This study aims to confirm the possibility of using the invasive alien plants in Jeju as a functional biomaterial. To achieve this purpose, 70% ethanol extract and solvent fractions were prepared for five invasive alien plants (Hypochaeris radicata, Rumex acetosella, Humulus japonicus Siebold & Zucc., Solanum viarum, Lactuca scariolar) and their antioxidant, antibacterial anti-inflammatory and anti-obesity effects were investigated. The DPPH radical scavenging activity of ethanol extract from invasive alien plants was shown in the order of Rumex acetosella > Hypochaeris radicata > Humulus japonicus. Antimicrobial activity of ethanol extract against food poisoning bacteria (4 species) and oral cavity-induced microorganisms (6 species) was measured. As a result, the extract of Humulus japonicus showed high antibacterial effects against food poisoning bacteria (E. coli, V. parahaemolyticus) and oral microbes (L. casei, S. epidermidis, E. faecalis). In LPS-induced RAW 264.7 cells, the anti-inflammatory effect of ethanol extract from invasive alien plants was investigated. As a result, the NO production inhibition activity was highest in the Rumex acetosella and the Humulus japonicus Siebold & Zucc. ethanol extract, and the NO production inhibition activity was concentration-dependent. In addition, the Rumex acetosella and the Humulus japonicus Siebold & Zucc. ethanol extract showed a concentration-dependent inhibitory effect on cytokine (IL-6) production. These extracts also showed inhibitory activity of COX-2, an inflammatory protein. This suggests that NO production inhibition activity by the extract of invasive alien plants is the result of inhibition of iNOS and COX-2 expression. Currently, organic solvent fractions of crude extract are manufactured and the investigation of active ingredients is continuing along with evaluation of biological activity such as anti-inflammatory. These results are expected to be a major data for the study on the separation and utilization of active ingredients with antioxidant, antibacterial and anti-inflammatory effects using foreign plant crude extract and solvent fractions, and are highly likely to be applied to the development of functional food and cosmetics materials.
This study was conducted to obtain the basic information on saline tolerance of plants. Fourteen plant species were grown under different salinity adjusted with NaCl. After 14 days treatment, physiological changes were investigated. Proline contents of tested plants were increased as salinity was increased. Acalvpha australis, Bidens bipinnata, Erechitites hieracifolia, Erigeron canadensis, Punicum dichotomiflorum, and Solanum nigrum showed drastic increase of proline contents in 200mM NaCl treatment. But Atriplex gmelini, Suaeda asparagoldes did not show drastic increase. As the NaCl concentration increased to 200mM in media, both contents of $Na^+$ and $Cl^-$ were increased. Acalypha australis, Digitaria sanguinalis, Erechitites hieracifolia, and Solanum nigrum showed drastic increase in $Na^+$ and $Cl^-$ contents under 200mM NaCl. But Suaeda asparagoides. Atriplex gmelini, and Spergularia marina were slightly increased. As salinity increased, the $K^+/Na^+$ ratio of halophytes, such as Suaeda asparagoides, Atriplex gmelini, and Spergularia marina were more slightly decreased than glycophytes.
Alfalfa mosaic alfamoviruses(AIMV) were isolated from infected potato (Solanum tuberosum) and azuki bean (Paseolus angularis) in Korea. Two AIMV isolated from potatoes were named as strain KR (AIMV-KR1 and KR2) and AIMV isolated from azuki bean was named as strain Az (AIMV-Az). Each isolated AIMV strain was characterized by using their host ranges, symptom developments, serological relations and nucleotide sequence analysis of coat protein (CP) gene. Strains KR1, KR2, and Az were readily transmitted to 20 of 22 inoculated plant species including bean, cowpea, tomato, tobacco, and potato. AIMV-KR1 and KR2 produced the typical symptoms like chlorotic or necrotic spots in Chenopodium quinoa and Solanum tuberosum cv. Superior. AIMV-Az caused bright yellow mosaic symptom and leaf malformation in Nicotiana glauca, which were different from the common mosaic symptom caused by AIMV-KR1 and KR2. Electron microscope observation of purified virus showed bacilliform virions containing a single-stranded plus-strand RNAs of 3.6, 2.6, 2.0 and 0.9 kbp in length, respectively, similar in size and appearance to those of Alfamovirus. In SDS-PAGE, the coat protein of the two viruses formed a consistent band that estimated to be about 24kDa. The CP genes of the AIMV strains, KR1, KR2, and Az have been amplified by RT-PCR using the specific primers designed to amplify CP gene from viral RNA-3, cloned and sequenced. Computer aided analysis of the amplified cDNA fragment sequence revealed the presence of a single open reading frame capable of encoding 221 amino acids. The nucleotide and peptide sequence of viral CP gene showed that strain KR1, KR2, and Az shared highest nucleotide sequence identities with AIMV strain 425-M at 97.7%, 98.2%, and 97.2%, respectively. CP gene sequences of two strains were almost identical compared with each other. Altogether, physical, serological, biological and molecular properties of the purified virus.
$Tomato$$spotted$$wilt$$virus$ (TSWV)-infected $Capsicum$$annuum$ plants were collected from open fields during June to July in 2010. The TSWV isolates were designated as Gneung, Ghang-Kjj, Gchang-Njc, Ghae, and Pap. The nucleotide sequence of the nucleocapsid protein (N) and movement protein (NSm) of the five isolates was determined. The pathogenicity of the five isolates was determined on 14 $C.$$annuum$ cultivars two times by using mechanical inoculation. The five isolates induced different response: Both Gneung and Gchang-Kjj did not infect any of the cultivars in the 2nd trial, while Gchang-Njc, Ghae and Pap infected 11, 6 and 13 of 14 cultivars, respectively. The five isolates also were tested on $Solanum$$lycopersicum$ breeding line TGC09-71 and three $Nicotiana$ species. $S.$$lycopersicum$ showed a similar response to the five isolates as did $C.$$annuum$. Both Gchang-Njc and Ghae infected systemically all three $Nicotiana$ species tested. While both Pap and Gneung did not infect any of the $Nicotiana$ species tested. In conclusion, five TSWV isolates induced different infection spectra in $C.$$annuum$ cultivars, $Nicotiana$ species and an $S.$$lycopersicum$ breeding line. Amino acid sequence analysis of the NSm gene could not support or explain the different infection spectra of the five isolates. This study indicated that various isolates must be used as virus inocula for evaluation of $C.$$annuum$ and $S.$$lycopersicum$ cultivars in breeding programs for TSWV resistance.
This study was carried out focused an ecological characteristics and habitat management of Abeliophyllum distichum subpopulations in the Byeonsanbando National Park, Korea. Among three subpopulations of the target species in the studied area, Gamaso region are known as translocation site due to the dam construction while the wild populations which is located at both the lower terrain of the Buandaem bank and Chongnim-ni, which is designated as Natural Monument No 370, is threat due to an intensive site management likes clearance of scrub amongst the Abeliophyllum plants. Vascular plant taxa in the studied sites were summarized as 182 taxa; 65 families, 143 genera, 164 species, 17 varieties and 1 form, and Gamaso area, the lower terrain of the Buandaem and Cheongnim-ni areas are summarized as 75, 102 and 89 taxa, respectively. The plant species likes persimmon tree, paulownia tree and wild mulberry tree are dominant species in the Gamaso subpopulation, while Quercus variabilis is dominant at the lower terrain of the Buandaem. The intensive site management was applied to the Cheongnim-ni subpopulation, one of the original wild habitats of the species. The population dymanics was varied; the one or two sprouting number of stems was over 87% both at Gamaso area and the lower terrain of the Buandam, while the Cheongnim-ni subpopulation recorded as 74.8%. Also three sprouting number recorded as 16% and as 9.2% for sprouting on numbers of four to eleven. The invaded species likes Chenopodium album, Rumex obtusifolius, Phytolacca americana, Pharbitis nil, Solanum lyratum, Erigeron canadensis and Erigeron annuus, were found at the wild habitat of the species, and those invaded species grow faster than Abeliophyllum plants. Among three habitats of the target species in the studied area, the habitat which in located at the lower terrain of the Buandam and Gamaso region are known as translocated due to the dam construction, and wild subpopulation at Chongnim-ni, which is designated as Natural Monument No 370, is under threatened due to an intensive site management likes clearance of scrub amongst the Abeliophyllumplants. It is one of the evidence that Cuscuta japonica at the population indicated an intensive site management likes clearance of scrub is not proper habitat management protocol. Thus, further researches should be followed to determine the effect of canopy removal and other disturbance factors as management techniques.
308 isolates of Ampelomyces quisqualis were isolated from powdery mildew fungi of 73 plant species in Korea from 1994 to 2004. Among them, the new mycohosts and new plant hosts of A. quisqualis were found in 13 species of powdery mildew fungi in 38 species of plant. The new hosts of A. quisqualis were Erysiphe heraclei on Heracleum moellenderfii; E. hommae on Elsholtzia splendins; E. glycines on Glycine max; E. lespedezae on Lespedeza biclor; E. pileae on Pilea mongolica; E. pisi on Phaseolus radiatus; E. polygoni on Rumex aquatica and Rumex crispus; Golovinomyce artemisiae on Artemisia princeps var. orientalis; G. cichoracearum on Rudbeckia laciniata var. hortensis; G. rubiae on Rubia akane; Oidium sp. on Valeriana fauriei, Lactuca indica var. laciniata, Carpesium triste var. manshuricum, Aster ageratoides var. turczaninow, Lufa cylindrica, Ixeris demtats Nakai, Phlox paniculata, Bidens bipinnata and Pathenocissus tricuspidata; Microsphaera alphitoides on Quercus aliema, M. pseudolonicerae on Cocculus trilobos; Podosphaera sp. on Ligustrum obtusifolium; Sphaerotheca aphanisi on Fragaria ananassa; S. balsaminae on Impatiens textori; S. fusca on Cucurbita pepo, Cucurbita maxima, Ligularia fischeri, Solanum melongena, Lagenaria leucantha, Cucumis melo var. makuwa, Acalypha australis, Cosmos bipinnatus and Aster scaber; Uncinuliella simulans on Rosa muliflora and Uncinula australiana on Lagerstroemia indica.
Injury caused by reactive oxygen species (ROS), known as oxidative stress, is one of the major damaging factors in plants exposed to environmental stress. Chloroplasts are specially sensitive to damage by ROS because electrons that escape from the photosynthetic electron transfer system are able to react with relatively high concentration of $O_2$ in chloroplasts. To cope with oxidative stress, plants have evolved an efficient ROS-scavenging enzymes such as superoxide dismutase (SOD) and ascorbate peroxidase (APX), and low molecular weight antioxidants including ascorbate, glutathione and phenolic compounds. To maintain the productivity of plants under the stress condition, it is possible to fortify the antioxidative mechanisms in the chloroplasts by manipulating the antioxidation genes. A powerful gene expression system with an appropriate promoter is key requisite for excellent stress-tolerant plants. We developed a strong oxidative stress-inducible peroxidase (SWPA2) promoter from cultured cells of sweetpotato (Ipomoea batatas) as an industrial platform technology to develop transgenic plants with enhanced tolerance to environmental stress. Recently, in order to develop transgenic sweetpotato (tv. Yulmi) and potato (Solanum tuberosum L. cv. Atlantic and Superior) plants with enhanced tolerance to multiple stress, the genes of both CuZnSOD and APX were expressed in chloroplasts under the control of an SWPA2 promoter (referred to SSA plants). As expected, SSA sweetpotato and potato plants showed enhanced tolerance to methyl viologen-mediated oxidative stress. In addition, SSA plants showed enhanced tolerance to multiple stresses such as temperature stress, drought and sulphur dioxide. Our results strongly suggested that the rational manipulation of antioxidative mechanism in chloroplasts will be applicable to the development of all plant species with enhanced tolerance to multiple environmental stresses to contribute in solving the global food and environmental problems in the 21st century.
Invasive alien species (IAS) can affect agricultural ecosystem negatively and critically. The geographical distributions of nine IAS plants in Korean orchards and uplands were investigated. Ambrosia artemisiifolia distributed widely in Korean peninsula and Ambrosia trifida distributed intensively in Gyeonggi-do. Aster pilosus distributed in Gyeonggi-Gangwon region and Lactuca scariola distributed linear region from the north-western to south-eastern part of Korea. Hypochaeris radicata distributed mainly in Jeju-do and Solanum carolinense distributed in eastern Gyeonggi-do. Rumex acetosella and Sicyos angulatus sporadically occurred in Korean croplands and Eupatorium rugosum was found in one region only. The distribution of IAS plants in croplands was very similar to that in non-croplands suggesting that ecological position of arable lands is not different from that of non-arable lands. Therefore, IAS plants in the croplands should be managed for not only prevention of yield reduction, but also protection or maintenance of original ecosystem. Development of management methods apposite to current Korean status are required to control IAS plants in croplands.
The studies were conducted to obtain the basic information of the effects of pollutants on plant species and to select the plant species showing specific responses to the pollutants. For these purposes, paraquat, ammonium, and cadmium as a source of oxidative stress, nitrogen toxicity, and heavy metal toxicity respectively were treated to the plant species. Among the tested plants, Lamiaceae, Brassicaceae, and Caryophyllaceae were tolerant to paraquat, whereas Poaceae and Asteraceae were sensitive. Especially Mosla dianthera of Lamiaceae, Hemistepta lyrata and Aster pilosus of Asteraceae, and Paspalum thunbergii of Poaceae showed higher tolerance than others. Paraquat resistance was related with life style, overwintering capacity, so perennial and biennial species showed higher tolerance than annual species. In response to ammonium, Poaceae showed higher resistance while Fabaceae and Caryophyllaceae showed sensitiveness. Weed species having tolerance to ammonium were Echinochloa crus-galli var. praticola, Panicum dichotomiflorum, Setaria glauca, Chenopodium album, and Solanum nigrum, while Mosla dianthera, Arenaria serpyllifolia and Perilla frutescens var. japonica showed sensitiveness. In the response of plant species to cadmium, Digitaria sanguinalis, Amaranthus lividus showed higher resistance, whereas Galinsoga parviflora, Plantago asiatica, Ambrosia trifida, and Paspalum thunbergii showed sensitiveness. The injured degree on germination stage by pollutants did not related with injured degree on matured stage. During germination, the root elongation was more sensitive than shoot elongation by pollutants, paraquat, ammonium, and cadmium.
Tomato spotted wilt virus (TSWV) was occurred on the three vegetables of egg plant (Solanum melongena), whole radish (Raphanus acanthiformis) and sugar loaf (Cichorium intybus) at Anyang area infested with TSWV. Whole radish was produced the symptoms of necrotic spots on the leaves, and necrosis and malformation on the roots by TSWV. Egg plant was induced the symptoms of typical multiple ring spots on the leaves and necrotic rings on the fruits. Sugar loaf was infected severely with the typical symptoms of ring spots on the leaves and stunt. The three isolates of TSWV could infect locally on the indicator plants of Chenopodium amaranticolor, C. quinoa and Nicotiana debney, and systemically on N. glutinosa, N. benthamiana and Datura stramonium. Two TSWV isolates from egg plant and sugar loaf were very similar in virulence. However, the virulence of TSWV from whole radish was very different as local infection on 5 Nicotiana species including N. tabacum 'Xanthi NC'.
