A report on 57 unrecorded bacterial species in Korea in the classes Betaproteobacteria and Gammaproteobacteria

  • In an investigation of indigenous prokaryotic species in Korea, a total of 57 bacterial strains assigned to the classes Betaproteobacteria and Gammaproteobacteria were isolated from diverse environments. Samples were collected from fresh water, natural caves, soil, paddy fields, lakes, sea water, jeotgal (fermented seafood), salt flats, soil from abandoned mines, plant roots, digestive organs of both Japanese crested ibis (Nipponia nippon) and Burmese python (Python molurus bivittatus) and tidal flats. From the high 16S rRNA gene sequence similarity (>98.7%) and formation of robust phylogenetic clades with closely related species, it was determined that each strain belonged to an independent and predefined bacterial species within either the Betaproteobacteria or Gammaproteobacteria. There is no official report or publication that describes these 57 proteobacterial species in Korea. Overall, in the class Betaproteobacteria there were 16 species in 12 genera of 4 families in the order Burkholderiales and two species in two genera of one family in the order Neisseriales. Within the class Gammaproteobacteia, there were five species in four genera of four families in the order Alteromonadales, 12 species in 11 genera of one family in the order Enterobacteriales, four species in four genera of three families in the order Oceanospirillales, 11 species in four genera of two families in the order Pseudomonadales, two species in the order Vibrionales and five species in five genera of one family in the order Xanthomonadales. Mechanism and Adsorption Capacity of Arsenic in Water by Zero-Valent Iron (수용액 중 영가 철의 비소흡착 및 반응기작 구명)

  • Objective of this research was to evaluate optimal conditions of arsenic adsorption in water by zero-valent iron (ZVI). Batch experiment showed that adsorption of arsenic by ZVI followed a Langmuir isotherm model. The masses of As(V) adsorbed onto ZVI were increased as decreasing pH of the reacting solution (pH 3: 2.05, pH 5: 1.82, pH 7: 1.24, pH 9: 1.03 mg As/g $Fe^0$) and as increasing the temperature ($15^{\circ}C$ : 1.59, $25^{\circ}C$ : 1.81, 35 : $1.93^{\circ}C$ mg As/g $Fe^0$). The SEM and EDS (energy dispersive X-ray spectrometer) analysis of morphology and structure of ZVI before and after reacting with arsenic in water revealed that a relatively smooth and large surface of ZVI was transformed into a coarse and small surface particle after the reaction. The EDS spectra on the chemical composition of ZVI demonstrated that arsenic was incorporated into ZVI by adsorption mechanism. The XRD analysis also identified that the only peak for $Fe^0$ in the ZVI before the reaction and confirmed that $Fe^0$ was transformed into $Fe_2O_3$ and FeOOH, and As into $FeAsO_4{\cdot}2H_2O$.

Changes in Physical and Chemical Properties of Coir Used as the Bag Culture Substrate of Greenhouse Tomatoes for Three Years

  • To identify causes for drastic decrease in yield of tomato with repeated culturing number of the bag culture substrate of greenhouse tomatoes we investigated the physical and chemical properties of a coir used as the bag culture substrate to grow tomato at the Booyeo tomato experimental institute located in Booyeo, Chungnam Province for three years from 2012 to 2014. The results showed that total porosity ranged from 65.4 to 73.1% for the bulk densities of coir ranging from 0.12 to $0.14g\;cm^{-3}$. The volumetric water contents measured at 0.01 bar as air entry point were 25% (before), 33% ($1^{st}yr$), 45% ($2^{nd}yr$), and 37% ($3^{rd}yr$). Organic matter contents ranged from 82.0 to 96.2% (highest in $1^{st}yr$). pH and EC ranged from 4.47 to 6.47 (highest in $2^{nd}yr$), and from 22.2 to $53.5dS\;m^{-1}$ (highest in $1^{st}yr$) and cation exchange capacity ranged from 71.0 to $191.7cmol\;kg^{-1}$ (highest in $3^{rd}yr$). The surface structure observed with electrical microscope showed that the number of large pores decreased with increasing cultivating time while the proportion of smaller pores increased, indicating that the coir was consistently decomposed. Therefore, we could conclude that these changes of all physical and chemical properties of the coir may influence the holding capacities of water and nutrients, resulting in deterioration of quality of culture substrate of greenhouse tomatoes.

Crystallinity and Chemical Reactivity of Bimessite(δ-MnO2) Influenced by Iron (철에 의한 버네사이트의 결정도 및 화학적 활성의 변화)

  • Manganese (Mn) oxides in soils have been a research subject since they react with nutrients and contaminants and Mn itself is an essential element for plant growth. Birnessite was synthesized in the presence of iron (Fe) in the precipitating solution. Influence of Fe, one of common elements in soils, on crytallinity, morphology, and chemical reactivity of birnessite was examined using X-ray diffraction (XRD), electron microscope, canon exchange capacity (CEC), and chromium (Cr) oxidation capacity. With increasing Fe concentration in the precipitating solution, crystallinity and crystal size decreased. Hexagonal plates of the birnessites formed at low Fe concentration were dominant and replaced more and more by aggregate of small particles with increasing the Fe concentration. There is no significant change in CEC with changing the Fe concentration. Chromium oxidation capacity of the birnessite increased with increasing the Fe concentration. Iron in the precipitating solution poisoned crystal growth by adsorption on the surface and increased nucleation. Since Fe is a common constituent under pedogenic environment and Fe and Mn oxides often coexist in Mn oxide nodules, the birnessite with small particle, low crystallinity, and high chemical reactivity is the form which is more likely to be formed in soils. The high CEC ($140cmol_ckg^{-1}$) and oxidation capacity of birnessite indicate that birnessite can be used in environment and agriculture.

Redescription of Newly Recorded Ciliate, Euplotes muscorum(Ciliophora: Polyhymenophora: Hypotrichida) and Comparison with Related Species from Korea (한국산 미기록 이끼토양자라하모충(섬모충문: 다막충강: 하모충목)의 재기재와 유사종의 비교)

  • The euplotid hypotrich collected from a puddle, Jeju-do in 2002 and cultured in the laboratory was identified as Euplotes muscorum Dragesco, 1970. The species is reported for the first time from Korea. The description was based on the observation of living specimens, protargol impregnation and biometrical analysis. This species is characterized by following diagnosis: 63-78 ${\mu}m$ in length, 40-52 ${\mu}m$ in width in vivo, 9 frontoventral cirri, 5 transverse cirri,4 caudal cirri, 1 micro- and macronucleus, adoral zone of membranelles with 32-36 adoral membranelles covering approximately 2/3 of body length, 8 dorsal kineties, mid-dorsal kinety with 20-24 cilia 3nd dargyrome complex type. This species with 9 frontoventral cirrotype is very similar to E. muscicola Kahl, 1932. The differences between these two species are: E. muscorum has 8 dorsal kineties and complex dargyrome type, while E. muscicola 9 dorsal kineties and multiple dargyrome type.

  • Microorganism producing glutaryl 7-aminodeacetoxycephalosporanic acid (GL-7-ADCA) acylase was screened from soil. The microorganism was identified as Alcaligenes sp. J-421 by its morphology and biochemical properties. Cultural conditions of Alcaligenes sp. J-421 were investigated for the production of GL-7-ADCA acylase. Optimum medium composition was 1% glucose, 1% beef extract, 0.5% yeast extract, 0.2% monosodium L-glutamate, 0.1% glutaric acid, 0.2% NaCl, 0.5% $K_2$ $HPO_4$, and 0.05% $CuSO _4{\cdot}5H_2O$. Optimum cultivation conditions for the production of the enzyme in 5 l jar fermentor were $37^{\circ}C$, tip speed 300 rpm, aeration 1 vvm. Optimum reaction pH of the enzyme was 8.0 and the enzyme was stable at pH7.0-11.0.

  • Nematophagous fungi can capture, kill, and digest nematodes using a specific capturing organ. Of the nematophagous fungi, while Arthrobotrys flagrans and A. superba have been described previously, certain characteristics have not been described. For a detailed description of the two nematophagous fungi, the fungi were isolated from soil samples and produced in a pure culture. Morphological characteristics, such as predatory ability (according to the nematode species), shape, and size of predatory organ, conidia, and chlamydospore were investigated and they were used for identification of the fungal isolates along with molecular phylogenetic analysis. Furthermore, this study provides the classification key for 21 nematophagous species.

  • As a subset investigation to discover indigenous prokaryotic species in Korea, a total of 21 bacterial strains assigned to the class Betaproteobacteria were isolated from a wide range of environmental samples which collected from fresh water, roots of plants, mineral water and soil from ginseng farm. Phylogenetic analysis based on 16S rRNA gene sequences indicated that 21 isolated strains were most closely related to the class Betaproteobacteria, with high 16S rRNA gene sequence similarity (>99.1%) and constructed a robust phylogenetic clade with the closest species in the class Betaproteobacteria. These isolated species have no previous report or publication in Korea; therefore 17 species in 14 genera of 6 families in the order Burkholderiales, 1 species in the order Methylophilales, 2 species in 2 genera of 1 family in the order Neisseriales are reported for betaproteobacterial species found in Korea. There is no official report that these 26 species have been described in Korea; therefore 14 strains for the order Flavobacteriales and two strains for the order Cytophagales were assigned to the class Bacteroidetes, and 8 strains for the order Bacillales and 4 strains for the order Lactobacillales were assigned to the class Firmicutes are reported for new bacterial species found in Korea.
