Nitrate enhances the secondary growth of storage roots in Panax ginseng

  • Background: Nitrogen (N) is an essential macronutrient for plant growth and development. To support agricultural production and enhance crop yield, two major N sources, nitrate and ammonium, are applied as fertilizers to the soil. Although many studies have been conducted on N uptake and signal transduction, the molecular genetic mechanisms of N-mediated physiological roles, such as the secondary growth of storage roots, remain largely unknown. Methods: One-year-old P. ginseng seedlings treated with KNO3 were analyzed for the secondary growth of storage roots. The histological paraffin sections were subjected to bright and polarized light microscopic analysis. Genome-wide RNA-seq and network analysis were carried out to dissect the molecular mechanism of nitrate-mediated promotion of ginseng storage root thickening. Results: Here, we report the positive effects of nitrate on storage root secondary growth in Panax ginseng. Exogenous nitrate supply to ginseng seedlings significantly increased the root secondary growth. Histological analysis indicated that the enhancement of root secondary growth could be attributed to the increase in cambium stem cell activity and the subsequent differentiation of cambium-derived storage parenchymal cells. RNA-seq and gene set enrichment analysis (GSEA) revealed that the formation of a transcriptional network comprising auxin, brassinosteroid (BR)-, ethylene-, and jasmonic acid (JA)-related genes mainly contributed to the secondary growth of ginseng storage roots. In addition, increased proliferation of cambium stem cells by a N-rich source inhibited the accumulation of starch granules in storage parenchymal cells. Conclusion: Thus, through the integration of bioinformatic and histological tissue analyses, we demonstrate that nitrate assimilation and signaling pathways are integrated into key biological processes that promote the secondary growth of P. ginseng storage roots.

  • In order to find out the application effect of commercial organic fertilizer on the germination rate of ginseng seed, growth of ginseng plant and changes of the physicochemical properties of soils, a Miwon organic fertilizer, byproduct of amino acid fermentation, was applied to the ginseng nursery bed. The application rates of Miwon organic fertilizer were 0kg-N from organic fertilizer and 30kg-N from wild grass compost per 10a as control(namely 0kg-N/10a), 2.5kg-N and 27.5kg-N/ 10a(2.5kg-N/10a) 5.0kg-N and 25kg-N/10a(5.0kg-N/10a), 10kg-N and 20kg-N/10a(10kg-N/10a), 15kg-N and 15kg-N/10a(15kg-N/10a) respectively, The obtained results are as follows: 1. The germination rate of ginseng seed were better in the plots that received 2.5kg, 5.0kg and 10kg-N/10a as compared with control plot. However no difference was found on germination rate between control and 15kg-N/10a, 2. The higher rate of missing plant was found in the 15kg-N/10a plot that showed poor growth. However control and 10kg-N/10a plot showed similar rate of missing plant. The best result, lower rate of missing plant, was obtained in the order of 5kg-N and 2.5kg-N/10a respectively. 3. The significant heavier ginseng root weight was obtained in 5kg-N/10a and 10kg-N/10a plot. However the root weight was decreased in the order of 2.5kg-N/10a, control, 15kg-N/10a. 4. In the physicochemical changes of soils between control and organic fertilizer treated soil before and after harvesting the increment of pH, C.E.C. and organic matter content along with the rate of organic fertilizer application were found. There were significant correlation between nitrate nitrogen and the rate of organic fertilizer application. 5. In the view points of germination rate, missing rate of plant and yield of ginseng root, it can be stated that the best result could be obtained from the plot that received 10kg-N/10a from Miwon commercial organic fertilizer and 10kg-N/10a from wild grass compost.

  • In order to get information on the ecology of rice false smut, germination ability and pathogenicity of sclerotia and chlamydospores of the pathogen, environmental conditions affecting the disease outbreak and varietal resistance have been investigated. 1. The degree of outbreak of rice false smut was higher in the upland rice in comparison with the paddy field rice in respect to the number of affected grains per ear, the size and weight of smut balls formed on affected grains as well as the ratio of sclerotial formation produced on smut balls. 2. Germination percentage and days required for germination of overwintered sclerotia placed on the soil surface in July were 81% and 19 days, respectively, while those of overwintered sclerotia treated in May were 60-70% and 41 days. Sclerotia placed on the soil surface or under 1 cm depth of the soil surface and incubated at $25-30^{\circ}C$ were germinated well, whereas those placed under 3 cm or 5 cm depth of the soil surface were not germinated at all. Germinability and stroma productivity of sclerotia were reduced when the sclerotia were cutted into small pieces. 3. The average number of stroma formed on a sclerotium was six and that of perithecia formed in a stroma was about 50 to 140. 4. Percentage of germination of chlamydospores on the yellow balls was very high and was decreased as the color of the balls being darken with maturation. 5. Panicle of rice plants were successfully infected by injection inoculation with suspention of ascospores and chlamydospores of the pathogen to the sheaths at the booting stages, while seeding infection by spraying with suspensions of chlamydospores was unsuccessful. 6. More number of infected grains was distributed on basal parts of an affected ear than that of infected ones distributed upper parts of the ear, when the affected ear was divided into five parts from its basal portion to the apical of the ear. 7. The occurrence of the disease was more severe in the late maturing varieties of rice in comparison with the early maturing varieties. 8. When the level of nitrogen applied was increased, the incidence of disease increased, and the infection percentage of the disease was increased as the transplanting date was delayed. 9. The weight of panicles and 1000 kernels and the ratio of ripenness were reduced, and the contamination degree of grains with chlamydospores were increased as the number of smut balls per panicle were increased.

Nitrogen Release from Hairy Vetch(Vicia villosa Roth) Residue in Relation to Different Tillages and Plant Growth Stage (생육단계 및 경운방법에 따른 헤어리베치 녹비의 질소방출 패턴의 변화)

  • Assessment of N release pattern from hairy vetch (Vicia villosa Roth) residue is an important step for the efficient use of the residue as a green manure and the specific management strategy development. Objective of this research was to determine the changes of chemical composition, especially in light of the N release, when the residues of hairy vetch were applied in the soil. hairy vetch was harvested at different growth stages (April 23, May 9, and May 28) and the residues were encapsulated in the nylon mesh (1 mm) bag. The bags were placed on the surface of no-tillage plots and buried at 15cm in depth in conventional tillage plots of the corn field in early June. The bags were removed for measurements of dry matter and N content at 1, 2, 3. 5, 8 and 12 weeks after placement. Dry matter and N content of hairy vetch were higher when harvested closer to maturity, as also found in C/N ratio, cellulose and hemicellulose contents. The rate of N releases from hairy vetch residue under conventional tillage condition was greater than that under no-tillage condition. Percentages of N remaining in residue over the initial N contents under conventional tillage condition were in average 40, 23, and 15% for 1, 2, and 3 weeks after placement of mesh bag, respectively, compared with respective values of 71, 31, and 21% under no-tillage condition. The rate of N release was slower as the harvest time was closer to maturity of hairy vetch under both tillage conditions. Most of N in residue was released within 3 and 5 weeks, under conventional and no-tillage conditions, respectively. Results indicated that amounts of N released from hairy vetch residue were more closely related to the variability of N contents in residue due to the harvest time than changes of chemical composition of plant or soil management.

Ecological Studies on Vegetation Recovery of Burned Field after Forest Fire (산화적지의 식생회복에 관한 생태학적 연구)

  • The recovery of vegetatiion and soil properties in the burned fields after forest fire were studied in Chung Cheong Buk Do area, Korea, from July 23 to 28, 1981. Miscanthus sinensis var. purpurascens, Carex humilis and Lespedeza bicolor were dominant species in the burned field of the second year after forest fire as well as unburned field. Especially, Lespedeza bicolor was gradually grown to the dominant species. Lespedeza bicolor, Carex humilis and Miscanthus sinensis var. purpurascens were the dominant species in the burned field of the fifth year after forest fire. Biological spectra of the burned fields were similar to the umburned fields with $H-e-D_1-R_5$ or $Ph-e-D_1-R_5$ from the second year after forest fire. Accordingly, biological spectra were recovered to the unburned fields from the second year. Degree of successiion was DS=423 in the burned field and DS=524 in the unburned field in 1981. The DS of the burned fields was gradually increased and recovered to be similar to the unburned from the second year. In the species diversities and evenness index, H,e and $\beta$ of the burned field in 1981 were higher and $\lambda$ was lower than the unburned field, but all of the indices were recovered to the unburned field from the second or third years. Accordingly, the vegetation of the first year was the complex community in view floristic composition, but it was recovered to the simple community as unburned field fromthe second or third years. In the soil preperties, pH, total nitrogen, available phosphorus, exchangeable potassium, exchangeable calcium and exchangeable magnesium were increased and organic matter was decreased due to forest fire, and then was recovered to the unburned field from the second or third years. The vegetation and soil properties of the burned field after forest fire were similary recovered to the unburned field from the second or third years. Accordingly, there was a close relationship between the trend of vegetation recovery and the changes of soil characteristics after forest fire.

Losses of Chemical Components by Infiltration Water during the Rice Cultivation at Silt Loam Paddy soil (영농기간 지하침투수에 의한 미사질양토 논의 화학성분 손실량)

  • Changes of chemical component and losses of chemical fertilizer by infiltration water from 0.5ha of paddy field in Chinan area of Chonbuk province during the rice cultivation were investigated. The infiltration water samples were collected in a ceramic porous cup which was a buried at the 30, 50, 70, 90㎝ of soil depth. pH of infiltration water ranged $6.64{\sim}7.90$ and EC showed $324{\sim}647{\mu}$S/cm. The content of total-N, $NH_4-N$ and $NO_3-N$ were $0.58{\sim}14.59$, $0.05{\sim}4.25$, and $0.15{\sim}7.71mg/L$ respectively. The content of total-P and ortho-P were $0.009{\sim}0.077mg/L$ and $0{\sim}0.029mg/L$ The content of $Ca^{++}$, $Mg^{++}$, $Na^{+}$ and $K^{+}$ showed $0.88{\sim}4.78$, $0.22{\sim}1.04$, $0.17{\sim}0.98$, and $0.84{\sim}3.19㎎/L$. These all at the first transplanting are higher than that of other periods. The content of $SO_4^{2-}$ showed $3.92{\sim}18.72mg/L$ and decreased with a soil depth. However $Cl^-$ of infiltration water ranged $9.03{\sim}19.97mg/L$ and no difference with a soil depth. When infiltrated $2,416.5m^{3}$ of an infiltration water from 0.5ha of paddy field during the rice cultivation, losses of chemical components were 20.34㎏/㏊ of total-N, 3.54㎏/㏊ of $NH_4-N$, 10.44㎏/㏊ of $NO_3-N$, 0.16㎏/㏊ of total-P and 0.028㎏/㏊ of ortho-P. Also $Ca^{++}$, $Mg^{++}$, $Na^+$, $K^+$, $SO_4^{2-}$ and $Cl^-$ were lost 10.24, 2.84, 2.84, 7.22, 50.04 and 62.20㎏/㏊ respectively. There were lost by infiltration water 9.35% of nitrogen, 0.59% of phosphorous and 22.79% of potassium in applied chemical fertilizer.

Effects of Fire on Forest Vegetation in Mt. Samma (산불이 삼마산의 삼림식생에 미치는 영향)

  • Authors report the changes of the community structure and secondary succession after fire in Mt. Samma (333 m). Approximately six ha of the red pine (Pinus densiflora) forest and its floor vegetation were burned and cutted down. The vegetation and soil properties were investigated in the burned and unburned sites from April to October, 1998. The dominant species based on SDR₃ in the burned site were Lespedeza cyrtobotrya (96.87), Quercus serrata (77.90), Cyperus amuricus (46.22) and Miscanthus sinensis var. purpurascens (38.33), whereas the dominant species in the unburned site were Pinus densiflora (100.00) and Q. variabilis (66.10) at the tree layer, Q. serrata (100.00) and Zanthoxylum schinifolium (29.64) at the shrub layer., and Q. serrata (76.30) and M. sinensis var. purpurascens (72.84) at the herb layer. The biological spectra based on SDR₃ were Th-D₁-R/sub 5/-e for the burned and H (M)-D₁-R/sub 5/-e type for the unburned site, respectively. The index of similarity (CCs) between the burned and unburned sites was 0.41. The degree of succession (DS) were 609 for the burned and 1168 for the unburned site, respectively. The species diversity (H) and evenness indices (e) of the burned site were lower than those of the unburned site, but the dominance indices (C) was higher in the burned site. In the analysis of soil properties, pH, the content of NO₃/sup -/-N, available phosphrous, and exchangeable cations (K/sup +/, Ca/sup 2+/, Na/sup +/, Mg/sup 2+/) in the burned site were higher than those in the unburned site, whereas the contents of organic matter, total carbon, total nitrogen and NH₄/sup +/-N were lower in the burned site. The results show that dominant species in the burned site change from Miscanthus-Lespedeza→Lespedeza→Quercus and finally to Quercus forest, and the changes of soil properties also affect the early vegetational succession after fire.

Effect of Immature Compost on Available Nutrient Capability and Heavy Metal Accumulation in Soil for Lettuce (Lactuca sativa L.) Cultivation (퇴비 내 영양소 및 중금속이 상추 재배에 미치는 영향)

  • The aim of this study was to evaluate effects of immature compost on the amount of nutrient content, heavy metal concentration, and application rate that were used for lettuce cultivation. The characteristics of the two composts (Compost A (CA) was immature compost and Compost B (CB) was mature compost) were evaluated upon mixing with commercial soil at 0%, 25%, 50%, and 75% (w/w). The poor chemical characteristics were appeared by use of immature compost as soil amendment; the 50% and 75% rates were weakly acidic at pH 5.39 and 5.50, respectively. The total carbon content at using of 75% of the immature compost and mature compost increased the most to 14.5 and 6.5% and it significantly increased concentrations of the total nitrogen and phosphorus compared to control. As for 75% mature compost rate increased significantly the concentrations of Cu ($128mg\;kg^{-1}$), Zn ($260mg\;kg^{-1}$), Pb ($0.32mg\;kg^{-1}$) and, Cd ($0.48mg\;kg^{-1}$) compared to control, and the highest As concentration increased significantly at 75% and 50% (6.69 and $6.28mg\;kg^{-1}$) including in 25% immature compost as $6.48mg\;kg^{-1}$. However, all of the high compost rates significantly decreased the shoot biomass of lettuce. The immature compost was potentially amended at an application rate of 25% due to a slight salinity and low risk to heavy metal uptake on lettuce growth. This use may be available if the rate is lower than that used in this trial.

Ecological Characteristics and Management Plan of Geumdangsil Pine Forest of Yecheon (예천 금당실 송림의 생태적 특성 및 관리방안)

  • The purpose of this study was to provide data for the basic research to found the effective conservation and management plan for the Geumdangsil Pine Forest of Yecheon designated as Natural Monument No. 469. Furthermore, this paper suggest efficient sustainable forest preservation and using. In order to achieve the sustainable forest preservation, this study was to analyse topography, land use, tree growth, soil environment, forest usage and forest management, etc. According to analysis the results, the site area is located in the flatlands where is from 130 to 140 m above sea level. The around forest was transformed into agricultural land. The 565 individuals of Pinus densiflora grows in the forest, whereas, 25 trees was cut down or died. There are signs of 25 stumps. The most of 565 trees' diameter at breast height(DBH) was centerized between 30 cm and 50 cm, moreover, the average life expectancy of trees were 85.4 years. The oldest age of tree was estimated to be 200 years. The Sample trees of rate of branch growth is from 4.3 cm to 5.1 cm per year. The middle branch which is more vigorous growth grow 24.2 cm for 3 years. Moreover, the result of soil physico-chemical properties analysis of 7 plots, 4 categories which is soil organic matter, total nitrogen, available phosphoric acid, specific electrical conductance was generally good, however, the 2 categories which is soil pH, exchangeable cation needed improvement. Currently, the site was not pressured by facilities and usage, however, there might be threaten by agriculture such as encroaching on forest. Therefore, there should establish comprehensive ecosystem management such as facility management, visitors management and operation management In this paper considered 4 fields that is ecosystem management, facility management, visitors management and operation management for sustainable management.

Studies on the Indigenous Vesicular-Arbuscular Mycorrhizal Fungi(VAMF) in Horticultural Crops Grown Under Greenhouse -III. Effects of the Indigenous VAMF Inoculation on the Early Growth and the Subsquent Growth after Transplanting of Greenhouse Grown Crops (시설원예(施設園藝) 작물(作物)에서 토착(土着) VA균근균(菌根菌)에 관한 연구(硏究) -III. 토착(土着) VA 균근균(菌根菌) 접종효과)

  • Effects of the indigenous Vesicular-arbuscular mycurrhizal fungi(VAMF) on early growth response of greenhouse grown crops were experimented. This study was done to evaluate the benefit of indigenous VAMF inoculation on the early growth and the subsequent growth after transplanting of some crops such as cucumber, tomato, hot pepper, eggplant, and melon. Leaf area, shoot dry weight, and plant length of mycorrhizal greenhouse crops showed the tendency of significant or no significant increase over control plants receiving no inoculation. The levels of VA mycorrhizal colonization were increased with plant growth, and infection rates of horticultural crop except hot pepper around one week after transplanting were decreased, while that of 8 weeks after emergence of mycorrhizal seedlings were increased again and infected by around 50% at harvesting time. In spore densities in the rhizosphere soil of craps experimented, the number of spore ranged from $72.7{\pm}26.3$ to $100{\pm}10.3g^1$ on dried soil basis and high density showed in both cucumber and tomato. Total nitrogen contents in shoots were lower in the mycorrhizal plants than non-mycorrhizal one, whereas P uptake in mycorrhizal hot pepper and tomato were highly ramarkable. The K contents in the shoots of mycorrhizal cucumber and eggplent were highly enhanced. Inoculation of the indigenous VAMF enhanced shoot Ca and Mg in both tomoto and melon. The contents of Fe, Zn, Mn and Cu in shoots of mycorrhizal crops were higher than non-mycorrhizal plants and vice versa in case of eggplent. Inoculation of the indigenous VAMF to horticultural crops were effective for alleviation of transplanting shock, and pretransplanting infection improved subsequent growth by reducing the time required for establishment of a functional mycorrhizal symbiosis following transplanting.
