Low-Z Particle Electron Probe X-ray Microanalysis법을 이용한 2001년 ACE-Asia 집중 측정 기간 중 제주도 고산에서의 입자상 물질의 특성분석 (Single Particle Characterization of Aerosol Particles Collected at Jeju Island, Korea, During 'ACE-Asia' Intensive Observation Period, Using Low-Z Particle Electron Probe X-ray Microanalysis)

  • Low-Z particle electron probe X-ray microanalysis (low-Z particle EPMA) was applied to characterize aerosol particles collected at Gosan in Jeju island during an IOP (intensive observation period) for an international ACE-Asia project in April, 2001. Various types of atmospheric particles such as organics, carbon-rich, aluminosilicates, silicon oxide, sodium nitrate, sodium chloride, and ammonium sulfate were observed. The reacted sea salt particles such as sodium nitrate were the most abundantly encountered, but original sea salt particles were rarely observed. Since the Low-Z particle EPMA can provide quantitative information on the chemical composition of aerosol particles, many different particle types on the basis of their chemical compositions were observed and identified. In this study, it is demonstrated that the Low-Z particle EPMA can provide detailed information on the chemical compositions for the aerosol particles collected for six consecutive days in April, 2001, at Gosan super-site.

산란노계육의 저장중 소금과 인산염 첨가가 pH, 수분, 연도 및 무기물에 미치는 영향 (Effects of Addition of Varied Levels of Sodium Chloride and Phosphates on pH, Tenderness, Moisture and Mineral Contents in Spent Layer Meat)

  • A study was conducted to investigate the effects of the addition of varied levels of NaCi and phosphates on the physicochernical properties of the breast meat of the spent layers(2 \pm 0.2 kg) which were stabilized for over 24 h before slaughter. Within 1 h after slaughter, breast meats were removed and treated with NaGl(0, 1, 2, 3%) and phosphates(0.25% and 0.5%) using a hot-salted method. The breast meat was stored at 4 \pm $1^{\circ}C$ for 3 d. The results obtained were summarized as follows. 1. The pH values of salt-treated groups were significantly higher than that of the control(P<0.05) ; the higher the salt level, the higher the pH. The pH values were significantly increased in both control and treatment groups during storage(P<0.05). Among salt-treated groups, the 0.5% phosphates level showed significantly high pH(P<0.05) compared to other levels of salt groups. 2. The moisture contents were significantly lower in all salt4reated groups than the control(P<0.05), and showed a negative relationship with the levels of salt. It decreased in control group gradually as the storage period extended, but not significantly changed in salt-treated groups. 3. The shear force values in salt-treated groups were lower than that of the control and showed a negative relationship with salt levels. At a constant level of NaCI, the shear force value was higher in 0.25% phosphates level than in 0.5% level. It decreased in both control and salt-treated groups during storage. 4. The salt treatments tended to increase the sodium content proportionately. The sodium content decreased in both control and salt treatment groups during storage(P<0.05). In addition, the combination of high levels of NaCl and phosphates rather than those of low levels of NaCI and phosphates resulted in elevated levels of sodium. 5. The phosphorus contents in salt-treated groups were higher than that of control. Between 0.5% and 0.25% phosphates levels this value showed significant difference(P<0.05). Its contents in both control and treatment groups were significantly decreased during storage (P<0.05).

Salt 스트레스에 의한 흰쥐 적출대동맥의 수축력 변화양상 (Changes of Vascular Contractility of isolated Rat Aorta treated with Salt Stress)

  • To examine whether salt stress would alter or not contractility of isolated rat aorta, under anesthesia with sodium pentobarbital(50 mg kg-1 i.p.), male Sprague Dawley rats(300-330 g) were subjected to 0, 50, and 150 mM of sodium chloride at 37$^{\circ}C$ for 60 min. where as the sham group was left at modified Krebs-bicarbonate solution. To measure contractile response of vascular ring preparation isolated from rat was determined in organ bath and was recorded on physiograph connected to isometric transducer. And the strip was checked for expression of heat shock protein(Hsp) by Western blotting. One, three and eight hours later, we measured vascular contractility of isolated rat aorta treated with KCI, phenylephrine from organ bath study. The dose-vascular responses of potassium chloride and phenylephrine showed a little augmentation by NaCl concentration in the strips exposed to NaCl for 8 hours. And the response of relaxation induced by nitroprusside and acetylcholine was not influenced by NaCl stress in isolated aorta ring for 8 hours, respectively. Expression pattern of Hsp 70 of vascular muscle in isolated rat aorta showed a little increase in 150 mM NaCl group at 8 hours after NaCl treatment but not at 3 hours, and Hsp 60 expression of rat aorta was markedly increased in 50 mM NaCl group at 8 hours after NaCl treatment. Taken together, NaCl induced dose-and time dependent accumulation of the Hsp but not affected contraction of rat aorta. These data suggest that short term high salt stress was not sufficient to induce hypertension of rat aorta.

A small-scale membrane electro-dialyser for domestic use

  • A small-scale electro-dialysis system was constructed for domestic use. It is composed of six compartments in which five special polystyrene ionic membranes are housed. A series of experiments on the transport of sodium and chloride ions through polystyrene membranes was performed and the effects of electric current and voltage on the pH of water were investigated. This electrodialyser could reduce the NaCl content to an acceptable level (5307 mg/L) when water containing 9945 mg/L of sodium chloride is fed to the electrodialyser. The reduction was by the action of direct current 60 mA/100 mA when a 15 V / 20 V potential is maintained across the membrane. The results showed that the pH of the treated water attained a value in the range of 7-8, with the chloride concentration of 5307 mg/L when the voltage was in the range of 20 volts. This was achieved when two of the small-scale electro-dialysers were placed in series and the solutions from the respective compartments were mixed. This is considered useful because this complies to the requirement of drinking water standard both in terms of chloride and pH. Therefore, this type electrodialyserhas the potential for domestic uses in isolated houses where potable water supply is not available.

Effect of salt on membrane protein Caveolin3 proved with NMR spectroscopy

  • Caveolin3, mainly expressed in muscle tissue types, is a structural scaffolding protein of caveolae which are microdomains of plasma membrane. To elucidate the relationship between structure and function, several studies on the structure of caveolins using NMR have been reported. Because the ionic strength can affect the electrostatic-driven association of proteins with ligand and protein structure, the effect of salt in the structural studies has to be considered. In this work, we observed that the chemical shifts of Cav3 in the LPPG detergent change depending on salt concentration. The R2 values also show salt concentration-dependent changes. Specifically, in the N-terminal region where conformational changes and various interactions occur, the R2 values decrease. Interestingly, the R2 values of residues expected to be located in the LPPG detergent are also influenced by the salt concentration. This work suggests that the concentration of NaCl can affect interpretation of NMR data from membrane proteins.

New Approaches to Production of Turkish-type Dry-cured Meat Product "Pastirma": Salt Reduction and Different Drying Techniques

  • In this study, the possible changes in the quality characteristics of pastirma, Turkish-type dry-cured meat product, produced by using two different salts (NaCl-KCl) in a curing mixture and two different production techniques (natural and controlled condition) were examined. Moisture, pH, salt, sodium, potassium, TBA, fat, water activity, instrumental colour, texture, and sensory analyses were implemented in order to determine the possible effects of these applications. Fat, aw, pH, colour, tiobarbituric acid (TBA), texture, salt, Na and K values may allow these desired modifications in pastirma production to be limited. The substitution of 15% KCl instead of NaCl was acceptable in terms of the sensorial properties of the pastirma. However, the sensory analyses did not allow for using a higher KCl instead of NaCl because both the hardness and chewiness in the texture of the pastirma samples salted with 30% of KCl were not scored positively. Besides this, negative effects, which may occur during the pastirma production under natural conditions, can be eliminated by the production being under controlled conditions.

Performance prediction of flat sheet commercial nanofiltration membrane using Donnan-Steric Pore Model

  • The rejection of sodium chloride (NaCl) and calcium chloride (CaCl2) single salt solutions were carried out for commercial nanofiltration NFDK membrane. Results showed that the NFDK membrane had a negative surface charge and had a higher observed rejection of 93.65% for calcium (Ca2+) ion and 78.27% for sodium (Na+) ions. Prediction of rejection for aqueous solutions of both salts was made using Donnan Steric Pore Model based on Extended Nernst-Planck Equation in addition to concentration polarization film theory. A MATLAB program was developed to execute the model calculations. Absolute Average Relative Error (% AARE) was found below 5% for real rejection of the NFDK membrane. This research could be used successfully to assess the membrane characterization parameter using a proposed procedure which can reduce the number of experiments.

멸치젓 숙성중 불휘발성아민의 함량 변화 (Change of Nonvolatile Amines During Fermentation of Anchovy)

  • 멸치를 물로 씻은 후 식염 10%, 15%, 20% 및 식염과 염화칼리 1:1 홈합염 10%를 각기 첨가하여 1~12주간 숙성할 때 불휘발성 아민의 함량과 미생물의 변화와 시중에 유통중인 멸치젓의 불휘발성아민 함량을 조사하였다. 불휘발성아민은 식염농도가 높을수록 생성량이 적었고 숙성기간별 생성량은 histamine의 경우 숙성 초기부터 증가하여 6주째에 식염 10%, 멸치젓이 657.8ppm으로 최고치를 나타낸 후 감소하기 시작하여 숙성 12주째는 식염 20%, 멸치젓이 210.8ppm으로 가장 적었고 혼합염 10% 멸치젓이 310.8ppm으로 가장 많이 생성되었다. Cadaverine과 putrescine은 숙성 8주째까지 생성량이 증가한 후 점차적으로 줄어들었고 tyramine은 숙성 10주째까지 RPthrwmd가한 뒤 숙성 12주째 현저히 감소하였다. 시중 유통품과 식염 함량이 비슷한 20% 멸치젓의 12주째 실험결과를 비교하면 시중 유통품의 불휘발성 아민 함량이 전반적으로 높게 나타났다. 미생물의 변화는 숙성 초기에는 Achromobacter 속, Aeromonas 속 및 Pseudomonas속이, 숙성 중기에는 Pediococcus속 , Microc....us 속 및 Lactobacillus 속이 많았으며 숙성 후기에는 Sacchromyces 속이 증가하였고 전 숙성기간 동안 미생물은 109주가 분리되었다.

The Pressure Effect on the Activity Coefficient of Sodium Chloride and Bromide Ions

  • A new paraffin salt bridge was developed for the concentration cell in high pressure system. The emf values of the concentration cells were measured to calculate the activity coefficients of the electrolytic ions depending upon the pressure of the system. The activity coefficients of sodium, chloride and bromide ions increase with the temperature of the cell and decreased, nearly half at 2500 bars, $20^{\circ}C$, with the pressure. These results can be explained to be attributed to the volume change of the hydrated ion due to the electrostriction. The volume change decreased with pressure due to an increase in the degree of the hydration.

이탈리안 라이그라스 품종별 NaCl 처리가 발아 및 초기생육에 미치는 영향 (Effects of Sodium Chloride Treatment on Seed Germination and Seedling Growth of Italian Ryegrass Cultivars)

  • 이탈리안 라이그라스의 발아 및 초기 생육단계에서 내염성 수준 정도를 조사하기 위하여 in vitro 조건에서 다양한 염농도 조건에서 그린팜, 플로리다80, 코원어리 및 화산101호 품종을 이용하여 염해에 대한 반응을 발아율과 산화 스트레스 관련 효소들의 활성을 통해 조사하였다. 저농도의 NaCl 처리구에서는 모든 품종에서 발아율의 차이가 1~3% 정도 감소하는 경향을 보였으며 250 mM 이상의 NaCl 처리구에는 화산101호 품종을 제외한 모든 품종에서 50% 이상의 발아율 감소를 보였다. 또한, NaCl 농도가 높아질수록 발아에 걸리는 시간이 지연되었으며, 350 mM NaCl이 첨가된 처리구에는 화산101호 품종만이 치상 10일후에 발아가 시작하여 12% 정도 발아하였다. NaCl 농도에 따른 shoot과 root의 길이와 생초와 건조 후 무게의 변화도 만생종으로 갈수록 더 생육이 우수한 것으로 타나났다. 품종별간의 잎 조직에서 항산화효소 활성을 분석 결과 고염 조건 (150 mM NaCl)에서 POD 활성은 대조군과 유사한 수준으로 확인 되었다. 반면 APX 활성은 플로리다80과 코윈어리 품종에서는 증가하는 경향을, 화산101 품종에서는 감소하는 경향이 나타났으나 유의적인 수준에서 차이는 확인되지 않았다. 반면 그린팜 품종에서는 유의적인 수준에서 고염 처리에 의한 APX 활성이 감소되는 경향이 나타났다. 지속적으로 in vitro screening을 통해 선발된 계통들은 향후 특성평가와 인공교배를 통해 내염성 이탈리안 라이그라스 개발에 필요한 유용한 육종모재로 활용될 수 있을 것으로 판단된다.
