Landfarming and a microbial agent, Bioil-D, were used to treat diesel-contaminated soil. The microbial agent was applied to the contaminated soil in a concentration of 5$\times$10$^2$ cfu/soil(g) and the total amount of microbial agent, 210$\ell$, was sprinkled on the soil four times for 24 days during 50 day-remediation period. The remediation goal, lower than 800mg/kg of TPH, was achieved from 5, 707 mg/kg of TPH within 50 days. The total number and activity of indigenous microorganisms were increased by 100 times after the microbial agent, Bioil-D, was applied to the contaminated soil.
Organic farming systems based on ecological concepts have the potential to produce sustainable crop yields with no decline in soil and environmental qualities. Recent expansion of sustainable agricultural systems, including organic farming, has brought about need for development of sustainable farming systems based on value judgments for key properties of importance for farming. Chemical and microbiological properties were chosen as indicators of soil quality and measured at soil depth intervals of 5-20 and 20-35 cm in conventional and organic-based apple orchards located in Yeongchun, Gyeongbuk. The orchards were two adjacent fields to ensure the same pedological conditions except management system. Soil pH in organic farming was around 7.5, whereas below 6.0 in conventional farming. Organic farming resulted in significant increases in organic matter and Kjeldahl-N contents compared to those found with conventional management. Microbial populations, biomass C, and enzyme activities (except acid phosphatase) in apple orchard soil of organic farming were higher than those found in conventional farming. Higher microbial quotient ($C_{mic}/C_{org}$ ratio) and lower microbial metabolic quotient for $CO_2(qCO_2)$ in organic farming confirmed that organic farming better conserves soil organic carbon. Biological soil quality indicators showed significant positive correlations with soil organic matter content. These results indicate organic-based farming positively affected soil organic matter content, thus improving soil chemical and biological qualities.

In our previous studies, we selected three antagonistic bacteria, KJ1R5, KJ2C12, and KJ9C8 against Phytophthora capsici, the casual agent of Phytophthora blight of pepper. For elucidating production, root colonization, and total microbial activity were investigated. The dual culture assay was accomplished to elucidate existence of antibiotics. In this assay, any antagonistic bacteria did not inhibit growth of six important fungal plant pathogens, suggesting that these antagonists do not produce antibiotics. root surface or rhizosphere soil colonizations were examined with spontaneous rifampicin-resistant mutants equal to antagonistic ability of wild types. KJ2C12 colonized consistently rhizosphere soil while yellowish colonies of KJ1R5 and KJ9C8 well colonized root surfaces and rhizosphere soil. Total microbial activity in pots treated with the antagonistic bacteria was measured using fluorescein diacetate hydrolysis. total microbial activity of three antagonistic bacteria treatments was significantly higher than that of buffer-treated control until 4days after treatment. However, total microbial activity of treatment of three antagonistic bacteria decreased after 7 days. These results indicate that the antagonistic bacteria, KJ1R5 and KJ9C8 colonized and protected roots well against Phytophthora blight of pepper through competition of infection courts, especially competitions.
The effect of oyster shell meal, which is made of a simple crushing and alkaline calcium materials, on soil microbial properties, microbial biomass C, N and P contents, and enzyme activities were evaluated in silt loam soil. The oyster shell meal fertilizer was added at the rates of 0, 4, 8, 12 and $16Mg\;ha^{-1}$. Microbial biomass C, N and P contents were significantly increased with increasing application of oyster shell meal. Soil enzyme activities, such as urease, ${\beta}$-glucosidase and alkaline phosphomonesterase were increased significantly by shell meal application, due to increased soil pH towards neutral range and increased nutrient availability in soil. In particular, the increased microbial biomass P content and phosphomonoesterase activities were strongly correlated with available P content in soil. Conclusively, oyster shell meal fertilizer could be a good supplement to improve soil microbial activities.
The relationship between pb, zn, cd, and the microbial biomass and activity were investigated in three public park soils of central and outer london. Variability with distance from the roadside and profile were studied. The heavey metal concentrations were the highest in hampstead heath and hyde park with high trafic density and the lowest in hainault. The highest concentrations of heavy metals were found adjacent to the roadside in the upper parts of the soil profile. Dehydrogenase activity, adenosine tri-phosphate and ergosterol contents used as indices of micrbial biomass and activity, were generally higher in hainadult, and also higher in the upper pats of the soil profile. Simple regression analysis indicated that the microbial biomass and activity were affected significantly by moisture content, water holding capacity, total organic carbon, total nitrogen, and organic mater rather than heavy metal concentration. Highest inputs of nitrogen and carbon were associated with high inputs of heavey metals, all three being derived from vehicle emissions adjacent to the road. The hyde park and hampstead heath microbial populations were able to respond to the c and n input positively by increase in biomass and activity, whereas the hainault populations could not. This rsult suggrsts adaptation in he former to heavy matals, but not in the latter.
Consumers' interest and government's support for the fruits rapidly increased organic fruit productions. This study was examined to compare the soil physicochemical and microbial properties of orchards soil in conventionally and organically management systems. Organic cultivation had lower soil bulk density, solid phase, and penetration resistance than the conventional cultivation. Soil pH and organic matter contents increased from March to August, and the values were greater in the organic cultivation than the conventional cultivation. Total nitrogen (N) and phosphorous concentrations decreased from March to August, and the organic soils had greater N but lower phosphorous concentrations than the conventional soils. Soil microbial carbon biomass increased 36% and 15% for organic and conventional cultivations, respectively, from March to August. Soil microbial N biomass was greater in June than March or August, and the organic cultivation had a greater biomass N compared to the conventional cultivation. Soil dehydrogenase and chitinase activities were greater in June than in March or August. ${\beta}$-glucosidase activity declined in both cultivations, while the phosphatase activity increased. Organic cultivation had greater enzyme activities in March, June, and August, except for the acid phosphatase activity in June.
The relationship between microbial activity and physico-chemical characteristics of golf course soils were investigated in Kyonggi Province with different open years. The increasing tendency of soil PH with depth seemed to the result of mineral leaching. Exchangeable $Ca^2$+ , $Mg^2$+ and K+ showed relatively higher concentration in lower horizon indicating the leaching of minerals. The minerals were heavily leached in older golf course. There was a significant positive correlation between pH and $Ca^2$+.The contents of organic matters, total nitrogen and phosphorus were high in higher horizon.Dehydrogenase activity(DHA) and the content of ATP were determined as an index of soil microbial activity. DHA value was significantly different with golf club as well as golf courses ac-cording todifferent open year. This indicates that DHA was affected by several fertilizertreatments rather than herbicide pesticide treatments. DHA was positively correlated with water holding capacity and total nitrogen. The content of ATP was high in fairway and in golf course with older open year, showing the similar tendency with the results of DHA.
The relationships between microbial activity and physico-KDICical characteristics of soils were investigated in three golf courses of Kwanak, Gold and Korea Country Clubs, with different open years. The soil samples were collected in tee, fairway and rough. There were ranges of 4.80-5.55 in pH, $25.55-98.50{\mu}S$ / cm in conductivity, 10.96-16.73% in moisture content, 0.18-0.36g / g in water holding capacity, 3.68-5.39% in organic matter, and 0.10-0.25% in total nitrogen. Dehydrogenase activity(DHA) as an index of soil microbial activity was determined. DHA values of soil were $69.83-314.43{\mu}$g / g in three courses and showed the order of Kwanak>Gold>Korea Country Club with open year. This indicates that DHA was affected by several fertilizer treatments rather than herbicide and pesticide treatments. DHA was significantly different with golf clubs as well as golf courses and positively correlated with water holding capacity and total nitrogen.
Soil enzyme and microbial activities are affected by fertilizer and compost applications and can be used as sensitive indicators of ecological stability. Microbial population and soil enzymes viz., dehydrogenase, urease, acid phosphatase and aryl-sulphatase were determined in the long-term fertilizer and compost applied paddy soil. Soil samples were collected from the four treatments (control, compost, NPK and compost+NPK). Long-term NPK+compost application significantly increased activities of urease, dehydrogenase and acid phosphatase than all other treatments. The compost application enhanced activities of urease, dehydrogenase and acid phosphatase than the NPK application. However, arylsulfatase activity was not significantly different between compost and fertilizer application. The highest microbial population was recorded in the NPK+compost treatment. The compost application also resulted in higher microbial population than the NPK application. The above results indicate that ecological stability could be maintained by application of compost alone or with NPK.
In the context of artificial groundwater recharge, a reactive soil column at pilot-scale (4.5 m depth and 3 m in diameter) fed by treated wastewater was designed to evaluate soil filtration ability. Here, as a part of this project, the impact of treated wastewater filtration on soil bacterial communities and the soil's biological ability for wastewater treatment as well as the relevance of the use of multi-bioindicators were studied as a function of depth and time. Biomass; bacterial 16S rRNA gene diversity fingerprints; potential nitrifying, denitrifying, and sulfate-reducing activities; and functional gene (amo, nir, nar, and dsr) detection were analyzed to highlight the real and potential microbial activity and diversity within the soil column. These bioindicators show that topsoil (0 to 20 cm depth) was the more active and the more impacted by treated wastewater filtration. Nitrification was the main activity in the pilot. No sulfate-reducing activity or dsr genes were detected during the first 6 months of wastewater application. Denitrification was also absent, but genes of denitrifying bacteria were detected, suggesting that the denitrifying process may occur rapidly if adequate chemical conditions are favored within the soil column. Results also underline that a dry period (20 days without any wastewater supply) significantly impacted soil bacterial diversity, leading to a decrease of enzyme activities and biomass. Finally, our work shows that treated wastewater filtration leads to a modification of the bacterial genetic and functional structures in topsoil.
