The Importance of Weathered Crude Oil as a Source of Hydrocarbonoclastic Microorganisms in Contaminated Seawater

  • This study investigated the hydrocarbonoclastic microbial community present on weathered crude oil and their ability to degrade weathered oil in seawater obtained from the Gulf St. Vincent (SA, Australia). Examination of the native seawater communities capable of utilizing hydrocarbon as the sole carbon source identified a maximum recovery of just $6.6{\times}10^1\;CFU/ml$, with these values dramatically increased in the weathered oil, reaching $4.1{\times}10^4\;CFU/ml$. The weathered oil (dominated by > $C_{30}$ fractions; $750,000{\pm}150,000mg/l$) was subject to an 8 week laboratory-based degradation microcosm study. By day 56, the natural inoculums degraded the soluble hydrocarbons (initial concentrations $3,400{\pm}700mg/l$ and $1,700{\pm}340mg/l$ for the control and seawater, respectively) to below detectable levels, and biodegradation of the residual oil reached 62% ($254,000{\pm}40,000mg/l$) and 66% ($285,000{\pm}45,000mg/l$) in the control and seawater sources, respectively. In addition, the residual oil gas chromatogram profiles changed with the presence of short and intermediate hydrocarbon chains. 16S rDNA DGGE sequence analysis revealed species affiliated with the genera Roseobacter, Alteromonas, Yeosuana aromativorans, and Pseudomonas, renowned oil-degrading organisms previously thought to be associated with the environment where the oil contaminated rather than also being present in the contaminating oil. This study highlights the importance of microbiological techniques for isolation and characterisation, coupled with molecular techniques for identification, in understanding the role and function of native oil communities.

DAF와 펜톤 산화 공정을 이용한 유류 오염 지하수 처리 (Treatment of Oil Contaminated Groundwater Using DAF and Fenton Oxidation Process)

  • 에너지원 및 다양한 화학제품의 원료로 유류의 소비량이 증가함에 따라 유류의 유출사고가 빈번하게 발생하고 있다. 유류오염 지하수 처리를 위해 용존공기부상법이 사용되어지고 있으나 용해성 기름을 효과적으로 제거하지 못하고 있는 실정이다. 따라서 지하수내 주오염원인 자유상 유류와 친수성(oil-in water) 또는 친유성 에멜젼(water-in oil) 상태의 기름과 용해성 기름을 효과적으로 저감하는 것이 필요하다. 본 연구에서는 용존공기부상법에 펜톤 산화 공법을 사용하여 효과적인 유류 오염 지하수 처리를 연구하였다. 펜톤 실험 결과 TPH(Total Petroleum Hydrocarbon) 제거를 위한 최적 조건은 pH 3, $H_2O_2$ 주입농도 25mM과 $Fe^{2+}$ 주입농도 25mM으로 나타났다. DAF와 펜톤 산화 실험을 수행한 결과 7분내에 TPH(Total Petroleum Hydrocarbon)와 COD(Chemical Oxygen Demand) 농도가 각각 1.5mg/L와 40.0mg/L 이하로 감소하는 것으로 나타났다. 하지만 펜톤 산화 반응으로 인하여 슬러지 농도가 약 5배 가량 증가하여 후단에 침전조를 설치하는 것이 바람직하다.

Soybean Oil 및 Flaxseed Oil 첨가 배양시 탄수화물 첨가수준에 의한 반추미생물의 Bio-hydrogenation과 CLA 생성에 미치는 효과 (Effect of the Level of Carbohydrates on Bio-hydrogenation and CLA Production by Rumen Bacteria When Incubated with Soybean Oil or Flaxseed Oil In vitro)

  • 본 실험은 대두유 또는 아마유를 in vitro 방법으로 배양 할 때, 탄수화물원의 첨가수준이 반추위 박테리아에 의한 bio-hydrogenation과 CLA 생성에 미치는 효과를 조사하였다. 4수준(0%, 0.3%, 0.6% 그리고 0.9%, w/v)의 혼합된 탄수화물원(glucose, cellobiose, soluble starch, 1:1:1, w/w/w)과 두 종류의 oil을 cellulose powder에 흡착시킨 형태로 각각 60mg씩 인공타액(120ml)과 반추위액(30ml)이 혼합된 배양액(150ml)에 넣은 다음 39℃에서 12시간동안 혐기적으로 배양하였다. 배양액의 pH와 암모니아 농도는 두 종류 oil을 첨가한 배양액 모두에서 탄수화물원의 첨가 수준이 높을수록 pH와 암모니아 농도가 낮았다(P<0.05). 탄수화물원의 첨가 수준이 증가할수록 total VFA 생성량 역시 증가되었으나(P<0.01) 첨가한 oil 간의 차이는 없었다. 배양시간이 경과됨에 따라 탄수화물원의 첨가수준이 높을수록 propionate의 조성비율이 증가된 반면(P<0.001) acetate와 butyrate의 조성비율은 감소되었다. 배양 후 3시간이 경과하였을 때 배양액 내 oleic acid의 조성비율은 대두유에 비하여 아마유를 첨가한 배양액에서 낮았으나(P<0.001) linoleic acid의 비율은 높았다(P<0.001). 이와는 달리 탄수화물원의 수준이 증가될수록 stearic acid(P<0.05), CLA(P<0.01) 및 cis-9, trans-11 CLA(P<0.001)의 조성비율은 감소되었으나, linoleic acid의 조성 비율은 증가되었다(P<0.05). Linolenic acid의 조성비율에 있어서는 첨가된 oil의 종류와 첨가된 탄수화물원의 수준간의 상호작용이 있는 것으로 나타났는데(P<0.001), 12시간의 배양종료 후 대두유 첨가구에 비해 아마유 첨가구에서 stearic acid(P<0.01), oleic acid(P<0.001), 그리고 trans-11C18:1(P<0.01)의 조성비율이 감소된 반면, linoleic acid(P<0.001)와 linolenic acid(P<0.01)의 조성비율은 증가되었다. 탄수화물원의 첨가수준이 증가될수록 stearic acid와 총 CLA의 조성비는 감소되었으나(P<0.01), trans-11-C18:1(P<0.05)와 linoleic acid(P<0.01)의 조성비율은 증가되었다. 배양 12시간 후 배양액 내의 oleic acid (P<0.05), linoleic acid(P<0.05) 및 linolenic acid(P<0.01)의 조성비율에 있어서는 첨가한 oil의 종류와 첨가한 탄수화물원의 수준간의 상호작용이 있었는데, 탄수화물원의 첨가수준이 감소됨에 따라 cis-9, trans-11 CLA와 trans-10, cis-12 CLA의 조성비 역시 감소되는 경향이었으나 첨가한 oil의 종류에 대한 영향은 거의 받지 않았다. 본 실험의 결과를 종합해 볼 때, 탄수화물의 첨가수준과 oil의 첨가는 반추미생물의 bio-hydrogenation 작용 및 CLA 생성에 영향할 수 있는 것으로 여겨진다.

Preparation of Highly Water Soluble Tacrolimus Derivatives: Poly(Ethylene Glycol) Esters as Potential Prod rugs

  • Tacrolimus (FK506), which is isolated from Streptomyces tsukubaensis, is a new potent immu-nosuppressant. Because of poor solubility in water, the conventional intravenous dosage forms of tacrolimus contain surfactants such as cremophor EL (BASF Wyandotte Co.) or hydroge-nated polyoxy 60 castor oil (HCO-60) which may cause adverse effects. This study relates to a polymer-tacrolimus conjugate, which can be dissolved in water, formed by chemically binding the sparingly soluble drug, tacrolimus, with the water soluble polymer, methoxypoly(ethylene glycol) (mPEG). Water soluble tacrolimus-mPEG conjugates have been synthesized and shown to be function in vitro as prodrugs. These conjugates are in the form of an ester wherein the 24-, 32- or 24,32-positions are esterified. The desired 24-, 32- or 24,32-esterified com-pounds were obtained by initially acylating of tacrolimus with iodoacetic acid at the 24-,32-, or 24,32-positions and then reacting the resulting acylated tacrolimus with a mPEG in the pres-ence of a base such as sodium bicarbonate. These conjugates were converted again into tac-rolimus by the action of enzymes in human liver homogenate, and the half-lives of the conjugates are approximately 10 min in the homogenate, indicating that the esterified tacroli-mus derivatives may be practically applicable as a prod rug for the immunosuppressant.

지용성 염료를 표지물로 사용한 인공오염포의 특성과 세척성에 판한 연구 (A Study on the Artificially Soiled Fabric Containing Oil Soluble Dye as an Indicator)

  • In order to evaluate the exact effect of detergency it is necessary both to measure the actual soil content by chemical analysis and to determine the degree of soil removal visually. Since it takes considerable time and effort to use both methods, usually one of the two methods is used. Many studies have been carried out through increasing the visibility of oily soil to evaluate detergency by measuring reflectance of fabrics. In this study Sudan Black B, an oil soluble dye was used as an indicator to increase the visibility of oily soil on cotton and polyester fabrics. The condition of artificially soiled fabrics and the method of evaluating detergency were investigated which represent the actual detergency of oily soil by measuring the reflectance only. Also the detergency of Sudan Black B and that of oily soil were compared with each other under various washing conditions, As a result, the K/S values converted from the reflectances showed a good correlation with the actual soil content. Linear relationship between K/S value and the actual soil content was obtained. The K/S values of washed fabrics were higher than those of unwashed fabrics which included same content of soil since the soil visibility changed during washing. But the difference was small when Sudan Black B was used. With the increase of soil content, detergency of cotton fabric decreased, but detergency of polyester fabric increased gradually. With regards to soiled fabrics, detergency of cotton fabric measured by K/S value was close to that of actual oily soil when Sudan black B was used as an indicator.

  • Because increased food delivery service and quick delivery service using motorcycle, registration numbers of motorcycles were sharply increased and it could contribute on worsening air quality. In this study, two models(50cc, 125cc) of motorcycle were tested by using three types of engine oil. Two motorcycles were tested with CVS-40 mode for emission characteristics such as CO, THC, NOx, Elemental Carbon(EC), Organic Carbon(OC), sulfate, soot and SOF(soluble organic fraction). Result of according to three types of lubricants which included phosphorus, sulfate ash impacted to particle matters so "C" lubricants is more higher PM than "A", "B" lubricants in this research.

Functional and Film-forming Properties of Fractionated Barley Proteins

  • Barley proteins are expected to have unique functional properties due to their high content of alcohol soluble protein, hordein. Since the barley proteins obtained by conventional isoelectric precipitation method cannot represent hordein fraction, barley proteins were fractionated to albumin, globulin, glutelin, and hordein with respect to extraction solvents. Functional properties and film-forming properties of solubility-fractionated barley proteins were investigated to explore their potential for human food ingredient and industrial usage. The 100 g of total barley protein comprised 5 g albumin, 23 g globulin, 45 g glutelin, and 27 g hordein. Water-binding capacities of barley protein isolates ranged from 140-183 mL water/100 g solid. Hordein showed the highest oil absorption capacity (136 mL oil/100 g), and glutelin showed the highest gelation property among the fractionated proteins. In general, the barley protein fractions formed brittle and weak films as indicated by low tensile strength (TS) and percent elongation at break (E) values. The salt-soluble globulin fraction produced film with the lowest TS value. Although films made from glutelin and hordein were dark-colored and had lower E values, they could be used as excellent barriers against water transmission.

  • Lignocellulosic biomass including acetosolv ricestraw and spruce lignin were liquefied and converted into liquid hydrocarbons by catalytic hydroliquefaction reaction. These experimental works were carried out in 1-liter-capacity autoclave using 50% tetralin and m-cresol solution respectively as soluble solvent and Ni. Pd. Fe and red mud as catalyst. $H_2$ gas was supplied into the reactor for escaltion of deoxhydroenolysis reaction. Catalyst concentrations were 1 % of raw material based on weight. The ratio between raw materials and soluble solvent are 1g and 10cc. The reaction conditions are 400-$700^{\circ}C$ of reaction temperature, 10-50 atms of reaction pressure. The highest yield of hydrocarbon, so called "product oil" showed 32% and 5.5% of lowest char formation when red mud was used as catalyst. The product oil yields from those of other catalysts were in the range of 20-29%. The influence of different initial hydrogen pressures was examined in the range d 30-50 atms. A minimum pressure of 35 atms was necessary to obtain a complete recovery of souble solvent for recycling.

면류용 알칼리제 처리가 건면의 조리특성에 미치는 영향 (Effect of Alkali Salts Adding on the Cooking Quality in Dried Noodles)

  • The effects of alkali salts adding on the cooking quality improving in dried noodles were investigated in the good texture maintaining for preventing solid soluble losses ,through chemical analysis and actual manufacturing practice ,the following results were obtained. Experiments were took a special flour of ASW:DNS=70:30, thickening agent(TA) composed of K2CO3 58%, Na2CO3 36% and Na4P2076%, and emulsified oil(EO) mixing of corn oil 44%, polysorbate 23%, emulsifier(ester of glycerin and fatty acids) 21%, soy lecithin 12%. When the mixing ratio of TA and EO to flour, is 0.03 and 1.5%(w/w) or morel than, satisfied the good quality. The water soluble solid matters content of the lowest 3.2% in the treating group that TA and EO is 0.03 and 1.5%(w/w) respectively, comparing to the 7.3% in the control group provides a excellent cooking quality. The research achieves the similar effects at specific gravity, water absorption ratio, weight increasing rate and volume expansion ratio. According to appearance test the more treating of TA turn the noodle into deeper yellow-green color. Turning to the deeper yellow color according to the increasing of EO provides better

콩의 종류와 건조 방법에 따른 비지 분말의 이화학적 특성 (Physicochemical Properties of Soybean Curd Residue Powder by Different Soybean and Drying Methods)

  • This study compared the physicochemical properties of soybean curd residue and black soybean curd residue produced by hot air-drying and freeze-drying. Regardless of drying method, the crude protein, crude ash, crude fiber contents, pH, L, a, b color values and water soluble index were higher in soybean curd residue, whereas total polyphenol contents and antioxidant activity were higher in black soybean curd residue. Significant differences in water absorption index, oil absorption capacity and emulsion activity were observed between soybean curd residue and black soybean curd residue in freeze-drying. On the other hand, the emulsion stability was not significant difference in both hot-air drying and freeze-drying. The crude protein and crude fiber contents of soybean curd residue were not significant difference between hot-air drying and freeze-drying. Freeze-drying resulted in higher crude ash contents, pH, water absorption index, water soluble index, oil absorption capacity, emulsion activity and emulsion stability than hot-air drying. Hot-air drying have caused significantly higher water contents, water activity, total polyphenol contents and antioxidant activity in soybean curd residue than freeze-drying. In conclusion, soybean type and drying methods affect the physicochemical and quality characteristics of soybean curd residue, which could be important factors in the manufacture of processed foods.
