The renal threshold for carnitine was similar between the MCT and the +MCT group (42.6 13.1 and $46.4{\pm}2.0{\mu}mol/L$, respectively), but the correlation between plasma free carnitine and urinary excretion was altered. Plasma free carnitine linearly increased with increasing carnitine dosage (-MCT group, $R^2=0.95$, p<0.001; +MCT group, $R^2=0.91$, p<0.001), but was decreased by 50% when medium-chain triglycerides were fed. The peak in plasma free carnitine concentration was depressed by medium-chain triglycerides feeding also. Therefore, the plasma and urinary short-chain/free carnitine ratio of the +MCT group was increased by 100% and 40%, respectively (p<0.01). Feeding of medium-chain triglycerides may delay plasma carnitine elevation via altering the kinetics of absorption. Similarly, the plasma and urinary short-chain/free carnitine ratio were affected by interaction between medium-chain triglycerides and time (p<0.01). The present study suggests that an oral carnitine dose over $480{\mu}mol/kg\;W^{0.75}$ may be needed to reach the free carnitine renal threshold within a short period, especially when provided together with medium-chain triglyceride.
Objectives : This study was performed to evaluate the efficacy of extract from Citri Pericarpium (CP) on lipid-lowering effect in semi-healthy subjects with increased serum levels of total cholesterol and triglyceride. Methods : A experimental group (n=45) and placebo group (n=46) were randomly established based on the plasma lipid profiles (triglyceride, total cholesterol etc.) in the semi-healthy subjects, then all subjects randomly took experimental or placebo capsules, which contain 1,200 mg of CP or the same dose of soluble starch per day with regular meals for 8 weeks by double-blind method. Results : One-hundred forty (140) subjects were screened, then 91 subjects enrolled in the study. No serious adverse events were reported for trial period. The CP treatment showed statistically significant decrease in serum levels of triglyceride both at 4-weeks (p<0.01) and at 8-weeks (p=0.0001). Futhermore, there was significant difference in the triglyceride levels between CP-treated and placebo group. The CP-treated but not placebo group, experienced decrease in serum levels of total cholesterol at 8-weeks. Then there was no significant difference in the total cholesterol levels between the two groups. In addition, statistically significant difference of AST, ALT, ${\gamma}$-GT serum levels wasn't shown before and after trial in two groups. Conclusion : These results demonstrate that CP can significantly suppress the elevated triglyceride but not total cholesterol level. Therefore, CP may play a role in the improvement of blood lipid levels, as an effective functional food.
To improve quality as a food source and functional properties of traditional fermented soybean paste (Doenjang), the Korean herb medicines (group I ∼ IV) extracted at 121$^{\circ}C$ for 2.5 hours were used as brine sources. Capsule Meju were fermented in cool brines with three different concentration (group A, B and C) of the extracts at room temperature for 3 months in large earthenware jars. The effect of traditional soybean Doenjang (refer as DHM below) supplemented with the extract of herb medicines on the food quality and chemical characteristics were examined. General composition of DHM (moisture 58.2%∼61.9%, pH 5.32∼5.56, total acidity 3.07∼3.70 ml, salt 11.7∼13.9%) did not differ from those of control (moisture 61.9%, pH 5.50, total acidity 3.25 ml, salt 12.6%). Concentration of reducing sugar, crude protein, crude lipid and amino type nitrogen were higher in DHM than those of control without addition of the extracts. Water insoluble-brown index showed no difference by the extracts. However, the former had higher water soluble-brown index than the latter, and within the DHM, groups I and II were higher than groups III and IV. Lightness values (L) of Hunter color index in groups I and II were 24.32 and 31.04, respectively. Redness values (a) of all DHMs were lower than that of control. Yellowness value (b) of groups III and IV were similar to that of control group, and Hunter b values of group I and II were lower than that of control.
In general, anionic and cationic surfactants are incompatible because their mixtures form insoluble complexes and precipitate in the water. There are, however, some equimolar complexes of anionic and cationic surfactant that are soluble and behave like regular surfactants, specifically like nonionic surfactants, thus named pseudo-nonionic surfactant complexes. Pseudo-nonionic complexes are more effective and efficient in surface activities than their ionic surfactant components as shown by their equilibrium and dynamic surface tensions. They pack at the interface more than their ionic components. When a novel cationic surfactant, diglyceryl dodecyl dimethyl ammonium chloride(DGDAC), having the polyhydroxyl group at the hydrophilic head group, was mixed with a conventional anionic surfactant (sodium dodecyl sulfate; SDS) at equimolar ratio, we found that the aqueous equimolar mixture showed strong positive synergism in which molecular interaction parameter ${\beta}^M$ was very low, -17.2. According to the studies of equilibrium phase behavior and microscopy, this mixed system could form homogenous solutions containing vesicles.
Objective: The aim of this study was to determine the effects of feeding starch sugar byproducts (SSBs) on in situ disappearance rate, performance, and carcass characteristics of Hanwoo steers in the late finishing stage. Methods: To determine the in situ disappearance rate, nylon bags filled with 5 g of SSB were inserted into the ventral sac of two cannulated Holsteins cows and incubated for 0, 2, 4, 8, 16, 24, and 48 h. A total of 30 Hanwoo steers were fed the experimental diets, which were basal diet (control) and 7% SSB on an as-fed basis (4.35% dry matter [DM]), formulated according to requirements of the Korean Feeding Standard for Hanwoo. The experiment was conducted over 80 days using a completely randomized block design. Results: Soluble fraction a of DM and organic matter (OM) was 44.20% and 64.60% DM, fraction b was 23.00% and 19.40% DM, and c values (the rate of degradation of fraction b) were 0.04 and 0.04/h, respectively. The effective degradability of DM at rumen solid outflow rates of 0.02, 0.05, and 0.08/h was 59.83, 54.75, and 52.16, respectively, and for OM was 77.78, 73.52, and 71.34, respectively. Initial and final body weight, average daily gain, DM intake, and gain:feed did not differ significantly between control and SSB groups during the entire experimental period. Carcass traits of Hanwoo steers with SSB supplementation were not significantly different between treatments except for dressing percentage, which was greater with SSB treatment. The content of saturated fatty acid (SFA) was greater and that of unsaturated fatty acids (UFA) was lower in the SSB group than in the control group. The ratio of UFA to SFA was significantly lower in the SSB group than in the control group. Conclusion: A total mixed ration containing less than 4.0% DM of SSBs can be used in Hanwoo steers without a decrease in productivity and carcass traits.
The Aster glehni extract has many therapeutic and medicinal values. Therefore, it is essential to set appropriate conditions for enzyme treatment to efficiently extract A. glehni. In this study, changes in the quality of A. glehni extract depending on the concentration and time of enzyme treatment was investigated to increase its effective utilization. Compared to the control, the pH of the extract of A. glehni its soluble solid content increased with the enzyme treatment. The color of the A. glehni extract changed from green-yellow to reddish-yellow with the increase in treatment duration. The fructose and sucrose contents of the extract were the highest at 7.73% and 6.78%, respectively, in the control group without the enzyme treatment. Glucose and maltose contents were 6.91% and 4.44% in the C group (3.2% enzyme concentration and 60 min for enzyme treatment), respectively. Total polyphenol content, which shows antioxidant activity, was the highest at 7.38 mg GAE/g in the E group (1.6% of enzyme concentration and 120 min for enzyme treatment). 2,2-diphenylpicrylhydrazyl (DPPH) and 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid (ABTS) showed the highest radical scavenging activity in the C group (3.2% of enzyme concentration and 60 min for enzyme treatment). These results enable setting appropriate conditions of enzyme treatment in terms of enzyme concentration and time for the production of dry powders using A. glehni extract.
Cu(II) binding mechanisms with water soluble organic fractions (WSOF) extracted from an agricultural soil (W), a soil treated with sludge for 6 years ($WS_6$), a sludge-soil mixture incubated for one week ($WS_1$), and sewage sludge (SS) were studied by electron spin resonance (ESR) spectroscopy and potentiometric titrations. Cu(II)-WSOF complexes produced $g_{11}$ values which were larger than $g_{\perp}$ values, indicating that the coordination of Cu(II) complex was an elongated octahedron. At liquid $N_2$ temperature (77K), the Cu(II)-W complex showed an anisotropic ESR spectrum while the Cu(II)-SS complex showed an isotropic spectrum. These spectral results suggest that the oxygen donor ligands of W may form relatively strong bonds with $Cu^{2+}$ due to extensive chelation while ligands of SS may form little or no chelate bonds with $Cu^{2+}$. The ESR spectra of Cu(II)-SS complex also suggest that each of four in-plane ligands (e.g., $COO^-$, $H_2O$, $Cl^-$, etc.) may act independently as monodentate ligands. Oxygen donor ligands such as aromatic carboxyl groups were probably the major Cu(II) binding sites in W. Sulfonate, aliphatic carboxyl group, and N-containing ligands were probably the major binding sites in SS at pH 5. The Cu(II) complexation with N-containing groups increased as sludge was added to the soil. Much higher (6x) pyridine concentrations were required to displace W from Cu(II)-W complex as compared to the Cu(II)-SS complex.
Chitosan as a natural polymer has superior physicochemical properties such as biocompatibility, biodegradability and nontoxicity, but application of chitosan for therapy of cancer and gene related-disease has been limited by poor solubility in aqueous solution. Therefore, low molecular weight water-soluble chitosan (LMWSC) with high reactivity and strong positive charge can be applied as a delivery system having function to carry in the specific tissue the bioactive material like poor solubility drug, or therapeutic gene and developed as a therapeutic system having good therapeutic efficiency. The most important factor for therapy of various diseases is to reveal the antigen or receptor expressed in specific lesion tissue and the antibody and ligand which can bind with antigen is to introduce at the biomaterials for enhancement the therapeutic efficiency. The studies for cationic synthetic polymer as drug or gene delivery have been actively performed, but it has many problems such as toxicity in the body, therapeutic efficiency. From this point of view, this article demonstrated the introduction of functional groups to target the specific tissue and therapeutic strategy using the modification of LMWSC with free-amine group. The development of these delivery system will provide a positive vision for cancer therapy.
A series of oleic acid esters containing mercaptobenzothiazole and dialkylenedithiocarbamate, which are potential environmentally friendly lubricating grease additives, were synthesized as 90% yield through several method such as nucleophilic substitution of dialkyl amine and carbon disulfide, reduction reaction, and condensation reaction. The structures of the additives were confirmed by $^1H$-NMR, FT-IR and EA analysis. The additives (1 wt%) were soluble in 100 N BO except C4-DTC-OE and soluble in soybean oil. The tribological properties as lubricating additives in 100 N BO were evaluated using 4-ball tester and the results showed as follows: Bz-thia-OE < C4-DTC-OE < Pyrro-DTC-OE < C8-DTC-OE. Whereas, the 4-ball anti-wear properties were not shown in soybean oil.
