Medium optimization for keratinase production by a local Streptomyces sp. NRC 13S under solid state fermentation

  • Thirteen different Streptomyces isolates were evaluated for their ability to produce keratinase using chicken feather as a sole carbon and nitrogen sources under solid state fermentation (SSF). Streptomyces sp. NRC 13S produced the highest keratinase activity [1,792 U/g fermented substrate (fs)]. The phenotypic characterization and analysis of 16S rDNA sequencing of the isolate were studied. Optimization of SSF medium for keratinase production by the local isolate, Streptomyces sp. NRC13S, was carried out using the one-variable-at-a-time and the statistical approaches. In the first optimization step, the effect of incubation period, initial moisture content, initial pH value of the fermentation medium, and supplementation of some agro-industrial by-products on keratinase production were evaluated. The strain produced about 2,310 U/gfs when it grew on chicken feather with moisture content of 75% (w/w), feather: fodder yeast ratio of 70:30 (w/w), and initial pH 7 using phosphate buffer after 8 days. Based on these results, the Box-Behnken design and response surface methodology were applied to find out the optimal conditions for the enzyme production. The corresponding maximal production of keratinase was about 2,569.38 U/gfs.

  • This study determined the level of fermented soybean meal (FSM) that could be substituted for fish meal in the diet for parrot fish Oplegnathus fasciatus. Fish meal (FM) was used the sole protein source in the control diet. FSM was substituted for 20, 40, 60, 80, and 100% of the fish meal in the experimental diets. The FSM resulted in increased crude protein, crude fat, and crude ash, but decreased crude fiber. The methionine and tryptophan contents in the FSM increased, while lysine and threonine decreased. The 18:2n-6 fatty acid content decreased from 55.30% to 28.67%. Fermentation increased 20:5n-3 (EPA) and 22:6n-3 (DHA) by 2.03% and 15.54%, respectively, although the differences were not significant. Based on growth performance, we concluded that FSM could replace up to 60% of FM for the maximum growth of juvenile parrot fish.

Limit load equations for partially restrained RC slabs

  • The expertise required in the judicious use of nonlinear finite element (FE) packages for design-assistance purposes is not widely available to the average engineer, whose sole aim may be to obtain an estimate for a single design parameter, such as the limit load capacity of a structure. Such a parameter may be required for the design of a proposed reinforced concrete (RC) floor slab or bridge deck with a given set of geometrical and material details. This paper outlines a procedure for developing design-assistance equations for carrying out such predictions for partially restrained RC slabs under uniformly distributed loading condition, based on a database of FE results previously generated from a large number of 'numerical model' slabs. The developed equations have been used for predicting the peak loads of a number of experimental RC slabs having varying degrees of edge restraints; with results showing a reasonable degree of accuracy and low level of scatter. The simplicity of the equations makes them attractive and their successful use in the field of application reported in this paper suggest that the outlined procedure may also be extended to other classes of concrete structures.

  • 본 연구에서는 선박의 안전항해의 관점에서 IMO조종성기준 중 변침 및 보침 성능에 관해 검토하였다. 침로불안정의 정도가 각각 다른 세척의 시리즈선박을 채택하여 지그재그시험에 관한 수치시뮬레이션을 수행하여 침로불안정의 정도와 오버슈트각 사이의 관계를 규명하였다. 그 결과 오버슈트각은 변침 및 보침 성능의 기준이 될 수 있음을 확인하였다. 그리고 자체개발한 시뮬레이터를 활용하여 시리즈선박의 항로항행에 관한 실시간 시뮬레이션 실험을 수행하여 선박의 조종난이도와 침로불안정의 정도 사이의 관계를 규명하였다. 아울러 수치시뮬레이션과 시뮬레이션실험 결과를 토대로 IMO조종성기준의 변침 및 보침 성능에 관한 새로운 안을 제시하였다.

Investigating vibrational behavior of graphene sheets under linearly varying in-plane bending load based on the nonlocal strain gradient theory

  • A study that primarily focuses on nonlocal strain gradient plate model for the sole purpose of vibration examination, for graphene sheets under linearly variable in-plane mechanical loads. To study a better or more precise examination on graphene sheets, a new advance model was conducted which carries two scale parameters that happen to be related to the nonlocal as well as the strain gradient influences. Through the usage of two-variable shear deformation plate approach, that does not require the inclusion of shear correction factors, the graphene sheet is designed. Based on Hamilton's principle, fundamental expressions in regard to a nonlocal strain gradient graphene sheet on elastic half-space is originated. A Galerkin's technique is applied to resolve the fundamental expressions for distinct boundary conditions. Influence of distinct factors which can be in-plane loading, length scale parameter, load factor, elastic foundation, boundary conditions, and nonlocal parameter on vibration properties of the graphene sheets then undergo investigation.

Protection of Polaromonas naphthalenivorans CJ2 from Naphthalene Toxicity by Extracellular Polysaccharide Capsules

  • Polaromonas naphthalenivorans CJ2, responsible for naphthalene degradation at a coal tar contaminated site, was isolated on MSB agar media supplied with naphthalene vapor as the sole carbon source at $10^{\circ}C$. The strain is not isolated under the same isolation condition using the same soil sediment at $20^{\circ}C$ although its optimum temperature is about $20^{\circ}C$. In this work we explored the reason why strain CJ2 could not have been isolated on MSB agar with naphthalene vapor at $20^{\circ}C$. Dispersed CJ2 cells in PBS buffer formed colonies on MSB agar with naphthalene vapor at $10^{\circ}C$ with low naphthalene vapor pressure, but not at $20^{\circ}C$ with high naphthalene vapor pressure. However, streaked cells without resuspension grew on MSB agar with naphthalene vapor at $10^{\circ}C,\;20^{\circ}C$, and even $25^{\circ}C$. Investigation of scanning electron microscopy showed that CJ2 cells formed extracellular polysaccharide (EPS) capsules, which were released easily from CJ2 cells by just dispersion. Therefore, it is concluded that strain CJ2 is able to overcome the naphthalene toxicity by forming a capsule-type barrier around the cells although it is susceptible to naphthalene toxicity at high temperature.

A Lipopeptide Biosurfactant Produced by Bacillus subtilis C9 Selected through the Oil Film-collapsing Assay

  • Bacillus subtilis C9 was selected by measuring the oil film-collapsing activity and produced biosurfactant in a medium containing glucose as a sole carbon source. The biosurfactant emulsified hydrocarbons, vegetable oils and crude oil, and lowered the surface tension of culture broth to 28 dyne/cm. A biosurfactant, C9-BS produced by B. subtilis C9 was purified by ultrafiltration, extraction with chloroform and methanol, adsorption chromatography, and preparative reversed phase HPLC. Structural analyses, IR spectroscopy, FAB mass spectroscopy, amino acid composition, and NMR analyses, demonstrated that C9-BS was a lipopeptide comprising a fatty acid tail and peptide moiety. The lipophilic part consisting of $C_{14}\;or\;C_{15}$ hydroxy fatty acid was linked to the hydrophilic peptide part, which contained seven amino acids (Glu-Leu-Leu-Val-Asp-Leu-Leu) with a lactone linkage.

Purification, Characterization, and cDNA Cloning of Xylanase from Fungus Trichoderma Strain SY

  • A xylanase-producing Trichoderma strain was isolated from soil. Xylanase from Trichoderma strain SY was purified 21-fold to an apparent homogeneity, with a $17.4\%$ yield. The optimum pH and temperature were determined to be 5.5 and $50^{\circ}C$, respectively, and its molecular weight was 21-kDa by SDS-PAGE. The corresponding gene, named xyl, was cloned by RT-PCR. DNA blot analysis of xyl showed that this gene is present as a single copy. The amino acid sequence of the Xyl protein showed similarity to those of other xylanases derived from various fungi. mRNA of xyl was highly expressed when this fungus was grown on cellulose or xylan as a sole carbon source, but undetectable when grown on sucrose. Extracts of Escherichia coli cells expressing xyl were found to have xylanase activity. It was confirmed that xyl from this isolate encodes xylanase.

Nitrogen Removal from Wastewaters by Microalgae Without Consuming Organic Carbon Sources

  • The possibility of microalgal nitrogen treatment was tested in wastewaters with a low carbon/nitrogen (C/N) ratio. Chlorella kessleri was cultured in the two different artificial wastewaters with nitrate as a nitrogen source: one contained glucose for an organic carbon source and the other without organic carbon sources. The growth rates of the two cultures were almost identical when the aeration rate was over 1 vvm. These results suggest that microalgae could successfully remove nitrogen from wastewater, as far as the mass transfer of $CO_2$, was not limited. Nitrate was successfully reduced to below 2 mg $NO_3^-$-N/ml from the initial nitrate concentration of 140 mg $NO_3^-$-N/ml in 10 days, even in the wastewater with no organic carbon source. Similar results were obtained when ammonium was used as the sole nitrogen source instead of nitrate. Higher concentrations of nitrogen of 140, 280, 560 and 1,400 mg/ml were also tested and similar amounts of nitrogen were removed by algal cultures without showing any substrate inhibition.

Microbial Basis for Enhanced Degradation of the Fumigant 1,3-Dichloropropene (1,3-D) in Soil

  • The differential enhanced degradation of cis- and trans-1,3-D was observed in the previous two studies performed by Ou et al. (1995) and especially Chung et al. (1999). This study was initiated to investigate the involvement of microorganisms in the differential enhanced degradation of the chemicals. As expected, microorganisms were responsible for the enhanced degradation of the chemicals. A mixed bacterial culture capable of degrading 1,3-D was isolated from an enhanced soil sample collected from a site treated with 1,3-D. Similar to the enhanced soil, the mixed culture degraded trans-1,3-D faster than cis-1,3-D. This mixed culture could not utilize cis- and trans-1,3-D as a sole source of carbon for growth. Rather, a variety of second substrates were evaluated to stimulate the differential enhanced degradation of the two isomers. As a result, the mixed culture degraded cis- and trans-1,3-D only in the presence of a suitable second substrate. Second substrates that had the capacity to stimulate the degradation included soil leachate, tryptone, tryptophan, and alanine. Other substrates tested, including soil extract, glucose, yeast extract, and indole (ailed to stimulate the degradation of the two isomers. Therefore, it appeared that the degradation of cis- and trans-1,3-D was a cometabolic process. The mixed culture was composed of four morphologically distinctive bacterial colonies.
