New record of five ciliates(Protozoa, Ciliophora) collected in eastern Gangwon-do Province, South Korea

  • We investigated five ciliate species from soil, estuary and marine habitats in eastern Gangwon-do Province, South Korea. Morphology was studied through observation of living specimens as well as silver preparations. The species were classified into five genera, five families, five orders and four classes. Aspidisca aculeata, Hemiurosoma similis, Litonotus alpestris, Stentor roeselii and Vorticella infusionum are first records in Korea, and among them, the genus Hemiurosoma is recorded for the first time. Here, we provide short descriptions with microphotographs of these species.

  • This study was a preliminary investigation of the influence of surface characteristics of substrates on the detergency of particulate soil. A PET film was surface modified with NaOH and DMF for different times. The surface morphology of the film was scanned by AFM and the surface energies were calculated from the measured contact angles between several solutions and film based on a geometric mean and the Lewis acid base method. The surface morphology of the PET film treated with NaOH and DMF became more etched and swelled with an increased treatment time, respectively. The surface roughness and surface area of film treated with NaOH enlarged with increased treatment time. However, the coefficient of friction of film treated with NaOH and coefficient of friction, surface roughness, and surface area of film treated with DMF increased and then decreased with increased treatment time. The contact angle of film treated with DMF decreased with increased treatment time in water and surfactant solution; however, the effect of treatment time on the contact angle was different in both solutions for film treated with NaOH. By the treatment of PET film with NaOH and DMF, the polar group of the surface energy increased and the nonpolar group decreased; however, the change of total surface energy was not significant.

  • The feasibility of earthworm protease as a new cosmetic material for human hair care was investigated. The effectiveness of the earthworm protease treatment was assessed by thickness change of hairs, optical microscope examination, aminoacid analysis, surface morphology, angular resolution analysis through methylene blue staining method and tensile strength change. The protease treated hair became thinner and the soil on the surface removed in experimental groups unlike control group. Tensile strength decreased in experimental group in which the enzymes may decompose polypeptide bonds.

  • During a survey of fungal species in South Korea, a species of Volutella ciliata was isolated and described based on the analysis of the internal transcribed spacer region of its rDNA and its morphological characteristics. This is the first record of Volutella ciliata isolated from crop field soil in Korea.

  • A previously unrecorded species, Metacordyceps chlamydosporia KNU14-22, was isolated from soil in Korea. Identification of the fungal species was based on morphological and molecular characteristics. This species has not been previously reported in Korea and herein we present data with detailed descriptions and figures.

Genome Analysis of Phage SMSAP5 as Candidate of Biocontrol for Staphylococcus aureus

  • In this study, we reported the morphogenetic analysis and genome sequence by genomic analysis of the newly isolated staphylococcal phage SMSAP5 from soil of slaughterhouses for cattle. Based on transmission electron microscopy evident morphology, phage SMSAP5 belonged to the Siphoviridae family. Phage SMSAP5 had a double-stranded DNA genome with a length of 45,552 bp and 33 % G+C content. Bioinformatics analysis of the phage genome revealed 43 open reading frames. A blastn search revealed that its nucleotide sequence shared a high degree of similarity with that of the Staphylococcus phage tp310-2. In conclusion, this study is the first report to show the morphological features and the complete genome sequence of the phage SMSAP5 from soil of slaughterhouses for cattle.

  • Oidiodendron flavum KNU13-6 was isolated for the first time from field soil in Korea and identified based on the internal transcribed spacer region (ITS) of rDNA and morphological characteristics. Based on phylogenetic analysis of ITS and morphological characteristics, the species has not been previously reported in Korea.

  • Pseudeurotium bakeri KNU14-21 isolated from crop field soil is a previously unreported fungal species in Korea. This fungal species was identified through morphological and molecular characteristics. Based on the shape and size of conidia and conidiophores as well as the internal transcribed spacer region of rDNA, the isolate was confirmed to be Pseudeurotium bakeri. This species has not been previously reported from Korea, and this is the first report of its incidence in Korea.

A New Record on Umbelopsis vinacea and Mucor hiemalis f. corticola Isolated from Korea

  • In the screening of fungal diversity, two strains were collected from the soil of Yeongcheon and dissected guts from the bodies of Chinese rice grasshopper (Oxya chinensis), Chinese grasshopper (Acrida cinerea), and Far eastern devil grasshopper (Oedaleus infernalis) from Daejeon in Korea. They were identified as Umbelopsis vinacea (KNU-YC-1801B) and Mucor hiemalis f. corticola (KNU-20F7, KNU-20F8, KNU-20F9). Multigene phylogenetic analyses of the internal transcribed spacer (ITS) regions, and large subunit (LSU) sequence data confirmed two unreported taxa along with their morphology. The results of molecular phylogeny firmly supported the detailed description and illustration for each taxon. As far as we know, both Umbelopsis vinacea and Mucor hiemalis f. corticola are the first reported taxa in Korea.

Seven New Records of Penicillium Species Belonging to Section Lanata-Divaricata in Korea

  • Penicillium species are known to be ubiquitous environmental saprophytes. In the survey of diversity of genus Penicillium, seven new records of Penicillium species belonging to section Lanata-Divaricata were isolated from freshwater and soil samples collected from different locations in Korea. Based on morphological characteristics and multilocus phylogenetic analysis of the rDNA internal transcribed spacer region (ITS), β-tubulin (BenA), and calmodulin (CaM) genes, the isolated strains were identified as P. annulatum, P. camponotum, P. echinulonalgiovense, P. globosum, P. limosum, P. onobense, and P. yunnanense, respectively. This study presents detailed phylogenetic analyses and morphological descriptions of these species that contribute to section Lanata-Divaricata in Korea.
