Pseudomonas putida FW에 의한 Aniline의 생물학적 분해 (Biodegradation of Aniline by Pseudomonas putida FW)

  • Aniline을 유일한 탄소원으로 한 최소배지에서 성장할 수 있는 균주를 활성오니로부터 분리하여 여러가지 특성을 조사하였다. 이 균주는 분류학적인 주성에 의하여 P. Putida biotype A로 동정되었으며 aniline을 유일한 탄소원으로 하여 배양하였을 때 10-20mM의 농도에서 최적의 성장을 나타내었으며 배양하는 동안에 pH의 변화는 일어나지 않았다. 이균주를 10mM의 aniline을 유일한 탄소원으로 하여 48시간 배양한 후 UV scanning spectrum, TLC, NMR을 이용하여 분석한 결과 aniline의 생분해에 의한 대사물질이 생성되는 것으로 추정되었다. 또한 이 균주는 streptomycin, trimethoprim, tetracycline, sulfanilamide에 강한 저항성을 나타내었으며 plasmid를 1개 가지고 있는 것으로 나타났다. Mitomycin C curing을 통하여 얻어진 여러개의 변이균주의 성질을 조사하여 본 결과 이 균주의 plasmid DNA는 aniline의 분해에 관여하는 것으로 추정되었다.

Sole Proprietorship Business Succession in Malaysia: A Perspective of Civil and Islamic Law

  • Issues relating to business inheritance are a primary focus for business owners. Business inheritance is critical to ensuring that the business can be managed by the next generation and family members can benefit from the earnings and revenues. Therefore, preliminary planning must be done while the business owner is still alive by selecting the appropriate business inheritance mechanism for Muslims in Malaysia. The mechanisms of business inheritance, notably faraid, hibah, amanah, and hibah amanah will be examined in this paper from both a civil and Islamic perspective. By reviewing written journals and previous research, this research utilizes the library method. According to this study, business succession can be carried out by naming a beneficiary as a partner in the company or by completing the ownership transfer process while the owner is still living. In the other situation, it can be done by anyone among the heirs agreeing to re-register the business as usual. Meanwhile, there are four main instruments in Islamic law that can be used: faraid, hibah, amanah, and commercial hibah. According to this study, each instrument has a different impact on business succession. Entrepreneurs should use these instruments in their firm succession planning.

  • As the first part of the body that strikes the ground during running, sports shoes are especially important for improving performance and reducing injuries. The use of new nanotechnology materials in the shoe's sole that can affect the movement angle of the foot and the ground reaction forces during running has not been reported yet. It is important to consider the material of the sole of the shoe since it determines the long-term performance of sports shoes, including their comfort while walking, running, and jumping. Running performance can be improved by polymer foam that provides good support with low energy dissipation (low energy dissipation). Running shoes have a midsole made of ethylene propylene copolymer (EPP) foam. The mechanical properties of EPP foam are, however, low. To improve the mechanical performance of EPP, conventional mineral fillers are commonly used, but these fillers sacrifice energy return. In this study, to improve the magnificence of physical education training with nanotechnology, carbon nanotubes (CNTs) derived from recycled plastics were prepared by catalytic chemical vapor deposition and used as nucleating and reinforcing agents. As a result of the results, the physical, mechanical, and dynamic response properties of EPP foam combined with CNT and zinc oxide nanoparticles were significantly improved. When CNT was added to the nanocomposites with a weight percentage of less than 0.5 wt%, the wear resistance, physical properties, dynamic stiffness, compressive strength, and rebound properties of EPP foams were significantly improved.

  • Pseudomonas putida BM01 was grown efficiently on glucose as the sole carbon source with a supply of a nitrogen source in pH-stat mode using a low setpoint limit. A final cell concentration of 100 g/l was obtained in 30 h of fed-batch cultivation by controlling glucose concentration within the range of 5-20 g/l and maintaining dissolved oxygen tension above 10$%$ saturation using pure oxygen. This high cell density culture technique is believed highly useful for the production of poly(3-hydroxyalkanoates) by this strain.

Chemokine Receptors in HIV-1 and SIV Infection

  • Seven transmembrane segment (7TMS) receptors for chemokines and related molecules have been demonstrated to be essential, in addition to CD4, for HIV and SIV infection. The beta-chemokine receptor CCR5 is the primary, perhaps sole, coreceptor for HIV-1 during the early and chronic phases of infection, and supports infection by most primary HIV-1 and many SIV isolates. Late-stage primary and laboratory-adapted HIV-1, HIV-2, and SIV isolates can use other 7TMS receptors. CXCR4 appears especially important in late-stage HIV infection; several related receptors can also be used. The specificity of SIV viruses is similar. Commonalities among these receptors, combined with analyses of mutated molecules, indicate that discrete, conformationally-depenclent sites on the chemokine receptors determine their association with the third variable and conserved regions of viral envelope glycoproteins. These studies are useful for elucidating the mechanism and molecular determinants of HIV-1 entry, and of inhibitors to that entry.

Degradation of Anthracene by a Pseudomonas strain, NGK1

  • Pseudomonas sp. NGK1, isolated by naphthalene enrichment culture technique, is capable of degrading anthracene as a sole source of carbon and energy. The organism degraded anthracene through the intermediate formation of 1,2-dihydroxyanthracene, 2-hydroxy-3-naphthoic acid, salicylate, and catechol. The intermediates were isolated and characterized by TLC, spectrophotometry, and HPLC analysis. The cell free extract of anthracene-grown cells showed activities of anthracene dioxygenase, 2-hydroxy-3-naphthylaldehyde dehydrogenae, 2-hydroxy-3-naphthoate hydroxylase, salicylate hydroxylase and catechol 2,3-dioxygenase. The formed catechol as a metabolite is degraded through meta-cleavage with the formation of ${\alpha}$-hydroxymuconic semi-aldehyde.

Roles of Glucose and Acetate as Carbon Sources in L-Histidine Production with Brevibacterium flavum FERM1564 Revealed by Metabolic Flux Analysis

  • The metabolic flux pattern for L-histidine production was analyzed when glucose and/or acetate were used as carbon sources. Total L-histidine production was enhanced when mixed substrate (glucose and acetate) was used, compared wish that when either glucose or acetate was used as the sole carbon source. Theoretical maximum carbon fluxes through the main pathways for L-histldine production, cell growth, and ATP consumption for cell maintenance were obtained by the linear programming (LP) method. By comparison of the theoretical maximum carbon fluxes tilth actual ones, it was found that a large amount of glucose was actually used for maintenance of cell viability. On the other hand, acetate was used for cell growth. After depletion of acetate in the mixed substrate culture, the flux for glucose to L-histldine synthesis was markedly enhanced. A strategy for effective L-histidine production using both carbon sources was proposed.

Isolation of Polyacrylamide-degrading Microorganisms from Soil

  • Two polyacrylamide degrading bacterial strains, No. 2 and No. 11, were isolated from soil, and identified as Bacillus sphaericus No.2 and Acinetobacter sp. No. 11, respectively. Both strains grew on medium containing polyacrylamide as sole carbon and nitrogen sources. B. sphaericus No. 2 and A. sp. No. 11 reduced by 16% and 19%of the initial polyacrylamide concentration, respectively. Optimal pH and temperature in growth of Acinetobacter sp. No. 11 were 8.0 and $37^{\circ}C$, respectively. After 14-day cultivation of A. sp. No. 11, the average molecular weight of polyacrylamide has been shifted from $2.3{\times}10^6\;to\;0.5{\time}106$.

  • A transposon Tn5 mutant of Rhodobacter sphaeroides 2.4.1 was isolated for its impaired ability of growth on minimal medium containing ${\beta}$-hydroxybutyric acid as a sole carbon source. The mutant, R. sphaeroides S7 showed approximately 6-fold decrease in ${\beta}$-hydroxybutyrate dehydrogenase activity compared with that of wild type. In R. sphaeroides S7 the Tn5 was located in DNA region corresponding to a 4.2-kb EcoRI DNA fragment of R. sphaeroides 2.4.1 chromosome.

Optimal Resolution of L-Carnitine from Racemic DL-Carnitine by Enterobacter sp. Assimilating D-Carnitine

  • In order to isolate a microorganism having preferential degradation of D-carnitine from DL-carnitine, a bacterium assimilating D-carnitine as a sole carbon and energy source was isolated from soil by enrichment culture and partially identified as Enterobacter sp. Also, a mutant having lessened L-carnitine decomposition rates was selected with nitrosoguanidine mutagenesis, which led to decrease the specific activities of carnitine dehydrogenase (7.6-fold) and ${\beta}$-hydroxybutyrate dehydrogenase (9.5-fold) as compared to the wild strain. Meanwhile, optimal culture conditions for optical resolution of DL-carnitine were investigated. Under optimal conditions, 3.53 g/l L-carnitine was obtained from 20 g/l DL-carnitine, which corresponded to 35.3% L-carnitine yield and 97.9% optical purity.
