Guidance on Estimating Soil Persistence and Degradation Kinetics from Environmental Fate Studies on Veterinary Pharmaceuticals for Environmental Risk Assessment (동물용의약품의 환경 중 위해성 평가를 위한 토양 잔류성 시험법 가이드라인)

  • BACKGROUND: To assess and prevent the environmental impacts and risks by veterinary pharmaceuticals, Guidance on Estimating Soil Persistence and Degradation Kinetics from Environmental Fate Studies on Veterinary Pharmaceuticals for Environmental Risk Assessment was proposed. METHODS AND RESULTS: Proposed guidance was coined by VICH, EU guideline, OECD guideline and soil dissipation studies for the purpose of international harmonizing. Guidance was also modified from pesticide soil persistence testing guidelines of US, EU, and Korea, with practical approaches adopting in-use test guideline for Korea. CONCLUSION(S): Proposed guidance are consisted of three parts; Laboratory Soil Experiment, Field Soil Dissipation Study, and Estimation of $DT_{50}/DT_{90}$. Proposed guidance is to be available for the requirement for registration of veterinary pharmaceuticals with fit for purpose in Korea.

Assessment of Soil Microbial Communities in Carotenoid-Biofortified Rice Ecosystem

  • This study was conducted to investigate the effect of Psy-2A-CrtI (PAC), a genetically modified (GM) rice with enhanced ${\beta}$-carotene, on the soil microbial community. The soil used to cultivate GM rice and its wild-type, Nakdong, was analyzed for population density, denaturing gradient gel electrophoresis (DGGE), and pyrosequencing. It was found that the bacterial, fungal and actinomycetes population densities of the PAC soils were within the range of those of the non-GM rice cultivar, Nakdong. The DGGE banding patterns of the GM and non-GM soils were also similar, suggesting that the bacterial community structures were stable within a given month and were unaffected by the presence of a GM plant. The pyrosequencing result showed a temporal difference in microorganism taxon and distribution ratio, but no significant difference between GM and non-GM was found. The persistence of the transgene DNA in the plant and surrounding soil were investigated for different time periods. There were differences in the persistence within the plant depending on the gene, but they could not be detected after 5 weeks. Also the transgenes were not detected in the surrounding soil. These results indicate that soil microbial communities are unaffected by the cultivation of a PAC rice within the experimental time frame.

Assessment of the Persistence of DNA in Decomposing Leaves of CMVP0-CP Transgenic Chili Pepper in the Field Conditions (포장 조건에서 CMVP0-CP 형질전환 고추 도입유전자의 지속성 조사)

  • This study was conducted to evaluate the persistence of DNA in the transgenic chili pepper resistant to cucumber mosaic virus (CMV) in the field condition. We analyzed the persistence of genes in the leaf samples obtained from two field conditions, below and above soil. Transgenic and non-transgenic leaf tissues were buried in the soil at a depth of 10 cm or placed on the soil surface. Qualitative and quantitative PCR analysis showed that the amount of transferred genes from the transgenic peppers below and above soil was dropped to 28.3-42.7% one month after buried and it was rapidly reduced to 0.9-3.3% after two months. The transgenes were not detected three to four month after buried. In addition, DNA of the leaves placed below soil decomposed about 8%more than those on soil surface. The gene transfer from decomposing leaves of the transgenic pepper to soil was investigated by PCR analysis with the soil attached to the samples. The PCR result indicated that the gene transfer from the transgenic pepper to soil was not occurred.

Influence of soil organic matter and moisture on the persistence of the herbicide mefenacet in soils (제초제 Mefenacet의 토양 중 분해에 미치는 토양유기물과 토양수분에 의한 영향)

  • In order to elucidate a degradation characteristics of herbicide mefenacet in soil, the persistence in soils was studied under laboratory conditions for $90\sim120$ days at $28^{\circ}C$. Mefenacet residues were determined from the two soils which pre-treated by sterilization and flooding, respectively. Non-sterilized upland soil was used as a control. When 70 days elapsed from application time, $55\sim63%$ of mefenacet applied were dissipated in control soils. However, $32\sim33%$ of mefenacet applied were dissipated in the sterilized soils and $33\sim35%$ was dissipated in the flooded soils. 까 lese results indicated that the degradation of mefenacet was assumed to be due to microorganism, especially aerobic microbes. In order to elucidate the influence of water content on the persistence of mefenacet in soil, water content in soils was adjusted to 20, 50, and 80% of the water-holding capacity(Field capacity, WHC). The half-life of mefenacet in soil containing 20% and 50% of WHC were 82 and 73 days, respectively, after incubation for 90 days. However, the half-life in soil containing 80% of WHC was shortened to 61 days. These results indicated that degradation of mefenacet in soil was influenced by the activity of soil microorganism, organic matter content and water content.

Persistence of Salmonella enterica, Escherichia coli O157:H7, and Listeria monocytogenes in Soil, Liquid Manure Amended Soil, and Liquid Manure

  • While searching for healthier diets, people became more attentive to agricultural organic products. However, organic foods may be more susceptible to microbiological contamination because of the use of livestock manure compost and liquid manure, potential sources of pathogenic bacteria. This study was undertaken to investigate the persistence of Salmonella enterica, Escherichia coli O157:H7, and Listeria monocytogenes in soil, liquid manure amended soil, and liquid manure. Loamy soil, liquid manure amended soil, and liquid manure were inoculated with S. enterica, E. coli O157:H7, and L. monocytogenes. Samples were incubated in consistent moisture content at $25^{\circ}C$. Samples had been periodically collected during 120 days depending on the given conditions. S. enterica and E. coli O157:H7 survived over 120 days in loamy soil and over 60 days in liquid manure amended soil, respectively. L. monocytogenes decreased faster than other pathogens in soil. S. enterica, E. coli O157:H7, and L. monocytogenes survived for up to 5 days in liquid manure. S. enterica and E. coli O157:H7 in soil decreased by 2 to $2.5log\;CFU\;g^{-1}$ for 120 days. S. enterica and E. coli O157:H7 in liquid manure amended soil decreased slowly for 21 days. However, S. enterica, E. coli O157:H7, and L. monocytogenes sharply decreased after 21 days. S. enterica, E. coli O157:H7, and L. monocytogenes in soil increased by 0.5 to $1.0log\;CFU\;g^{-1}$ for 7 days. Foodborne pathogens in soil and liquid manure amended soil gradually decreased over time.

Effect of Repeated Application of IBP on the Degradation of Pesticides in Flooded Soil (IBP의 반복처리(反復處理)가 담수토양중(湛水土壤中) 농약(農藥)의 분해(分解)에 미치는 영향(影響))

  • This experiment was conducted to see the effect of repeated application of IBP granular formulation(17%, 0,0-diisopropyl-S-benzyl thiophosphate) on the biodegradation of IBP and diazinon〔0,0-diethyl 0-(2-isopropyl-4-methyl-5-pyrimidinyl) phosphorothioate〕 in silt loam soil with 2.1% organic matter under flooded condition. The persistence of IBP in the soil was shortened by increasing the frequencies of application of the chemical. Enhanced degradation ability in the soil caused by repeated application of IBP was prolonged about 53 days, while the ability did not influence diazinon persistence in the soil. The half-lives of IBP in sterilized soil autoclaved at $121^{\circ}C$ for 30 minutes were about 3 times longer than those in viable soil, suggesting that microbial process was a major factor for IBP degradation in the soil. The total colony number of soil microbes showed little difference between the soils with and without repeated application of IBP. A possible concern of specific soil microorganisms on the pesticide degradation in soil was discussed.

Optimum Soil Incorporation Time of Chinese Milk Vetch(Astragalus sinicus L) for its Natural Re-seeding and Green Manuring of Rice in Gyeongnam Province, Korea

  • To develop a natural re-seeding technology, the optimum soil incorporation time of Chinese milk vetch(CMV) plant residues, seed persistence in soil, biomass production and subsequent rice yield were determined from 2005 to 2007 in rice with CMV green manure in southern Korea. Generally, insufficient seedling stand was regenerated with CMV incorporation to the soil at 20 and 25 days after flowering(DAF), while high regeneration of greater than 400 seedlings per $m^2$ was observed at 35 and 40 DAF. High re-seeding stand of CMV incorporated at 35 DAF or later was related with high seed viability and heavy seed weight. Appreciable number of CMV seeds remained 4 month after burial in soil and a good number of CMV seedlings regenerated from this seed bank at rice harvest time in the fall. Based on the relationships among re-seeding number of CMV plants, seed weight and seed viability, CMV plants should be incorporated into the soil 35 DAF(May 30) or later when CMV seeds were sufficiently matured. The natural re-seeding stand for the 3-year trials was stable ranging from 437 to 700 plants per $m^2$ and the biomass production was sufficient to supply nitrogen for rice growth. The use of re-seeding CMV plant can produce similar rice yield like that of rice without CMV green manure.

Persistence of IBP and Isoprothiolane in Rice Plant (수도체(水稻體)중 IBP 와 Isoprothiolane의 잔류소장(殘留消長))

  • Effect of the application time on the persistence of IBP and isoprothiolane in rice plant was studied in the field and effects of the water depth and soil texture on their persistence were also tested as a pot experiment. When granules were applied to the rice paddy water, two fungicides were readily absorbed through the root system and rapidly translocated to the upper parts of the plant. The concentrations of two fungicides in rice plant reached to the maximum within 24 hr regardless of the application time. When applied at the maximum tillering stage, the persistence pattern of two fungicides in plant showed similar trends; that is, residue levels of two compounds declined rapidly upto 7 days after application but more slowly thereafter. When applied at the heading stage, the persistence pattern of IBP in plant was similar to the maximum tillering stage while isoprothilane was quite different; 3 ppm reached on 3rd days after application was maintained almost constant for further 25 days. There was no effect of the water depth on the persistence of two compounds in plant and IBP concentration in plant was also not affected by soil texture. However, isoprothiolane in plant was higher in sandy loam than in loam and clay loam. Isoprothiolane residues in plant were much higher than those of IBP.

Effects of Soil Organic Matter Content on Activity Change, Vertical Migration, and Persistence of Two Nematicides, Carbofuran and Ethoprophos, to Root-Knot Nematode, Meloidogyne incognita (토양중 유기물함량 차이에 따른 고구마뿌리혹선충(Meloidigyne incognita)에 대한 Carbofuran과 Ethoprophos의 효력변동, 수직이동성 및 잔효성조사)

  • Effects of organic matter content in soil on activity. vertical migration. and persistence of two nernaticides. carbofuran and ethoprophos. to root-knot nematode, Mrloitlog!~iei ~lcognita.w ere investigated. As the organic matter content increased. activity of the nernaticides tended to be reduced. Both nematicides exhibited control values of more than 80'k' to M. iix.o,yilit~iln 0-2 cm depth soil layer from the surface. regi~rdless of organic matter content in soil. In 2-4 cm depth soil layer. however. the control value of the neniaticides varied with the organic niatter content in soil. The control value of carbofuran in the soil layer was ranged from I0 to 30'2,. depending on the soil organic niatter content. In contrast. ethoprophos had no control value against M. i/ic.o,ytlitcr in the \oil layer, except that the nematicide had a control value of 30% when the organic matter content was 0.4%. Furthermore. ethoprophos had no effect on controlling M. i/ic.o,gtiitrr in soil layer of below 4cm. whereas control values of carbofuran were approximately from 5 to 20% in all test soils having different organic matter contents. These results indicate that carbofuran has more vertical migration effect than ethoprophos. Persistence of the two neniaticides was also decreased with increasing soil organic matter content. Half life of carbofuran was 2-3 weeks in soil containing 0.4% organic matter, whereas it was found to be I week in soils containning 0.8 ant1 1.6% of organic matter. On the other hand. activity of ethoprophos was reduced to half in 3-3 weeks and in 2-3 weeks in soil containing 0.4 and 0.8%, and 1.6%) of organic matter, respectively. However, no activity of the both nernaticides was found in soil containing 3.2% of organic matter.
