Changes in land-use type can affect soil and water properties in stream ecosystems. This study examined the effects of different land-use types on biogeochemical properties and microbial activities of a stream. We collected water and sediment samples in a stream at three different sites surrounded by varying land-use types; a forest, a radish field and a rice paddy. Nitrogen contents, such as nitrate, nitrite and total nitrogen in the stream water body, showed significant differences among the sampling sites. The highest nitrogen values were recorded at the site surrounded by cropland, as fertilizer runoff impacted the stream. Soil organic matter content in the sediment showed significant differences among sites, with the highest content exhibited at the forest mouth site. These differences might be due to the organic matter in surrounding terrestrial ecosystems. Microbial activities determined by extracellular enzyme activities showed similar values throughout all sites in the water body; however, the activities in the sediments exhibited the highest values near the forest site and mirrored the soil organic matter content values. From these results, we conclude that different land-use types are important factors affecting water and sediment properties in stream ecosystems.
A laboratory experiments were carried out to study the urea transformation and kinetics of soil urease in paddy soils with different properties. Urea transformation in paddy soils followed first-order kinetics, the rate of urea hydrolysis and the first-order constant was higher in the soil with high total urease activity (TUA) and accumulated urease activity (AUA) than those with low TUA and AUA. The values of Km and Vmax indicated that the Km values of accumulated urease in the soils were different in each soils and lower than that of microbial urease. However, the Km values of microbial urease were nearly same each other.
The study was initiated to compare crop productivity as affected by a long-term application with homemade liquid fertilizers in leading organic cucumber (Cucumis sativus L.) farms in Suncheon and Kimcheon provinces. A Suncheon farm have applied an EM (effective organism) liquid fertilizer for one year and fifteen years, designating as EM 1-year and EM 15-year plots, respectively, with 4-year and 5-year application of native microbes-liquid fertilizer in Kimcheon farm, designating as Micro 4-year and Micro 5-year plots, respectively. pH in the EM-liquid fertilizer was high to approximately 7.7, and EC in the Micro-liquid fertilizer was 0.1 dS/m higher than those of EM-liquid fertilizer, with similar macro-nutrient concentrations observed in the both liquid fertilizers. Soil EC was the highest to the 10.0 dS/m for the liquid fertilizer with EM 1-year and showed less than 1.5 dS/m for other liquid fertilizer plots. Micro-liquid fertilizer plots had soil OM contents less than 20 g/kg, which was approximately two times less than those of EM plots. Soil microbial properties were not significantly different among the liquid fertilizer plots. SPAD and PS II values were significantly increased by EM 15-year plots with high levels of soil OM and EC. Liquid fertilizer plot with EM 1-year had high concentrations of T-N, Ca, and Na in the cucumber crops but low concentrations of P and Mg, in particular for low K of 1.2% which was two times less than those of desired level for an optimum cucumber growth. The lowest fruit yield was observed for the liquid fertilizer plot with EM 1-year with the highest soil EC accumulated. Liquid fertilizer plot with EM 15-year produced the expanded volume of crop canopy and increased fruit yield. Therefore, long-term of continuous application with an organic liquid fertilizer would have sustainably improved soil stability and the crop productivity.
Studies were conducted during 2 months from May of 1997 to evaluate the effects of pig manure compost(PMC) on soil microbial flora. To do so, a field experiment of Chinese cabbage(Brassica campestris L.) was conducted in a randomized block design on a sandy loam soil and microbial floral characteristics in soils were analyzed. Treatments to control included the application of PMC at (A) $8Mg\;ha^{-1}$CM-8), (B) $29Mg\;ha^{-1}$(CM-2,9), and (C) $57Mg\;ha^{-1}$(CM-57), and of chemical fertilizer(D) at $320N-80P_2O_5-200K_2O\;kg\;ha^{-1}$(NPK). In each treatment, the rhizosphere and non-rhizosphere soils were tested for the analysis of microbial populations. The populations of bacteria, actinomycetes, and fungi increased in soils with the applications of PMC and chemical fertilizer, but that of Bacillus sp. decreased. However, the population of fluorescent Pseudomonas sp. was reduced in NPK plots only. With increasing application rates of PMC, the number of colony forming units(cfu) of bacteria (Pseudomonas sp. and actinomycetes) and fungi increased. in all PMC-treated plots, the population density peaked at early growth stage for bacteria(including Bacillus sp.), at late growth for fluorscent Pseudomonas sp., and at harvest for fungi and actinomycetes. The rhizosphere effect was greatest for fluorscent Pseudomonas sp. As the application rates of PMC increased, Total N, organic matter, available phosphate, and exchangeable -K, -Ca, and -Mg increased compared to control, but soil pH was lowered. In NPK plots, EC was 3.4-fold and exchangeable K was 5-fold higher than control.

This study was carried out to evaluate the application effects of fish meal liquid fertilizer on soil characteristics and growth of cucumber for organic cultivation. Cucumber in greenhouse was transplanted on March $31^{th}$ in 2016, and the experimental treatments involve six treatments: No fertilizer, 0, 25, 50, and 100 mg/L N application by fish meal liquid fertilizer and chemical fertilizer. In the results of soil chemical property, application of 100 mg/L of fish meal liquid fertilizer showed a significant differences in pH, K, and Mg contents. The soil microbial community varied in relation to the fish meal liquid fertilizer treatments. Microbial biomass was lower in the chemical fertilizer than in the liquid fertilizer treatment. Result of principal component analysis obtained from Ecoplate showed that fish meal liquid fertilizer treatments, no liquid fertilizer, chemical fertilizer, and no fertilizer were divided into distinct groups, with the no fertilizer treatment located furthest from the other treatments. There were no significant differences in plant height of cucumber between the fish meal liquid fertilizer treatments and chemical fertilizer treatments. Also, the cucumber yield did not vary significantly between the concentrations of liquid fertilizers, and there were also no significant differences in the yield among the fish meal liquid and chemical fertilizer treatments. In conclusion, it is suggested that the application of fish meal liquid fertilizer can be used as a additional fertilizer for cucumber production with organic culture in greenhouse.
Nutrient availability is a critical component of agroecosystems, and is relevant to both above- and below- ground interactions. The principal objective of this study was to determine how the cessation of fertilization affects the communities of weeds and soil organisms in a corn/wheat field. Changes in dominant weed species, substrate-induced respiration, and the population density of nematodes and microarthropods were evaluated. Microbial substrate-induced respiration (SIR) and the population density of microarthropods decreased following the cessation of fertilization and were partly correlated with the aboveground weed biomass. The cessation of organic fertilizer application but continuing application of inorganic fertilizer reduced the population density of nematodes. In response to the cessation of fertilization, weed communities were dominated by species with little dependency on fertilization. Amaranthus retroflexus was identified as the most dominant species in the corn field; however, it was replaced by Digitaria ciliaris after the cessation of fertilization. In the wheat field, the cessation of fertilization led to a rapid reduction in the biomass of most weeds, except for Vicia angustifolia, supposedly as the result of symbiotic nitrogen fixation. Additionally, the fact that weed biomass was partially correlated with SIR or the population density of microarthropods may reflect a mutual feedback between soil organisms and weeds. The results indicate that the cessation of fertilization alters communities of weeds and soil organisms through changes in weed biomass and interactions with symbiotic microorganisms.
In order to study a relationship between soil nutrients and rice brown spot occurrence, paddy field soils, rice grains and straws collected from different paddy fields with different disease degrees of brown spots were analyzed for inorganic nutrients. Brown spot was prevalent in the rice grown in nutrient-deficient soils, which is especially low in macronutrient elements (phosphoric acid, potassium, silicic acids) and micronurients (calcium, magnesium). The soil, however, was high in sodium while organic nutrients and pH level were similar to others. The rice straws with severe brown spot were low in inorganics such as ferrous, copper, T-N, and $P_{2}O_{5}$ while the rice grains with brown spot were low in ferrous, MgO, Zn, and Mn. In the analysis of field type and nitrogen level, the highest disease severity was found in sandy-type field soil, followed by salty-type field soil and disease severity decreased as application level of nitrogen fertilizer increased. As a summary, the most important factor for effective brown spot control in rice is maintenance of proper nutrients in sandy-type field and control of sodium level in salty-type field soil.
T-RFLP analysis and clone sequencing analysis based on bacterial 16S rDNA were conducted to assess bacterial community structure and diversity in Zoysia japonica soil treated with liquid fertilizer containing amino acids(LFcAA) after spray with herbicide. The results of T-RFLP (terminal restriction fragment length poly-morphism) analysis using restriction enzyme Hae III showed that the T-RFs of various size appeared evenly in the 32 clones of KD3 and 38 clones of KD4 respectively that had been treated with liquid fertilizer containing amino acid(LFcAA) compared to 23 clones of KD2 hat had not been treated with LFcAA. The microbial com- munity structure in KD2 appeared less diverse than those in KD3 and KD4. Analysis of partial sequences for 110 clones from KDI (control), KD2 (non-treated), KD3 (LFcAA 1X), KD4 (LFcAA 2X), respectively, revealed that most bacteria were related with uncultured bacteria in a 16S rDNA sequence similarity range of 91-99% through blast search. Otherwise, the other clones were members of proteobacteria, Acidobacteria, Act-inobacteria, Sphingobacteria and Planctomyces groups. Especially in KD4, members of Alpha Proteobacteria, Rhizobiales, Sphigomonadales, Caulobacterales, Gamma Proteobacteria, the genus Pseudomonas, Betapro-teobacteria, Nitrosomonadales and genus Nitrosospira appeared to be dominant. In addition, Acidobacteria group, Actinobacteria group, Planctomycetacia and Sphingobacteria were also shown. The microbial com-munity structure in Z. japonica soil sprayed with herbicide was affected by LFcAA.
This study was conducted to investigate the effect of pyroligneous acids on urea hydrolysis for the purpose of inhibiting ammonia volatilization during urea fertilizer application. Different types of synthetic urease inhibitors have been searched and developed, but their use is limited due to varying inhibition effects on soil urease, and environmental problems. In this study, the effect of pyroligneous acids, a natural substance, on urea hydrolysis in soil was evaluated by analyzing inhibition of urease activity. Pyroligneous acids inhibited plant urease and microbial urease activity, as well as soil urease with various urease complex. In addition, pyroligneous acids exhibited non-competitive urease inhibition effect through urease kinetics and inhibited urea hydrolysis in the soil. This study showed that pyroligneous acids treatment with urea fertilizer decreases the loss of urea fertilizer, improves the efficiency of nitrogen application on plant and reduces the amount of nitrogen fertilizers applied in soil.
The sustainability of conventional agriculture which is characterized by input dependent and ecologically simplified food production system is vague. Chemicals and present practices used in agriculture are not only costly but also have widespread implications on human and animal health, food quality and safety and environmental quality. Thus there is a need for alternative farming practices to sustain food production for the escalating population and conserve environment for future generations. The present research scenario in the area of plant microbe interactions for maintaining sustainable agriculture suggests that the level of internal regulation in agro-ecosystems is largely dependent on the level of plant and microbial diversity present in the soil. In agro-ecosystems, biodiversity performs a variety of ecological services beyond the production of food, including recycling of nutrients, regulation of microclimate and local hydrological processes, suppression of undesirable organisms and detoxification of noxious chemicals. Controlling the soil microflora to enhance the predominance of beneficial and effective microorganisms can help improve and maintain soil chemical and physical properties. The role of beneficial soil microorganisms in sustainable productivity has been well construed. Some plant bacteria referred to as plant growth-promoting rhizobacteria (PGPR) can contribute to improve plant growth, nutrient uptake and microbial diversity when inoculated to plants. Term PGPR was initially used to describe strains of naturally occurring non-symbiotic soil bacteria have the ability to colonize plant roots and stimulate plant growth PGPR activity has been reported in strains belonging to several other genera, such as Azotobacter, Azospirillum, Arthrobacter Bacillus, Burkhokderia, Methylobacterium, and Pseudomonas etc. PGPR stimulate plant growth directly either by synthesizing hormones such as indole acetic acid or by promoting nutrition, for example, by phosphate solubilization or more generally by accelerating mineralization processes. They can also stimulate growth indirectly, acting as biocontrol agents by protecting the plant against soil borne fungal pathogens or deleterious bacteria. Present review focuses on some recent developments to evolve strategies for better biotechnological exploitation of PGPR's.
