버섯류의 미양금속, 중금속 및 유기산 함양에 관한 연구 (Studies on the Trace Elements, Heavy Metals and Organic acids Content of Edible Mushrooms)

  • To investigate on the element content of twelve edible mushrooms and Aloe arborescent, i.e., Lentinus edodes, Ganoderma lucidum (culturing in wood and soil), Tricholoma matsutake, Agaricus auricula-Jude (produced in Korea and China), sarcodon asparatus, Pleurotus ostreatus, Coriolus bisporus, Cyrophora esculenta, Auricularia auricula-Jude (produced in Korea and China), Sarcodon asparatus, Pleurotus ostreatus, Coriolus rersicolor, Smilax rotundifolia and Aloe arborescenstyl were analyzed by Atomic absorption spectrometer. The obtained results were summarized as follows : 1. Potassium, sodium, magnesium and iron content for the most part samples were in large quantities, especi phosphorus content of those was highest ammount for the all samples. 2. Sodium content f·as much ammount in the Lentinus edodes(39 mg, and Ganoderma lucidum(20 mg) , Culturing in wood and soil, while potassium was very high ammount in the Aloe arborescens and other samples. Mush ammount of magnesium as compared with others was Lentinus edodes(144 mg) , Ganoderma lucidum(128 mg), Aloe arborescens(50 mg) and Pleurotus ostreatus (60 mg) . 3, Phosphorus content of Lentinus edodes, Gyrophora esculenta, Auricularia polytricha and Agaricus bisporus was much ammount while iron content of all samples equality higher ammount. Sodium content of Aloe arborescens was not analyzed out for almost all, its potassium(82 mg), magnesium(50 mg) and iron(18 mg) content comparatively higher quentity than others nllnerals and phosphorus volume (4.9 mg) as compared with others, was conspicuously lower detect. 4. Cadimium and lead content of harmful metal element were detected on trace quantity for the most part samples. 5. Organic acids of samples i.e., Lentinus edodes, Agaricus bisporus, Pleuritus ostreatus and Ganoderma lucidum were Citrate, Malate, Fumalate, Succinate, Oxalate, Acetate, Lactate, and Tartarate and Citrate, Malate and Fumarate contents were higher amount remarkbly than other organic acids. Tartarate content was trace amount.

Purification and Characterization of a Novel Alkaline Protease from Bacillus horikoshii

  • An investigation was conducted on the enhancement of production and purification of an oxidant and SDS-stable alkaline protease (BHAP) secreted by an alkalophilic Bacillus horikoshii, which was screened from the body fluid of a unique Korean polychaeta (Periserrula leucophryna) living in the tidal mud flats of Kwangwha Island in the Korean West Sea. A prominent effect on BHAP production was obtained by adding 2% maltose, 1% sodium citrate, 0.8% NaCl, and 0.6% sodium carbonate to the culturing medium. The optimal medium for BHAP production contained (g/l) SBM, 15; casein, 10; $K_2HPO_4$, 2; $KH_2PO_4$, 2; maltose, 20; sodium citrate, 10; $MgSO_4$, 0.06; NaCl, 8; and $Na_2CO_3$, 6. A protease yield of approximately 56,000 U/ml was achieved using the optimized medium, which is an increase of approximately 5.5-fold compared with the previous optimization (10,050 U/ml). The BHAP was homogenously purified 34-fold with an overall recovery of 34% and a specific activity of 223,090 U/mg protein using adsorption with Diaion HPA75, hydrophobic interaction chromatography (HIC) on Phenyl-Sepharose, and ion-exchange chromatography on a DEAE- and CM-Sepharose column. The purified BHAP was determined a homogeneous by SDS-PAGE, with an apparent molecular mass of 28 kDa, and it showed extreme stability towards organic solvents, SDS, and oxidizing agents. The $K_m$ and $k_{cat}$ values were 78.7 ${\mu}M$ and $217.4s^{-1}$ for N-succinyl-Ala-Ala-Pro-Phe-pNA at $37^{\circ}C$ and pH 9, respectively. The inhibition profile exhibited by PMSF suggested that the protease from B. horikoshii belongs to the family of serine proteases. The BHAP, which showed high stability against SDS and $H_2O_2$, has significance for industrial application, such as additives in detergent and feed industries.

  • 식육 및 야생쥐의 장으로부터 식품매개성 질병에 관여하는 Enterobacter를 분리하여 그 특징을 구명하므로 공중위생 및 식품위생학적으로 기초자료를 제시하고저 본 연구를 실시하여 다음과 같은 결론을 얻었다. Enterobacter는 우육에서 1주(E. aerogenes), 돈육에서 2주(E. aerogenes, E. intermedium), 야생쥐의 소장에서 2주(E. aerogenes, E. cloacae)가 동정되어 총 분리된 89균주에 대한 분리율은 5.62%이었다. 분리된 Enterobacter를 각종 화학기질이 첨가된 배지에서 $55^{\circ}C$, 35분간 열처리한 결과, 전반적으로 0.1M glycine, 5% mannitol 그리고 5% sorbitol이 첨가된 배지에서는 열저항성이 강하였고 1% sodium citrate, 1% casein, 10% NaCl 그리고 0.1M cystein이 첨가된 배지에서는 열저항성이 약하였다.

Enhancement of L-Lactic Acid Production in Lactobacillus casei from Jerusalem Artichoke Tubers by Kinetic Optimization and Citrate Metabolism

  • Efficient L-lactic acid production from Jerusalem artichoke tubers, by Lactobacillus casei G-02, using simultaneous saccharification and fermentation (SSF) in a fed-batch culture, is demonstrated. A kinetic analysis of the SSF revealed that the inulinase activity was subjected to product inhibition, whereas the fermentation activity of G-02 was subjected to substrate inhibition. It was also found that the intracellular NADH oxidase (NOX) activity was enhanced by the citrate metabolism, which dramatically increased the carbon flux of the Embden-Meyerhof-Parnas (EMP) pathway, along with the production of ATP. As a result, when the SSF was carried out at $40^{\circ}C$ after an initial hydrolysis of 1 h and included a sodium citrate supplement of 10 g/l, an L-lactic acid concentration of 141.5 g/l was obtained after 30 h, with a volumetric productivity of 4.7 g/l/h. The conversion efficiency and product yield were 93.6% of the theoretical lactic acid yield and 52.4 g lactic acid/l00 g Jerusalem artichoke flour, respectively. Such a high concentration of lactic acid with a high productivity from Jerusalem artichokes has not been reported previously, making G-02 a potential candidate for the economic production of L-lactic acid from Jerusalem artichokes on a commercial scale.

탈지산양유(脫脂山羊乳)가 우정자보존(牛精子保存)에 미치는 영향(影響) (Effects of Skimmed Goat Milk as a Semen Extender on Preservation of Bull Spermatozoa)

  • Skimmed goat milk heated at $92^{\circ}C$ for 10 minutes was used as a basal extender for bull semen. The extenders for liquid semen were prepared by adding simultaneously at various ratio of 5% dextrose solution and egg yolk to skimmed goat milk. After bull seven was diluted with the extenders at the rate of 20 million spermatozoa per ml of the extenders. The extenders were stored at $5^{\circ}C$ and the survival rates of spermatozoa were examined at 4 and 24 hours, and 3, 5 and 7 days after dilution. The extenders for frozen semen were prepared by adding various ratlo of glycerol to skimmed goat milk containing 20 parts of 5% dextrose solution and 3 parts of egg folk to 77 parts of skimmed goat milk. After bull semen was diluted with the extenders at the rate of 40 million spermatozoa per ml of the extenders, the extenders were frozen in liquid nitrogen tank. The frozen extenders were thawed at $40^{\circ}C$ for 2 minutes, and the revival rates of the spermatozoa in the extenders were examined. These thawed extenders were stored at $5^{\circ}C$ and the survival rates of the spermatozoa were examined at 10 minutes and 24 hours and 3 and 5 days after thawing. The results obtained were as follows: 1. Among the extenders stored at $5^{\circ}C$, the survival rate of the sperm was the highest in the extender including 20 parts of 5% dextrose solution and 3 parts of egg yolk to 77 parts of skimmed goat milk, and the survival rate was significantly higher that of the spermatozoa in egg folk-2.9% sodium citrate (1 : 4) extender. (P<0.05) 2.Among the extenders frozen in liquid nitrogen tank, the revival rate of the spermatozoa was the highest in the extender containing 7ml of glycerol per 100ml of the extender with consisted of 77 parts of skimmed goat milk, 20hparts of 5% dextrose solution and 3 parts of egg yolk, and the revival rate was significantly higher than that of the spermatozoa in egg yolk-2.9% sodium citrate (1 : 4) extender containing 8ml of glycerol per 100ml of the extender (p<0.01). 3. Among the extenders stored at $5^{\circ}C$ after thawing, the survival rate of the spermatozoa was the highest in the extender containing 7ml of glycerol per 100ml of extender which consisted of 77 parts of skimmed goat milk, 20 parts of 5% dextrose solution and 3 parts of egg yolk, and the survival rate was significantly higher than that of the spermatozoa in egg yolk -2.9% sodium citrate (1 : 4) extender containing 8ml of glycerol per 100ml of the extender (p<0.01).

식품중 Nitrosamine 관련물질의 함량과 시험관내에서 NDMA의 생성조건 (Contents of Nitrosamine Related Compounds in Some Foods and Condition for NDMA Formation in Vitro)

  • This study was carried out to investigate contents of nitrosamine precursors such as trite and dimethylamine( DMA ) in some foods. The diazo and Cu- dithiocarbamate melt were used for determination, respectively. The major affecting factors of Nnitrosodimethylamine( NDMA ) formation such as pH, contents of DMA and NaNO$_{2}$, other chemicals, and UV- ray in beverage were investigated in vitro. The contents of nitrite in meat sausage and meat ham were 6.44 ∼ 18.66ppm and 12.85- 39.95pp% respectively, And extremely low level was detected in a certain kind of fish sausage. The contents of DMA in meat sausage meat ham and fish sausage were 3.34∼15. 85ppm, 1.20∼7.10ppm and 7.38∼12.28ppm, respectively. The optimum pH for NDMA formation in vitro was 3.0. NDMA formation was rapidly occurred at high temperature and formed above 80% within 1 hour reaction. The formation of NDMA was increased in proportion to the concentration of DMA and the square of the nitrite concentration. 0.1 M of sodium citrate, sodium tartarated and sodium taiocyanate enhanced NDMA formation. But sodium chloride did not affect. However, 0.3M of ascorbic acid, erythroid acid, ascorbic, palmitate and propy, gallate inhibited NDMA formalion approximately 78%,81%,86% and 85%, respectively. Cow milk and soybean milk inhibited 35 ∼47% of NDMA formation but orange juice and apple juice enhanced 15 ∼64% of NDMA formation. The peak in HPLC for NDMA disappeared by irradiation of UV to prior formed NDMA This result suggest that NDMA was destroyed by UV irradiation.

무전해도금에 의한 Ni-Tl-P 피막형성에 관한 연구 (A Study on Formation of Ni-Tl-P deposits by Electroless Plating)

  • This study investigated the bath compositions and plating conditions and crystal structure used for achieving nickel-thallium-phosphorus deposits by means electroless plating. The electroless nickel-thallium-phosphorus deposits were achieved with a bath using sodium hypophosphite as the reducing agent and sodium citrate as the complexing agent. The depositing rate was 10.5mg.$cm^{2-1}$ .$hr^{-1 }$ from the optimistic bath composition, 0.1M nickel sulfate, 0.005M thallium sulfate, 0.2M sodium hypophosphite, and 0.05M sodium citrat and the recommended plating conditions, pH 5.5 and $90^{\circ}C$. The composition of alloy deposits determined by X-ray analysis (EDS) that the Thallium was increased with major increasing concentration of complexing agent and thallium ion in bath solution, it decreased according to the increasing concentrations of reduction agent in the bath solution, Bit Phosphorus showed a contrary to the thallium. It was observed from X-ray diffraction analysis, Scanning Electron Microscopy and Transmission Electron Microscopy. The crystalline structure of deposits was amorphous at the first deposited state but it was changed $Ni-T1-Ni_{5}$ $P_2$ polycrystalline when subjected to 1 hour heat treatment of more than $350^{\circ}C$. TEM observation demonstrated that the microstructure was identical to the result of the XRD at as deposited but it became $Ni-Tl-Ni_{5}$ $P_2$ polycrystalline when heated. And grain size was 10-50nm.

Biochemical Characteristics of Micrococcus varians, Staphylococcus carnosus and Staphylococcus xylosus and Their Growth on Chinese-Style Beaker Sausage

  • This study was conducted to investigate protein and carbohydrate utilization of Micrococcus varians, Staphylococcus carnosus and Staphylococcus xylosus. Sensitivity to pH, sodium chloride, potassium sorbate and sodium nitrite of these strains was also determined. In Chinese-style beaker sausage manufacturing, the growth rate of these strains during the curing period ($20^{\circ}C$ and $30^{\circ}C$) was evaluated. The results indicated that no strains could hydrolyze azo-casein and sarcoplasmic protein and only S. xylosus could hydrolyze gelatin at $30^{\circ}C$. All of these strains could oxidize and ferment fructose and mannitol. S. carnosus and S. xylosus could slightly oxidize lactose and utilize citrate. Arabinose was oxidized by S. xylosus and sorbitol was oxidized by S. carnosus. Growth of M. varians was restricted at pH 5.0 and S. carnosus and S. xylosus were restricted at pH 4.5. S. xylosus and S. carnosus were able to grow with 0.1~0.5% potassium sorbate, 50~200 ppm sodium nitrite or 1~15% sodium chloride. S. xylosus had a higher growth rate than the other strains. Staphylococcus species grew well during curing period of Chinese-style beaker sausage then followed by Micrococcaceae.

연료전지의 수소저장용 마그네슘계 합금의 표면제어에 의한 전기화학적 수소화 거동 연구 (Electrochemical Hydrogenation Behavior of Surface-Treated Mg-based Alloys for Hydrogen Storage of Fuel Cell)

  • [ $Mg_2Ni$ ]계 합금 입자의 수소저장 특성에 대한 표면처리 효과가 마이크로 전극 측정법에 의해 검토되었다. 카본-섬유로 된 마이크로 전극을 KOH 수용액 속에서 조정자를 사용하여 수소 단일입자에 접촉시켰다. 상온에서 $Mg_2Ni$ 합금의 수소저장 특성은 니켈 도금용액에 의한 표면 처리에 의해 크게 개선되었다. 니켈 도금용액 속에 있는 함유된 나트륨염(sodium phosphate 및 sodium dihydrogen citrate)이 합금을 아몰퍼스와 같은 형태로 만들었으며, 그 결과 상온에서 수소 흡장/방출 용량이 최초의 17[mAh/g]에서 150[mAh/g]로 향상되었다. 합금 입자 내에서 수소원자의 겉보기 화학적 확산계수를 계산하기 위하여 Potential-step 실험을 실시하였으며 데이터 해석을 위해 구형확산 모델을 적용하였다. 실험결과로서 겉보기 확산계수($D_{app}$)는 수소 흡장 및 방출되는 전 과정에서 $10^{-8}{\sim}10^{-9}[cm^2/s]$ 수준인 것으로 확인되었다.

키토산과 유기산염 첨가가 배추김치의 저장성과 펙틴분획에 미치는 영향 (Effects of Chitosan and Organic Acid Salts on the Shelf-life and Pectin Fraction of Kimchi during Fermentation)

  • 배추김치에 0.25% chitosan, 0.5% chitosan, 0.5% chitosan+0.04M Na-malate, 0.5% chitosan+0.04M Na-lactate, 0.5% chitosan+04M Na-citrate를 첨가하여 김치의 숙성 과정 중 pH, 산도, pH/TA ratio, ascorbic acid 함량 및 Pectin질의 변화에 대한 영향을 알아보았다. 그 결과는 다음과 같다. 1. 김치의 숙성과정 중 pH는 감소하는 경향을 보였으며, 대조군, 0.25% chitosan 첨가군, 0.5% chitosan 첨가군, 0.5% chitosan+Na-citrate 첨가군, 0.5% chitosan+Na-malate 첨가군, 0.5% chitosan+Na-lactate 첨가군의 순으로 감소하였다. 2. 김치의 숙성과정 중 산도는 증가하는 경향을 보였으며, 대조군, 0.5% chitosan+Na-malate 첨가군, 0.25% chitosan 첨가군, 0.5% chitosan+ Na-citrate 첨가군. 0.5% chitosan 첨가군, 0.5% chitosan+Na-lactate 첨가군의 순으로 증가하였다. pH/TA ratio는 김치의 숙성 중 감소하는 경향을 보였으며. 대조군, 0.25% chitosan 첨가군, 0.5% chitosan+Na-malate 첨가군, 0.5% chito-san+Na-citrate 첨가군, 0.5% chitosan+Na-lactate 첨가군의 순으로 감소하였다. 3. Ascorbic acid 함량은 3일째에 가장 높은 함량을 보였으며, 0.5% chitosan에 유기산염 병용첨가군이 다른 군들보다 월등히 높았다. 그 후 숙성기간이 경과할수록 감소하였다. 4. AIS 함량의 변화는 숙성기간이 지날수록 모두 감소하는 경향을 보였으며, 대조군, 0.25% chit-osan 첨가군, 0.5% chitosan 첨가군, 0.5% chito-san+Na-citrate 첨가군, 0.5% chitosan+Na-lactate 첨가군, 0.5% chitosan+Na-citrate 첨가군 순으로 감소하였다. 5. AIS로부터 펙틴을 추출하여 HWSP, HXSP, HCISP로 분획한 함량비는 대조군이 숙성이 진행됨에 따라 HWSP가 증가하고 HCISP는 감소하는 경향을 보인 반면, chitosan 첨가군은 숙성이 진행됨에 따라 HWSP가 감소하고 HCISP는 증가하는 경향을 나타내었다. 위와 같은 결과로부터 chitosan의 첨가는 김치의 보존성 향상에 효과를 보였으며. 유기산염과 병용첨가군이 단독첨가군보다 더욱 효과적이었다. 유기산염의 종류에 따른 병용에 있어서는 Na-lactate 첨가군이 Na-citrate와 Na-malate 첨가군보다 효과적으로 나타났다.
