Isolation of Bacillus sp. Producing Xylanase and Cellulase and Optimization of Medium Conditions for Its Production. (Xylanase, Cellulase의 생산성이 높은 Bacillus sp.의 분리 및 효소생산을 위한 배지조건의 최적화)

  • A bacterium producing the extracellular xylanase and CMCase was isolated from soil and has been identified as Bacillus sp. The isolate, named Bacillus sp. A-7, was shown to be very similar to Bacillus licheniformis on the basis of its biochemical and physiological properties. The maximum xylanase and CMCase production were obtained when 2.0% (w/v) glucose and 0.3% (w/v) yeast extract were used as carbon source and nitrogen source, respectively. The best mineral conditions for xylanase and CMCase production were 0.1%(w/v) $CaC1_2$. Among the various feedstuffs, 1.0%(w/v) soybean meal was selected for the best xylanase and CMCase production.

Selecting Target Sites for Non-point Source Pollution Management Using Analytic Hierarchy Process (계층분석적 의사결정기법을 이용한 비점원오염 관리지역의 선정)

  • This paper suggests a hierarchial method to select the target sites for the nonpoint source pollution management considering factors which reflect the interrelationships of significant outflow characteristics of nonpoint source pollution at given sites. The factors consist of land slope, delivery distance to the outlet, effective rainfall, impervious area ratio and soil loss. The weight of each factor was calculated by an analytic hierarchy process(AHP) algorithm and the resulting influencing index was defined from the sum of the product of each factor and its computed weight value. The higher index reflect the proposed target sites for nonpoint source pollution management. The proposed method was applied to the Baran HP#6 watershed, located southwest from Suwon city. The Agricultural Nonpoint Pollution Source(AGNPS) model was also applied to identify sites contributing significantly to the nonpoint source pollution loads from the watershed. The spatial correlation between the two results for sites was analyzed using Moran's I values. The I values were $0.38{\sim}0.45$ for total nitrogen(T-N), and $0.15{\sim}0.22$ for total phosphorus(T-P), respectively. The results showed that two independent estimates for sites within the test water-shed were highly correlated, and that the proposed hierarchial method may be applied to select the target sites for nonpoint source pollution management.

Formation of Sedimentation Pool within Irrigation Reserviors for Water Quality Improvement (저수지 수질개선을 위한 저수지 내 침전지 조성)

  • Large quantitive of polllutants are washed into reservoirs during storm events. These polllutants contribute to eutrophication, such as algal blooms and fish kills. This study was conducted for the purpose of assessing the pollutant removal possibilities of sedimentation pool formed by deep dredging of a reservoir inlet. Water quality data were collected in the Masan reservoir, whose inlet has been dredged deep like sedimentation pool. The average concentration of chemical oxygen demand(COD) , toatal nitrogen(T-N) and total phosphrous(T-P) in the deep dredged area were 8.7 ~20.5mg/ι (T-N), 0.17~0.84mg/ι(T-P), which were 4.9%(COD), 29.0%(T-N) and 44.8%(T-P) higher than those of middle part of the reservior. The texture of sediment in the dredged area was silty loam, while that of the middle part was sandy clay loam. Organic matter contents, T-N and T-P of the bottom soil in the dredge area showed higher values than the middle part of the reservoirs. From these results, it was considered thedeep dredged area in the inlet of reservoir might play a key role to settle pollutant particulate. Based on the result of water quality analysis, deep dredging of the reservoir inlet could be assessed to reduce T-N and T-P of the reservoir about 6.5% , 8.3%, respectively. However, the effect of the sedimentation pool would be raised if the settled particles were taken into account in assessing water quality improvement for the reservoir. Accordingly, dredging of a reservoir inlet to make a shape of sedimentation pool is recommended for water quality improvement of reservoir in the stage of dredging plan.

Effects of Fertilizer Types on Pollutant Loadings from Rice Paddy Fields (비종에 따른 논에서의 배출부하량 영향평가)

  • The main objective of this study was to evaluate the effects of control-release fertilizer (CRF) on pollutant loadings from a small watershed. The Baran watershed, 386 ha in size, was selected as the study site, and the AGNPS (Agricultural Non-point Source Pollution) model was used to evaluate the effects of fertilizer types. Digital maps of digital elevation (DEM), slope distribution, channel, flow direction, landuse, soil, and curve number were extracted from the study watershed. Model parameters related to hydrology and water quality were calibrated and validated by comparing model predictions with the observed data collected for 2 years (1999 to 2000). Calibration and validation resulted in $R^2$ values of 0.75-0.91 for all the water quality parameters. All the paddy fields (21.2 %) of the study watershed were sprayed by either CRF or NPK (standard fertilizer). In CRF application, total nitrogen (TN) load was 4.9% less than NPK application, however total phosphorus (TP) load was 0.7 % more than NPK application. In CRF application, considering only paddy fields in the study area, TN load was 38.7 % less than NPK application. Using CRF in paddy fields could be one of the ways to reduce pollutant loadings from agricultural watersheds, however, in order to confirm it, more researches about effects of using CRF are necessary.

  • Studies were carried out to investigate the optimal culture conditions for the production of penicillinase using a strain of Streptomyces sp. isolated from soil, YS-40. Among the carbon and nitrogen sources, glucose and L-asparagine increased the peniciilinase production. The addition of M $n^{++}$, $Ca^{++}$ and L $i^{+}$ increased the enzyme production, but depressed by F $e^{+++}$, F $e^{++}$, $Mg^{++}$, Z $n^{++}$, A $g^{++}$, $Ba^{++}$ and S $n^{++}$. L-Leucine slightly increased the enzyme production but L-histidine, L-methionine depressed. Among the vitamins riboflavine, i-inositol, hesperidine, niacin-amide, biotin, folic acid, DL-$\alpha$-lipoic acid increased the enzyme formation. The addition of cephradine, cephalexin, ampicillin, cloxacillin more increased the enzyme formation than that of other$\beta$-lactam antibiotics and antibiotics. Optimal pH and temperature on the enzyme formation was pH 7.0 and 28$^{\circ}C$ respectively Amount of the enzyme production reached at maximum with incubation for 3 days on the optimal condition.

  • A highly cyclodextrin glucanotransferase producing strain of Bacillus sp. was isolated from soil, and basic studies on the characteristics of the strain and its enzyme, conditions for the enzyme production, and the enzyme utilization were carried out. The isolated strain was aerobic, motile, endospore-forming and rod-shaped bacterium. Optimum pH and temperature for the enzyme action were 6.0 and 45$^{\circ}C$, and the enzyme was stable within 5$0^{\circ}C$, and between pH 6.0 and 10.0. The highest yield of the enzyme was obtained using the medium containing 2% corn starch as a carbon source, and 5% corn steep liquor, 0.1% urea and 0.25% ammonium sulfate as nitrogen sources. The fermentation conditions for the enzyme production in a jar fermentor were cetermined to be 3$0^{\circ}C$, 200rpm, 0.6vvm and 60hr cultural period. Stevioside transglycosylation catalyzed by this enzyme was identified by high performance liquid chromatography.

  • A strain of Penicillium sp. which produces considerable amount of $\beta$-galactosidase was selected from extracellular $\beta$-galactosidase-producing fungi isolated from soil. The enzyme was found to be very stable in neutral pH range. Maximum enzymatic activity was reached after 72 hr of incubation in a wheat bran medium at 3$0^{\circ}C$. Productivity of the enzyme appeared not to be affected by the addition of carbon sources to the medium but the activity of the enzyme was increased from 14% to 85% by the addition of various nitrogen sources. The enzyme extracted from the wheat-bran culture of the Penicillium sp. was purified to 5050-fold by ammonium sulfate fractionation, SP-Sephadex C-50 chromatography, Ultrogel AcA 44 filtration and hydroxyapatite chromatography. The purified $\beta$-galactosidase was homogeneous on ultracentrifugation and disc electrophoresis.

Isolation and Characterization of a Feather Degrading Alkalophilic Streptomyces sp. TBG-S13A5 and its Keratinolytic Properties

  • Keratinases are of particular interest because of their action on insoluble keratins and generally on a broad range of protein substrates. Alkalophilic and neutrophilic actinomycete strains isolated from different soil samples, rich in keratinaceous substances were screened for keratinolytic activity. An alkalophilic isolate, TBG-S13A5, was found to possess good keratinolytic activity and was able to utilize feather as the sole carbon and nitrogen source. TBG-S13A5 exhibited an off-white aerial mass color with a rectus-flexibilis type of spore chain. The morphological, microscopical and biochemical characters were comparable with that of Streptomyces albidoflavus. Fatty acid methyl ester profiling (FAME) and 16S rDNA sequence analysis confirmed its identity as a strain of S. albidoflavus. Under submerged fermentation conditions, maximum protease production was recorded on the $5^{th}$ day of incubation at $30^{\circ}C$, using basal broth of pH 9.0 with 0.25% (w/v) white chicken feather. This strain could affect feather degradation when the initial pH was 8 and above and maximum protease production was recorded when the initial pH was around 10.5. The effectiveness of the crude enzyme in destaining and leather dehairing were also demonstrated.

Application of Analytical Instrument Method for Determining Level of Malodorous Sulfur Compounds. (악취성분중 황화합물에 대한 기기분석법의 적용)

  • The analytical instrument method was applied to analyze malodorous sulfur compounds emitted from industrial fields. Six factories and two sites which release malodorous substances into ambient air were selected to determine the level of hydrogen sulfide($H_2S$), methylmercaptan(MeSH), dimethyl sulfide($Me_2S$), and dimethyl disulfide($Me_2S_2$) using automated thermal desorption system (STD400) and GC-FPD in summer and fall seasons of 1999. The Air sampler for ATD400 uses a small pump to draw sample and a mass flow controller to adjust sample amount without using a dilution apparatus. The trap temperature of ATD400 reached to $-80^{\circ}$ by supplying liquid nitrogen and $H_2S$ can be analyzed under this condition. The recovery rates of $H_2S$, MeSH, $Me_2S$, and $Me_2S_2$ of odorous sulfur compounds standard were shown 98.2%, 93.6%, 98.2%, 99.4% respectively. The concentrations of $Me_2S$ at outside boundary of G market, L factory, and J factory were 0.018ppm, 0.021ppm, 0.032ppm in summer, respectively. The concentration of $H_2S$ at Nanjido landfill was 1.167ppm in summer, but that of $H_2S$ was not detected in fall because of soil covering. The concentration of H2S and $Me_2S_2$ at inside of Chonggye stream were 0.564ppm and 1.045ppm in summer, while those of H2S and Me2S2 were 0.285ppm and 0.465ppm in fall, respectively.

Production of protein-bound polysaccharides by solid-state fermentation of Coriolus versicolor (구름버섯의 고체발효에 의한 단백다당류 생산)

  • The possibility of solid-substrate fermentation of Coriolus versicolor for the production of protein-bound polysaccharides(PBP) was studied. Zeolite and orchid-pot soil were used as solid materials for the culture because of the desirable physical properties. Glucose, sucrose and starch showed to be good carbon sources for the production of PBP by the solid-substrate fermantation of C. versicolor. Among the nitrogen sources, bactosoyton and peptone were very effective for the PBP production. The optimum pH for solid-substrate culture for the production of PBP was at the range of 5-6. The yields of PBP reached to 5-6 mg per 100 g solid-substrate.
