Detemination of Short-term Bioconcentration Factor on Dichlorvos, Methidathion and Phosalone in Brachydanio rerio and Xiphophorus hellieri (Brachydanio rerio와 Xiphophorus hellieri를 이용한 Dichlorvos, Methidathion 및 Phosalone의 단기간 생물농축계수의 측정)

  • This study was performed to investigate the bioconcentration of dichlorvos, methidathion and phosalone in zebrafish (brachydanio rerio), red sword tail(Xiphophorus hellieri). The fishes were exposed to 0.05 ppm, 0.01 ppm, 0.50 ppm, one-hundredth concentration of 96-hrs LC$_{50}$ and one-thousandth concentration of 96-hrs LC$_{50}$ and test periods were 3, 5 and 8 days. The deputation rate of each pesticide from the whole body of fish was determined over the 24-hr period after treatment. Obtained results are summerized as follows: In the case of dichlorvos, dichlorvos concentration in zebrafish extract and BCF$_{s}$ of dichlorvos were increased as increasing test concentration. In the case of same experimental concentrations, dichlorvos concentration in zebrafish extract and BCF$_{s}$ of dichlorvos were decreased as proloning test periods, especially dropped after 5days. Dichlorvos concentration in red sword tail extract were increased as increasing test concentration, lyat BCF$_{s}$ in concentration of 0.05 ppm, 0.01 ppm and one-hundredth of 96-hrs LC$_{50}$ were decreased. Methidathion and phosalone concentration in zebrafish extract in zebrafish extract were increased as increasing test concentration, but there was little difference in BCF$_{s}$. In the case of same experimental concentrations, there were little differences in BCF$_{s}$ and concentration in zebrafish extract. In the case of red sword tail, it was impossible to calculate on BCF$_{s}$ data because test concentration was under the detecting limit on GC or test fish were die. Determined deputation rate conatant were highest on dichlorvos, and followed by methidathion, and phosalone. The results of determining depuration rate of these pesticides showed that the high BCF in fish might be due to the slow depuration rate in fish, it is thought to be responsible for vapor pressure, water solubility and partition coefficient. It is suggested that one-hundredth concentration of 96-hrs LC$_{50}$ will be proper test concentration because one-thousundth of LC$_{50}$ was under the detecting limit on GC. Dichlorvos, methidathion and phosalone, organophosphorous pesticides, were examined to their BCF$_{s}$ and depuration rates by means of fish test.

Solid Dispersion of an HIV Protease Inhibitor

  • LB71350 is an HIV protease inhibitor with poor aqueous solubility and extensive first pass effect. The purpose of the present study was to test the feasibility of solid dosage form of LB71350 with improved bioavailability utilizing solid dispersion. Three different compositions with varying ratio of (LB71350: Gelucire 44/14: Tween 20) were studied. Capsule filling of these solid dispersion compositions was tested using a semi-automatic capsule filling system. Oral bioavailability in dog was tested. Chemical and physical stability at 4, 25 and $40^{\circ}C$ was monitored by HPLC assay, dissolution test, powder XRD and microscopy. The capsule filling system yielded uniform products of drug loading up to 10%. Oral bioavailability in dog was improved compared to the aqueous suspension of crystalline LB71350. Capsules were chemically stable for up to 6 months at $40^{\circ}C$. However, there were temperature and composition dependent physical changes. Decrease in dissolution rates after storage at $40^{\circ}C$ was due to the polymorphic change. In conclusion, manufacturing process, bioavailability, and physico-chemical stability have been considered to propose a solid dispersion capsule formulation for the HIV protease inhibitor with poor physico-chemical properties. A new less soluble crystalline form identified during the physical stability test warrants further study.

  • To improve the solubility and adhesion strength of diamine-quinone polymers, urethane group was introduced into the polymer backbone. Urethane group containing polymeric diamines(UDA) were prepared firstly by the decarboxylation of the NCO group in NCO terminated prepolymer obtained from the reaction of MDI and 1.6-hexandiol. The reaction mole ratio of NCO/OH was changed from 1.2 to 2.1 to increase the molecular weight of diamine. From the addition reaction of UDA and p-benzoquinone in THF or DMSO solvent, five kinds of urethane group containing diamine/quinone polymers(PUAQ) were prepared according to the diamine used. IR, NMR, UV-visible spectroscopy and GPC were used to characterize the molecular structure and molecular weight of UDAs and PUAQs. Thermal analysis of polymer by DSC and TGA were carried out according to the molecular weight differency. Also solubility test of UDA and PUAQ with 10 different solvents were carried out to predict the coacting property of the polymers.

Improvement of Hydrolysis and Bioavailability of Coprecipitated Products of Coptidis Rhizoma and Scutellariae Radix by β-Glucuronidase (베타-글루쿠로니다제에 의한 황련.황금 공침물의 가수분해 및 생체이용률 증가)

  • During the preparation of decoction from the mixture of Coptidis Rhizoma and Scutellariae Radix, insoluble copreciptate was formed. The coprecipitated product (COP) was composed of berberine and baicalin which was the active ingredient of Coptidis Rhizoma and Scutellariae Radix, respectively. COP was slightly soluble in water and could not be well absorbed after oral administration. This poor bioavailibility might be associated with its poor aqueous solubility. With the purpose of increasing the solubility and bioavailibility of COP, hydrolysis of COP by ${\beta}-glucuronidase$ was carried out. Hydrolyzed products (HOP) of COP were identified and assayed for active ingredients. The partition coefficient study, in situ absorption test, and pharmacokinetic study after oral administration were also performed. COP was found to be consisted of berberine and baicalin with molecular ratio of 1 to 1. This compound was hydrolyzed to berberine and baicalin by ${\beta}-glucuronidase$. The rate of hydrolysis was higher at higher temperature up to $50^{\circ}C$ and higher concentration of ${\beta}-glucuronidase$ up to 2500 unit under our experimental conditions. Baicalein, which is more liphophilic than baicalin, showed greater absorption in small intestine than baicalin did. The plasma concentrations of berberine and baicalein after oral administration of HOP were significantly higer than those of COP. The possible mechanism of increased bioavailibility of berberine and baicalein could be the hydrolysis of COP by ${\beta}-glucuronidase$. On the basis of the above results, it might be said that HOP should be a suitable preparation for increasing the bioavailibility of Coptidis Rhizoma and Scutellariae Radix.

Effects of thermoforming on the physical and mechanical properties of thermoplastic materials for transparent orthodontic aligners

  • Objective: The aim of this systematic multiscale analysis was to evaluate the effects of thermoforming on the physical and mechanical properties of thermoplastic materials used to fabricate transparent orthodontic aligners (TOAs). Methods: Specimens were fabricated using four types of thermoplastic materials with different thicknesses under a thermal vacuum. Transparency, water absorption and solubility, surface hardness, and the results of three-point bending and tensile tests were evaluated before and after thermoforming. Data were analyzed using one-way analysis of variance and Student's t-test. Results: After thermoforming, the transparency of Duran and Essix A+ decreased, while the water absorption ability of all materials; the water solubility of Duran, Essix A+, and Essix ACE; and the surface hardness of Duran and Essix A+ increased. The flexure modulus for the 0.5-mm-thick Duran, Essix A+, and eCligner specimens increased, whereas that for the 0.75-/1.0-mm-thick Duran and eClginer specimens decreased. In addition, the elastic modulus increased for the 0.5-mm-thick Essix A+ specimens and decreased for the 0.75-mm-thick Duran and Essix ACE and the 1.0-mm-thick Essix ACE specimens. Conclusions: Our findings suggest that the physical and mechanical properties of thermoplastic materials used for the fabrication of TOAs should be evaluated after thermoforming in order to characterize their properties for clinical application.

토양내 총 NAPL과 공기접촉 NAPL의 측정을 위한 분별 NAPL 분배 추적자 기술의 개발

  • Gaseous partitioning tracer test has been used for determining the volume and spatial distribution of residual non-aqueous phase liquid (NAPL) in the unsaturated soils. In this study, an experimental method for measuring the content of gas-exposed NAPL as well as that of total NAPL in a sand during air sparging was developed. Two different gaseous phase NAPL-partitioning tracers were used; n-pentane, with very low water solubility, was used as the tracer that partitions into NAPL that is only in contact with the mobile gas, and chloroform, with fairly good water solubility, was selected for detecting total NAPL content in the sand. Helium and difluromethanewere used as the non- reactive tracer and water-partitioning tracers, respectively. Using n-decane as a model NAPL (NAPL saturation of 0.018), 25.6% of total NAPL was detected by n-pentane at the water saturation of 0.68. Oniy 9.1% of total NAPL was detected by n-pentane at the water saturation of 0.84. This result implies that the quantity of gas-exposed NAPL increased about three times when the water saturation decreased from 0.84 to 0.68. At the water saturation of 0.68, more than 90% of total NAPL was detected by chloroform while 65.8% of total NAPL was detected by chloroform at the water saturation of 0.84. Considering that the removal rate of NAPL during air sparging for NAPL-contaminated aquifer is expected to be greatly dependent upon the spatial arrangement of NAPL phase with respect to the mobile gas, this new approach may provide useful information for investigating the mass transfer process during air-driven remedial processes fer NAPL-contaminated subsurface environment.

Effect of Sodium Bicarbonate Injection in Pre-rigor Porcine M. Longissimus lumborum on Pork Quality

  • Effects of sodium bicarbonate (SBC) injection on meat quality and functionality of porcine M. longissimus lumborum were investigated. Fifteen pigs ($100{\pm}5kg$) were randomly selected at a commercial slaughter plant. After slaughtering the loins were dissected from the carcass before chilling at approximately 30 minutes post mortem. The loins were divided into four cuts for sample treatment, and SBC of 0.25 M, 0.40 M and 0.75 M was injected (2% w/w) using a syringe. As SBC injection level was increased, muscle pH increased significantly (p<0.05). SBC injection decreased lightness ($L^*$) values on the surface of muscle. Moreover, with injection of SBC, drip loss %, cooking loss % and shear force were significantly (p<0.05) decreased, whereas WHC and $Na^+$ content were significantly (p<0.05) increased. From panel testing of uncooked pork loin, no significant differences (p>0.05) were found in aroma, off-flavor and drip between injection of SBC at different levels and the control, although color and acceptability were significantly lower (p<0.05) in control pork loin compare with injection of SBC at 0.75 M. In cooked pork loin from the panel test, aroma, flavour, off-flavour and juiciness were found to be similar (p>0.05) on all treatments, but tenderness and acceptability were significantly higher (p<0.05) with injection of SBC at 0.75 M than for control loin. Myofibrillar protein solubility of muscles treated with SBC was significantly (p<0.05) higher than that of the control, although no significant differences (p>0.05) were found in sarcoplasmic protein solubility between the treatments. These results indicated that SBC injection into pre-rigor porcine M. longissimus lumborum could improve ultimate pork quality characteristics such as meat color, water-holding capacity, and could inhibit muscle protein denaturation due to an increase in muscle pH.

A Study on the Characteristic of Gas Pore Formation in Lost Foam Casting of Mg alloy;Comparison with Al alloy (마그네슘 합금의 소실모형주조 시 기포형성 특성에 관한 연구;알루미늄 합금과 비교)

  • The pore formation characteristic of Mg alloy during Lost Foam Casting(LFC) was investigated with reduced pressure test and real casting, which was compared with the results of previous work for Al alloy. Cast Mg alloys in LFC had much lower porosities in comparison with those of Al alloys. Also, the proper pouring temperature gave the minimum porosity like Al alloy although it was higher than that of Al alloys due to the worse fluidity of Mg alloy. The pore formation mechanism of Mg alloy in LFC was similar to that of AI alloy but the critical temperature showing the different mechanism is higher than that of Al alloy as much as $30{\sim}50^{\circ}C$. The result that Mg alloy in LFC had the lower porosity comparing with Al alloy was due to the extra solubility of hydrogen gas although the solubility of Al alloy was easily exceeded by the external sources like pyrolyzed polystyrene products. The mold evacuation gave the lower porosity due to the removal of polystyrene pyrolysis products, and reduced shrinkage defects. Also, there was a proper evacuation pressure that gave a porosity of almost 0vol%. But much higher vacuum degree than this proper pressure caused the severe entrapment of polymer pyrolysis products that gave the large porosity.

  • This study was designed to evaluate the influences of evaporation of liquid of dental cements by drying during long term using. Zinc phosphate cement, polycarboxylate cement, and glass ionomer cement were used, and evaluated the properties as follows; consistency, setting time, film thickness, solubility, and compressive strength according to the ADA specification. The specimens of control group were made by mixing the newly opened liquid using the powder-liquid ratio recommended by the manufacturer, and the specimens of ES groups were made by mixing the 10% evaporated liquid by drying with the powder-liquid ratio recommeded by the manufacturer, and the specimens of EM group were made by mixing the 10% evaporated liquid with the powder-liquid ratio modified for standard consistency. The following conclusions were drawn ; 1. The viscosity of mixture of all kinds of cements were increased by the evaporation of liquid, especially the viscosity of glass ionomer cement were influenced significantly. 2. The amount of liquid should be increased to get a standard consistency at the using of evaporated liquid of cement, the most significant increase of liquid amount was required on Ketac-Cem. 3. The setting times were increased at both cases of mixing of evaporated liquid with powder - liquid ratio recommended by manufacturer or modifided through consistency test. 4. At an experimental group of mixing of the evaporated liquid with powder-liquid ratio recommended by manufacturer, solubility was decreased and film thickness was increased. 5. By the result of evaporation of cement liquid, the compressive strength of polycarboxylate cement was increased slightly and it of glass ionomer cement was increased, however, by the increase of amount of liquid to be possible to manipulate the compressive strength were decreased.

Synthesis of Dodecyl Phenol Novolac Epoxy Resin and Physical Properties of Coatings (Dodecyl phenol novolac 에폭시수지의 합성과 도막물성)

  • In the paper, mild solvent soluble alkyl group modified epoxy resins were prepared via a three-step method; (1) the condensation reaction of dodecyl phenol (DP) and formaldehyde, (2) the crosslinking reaction of dodecyl phnol novolac compound (DPC) and bisphenol A diglycidyl ether, (3) the dodecyl phenol novolac epoxy resins containing fatty acid (DPFA) was prepared by introducing fatty acid to DPC. Equivalent ratios of DP and formaldehyde were 1.25~1.333/1.0. Equivalent ratio of DPC and bisphenol A diglycidyl ether (YD-128) was 1.0/2.0. Reactivity, viscosity, molecular weight, solvent solubility, and physical properties of DPFA were investigated. The result show that as the number of aromatic ring of DPFA increased, viscosity increased and solvent solubility improved. When we test the properties of coatings by blending the synthesized DPFA with a white pigment, DPFAC-5 using triphenylphosphine (TPP) as a ring-open catalyst showed optical performance for drying time, adhesion, hardness, impact resistance, acid resistance and storage stability.
