Production of DNA polymerase from Thermus aquaticus in recombinant Escherichia coli

  • Among dozens of DNA polymerases cloned from thermophilic bacteria, Taq DNA polymerase from Thermus aquaticus has been most frequently used in polymerase chain reaction (PCR) that is being applied to gene cloning, DNA sequencing, gene expression analysis, and detection of infectious and genetic diseases. Since native Taq DNA polymerase is expressed at low level in T. aquaticus, recombinant Escherichia coli system was used to produce Taq DNA polymerase in a large amount. Taq DNA polymerase was expressed as a soluble form under the control of tac promoter in E. coli, and purified by heat treatment and ion exchange chromatographies. The purified Taq DNA polymerase was nearly homogeneous and exhibited a similar DNA amplification activity with a commercial Taq DNA polymerase.

  • 배추흰나비의 변태과정에 따른 중장내 trehalase의 변화와 분포 및 특성을 측정, 비교하였으며, 5영말유충의 중승내 trehalase를 정제하여 다른 기관과의 상호관계를 면피학적 방법으로 구명하여 다음과 같은 결과를 얻었다. 중장조직의 trehalase 활성은 5영초보다 5영말에서 높았으며 trehalase는 암, 수 모두 중장 후부에 많이 존재하였다. 전라초부터 우화직전까지는 중장내용물에서 활성이 나타났는데 누직후에 가장 높았으며 중장의 trehalase는 soluble form 이었다. 혈림프에서도 trehalase가 확인되었는데 나화직전에 높게 나타났다가 나화직후에 급진적으로 액소하였다. 중승 trehalase의 최적 P광는 6.0으로 전변태단계에 걸쳐 일정하게 나타났으며, Km값은 2.86$\times$10-3M이었다. 최종 정제된 나소의 순도는 약 9.2이며, Rm값은 0.3이었다.

  • The effect on the hydration of cement was that Cu and Pb reacted with alkali to form soluble hydrates at theinitial stage and then there followed a slow reaction forming insoluble metal hydroxides. These hydroxides were deposited on the surface of cement particles providing a barrier against further hydration. But as a slow reaction continued, the insoluble layers were eventually destroyed and the hydration reaction resumed. Thereafter, another retardation occured by restricting the polymerization of silicates, shown by FT-IR spectroscopy analysis. In the case of Cr, as its reaction with cement caused H2O, the coordinator of Cr complex, to replace or polymerize with OH-, the formation of Cr complex promoted the leakage of OH- and increased the heat of dissolution. So the total heat evolution during hydration was larger than that in the case of Pb or Cu. The retarding effect of heavy metal ions was in the order Pb>Cu>Cr.

  • A surface equation of state for globular proteins at air-water interface accounting for the molecular structure, segment-segment, segment-solvent, and electrostatic interactions was proposed and compared to C-14 isotope experiments. This lattice model comprised a simplifying assumption that all adsorbed segments are in the form of trains. The number of segment adsorbed per molecule in case of bovine serum albumin linearly depended on the surface concentration whereas the lysozyme segments adsorbed at the interface were independent of surface concentration. The segment-solvent(water) interaction for both of proteins were found to be unfavorable owing to the proteins unfolding. From comparison of model computation and experimental data, BSA unfolded more than lysozyne because of the larger surface area of contact.

  • A sequential copolypeptide with repeating unit sequences, in which a glycyl residue is flanked progressively by two ${\gamma}$-benzyl-L-glutamyl residues, has been synthesized by poymerizing p-nitrophenyl ester of ${\gamma}$-benzyl-L-glutamyl-${\gamma}$ -benzyl-L-gutamyl-glycine. Polymers obtained were characterized by viscosity and infra-red spectral data. The highest molecular weight obtained was 21,000. Molecular conformation in solid state was found to be a mixed form of and |${\beta}$-structure. Polymers obtained were insoluble in the most of the organic solvents except in a strong acid like dichloroacetic acid, but in binary mixtures of solvents such as dichloroacetic acid-ethylene dichloride or dichloroacetic acid-chloroform, they were soluble within certain ranges of solvent compositions.

  • Poly(2-cyano-5-methoxy-1,4-phenylenevinylene), PCMPV and a series of PPV copolymers containing 2-cyano-5-methoxy-1,4-phenylenevinylene (CMPV) units were prepared via the water-soluble precursor method. They were obtained in films form and were easily doped with $FeCl_3$. Doping of undrawn and drawn films of PCMPV homopolymer with $FeCl_3$ led to conductivity of $10^{-5}-10^{-4}\;Scm^{-1}$. Conductivity of $FeCl_3$-doped copolymer films ranged from $10^{-3}$ 4.0 Scm$^{-1}$ depending on composition. As the content of CMPV units in the copolymer increased further, the electrical conductivity steadly decreased. Electronic effect by the CN substituent and morphological changes brought by copolymerization appear to interplay intricately resulting in the observation of a maximum conducting composition.

  • The sludge, a waste of brewery industries, was examined for potentials as a natural organic fertilizer (or soil conditioner) for lawn. Trial products were measured for changes of physical, chemical properties in laboratory and seed germination and seedling growth in green house were also tested. The results are as the following:1The sludge from distilled liquor brewery contained high quantity of organic matter which had proper physical and chemical properties for lawn fertilizer (natural organic fertilizer, soil conditioner, top-dressing mix) . It showed good characteristics in handling and capabilities to be developed as commercial products for golf courses. 2.Sludge from beer company needs proper treatment to improve physical properties for futher degradiation. It is because aggregation of the sludge particles prevented microbial activities and changing to soluble form. 3.Green carbon can be used as carbon source for organic fertilizer production using brewery sludge, but it should not contain wood extract which inhibit seed germination and seedling growth.

Soluble Complex Formation of Rifampicin with Arginine (Rifampicin과 Arginine간의 가용성 Complex 형성에 관한 연구)

  • Rifampicin-arginine complex was prepared to increase the solubility and dissolution rate of rifampicin. Solvation method was applied to make the complex and its formation was identified by the solubility method, powder x-ray diffractometry, differential thermal analysis and spectroscopic determination. The complex was formed in the molar ratio of 1 : 1 which was proved by the slope ratio method and continuous variation method. The complex was a non-crystalline form determined by the x-ray powder diffractometric analysis. The solubility of complex in water was significantly higher than that of rifampicin itself. The stability constant and thermodynamical properties of the complex were determined to investigate the solubilization phenomena. The thermodynamic data showed that the complexation between rifampicin and arginine was an exothermic and spontaneous reaction. Simulated absorption studies carried out through the artificial lipid barrier in artificial gastric and intestinal juices. The results showed that the complex had an enhanced absorption rate of rifampicin nearly twice compared with that of rifampicin itself.

Preparation and Thermal Properties of Poly(enaminonitriles-ester)s Derived from Dicyanovinyl-Containing Bis-Hydroxy Monomers

  • Dicyanovinyl-containing bis-hydroxy monomers, p-bis[1-(4-hydroxypiperidinyl)]-2,2-dicyanovinyl]benzene (2), p-bis[1-[1-(2-hydroxyethyl)piperazinyl]-2,2-dicyanovinyl]benzene (3), p-bis[1-(4-hydroxyphenylamino)-2,2-dicyanovinyl]benzene (4) and p-bis[1-[N-methyl-(N-hydroxyethyl)amino]]-2,2-dicyanovinyl]benzene (5) were prepared from p-bis(1-chloro-2,2-dicyanovinyl)benzene (1) and the corresponding amino alcohol. The poly(enaminonitriles-ester)s with a variety of chemical structures in the main chain were prepared from them. The chemical structure of polymers was confirmed through the syntheses of their corresponding model compounds. The polymers are easily soluble in polar aprotic solvents such as DMF, DMSO and NMP. Brittle and hard films can be cast from DMF solutions of polymers. Most polymers showed a large exotherm in DSC analyses and undergo a curing reaction around 350 ℃ to form insoluble materials. The polymers consisting of rigid aromatic moieties show 80-88% residual weight at 500 ℃ under nitrogen.

  • In order to confirm the biological function of ORF10 in cephabacin biosynthesis gene cluster of Lysobacter lactamgenus as an ATP-binding-cassette (ABC) transporter, the gene for ORF10 was amplified and subcloned into pET-28a(+) expression vector. After gene induction with 0.5 mM IPTG at 30~! and further cultivation at $30^~$ !. for 8 hr, a lot of the recombinant ORF10 protein was produced as soluble form in cytoplasmic fraction as well as a membrane protein in the membrane fraction as likely as other ABC transporters.
