  • New anionic oligomeric surfactants, sodium bis(alkyl decaoxyethylene) sulfonated succinates, had been synthesized through the addition reaction of sodium hydrogen sulfite to bis(alkyl decaoxyethylene) maleates. Bis(alkyl decaoxyethylene) maleates were obtained by esterification with maleic anhydride and long chain alkyl decaoxyethylene ethers which were also obtained by addition ethylene oxide 10 mole to straight long chain alcohol with alkyl group having from 10 to 18 carbon atoms, their structure of the synthetic compounds have been characterized with IR. $^{1}H$ NMR and elemental analysis respectively.

Thiosulfate Oxidation and Mixotrophic Growth of Methylobacterium goesingense and Methylobacterium fujisawaense

  • The mixotrophic growth with methanol plus thiosulfate was examined in nutrient-limited mixotrophic condition for Methylobacterium goesingense CBMB5 and Methylobacterium fujisawaense CBMB37. Thiosulfate oxidation increased the growth and protein yield in mixotrophic medium that contained 150mM methanol and 20mM sodium thiosulfate, at 144 h. Respirometric study revealed that thiosulfate was the most preferable reduced inorganic sulfur source, followed by sulfite and sulfur. M. goesingense CBMB5 and M. fujisawaense CBMB37 oxidized thiosulfate directly to sulfate, and intermediate products of thiosulfate oxidation such as polythionates, sulfite, and sulfur were not detected in spent medium and they did not yield positive amplification for tested soxB primers. Enzymes of thiosulfate oxidation such as rhodanese and sulfite oxidase activities were detected in cell-free extracts of M. goesingense CBMB5, and M. fujisawaense CBMB37, and thiosulfate oxidase (tetrathionate synthase) activity was not observed. It indicated that both the organisms use the "non-S4 intermediate" sulfur oxidation pathway for thiosulfate oxidation. It is concluded from this study that M. goesingense CBMB5, and M. fujisawaense CBMB37 exhibited mixotrophic metabolism in medium containing methanol plus thiosulfate and that thiosulfate oxidation and the presence of a "Paracoccus sulfur oxidation" (PSO) pathway in methylotrophic bacteria are species dependant.

생쥐난자에 있어서 투명대 경화현상이 체외수정에 미치는 영향 II. 투명대 경화 현상을 유도하는 원인물질의 구명 (Effect of Zona Hardening on In Vitro Fertilization in Mouse Oocytes II. Analysis of Materials Causing Zona Hardening)

  • In order to demonstrate whether ovoperoxidase hardens the zona of oocytes activated by incubating in M-S buffer supplemented with 20$\mu$M of Ca-ionophore A 23187, the effect of peroxidase inhibitors(250mM pheylhydrazine, 28mM sodilum sulfite, 350mM glycine ethyl ester and 50mM sodium azide), tyrosine analogue(12.5mM tyramine) and exogeneous peroxidase(50$\mu\textrm{g}$/ml horseradishperoxidase ; HRP) on zona hardening in ionorphore-treated oocytes were investigated. The results obtained from thses experiments were summarized as follows : 1. The zona solubility (t50) of ionophore-activated and DMSO-treated oocytes at 1, 2 and 3 hr of culture were 25.0, 31.6 and 40.6min., and 9.7, 10.8 and 15.5 min., respectively. The longest time required for zona lysis of ionophore activated oocytes at 1 hr after onset of ionophore treatment. The diferences int50 for zona was significantly greater as compared to DMSO-treated controls(P<0.01). 2. The inhibition rates of hardening in the oocytes treated with the phenylhydrazine, sodium sulfite, glycine ethyl ester and sodium azide, were 23.8, 61.9, 95.2 and 23.8%l, respectively, and the tyramine, was 14.3%. Several known peroxidase inhibitors and tyrosine analogue were blocked zona hardening in ionophore activated oocytes. 3. The treatment of exogeneous peroxidase promoted the zona hardening of activated oocytes but not unactivated oocytes. These resuls indicate that the ovoperoxidase apparently catalyzes the hardening of the zona following ionophore activation of mouse oocytes.

우렁쉥이 이용에 관한 연구 -4. 우렁쉥이 육의 갈변 및 그 방지- (Utilization of Ascidian, Halocynthia roretzi -4. Browning of Ascidian meat, Halocynthia roretzi and Its Prevention-)

  • 최근 과잉생산단계에 이르고 생식이외의 소비방법이 없는 우렁쉥이는 봄부터 여름 사이에 집중적으로 대량출하되므로서 선도의 보존과 소비에 한계가 들어나고 있다. 또한 젓갈 등의 제품가공시에는 처리중에 일어나는 변색 즉 갈변으로 인한 품질의 저하가 문제가 되고 있다. 따라서 본 연구에서는 (1)갈변의 원인을 밝히고, (2)그에 관여하는 여러가지 반응요인, 그리고 (3)갈변을 방지하기 위한 처리방법 등에 관해서 검토하였다. 1. 우렁쉥이 육과 내장에서 추출한 조효소액을 L-tyrosine과 catechol을 기질로 하여 polyphenol oxidase 활성을 측정한 결과 L-tyrosine의 경우 육과 내장에서 20.6 및 63.5 unig/mg의 비활성도를 각각 나타냈으며 특히 내장의 경우가 육보다도 3배정도 높았다. 이 결과 보아 갈변의 주된 원인이 tyrosinase의 작용이 아닌가 추정된다. 2. 우렁쉥이 조효소액의 tyrosinase활성 최적 pH는 6.0, 최적온도는 $30^{\circ}C{\sim}35^{\circ}C$였으며, 열안정성은 $70^{\circ}C$에서 9분, $80^{\circ}C$에서 3분 그리고 $90^{\circ}C$$100^{\circ}C$에서 1분 처리로 완전히 실활하였다. 3. 효소활성 저해효과에 있어서 육의 경우 0.5mM의 농도에서는 ascorbic acid, sodium bisulfite, cystein, citric acid, cyanide 등이 효과가 있었고, 0.1mM의 농도에서 ascorbic acid, sodium bisulfite, cystein만이 유효하였다. 그러나 내장의 경우와 같이 효소활성이 클 때는 sodium bisulfite만이 tyrosinase활성을 저해했다. 4. 갈변방지를 위하여 산, buffer, sulfite, 기타 갈변억제처리 효과를 검토한 결과 0.2M citric acid에 5분, 또는 $0.2\%\;NaHSO_3$에 1분 침지한 것이 갈변방지에 좋은 효과를 나타내어 젓갈 등의 가공제품의 개발에 있어서도 유용한 전처리 방법이라 생각되었다. 또 $0.2\%\;NaHSO_3$에 1분 침지한 경우의 잔존 $SO_2$양은 100 ppm 이하였으며 $NaHSO_3$에 침지후 상온숙성의 경우는 약 30일 정도, 저온숙성의 경우는 50일까지 갈변발생을 억제할 수 있었다.

벼 바이러스(RSV, RBSDV)와 애멸구의 간편한 VC/RT-PCR 유전자 진단기술 (Convenient Genetic Diagnosis of Virion Captured (VC)/RT-PCR for Rice Viruses (RSV, RBSDV) and Small Brown Plant Hopper)

  • 우리나라 벼에 발생하는 주요 바이러스 중 애멸구에 의하여 전염하는 벼줄무의잎마름병(RSV)과 벼검은줄오갈병(RBSDV)에 대한 간편한 유전자 진단법인 VCHT-PCR 방법을 개발하였다. 벼 잎의 경우 즙액 추출 완충액은 0.5% sodium sulfite를 첨가한 0.01 M 인산완충액(pH 7.0)을 기본 완충액으로 이용하고 애멸구를 진단할 경우에는 기본 완충액에 2% PVP을 첨가하였을 때 VC/RT-PCR 진단이 잘 되었다. VC/RT-PCR을 이용한 진단에 적합한 RSV와 RBSDV 프라이머를 선발하였고 이것을 이용하여 동시진단으로 경기도 김포, 평택, 시흥지역에서 채집한 애멸구의 RSV와 RBSDV의 보독충을 쉽고 경제적으로 진단 할 수 있었다. ELISA에 의한 진단결과와 비교할 때 세 지역을 합하여 RSV에 대한 평균 보독충률은 9.2%로 동일하였으나 보독충 중 일부가 ELISA와 VC/RT-PCR 두 방법에 의한 진단결과가 다르게 나온 것은 RSV의 혈청학적, 유전적 계통 존재 가능성을 제시하고 있다. 포장에서 채집한 애멸구의 VC/RT-PCR 진단효율은 RSV와 RBSDV를 동시에 진단하므로 써 RSV만 진단이 가능한 ELISA 결과 보다 3.2% 높았다.

견층(繭層) Sericin에 대(對)한 몇 가지 화학약제(化學藥劑) 처리(處理)의 효과(效果) (Effects of Several Chemicals Treatment for Cocoon Sericin)

  • 본(本) 실험(實驗)은 견층(繭層) seicin에 대(對)한 몇가지 화학약제(化學藥劑) 처리(處理)의 온도별(溫度別), 농도별(濃度別) seicin 용해성(溶解性)에 미치는 영향(影響)을 조사(調査)하였던 바 다음과 같은 결과(結果)를 얻었다. 1. 공시(供試) 해서촉진제중(解舒促進劑中) 견층(繭層) seicin의 용해촉진력(溶解促進力)이 가장 큰 약제(藥劑)는 sodium peroxide($Na_2O_2$)였으며, sodium carbonate($Na_2CO_3$), sodium sulfite($Na_2SO_3$) ammonia water($NH_4OH$)의 순(順)으로 용해촉진력(溶解促進力)이 작았다. 2. 공시(供試) 해서억제제중(解舒抑制劑中) 견층(繭層) sericin의 용해억제력(溶解抑制力)이 가장 큰 약제(藥劑)는 tannic acid($C_{14}H_{10}O_9$)였으며, stannic acid($S_n(OH)_4$), formic acid(HCOOH)< methyl alcohol($CH_3OH$)의 순(順)으로 용해억제력(溶解抑制力)이 작았다. 3. 공시(供試) 해서억제제중(解舒抑制劑中)에서 formic acid와 stannic acid는 $100^{\circ}C$의 고온(苦蘊)에서는 견층(繭層) sericin의 용해(溶解)를 크게 촉진(促進)하였다. 4. 공시(供試) 해서억제제중(解舒抑制劑中) methyl alcohol의 경우(境遇)는 저농도(低濃度)에서는 용해억제력(溶解抑制力)이 전혀 없었다.

L-Ascorbic Acid-2-Phosphate Mg염의 합성 및 응용

  • Purely synthesized L-ascorbic acid 2 phosphate Mg salt (1 AsA PMg) improved the weak point of ascorbic acid which is easily decomposed in water solution. This compound is hydrolyzed with phosphatase of skin to corresponding ascorbic acid giving Vitamine C activities. The buffer solution of potassium acetate 0.5% and citric acid 0.005% and the sodium sulfite respectively showed good stabilizing effect of the AsA PMg solution. Compared to the other ascorbic acid derivatives the good solubility of AsA PMg gives broad application to cosmetic field.

Influence of Processing Method on Ileal Digestibility of Nutrients from Soybeans in Growing and Finishing Pigs

  • Eight crossbred barrows (four growing and four finishing pigs with average initial BW of 40 and 82 kg, respectively) were fitted with T-cannulas at the distal ileum and used in a 36 d metabolism experiment ($4{\times}4$ Latin squares) to determine the effects of roasting and extruding full-fat soybeans on nutrient utilization. Treatments were: 1) soybean meal; 2) roasted soybeans; 3) extruded soybeans; and 4) soybeans extruded with an extrusion enhancer (sodium sulfite). The control diet was corn starch-based with 0.90% lysine, 0.65% Ca and 0.55% P for the growing pigs and 0.75% lysine, 0.55% Ca and 0.45% P for the finishing pigs. For the growing pigs, apparent total tract digestibilities of DM (p<0.04) and GE (p<0.008) were greater for soybean meal than full-fat soy products. However, ileal digestibilities of DM, GE, N and most amino acids were, in general, greatest for extruded soybeans and lowest for roasted soybeans, with soybean meal intermediate. For finishing pigs, trends in digestibilities of nutrients were very similar to those for the growing pigs. Total tract digestibilities of DM (p<0.03) and GE (p<0.001) for soybean meal were greater than for the full-fat soy products and ileal digestibilities of DM, GE, N and most amino acids were greater for the extruded soybeans than for the roasted soybeans. In conclusion, nutrient digestibilities and availabilities of indispensable amino acids tended to be greatest in extruded soybeans, intermediate in soybean meal and lowest in roasted soybeans for growing and finishing pigs.

파파인 가공한 양모/폴리에스터 혼방직물의 정련 및 염색성 (Dyeing Properties and Scouring of Wool/Polyester Blend Fabrics Using Papain from Carica Papaya)

  • This study provides the optimum papain treatment method and its effect on wool/polyester blend fabrics. The enzymatic treatment condition is optimized depending on its pH level, temperature, concentration of enzyme, treatment time and concentration of activators. The characteristics of samples treated with the papain are measured using weight loss, tensile strength, whiteness, WCA, dyeing property and surface micrographs. The results are described as follows: According to measuring weight loss, tensile strength and whiteness, a pH level of 7.5, $70^{\circ}C$, 10% papain(o.w.f.) and 60minutes of treatment time are optimized for papain treatment. L-cysteine and sodium sulfite are able to activate the papain. The optimum concentrations of them are 10mM and 50mM respectively. The WCA of fabrics is decreased since papain treatment makes wool/polyester blend fabrics more hydrophilic. Scouring with papain treatment improves whiteness and dyeing property of fabrics. The dyeing property of papain-treated fabrics is enhanced simply by a single step dyeing process using a basic dye. The surface of wool treated with papain in the presence of L-cysteine shows to be descaled. The surface of wool fibers in the presence of sodium sulfite, however, shows it is hydrolyzed evenly instead of being descaled. The surface of papain treated polyester fibers shows cracks and voids.

도금폐수내 유리시안과 착염시안 및 중금속의 처리특성 (I) (Treatment Characteristics of Plating Wastewater Containing Freecyanide, Cyanide Complexes and Heavy Metals (I))

  • The mean pH of wastewater discharged from the plating process is 2, so a less amount of alkali is required to raise pH 2 to 5. In addition, if sodium sulfite is used to raise pH 5 to 9 in the secondary treatment, caustic soda or slaked lime is not necessary or only a small amount is necessary because sodium sulfite is alkali. Thus, it is considered desirable to use only $FeSO_4{\cdot}7H_2O$ in the primary treatment. At that time, the free cyanide removal rate was highest as around 99.3%, and among heavy metals, Ni showed the highest removal rate as around 92%, but zinc and chrome showed a low removal rate. In addition, the optimal amount of $FeSO_4{\cdot}7H_2O$ was 0.3g/L, at which the cyanide removal rate was highest. Besides, the free cyanide removal rate was highest when pH value was 5. Of cyanide removed in the primary treatment, the largest part was removed through the precipitation of ferric ferrocyanide: $[Fe_4(Fe(CN)_6]_3$, and the rest was precipitated and removed through the production of $Cu_2[Fe(CN)_6]$, $Ni_2[Fe(CN)_6]$, CuCN, etc. Furthermore, it appeared more effective in removing residual cyanide in wastewater to mix $Na_2SO_3$ and $Na_2S_2O_5$ at an optimal ratio and put the mixture than to put them separately, and the optimal weight ratio of $Na_2SO_3$ to $Na_2S_2O_5$ was 1:2, at which the oxidative decomposition of residual cyanide was the most active. However, further research is required on the simultaneous removal of heavy metals such as chrome and zinc.
