Inhibitory Substance Produced by Aspergillus sp. on the Snake Venom Proteinase - Isolation of Microorganism and Biological Activities of the Inhibitor - (Aspergillus 속 균주가 생성되는 사독 Proteinase에 대한 저해물질 - 균의 분리 및 저해물질의 생물학적 작용상 -)

  • Aspergillus sp. (MK-24) producing a biological active substance that inhibited the venom proteinase activity was isolated from soil. The substance also inhibited the activity of trypsin and coagulation of blood, but did not inhibit papain, $\alpha$-chymotrypsin and pepsin. The substance was partially purified from culture filtrate by precipitaion with acetone, and by chromatography of DEAE-Sepadex A-50 column and Amberlite IRC-50 ion exchange. The inhibitory substance was stable in the wide pH range from 2.0 to 12.0 at 37$^{\circ}C$, but not stable at $65^{\circ}C$ in the alkaline pH. Only 12% of the activity was decreased by the heat treatment at 10$0^{\circ}C$ for two hours. The inhibition on venom proteinase (Agkistrodon bromohoffi brevicaudus) was a mixed type. The inhibitory activity depended on the preincubation time and completely depressed by cupric, zinc and cobalt ions. The inhibition on the venom proteinase was appeared strongly on casein but not on ovalbumin or hemoglobin as a substrate.

A Study on the Selection of Vegetation and Method for the Revegetation on the Highway Slope in Consideration of the Surrounding Environment (고속도로(高速道路) 비탈면(面)에서 주변환경(周邊環境)을 고려(考慮)한 녹화공법(綠花工法)과 식생선발(植生選拔)에 관(關)한 연구(硏究))

  • This study was carried out from March 1995 to February 1996 to select suitable vegetation and measures for revegetation on the highway cut-slope in consideration of the surrounding environment in the middle region of Korea. The numbers of revegetation-measures type surveyed were eighteen measures as a whole in descending order of the Seed-spraying measures, Block-sod pitching measures, and Straw-mat mulching measures. The numbers of vegetation species by highway line were 243 in Chungbu, 223 in Jungang, 210 in Kyongbu 2, 199 in Kyongbu 1, 179 in Sohaean, 163 in Seoul circulation. 148 in Shingal-ansan, 141 in Second kyongin and 123 in Kyongin respectively. The highest index of species diversity(H') was in Kyongbu 1-line and the evenness(E') on upper site was greater than that of the lower site. The revegetation measures were selected in consideration of nature of soil, gradient, area and situation, and 83 species were selected as those for revegetation measures in highway cut-slope.

Classification and Characteristics of the Roadbed Plant Communities in Daegu, Korea (대구지역 노상식물군락의 분류와 분포 특성)

  • An important theme of this paper is to search for more genelalities of diversity and distribution pattern on the trampled plant communities in Daegu area. A total of 50 phytosociological releves were analyzed by the Zurich-Montpellier School's method and generalized habitat characteristics by using Euclidean coefficient and PCoA (Principal Coordinates Analysis). Five plant communities were classified and matched into largely two physiognomic types: grass-types such as Eragrostis ferruginea community, Eleusine indica community, Setaria viridis community, Digitaria ciliaris community, and forb-type of Plantage asiatica community only. These plant communities correspond to quite distinctive four habitat types, with both soil stability and moisture in microhabitat conditions: stable-moderate, unstable-moderate, stable-dry, and unstable-dry. Top-ten species showing the highest relative net contribution degree(r-NCD) were Digitaria ciliaris, Eleusine indica, Eragrostis ferruginea, Plantago asiatica, Echinochloa crus-galli, Trifolium repens, Polygonum aviculare, Setaria viridis, Setaria glauca and Artemisia princeps. The front three species showing more than 50% in percent r-NCD possess florescence after monsoon season. It is critically pointed out that the regional ecological differentiations i.e. the Daegu bioclimatic division, the $C_4$-plant dominant season(post-monsoon) of Korean peninsula, and species composition different from Japan's Plantaginetalia asiaticae, are the basis for deriving characteristics of the roadbed trampled plant communities of Daegu.

Estimations of Forest Growing Stocks in Small-area Level Considering Local Forest Characteristics (산림의 지역적 특성을 고려한 시군구 임목축적량 통계 산출 기법 개발)

  • Forest statistics of local administrative districts have many social needs, nevertheless we have some difficulties for working out an accurate statistics because of insufficient data in small-area level. Thus, new small-area estimation method has to set aside additional data, decrease errors of statistics and consider the local forest characteristics at the same time. In this study, we researched the spatial divisions that can set aside additional data for statistics production and satisfy the major premise, which is "forest characteristics of spatial divisions have to be equal to that of small-area". And we compared synthetic estimation methods based on three different spatial divisions(provinces, neighbor districts and new expanded districts). New expanded districts were divided based on the criteria of climate, soil type and tree species composition that affects local forest characteristics. Small-area statistics were assessed in terms of the ability to estimate local forest characteristics and consistency within large-area statistics. As a result, new expanded districts synthetic estimation was assessed to calculate statistics that reflects local forest characteristics better than other two estimation methods. Moreover, this synthetic estimation method produced the statistics that was included within 95% confidence interval of large-area statistics and was the closer to large-area statistics than the neighbor districts synthetic estimation.

Geotechnical Engineering Progress with the Incheon Bridge Project

  • Incheon Bridge, 18.4 km long sea-crossing bridge, will be opened to the traffic in October 2009 and this will be the new landmark of the gearing up north-east Asia as well as the largest & longest bridge of Korea. Incheon Bridge is the integrated set of several special featured bridges including a magnificent cable-stayed girder bridge which has a main span of 800 m width to cross the navigation channel in and out of the Port of Incheon. Incheon Bridge is making an epoch of long-span bridge designs thanks to the fully application of the AASHTO LRFD (load & resistance factor design) to both the superstructures and the substructures. A state-of-the-art of the geotechnologies which were applied to the Incheon Bridge construction project is introduced. The most Large-diameter drilled shafts were penetrated into the bedrock to support the colossal superstructures. The bearing capacity and deformational characteristics of the foundations were verified through the world's largest static pile load test. 8 full-scale pilot piles were tested in both offshore site and onshore area prior to the commencement of constructions. Compressible load beyond 30,000 tonf pressed a single 3 m diameter foundation pile by means of bi-directional loading method including the Osterberg cell techniques. Detailed site investigation to characterize the subsurface properties had been carried out. Geotextile tubes, tied sheet pile walls, and trestles were utilized to overcome the very large tidal difference between ebb and flow at the foreshore site. 44 circular-cell type dolphins surround the piers near the navigation channel to protect the bridge against the collision with aberrant vessels. Each dolphin structure consists of the flat sheet piled wall and infilled aggregates to absorb the collision impact. Geo-centrifugal tests were performed to evaluate the behavior of the dolphin in the seabed and to verify the numerical model for the design. Rip-rap embankments on the seabed are expected to prevent the scouring of the foundation. Prefabricated vertical drains, sand compaction piles, deep cement mixings, horizontal natural-fiber drains, and other subsidiary methods were used to improve the soft ground for the site of abutments, toll plazas, and access roads. Light-weight backfill using EPS blocks helps to reduce the earth pressure behind the abutment on the soft ground. Some kinds of reinforced earth like as MSE using geosynthetics were utilized for the ring wall of the abutment. Soil steel bridges made of corrugated steel plates and engineered backfills were constructed for the open-cut tunnel and the culvert. Diverse experiences of advanced designs and constructions from the Incheon Bridge project have been propagated by relevant engineers and it is strongly expected that significant achievements in geotechnical engineering through this project will contribute to the national development of the longspan bridge technologies remarkably.

Inversion of Rayleigh-wave Dispersion Curves for Near-surface Shear-wave Velocities in Chuncheon Area (춘천지역의 천부 횡파속도를 구하기 위한 레일리파 분산곡선 역산)

  • To evaluate methods of determining near-surface shear-wave velocities (${\nu}_s$), we derived dispersion curves of Rayleigh waves generated by both passive and active sources in Chuncheon, Korea. Microtremors were recorded for 5 minutes in each of four triangular arrays with radii of 5 ~ 40 m. Those data were analyzed using the Spatial Autocorrelation method. Rayleigh waves were also generated by a hammer source and recorded in the same area for 2 s using 24 4.5-Hz geophones. Multichannel Analysis of Surface Waves was applied to those data. Velocity spectra were derived with relatively high signal-to-noise ratios in the frequency ranges of 7 ~ 19 and 11 ~ 50 Hz for the microtremors and synthetically generated Rayleigh waves, respectively. The resultant dispersion curves were combined as one and then input to inversion to derive shear wave velocities that were compared with a lithology log from a nearby well. Shearwave velocities in the top soil and soft-rock layers are almost constant with values of 221 and 846 m/s, respectively; while the inverse-modeled ${\nu}_s$ increases linearly in the gravelly sand, cobbles, and weathered-rock layers. If rock type is classified based on shear-wave velocity, the inversion-derived boundary between weathered-rock and soft rock may be about 5 m deeper than in the well log.

Region of Interest (ROI) Selection of Land Cover Using SVM Cross Validation (SVM 교차검증을 활용한 토지피복 ROI 선정)

  • This study examines machine learning cross-validation to utilized create ROI for classification of land cover. The study area located in Sejong and one KOMPSAT-3A image was used in this analysis: procedure on October 28, 2019. We used four bands(Red, Green, Blue, Near infra-red) for learning cross validation process. In this study, we used K-fold method in cross validation and used SVM kernel type with cross validation result. In addition, we used 4 kernels of SVM(Linear, Polynomial, RBF, Sigmoid) for supervised classification land cover map using extracted ROI. During the cross validation process, 1,813 data extracted from 3,500 data, and the most of the building, road and grass class data were removed about 60% during cross validation process. Based on this, the supervised SVM linear technique showed the highest classification accuracy of 91.77% compared to other kernel methods. The grass' producer accuracy showed 79.43% and identified a large mis-classification in forests. Depending on the results of the study, extraction ROI using cross validation may be effective in forest, water and agriculture areas, but it is deemed necessary to improve the distinction of built-up, grass and bare-soil area.

Myxococcus xanthus socD500 mutation causes Sporulation and Induction of two C-signal Specific Genes (Myxococcus xanthus socD500에 의한 포자 형성 및 CsgA신호에 특이적 유전자의 발현에 관한 연구)

  • Myxococcus xanthus is a Gram negative, rod-shaped, soil bacterium that displays a social behaviors, and multicellular development upon nutrient deprivation. The csgA gene encoding a cell surface protein is essential for developmental behaviors including rippling, aggregation, fruiting body formation and sporulation. csgA mutants show normal vegetative growth, but lack all these developmental phenotypes. Expression of the CsgA (C-signal) specific genes are eliminated or dramatically reduced in csgA mutants. In order to identify components of C-signal transduction pathway, second site mutations were introduced into csgA mutants and were identified which can fully or partially restore development of csgA mutants (Rhie, H. G. et. al. 1989. J. Bacteriol. 171, 3268-3276). One of such csgA suppressor mutations, socD500 restores only sporulation to csgA mutants at 15$^{\circ}C$. The socD500 mutaion however eliminates the three basic developmental requirements, starvation, high cell density and a solid surface. Only sporulation, not accompanied with fruiting body formation is induced simply by shifting the temperature of vegetatively growing cells from $32^{\circ}C$ to $15^{\circ}C$. Spores induced by socD500 mutation is not as thick as that of wild-type fruiting body. In socD500 genetic background, two of ten C-signal dependent genes, $\Omega$DK4506 and $\Omega$DK4406 are more highly expressed in growing cells at $15^{\circ}C$. These results indicate that the socD500 mutation may be partly involved in the regulation of expression of two C-signal dependent genes and genes for sporulation in this transduction pathway.

Comparative Study of Cost Estimate System in Landscape Architectural Construction - Comparison of Unit Price between Actual Construction Cost and Standard Quantity per Unit - (조경공사 적산방식의 비교연구 - 실적공사비와 표준품셈의 단가비교 -)

  • This study aims to find a proper construction cost calculation method by comparing unit prices of cost estimate in landscape construction among other public ordering construction projects. There were 7 out of 12 items from the actual work cost in the first half of 2011 were compared. The 12 items were classified as landscape Architectural construction and the 7 items were the approximate standard. As applied construction types, the comparable 35 items out of the 80 actual work cost items applied to 5 sites were compared to the unit price of the standard quantity per unit in March 2011, which was the approximate standard. Actual construction rate of the 7 items in the category of landscape sector was 104.86% for each item and 92.09% as a total construction cost. The high actual construction rate was caused by the high rate of seed spray depending on the status of applying rocks. However, there were more fundamental reasons for the cost generated from aslope treatment for grass and seed spray. So, it requires more detailed regulations on the modification factors for each soil type, the standard and needs to improve theillogical standard quantity per unit system. Actual construction rate of the 35 items in the applied sectors of civil engineering and architecture was 78.65% for each item and 71.31% (70.17%) as a total construction cost. This shows that actual unit cost cannot reflect actual cost structurally and standard quantity per unit system lacks practicality in terms of construction due to thelabor force. 85.1~91.2 % actual construction rate announced by the Ministry of Land and Maritime Affairs referred to the newly switched items. So the result was estimated as actual construction rate. This requires supplementation after verification in order to make the actual work cost produce at a proper rate. Also, standard quantity per unit system needs complementation with these actual data and so on.

Structural Behavior of Composite Basement Wall According to Shear Span-to-Depth Ratio and FE Analysis Considering the Condition of Contact Surface (전단경간비에 따른 합성지하벽의 거동과 접촉면의 조건을 고려한 유한요소 해석)

  • The objective of this paper is to study the structural behavior of Composite Basement Wall (CBW) according to shear span-to-depth ratio through an experiment and predict the nonlinear behavior of CBW by using ADINA program widely has been being used for FE analysis. Especially, this study focuses on the part of CBW in which the Reinforced Concrete (RC) is under compression stress; At the region of CBW around each floor, RC part stresses by compressive force when lateral press by soil acts on the wall. The contact condition between RC wall and steel (H-Pile) including stud connector is main factor in the analysis since it governs overall structural behavior. In order to understand the structural behavior of CBW whose RC part is under compressive stress, an experimental work and finite element analysis were performed. Main parameter in the test is shear span-to-depth ratio. For simplicity in analysis, reinforcements were not modeled as a seperated element but idealized as smeared to concrete. All elements were modeled to have bi-linear relation of material properties. Three type of contact conditions such as All Generate Option (AGO), Same Element Group Option with Tie(SEGO-T) and Same Element Group Option with Not tie(SEGO-NT) were considered in the analysis. For each analysis, the stress flow and concentration were reviewed and analysis result was compared to test one. From the test result, CBW represented ductile behavior by contribution of steel member even if it had short shear span-to-depth ration which is close to "1". The global composite behavior of CBW whose concrete wall was under compressive stress could be predicted by using contact element in ADINA program. Especially, the modeling by using AGO and SEGO-T showed more close relation on comparing with test result.
