We investigated the changes of composition in immature green persimmon during cold(7$^{\circ}C$) and freezing (-2$0^{\circ}C$) storage for 30 days. Moisture, protein, fat and carbohydrate were not significantly different by storage temperature and storage days. Crude fiber had little change during freezing storage, but in case of cold storage, as storage time goes by, it slightly deceased. Content of polyphenol was increased during cold and freezing storage, especially during cold storage it was remarkably increased. Content of total pectin was decreased, while content of soluble pectin was increased during cold storage. Content of total pectin and soluble pectin was slightly decreased or increased during freeze storage. Content of tannin had little change during freezing storage but it significantly decreased during cold storage.

The experiment were carried out to determine the rate of regrowth, the changes of carbohydrates contained in the samples right after cutting, plant part for storage of carbohydtates and top-dressing time of ladino clover and orchardgrass. Plants were grown in the simple growth chamber as well as in the fields. The temperature, light intensity and light period were controlled to be 15-20$^{\circ}$, 4,250 Lux, and 15 hours in the simple growth chamber. The results obtained are as follows ; 1. The period of recovery to initial stands after cutting was about 3 weeks in ladino clover and 4-5 weeks in orchard grass in terms of dry matter and carbohydrates. 2. The content of total water-soluble carbohydrates of plant parts was the hightest in stolon of ladino clover and in sheath of orchardgrass, and the lowest in roots of ladino clover and orchardgrass. 3. Cutting resulted in a temporary decrease of total water-soluble carbohydrates in the shoots. Similar trends were observed when the plants were left intact with top-dressing. 4. The top-dressing applied simultalliousely with the cutting was more effective to stimu1ate regrowth than that applied after cutting, however, both of the simultaneous and after cutting application were still better than application prior to cutting.
Yam(Dioscorea) tubers were used as one of the dietary starches in Korea, China and Japan because of its characteristic properties. Yam tubers were powdered after freeze drying the sliced tubers to investigate carbohydrates. Chemical properties such as proximate components and carbohydrates were studied using three varieties of Korean yams, namely. D. batatas, D. aimadoimo and D. japonica. The proximate components of yam tubers showed that the major components of Korean yam tubers were carbohydrates and moisture. The main components of free sugars in yams were identified as fructose, glucose, sucrose and maltose. In addition to these four sugars, there were two unidentifiable peaks whose areas are too big to ignore. These results were obviously different from other reported data of free sugars in yams. The main components of total sugars were mannose and glucose.

In the present study, a new analytical method was devised for the simultaneous determination of soluble carbohydrates in soybean seeds using high performance liquid chromatography/evaporative light scattering detection (HPLC/ELSD). The limit of quantification (LOQ) for soybean soluble carbohydrates ranged from 5.6~7.6 mg/kg using the HPLC/ELSD method and from 16.2~33.9 mg/kg using the high performance liquid chromatography/refractive index detection (HPLC/RID) method. Therefore, the HPLC/ELSD method was more sensitive than HPLC/RID. The precision values for retention time and peak area of the HPLC/ELSD method were evaluated by inter-day (n=5) and intra-day (n=10) assays using a standard solution. All precision values (CV<2.5%) for soybean soluble carbohydrates were acceptable and fulfilled international acceptance criteria. All linear calibration curves were obtained with a correlation coefficient of $R^2$ >0.999. The contents of soluble carbohydrates for the "Shingikong" (yellow soybean) and "Cheongjakong 3" (black soybean) samples were analyzed using the HPLC/RID and HPLC/ELSD methods. The difference in carbohydrate contents between the two detection methods was significant. Carbohydrate contents in the HPLC/ELSD method were higher than those in the HPLC/RID method. Overall, the HPLC/ELSD method showed satisfactory resolution with a favorable LOQ and reproducibility. Therefore, these results indicate that the HPLC/ELSD method may be applied to determine the contents of soluble carbohydrates in soybean seeds and related food stuffs.

This experiment was carried out to investigate the effects of the different nitrogen rates and cutting heights on the dead stubble after cutting, and the carbohydrate reserved in stubble of sorghum-sudangrass hybrid (Sorghum bicolor (L.) Moench) Pioneer 988. The experiment was undertaken in the Experimental Livestock Farm of Agriculture Coll., Seoul Nat'l Univ. In Suweon, 1981 and 1982. The results obtained are summarized as follows: 1. The dead stubble after cutting was found to be great with high rate of nitrogen fertilizer and low cutting height. It was also learned that the rainfall during forage cutting period made the dead stubble increased. 2. A significant higher (p<0.05) carbohydrate reserves in the stubble was observed in the high stubble height at the first cutting time and 6th through 7th day after the first cutting. The results indicate that the high stubble height reserves more carbohydrate for the early regrwoth stage after the first cutting when comparing with the low stubble. 3. The content of carbohydrate reserves was influenced by climete. Drought is caused to high content of carbohydrate, whereas, rain is caused to low content of that. The critical soluble carbohydrate content causing the death of stubble supposed to be 3 to 6% at least. 4. It is suggest that carbohydrate reserves in plant do not play a distinctive role for the regrowth in a summer annual forage like sorghum-sudangrass hybrid, but it might be rather influenced by the other factors, for example, environmental conditions at harvest and new bud.
To investigate the solute pattern of salt marsh plants in Suncheon Bay in Korea, plants and soil samples were collected at three sites from July to September 2011. The soil pH around the investigated species was weakly alkaline, 6.9-8.1. The total ion and Cl- content of site 1 gradually increased, while those of site 2 and site 3 were lowest in August and highest in September. The exchangeable $Ca^{2+}$, $Mg^{2+}$ and $K^+$ in the soil were relatively constant during the study period, but the soil exchangeable $Na^+$ content was variable. Carex scabrifolia and Phragmites communis had constant leaf water content and very high concentrations of soluble carbohydrates during the study period. However, Suaeda malacosperma and S. japonica had high leaf water content and constant very low soluble carbohydrate concentrations. Carex scabrifolia accumulated similar amounts of $Na^+$ and $K^+$ ions in its leaves. Phragmites communis contained a high concentration of $K^+$ ions. Suada japonica and S. malacosperma had more $Na^+$ and $Cl^-$ ions than $K^+$ ions in their leaves. Suaeda japonica had higher levels of glycine betaine in its leaves under saline conditions than C. scabrifolia and P. communis. Consequently, the physiological characteristics of salt marsh chenopodiaceous plants (S. japonica and S. malacosperma) were the high storage capacity for inorganic ions (especially alkali cations and chloride) and accumulation of glycine betaine, but monocotyledonous plant species (C. scabrifolia and P. communis) showed high $K^+$concentrations, efficient regulation of ionic uptake, and accumulation of soluble carbohydrates. These characteristics might enable salt marsh plants to grow in saline habitats.

A suitable bioreactor culture system for shoot proliferation and bulblet formation of Allium victorialis var. platyphyllum Makino was established. Uptake of soluble carbohydrates in different bioreactor culture systems was also analyzed during the entire culture period. Optimal conditions for multiple shoot formation were determined in raft culture (RC) and modified raft culture system (MRC) (13-15 per explant) in which the explants were placed on a net contacting liquid medium. For bulblet formation and enlargement, 93.4% of shoot clumps formed bulblets at the basal part. Furthermore, they were uniform in size when cultured with ebb & flood system (E&FS). Bulblets harvested from RC and MRC showed vigorous rooting, however, their growth was not uniform. Whereas soluble carbohydrate contents in the bulblets cultured in E&FS were low, starch content was high. Sucrose, glucose and fructose concentrations in the medium of E&FS culture system decreased as bulblet formation and enlargement proceeded, suggesting that external sucrose is taken up to by the cells before it is hydrolyzed.

Insect cell cultures have become important tools in the production of biological substances for use in a variety of research, human and veterinary medicine, and pest control applications. These applications often require the introduction of foreign DNA into the cells and have generally used methods originally developed for use with human and other mammalian cell cultures. While these methods can be successfully employed, they are often less efficient with insect cells and frequently involve complex procedures or require specialized equipment. Even when they do work, they may require substantial modification because of differences in the culture medium or growth patterns of insect cells. In this study, We have optimized transfection conditions of Sf9 cell line using insect expression vector pIZT/V5-His which expresses green fluorescent protein effectively. Human stem cell factor (hSCF) is a glycoprotein that plays a key role in hematopoiesis acting both as a positive and negative regulator, often in synergy with other cytokines. It also plays a key role in mast cell development, gametogenesis, and melanogenesis. It can exist in membrane-bound form and in proteolytically released soluble form. As determined by an enzyme-linked immunosorbent assay performed, hSCF level in supernatant averaged 995ng/ml. The human hSCF was partially purified by immunoaffinity chromatography and analyzed with sodium dodecyl sulfate-polyacrylamide gel electrophoresis and immunoblotting. The results show that the hSCF has N-linked carbohydrate and corresponds to the soluble form, at or about 223 amino acids in length. The findings suggest functional importance for soluble hSCF in cells.
Soybeans were soaked in water for 3 hrs and germinated at $25^{\circ}C$ for 5 days followed by hot water extraction and boilding for 30 min. The prepared soymilk was analyzed for chemical composition, phytic acid and soluble minerals. Germination resulted in a slow decrease in total solids while protein recovery increased to a maximum of 92.5% after 2 days. Carbohydrate content was reduced to 65% by the 5th day of germination. The intial phytic acid content in soymilk of 1.26g recovered from 100g dry soybean was decreased by 41.3% while inorganic phosphorus increased more than four times from 31.3mg to 135.0mg. Total soluble minerals was also steadily increased.
A gene (sll0158) putatively encoding a glycogen branching enzyme (GBE, E.C. 2.4.1.18) was cloned from Synechocystis sp. PCC6803, and the recombinant protein expressed and characterized. The PCR-amplified putative GBE gene was ligated into a pET-21a plasmid vector harboring a T7 promoter, and the recombinant DNA transformed into a host cell, E. coli BL21(DE3). The IPTG-induced enzymes were then extracted and purified using Ni-NTA affinity chromatography. The putative GBE gene was found to be composed of 2,310 nucleotides and encoded 770 amino acids, corresponding to approx. 90.7 kDa, as confirmed by SDS-PAGE and MALDI-TOF-MS analyses. The optimal conditions for GBE activity were investigated by measuring the absorbance change in iodine affinity, and shown to be pH 8.0 and $30^{\circ}C$ in a 50 mM glycine-NaOH buffer. The action pattern of the GBE on amylose, an $\alpha$-(1,4)-linked linear glucan, was analyzed using high-performance anion-exchange chromatography (HPAEC) after isoamylolysis. As a result, the GBE displayed $\alpha$-glucosyl transferring activity by cleaving the $\alpha$-(1,4)-linkages and transferring the cleaved maltoglycosyl moiety to form new $\alpha$-(1,6)-branch linkages. A time-course study of the GBE reaction was carried out with biosynthetic amylose (BSAM; $M_p{\cong}$8,000), and the changes in the branch-chain length distribution were evaluated. When increasing the reaction time up to 48 h, the weight- and number-average DP ($DP_w$ and $DP_n$) decreased from 19.6 to 8.7 and from 17.6 to 7.8, respectively. The molecular size ($M_p$, peak $M_w{\cong}2.45-2.75{\times}10^5$) of the GBE-reacted product from BSAM reached the size of amylose (AM) in botanical starch, yet the product was highly soluble and stable in water, unlike AM molecules. Thus, GBE-generated products can provide new food and non-food applications, owing to their unique physical properties.
