Biological and Physico-chemical Properties of Antifungal Cyclic Lipopeptides Produced by Pseudomonas cepacia Strains (Pseudomonas cepacia 균주가 생산하는 항진균성 Cyclic Lipopeptide의 생물학적 및 물리 화학적 특성)

  • Five strains AF027, AF069, AF2001, AF2011 and SD02 of Pseudomonas cepacia were isolated from soil, and the antifungal cyclic lipopeptides(CLP) i.e, CLP027A, CLP069A, Cepacidine A, CLP2011A and CLP02A were produced from each strains, respectively. Nitrogen and carbon sources in media were proved to be important factors for the production of CLP and among them, polypeptone-S, glucose and fructose were the most effective. It appeared that compounds CLP027A and CLP069A were identical with Cepacidine A and Xylocandine A, respectively. contain aspartic acid as amino acid component, are differentiated from Xylocandine A containing asparagine. Although molecular weight, amino acid composition and UV spectrum of CLP2011A and CLP02A are same with those of Cepacidine A, it is postulated that these compounds are not identical with Cepacidine A when the antifungal spectra and antifungal activity were compared to those of Cepacidine A.

Remediation of Arsenic Contaminated soils Using a Hybrid Technology Integrating Bioleaching and Electrokinetics (생용출과 전기동력학을 연계한 통합기술을 이용한 비소 오염 토양의 정화)

  • The objective of the study was to develop a hybrid technology integrating biological and physicochemical technologies to efficiently remediate arsenic contaminated lands such as abandoned mine area. The tailing soil samples contaminated with As at a high level were obtained from Songchon abandoned mine, and the content of arsenic and heavy metals as well as physicochemical properties and mineral composition were investigated. In addition, two sets of sequential extraction methods were applied to analyze chemical speciations of arsenic and heavy metals to expect their leachability and mobility in geoenvironment. Based on these geochemical data of arsenic and heavy metal contaminants, column-type experiments on the bioleaching of arsenic were undertaken. Subsequently, experiments on the hybrid process incorporating bioleaching and electrokinetics were accomplished and its removal efficiency of arsenic was compared with that of the individual electrokinetic process. With the results, finally, the feasibilty of the hybrid technnology was evaluated. The arsenic removal efficiencies of the individual electrokinetic process (44 days) and the hybrid process incorporating bioleaching (28 days) and electrokinetics (16 dyas) were measured 57.8% and 64.5%, respectively, when both two processes were operated in an identical condition. On the contrary, the arsenic removal efficiency during the bioleaching process (28 days) appeared relatively lower (11.8%), and the result indicates that the bioleaching process enhanced the efficacy of the electrokinetic process as a result of mobilization of arsenic rather than removed arsenic by itself. In particular, the arsenic removal rate of the electrokinetics integrated with bioleaching was observed over than 2 times larger than that obtained by the electrokinetics alone. From the results of the study, if the bioleaching which is considered a relatively economic process is applied sufficiently prior to electrokinetics, the removal efficiency and rate of arsenic can be significantly improved. Consequently, the study proves the feasibility of the hybrid process integrating both technologies.

Inhibitory Substance on the Snake Venoms Produced by Penicillium sp. (사독의 조해물질에 관한 연구)

  • One strain of Penicillium sp. (175-66-B), isolated from soil, was able to produce a substance that has a strong inibition activity against the Agkistrodon and Trimeresurus venoms. In this experiment, the chemical and biological properties of the sample were investigated. As an inhibitory substance, it was effective to the proteinase, hemorrhagic and lethal factors of Agkistrodon and Trimeresurus venoms, and also effective to several fractions of the proteinases and hemorrhagic factors of Agkistrodon halys blomhoffi venom. Moreover, in the addition of prednisotone, it was more effective for the cure of the mouse envenomated with the venom amount of two fold of MLD$_{100}$. This substance was very stable to the acid, alkali and heat. Its melting point was high enough to sublime at 222$^{\circ}C$ without any decomposition. This sample was easily dissolved only in hot water, but not in several organic solvents except for a little dissolution in elate. It did not have the chelating activity. It had very strong specificity to the snake venoms. but its activity was depressed by the addition of zinc or cupric salts. This sample had no acute toxicity to the mouse. Its chemical formula was $C_{16}$ $H_{12}$$N_2$ $O_{10}$ with the molecular weight of about 392. It has two epoxy groups and four carboxyl radicals, but amino, nitrite and nitrate radicals, unsaturated bonds and aromatic ring were not detected. Effects of streambed geomorphology on nitrous oxide flux are influenced by carbon availability (하상 미지형에 따른 N2O 발생량 변화 효과에 대한 탄소 가용성의 영향)

  • Denitrification in streams is of great importance because it is essential for amelioration of water quality and accurate estimation of $N_2O$ budgets. Denitrification is a major biological source or sink of $N_2O$, an important greenhouse gas, which is a multi-step respiratory process that converts nitrate ($NO_3{^-}$) to gaseous forms of nitrogen ($N_2$ or $N_2O$). In aquatic ecosystems, the complex interactions of water flooding condition, substrate supply, hydrodynamic and biogeochemical properties modulate the extent of multi-step reactions required for $N_2O$ flux. Although water flow in streambed and residence time affect reaction output, effects of a complex interaction of hydrodynamic, geomorphology and biogeochemical controls on the magnitude of denitrification in streams are still illusive. In this work, we built a two-dimensional water flow channel and measured $N_2O$ flux from channel sediment with different bed geomorphology by using static closed chambers. Two independent experiments were conducted with identical flume and geomorphology but sediment with differences in dissolved organic carbon (DOC). The experiment flume was a circulation channel through which the effluent flows back, and the size of it was $37m{\times}1.2m{\times}1m$. Five days before the experiment began, urea fertilizer (46% N) was added to sediment with the rate of $0.5kg\;N/m^2$. A sand dune (1 m length and 0.15 m height) was made at the middle of channel to simulate variations in microtopography. In high- DOC experiment, $N_2O$ flux increases in the direction of flow, while the highest flux ($14.6{\pm}8.40{\mu}g\;N_2O-N/m^2\;hr$) was measured in the slope on the back side of the sand dune. followed by decreases afterward. In contrast, low DOC sediment did not show the geomorphological variations. We found that even though topographic variation influenced $N_2O$ flux and chemical properties, this effect is highly constrained by carbon availability.

  • Along with Cs-137 (half-life: 30.17 years), Sr-90 (half-life: 28.8 years) is one of the most important environmental monitoring radioactive elements. Rapid and easy monitoring method for Sr-90 using Laser-Induced Breakdown Spectroscopy (LIBS) has been studied. Strontium belongs to a bivalent alkaline earth metal such as calcium and has similar electron arrangement and size. Due to these similar chemical properties, it can easily enter into the human body through the food chain via water, soil, and crops when leaked into the environment. In addition, it is immersed into the bone at the case of human influx and causes the toxicity for a long time (biological half-life: about 50 years). It is a very reductive and related with the specific reaction that makes wet analysis difficult. In particular, radioactive strontium should be monitored by nuclear power plants but it is very difficult to be analysed from high-cost problems as well as low accuracy of analysis due to complicated analysis procedures, expensive analysis equipment, and a pretreatment process of using massive chemicals. Therefore, we introduce the Laser-Induced Breakdown Spectroscopy (LIBS) analysis method that analyzes the elements in the sample using the inherent spectrum by generating plasma on the sample using pulse energy, and it can be analyzed in a few seconds without preprocessing. A variety of analytical plates for samples were developed to improve the analytical sensitivity by optimizing the laser, wavelength, and time resolution. This can be effectively applied to real-time monitoring of radioactive wastewater discharged from a nuclear power plant, and furthermore, it can be applied as an emergency monitoring means such as possible future accidents at a nuclear power plants.
