Expression of Pea Superoxide Dismutase Gene in Transgenic Cucumber (Cucumis sativus L.) Plants (형질전환 오이(Cucumis sativus L.) 식물체에서 완두 Superoxide Dismutase 유전자의 발현)

  • To develop the fruits of cucumber (Cucumis sativus L.) producing high yields of superoxide dismutase (SOD), the MnSOD cDNA from pea (Pisum sativum) under the control of the cauliflower mosaic virus 35S promoter was introduced into cucumber using Agrobacterium tumefaciens (strain LBA 4404)-mediated transformation. The kanamycin-resistant shoots were selected on the selection medium containing MS basal salt, 1.0 mg/L zeatin, 0.1 mg/L IAA, 300 mg/L claforan, and 100 mg/L kanamycin. After 6 weeks of culture on the selection medium, the shoots were transferred to MS medium containing 0.2 mg/L NAA to induce roots. PCR analysis using the primers for neomycin phosphotransferase (NPTII) gene revealed that three plantlets were transformed. The fruits of one transgenic plant had approximately 3.2-fold higher SOD activity than those of non-transgenic plants. MnSOD isoenzyme band was strongly detected on native gel in fruits of transgenic plants.

Optimization for Functional Properties of Cabbage Extracts (썸머파워 양배추 추출물의 기능적 특성 최적화)

  • This study was conducted to optimize the extraction conditions from cabbage by a response surface methodology. In extraction conditions based on the central composite design with variations, the ratio of solvent to sample ($10\~30$mL/g), ethanol concentration ($0\~100\%$) and extraction temperature ($35\~95^{circ}C$) coefficients of determinations ($R^2$) were 0.8162(p<0.1), 0.8173(p<0.1), 0.9374(p<0.01) and 0.9116(p<0.05) in extraction yield, electron donating ability, tyrosinase inhibition and SOD-like ability, respectively. Estimated extraction conditions for the maximizing yield, electron donating ability and SOD-like ability were $15\~30$ mL/g in ratio of solvent to sample, $40\~80\%$; ethanol concentration, and $50\~90^{\circ}C$ ; extraction temperature. Predicted values at the optimum condition (25 mL/g solvent to sample, $50\%$ ethanol concentration and $70^{\circ}C$ in extraction temperature) were in good agreement with observed values.

  • To develop new anti-aging foods or cosmetics by using antioxidants, SOD activator and elastase inhibitor, both potent anti-aging substances, were screened from various extracts of medicinal Plants and its optimal extraction conditions were investigated. Antioxidant activity has showed the highest in methanol extracts of Prunus persica (seed; 98.0$\%$). Methanol extracts of Morus alba (leave; 41.0$\%$) showed the highest elastase inhibitory activity while Lycium chinense (fruit; 197$\%$) showed the highest activation effect in SOD activity. The Prunus persica extract that exhibited the highest activity was extracted by treatment of Prunus persica powder with methanol at 40$^{\circ}C$ for 18 h and the SOD activity was maximum with extract from Lycium chinense extracted with deionized water at 30$^{\circ}C$ for 12 h. Elastase inhibitory activity of Morus alba was maximally extracted when it was treated with 70$\%$ methanol at 50$^{\circ}C$ for 12 h.

Effect of Maengjong-Juk (Phyllostachys Pubescens) Extract Coated Rice Diet on Antioxidative System of C57BL/6 Mice Fed Atherogenic Diet (맹종죽(Phyllostachys Pubescens)추출물 코팅쌀이 Atherogenic 식이를 섭취한 C57BL/6 마우스의 항산화 시스템에 미치는 영향)

  • To evaluate the antioxidative effect of maengjong-juk (Phyllostachys pubescens) extract coated rice in vivo system, maengjong-juk extract coated rice diets were fed to C57BL/6 mice for 16 weeks. Plasma total antioxidative capacity, hepatic lipid peroxidation, protein oxidation, activities of antioxidative enzymes and total glutathione content were measured. Plasma total antioxidative capacity was elevated significantly in maengjong-juk extract diets supplemented group in a dose dependant manner. Hepatic TBARS contents were significantly decreased in maengjong-juk extract diets supplemented group compared to high cholesterol group. Maengjong-juk extract coated rice diets suppressed the protein oxidation significantly in liver. Activities of hepatic antioxidative enzymes such as total SOD, CuㆍZn-SOD, Mn-SOD, GSH-Px and catalase activities of maengjong-juk extract coated rice diets were significantly higher than those of high cholesterol diet. Total hepatic glutathione content was significantly increased by maengjong-juk extract coated rice diets administration. According to this study, numerous antioxidative materials and phytochemicals containing in maengjong-juk extracts appear to protect antioxidative systems in C57BL/6 mice fed bamboo extract coated rice diet. Effects of Radix Puerariae, Flos Puerariae and Bamboo+Radix Puerariae Water Extracts on the Ethanol-administered Mice (갈근, 갈화, 갈근과죽력의 추출물이 알코올을 투여한 생쥐에 미치는 영향)

  • This study was performed to investigate the effects of Radix Puerariae (RP), Flos Puerariae (FP) and Caules in Liquamen Phyllostachyos+Radix Puerariae water extracts on the alcohol dehydrogenase (ADH). transaminase (GOT, GPT) activities, and two hepatic antioxidant enzyme (SOD, catalase) activities in acute ethanol administered mice, and we have investigated the morphological changes that occur in hepatocytes of the experimental mice. The activities of ADH decreased compared with control group in the A1(66%), C1(57%), C2(54%) groups. The transaminase activites increased in the control groups compared with experimental groups. Ethanol treatment group without the RP or FP administration significantly lowered the activities of hepatic SOD and catalase, whereas MnSOD increased in the A1(27%) and B2(43%)groups, CuZnSOD increased in the B2(25%) and C2 groups. The catalase activites were increased in the A1(270%), A2(478%), B2(487%) and B1(770%) compared with control group, A lot of PAS-positive granules were observed in the A1, A2, C1 and C2 groups compared with the other groups. These results suggested that RP, FP and RP+Bamboo extracts administration may be prevent from liver damage in the alcohol treatment mice.

Induction of Immune Responses by Two Recombinant Proteins of Brucella abortus, Outer Membrane Proteins 2b Porin and Cu/Zn Superoxide Dismutase, in Mouse Model

  • The diagnosis of Brucella abortus is mainly based on serological methods using antibody against LPS, which has diagnostic problems. Therefore, to solve this problem, we evaluated two proteins of B. abortus, Cu/Zn superoxide dismutase (SodC) and outer membrane proteins 2b porin (Omp2b). The genes were cloned and expressed in a pMAL system, and the recombinant proteins, rOmp2b and rSodC, were purified as fusion forms with maltose-binding protein. The identity of the proteins was confirmed by SDS-PAGE and Western blot analysis with sera of mice infected with B. abortus. Production of cytokines and nitric oxide (NO) was investigated in RAW 264.7 cells and mouse splenocytes after stimulation with the proteins. Moreover, cellular and humoral immune responses were investigated in BALB/c mice after immunization with the proteins. TNF-${\alpha}$, IL-6, and NO were significantly inducible in RAW 264.7 cells. Splenocytes of naive mice produced IFN-${\gamma}$ and IL-4 significantly by stimulation. Moreover, number of IgG, IFN-${\gamma}$, and IL-4 producing cells were increased in immunized mice with the two proteins. Production of IgG and IgM with rOmp2b was higher than those with rSodC in immunized mice. These results suggest that the two recombinant proteins of B. abortus may be potential LPS-free proteins for diagnosis.

The Effects of Exercise and Other Relating Factors on the Activity of Erythrocyte Antioxidant Enzymes and Plasma TRAP Levies in Male College Students (남자 대학생의 적혈구 항산화 효소 활성 및 혈장 TRAP수준에 대한 운동량 및 기타 관련 요인의 영향)

  • This study was conducted to evaluate the effect of regular exercise and other relating factors on the activities of erythrocyte antioxidant enzymes and plasma total radical-trapping antioxidant potential (TRAP) in 61 healthy male college students. The study population were divided in two groups ; small amount of exerciser (exorcise time less than 30min/d) and moderate amount of exerciser (exorcise time more than 30min/d) according to their physical exercise habits measured by a questionnaire. Dietary intake of vitamin C and vitamin E, Plasma lipid Profiles, erythrocyte superoxide dismutase(SOD), glutathione peroxidase(GSH-Px) and catalase activities, as well as plasma TRAP levels were determined. Plasma TRAP level was significantly higher in moderate amount of exercisers than that in small amount of exercisers. No significant differences were observed in erythrocryte SOD, catalase and GSH-Px between the two groups. Mean exercise time was positively correlated with the plasma level of TRAP significantly, and amount of alcohol consumption was negatively correlated with the erythrocyte SOD activity, Dietary vitamin C and I intakes did not correlated with either erythrocyte enzyme activities or plasma TRAP levels. There were positive correlations between plasma HDL-cholesterol, and erythrocyte GSH-Px or plasma TRAP levels. Plasma vitamin C concentrations was negatively correlated with plasma TRAP levels and erythrocyte SOD activity, however plasma vitamin C concentration was positively correlated with erythrocyte GSH-Px activity, The results would suggest that regular moderate exercise, nonsmoking, high HDL-cholesterol and high plasma vitamin E concentration enhance antioxidant defences against reactive oxygen species and may increase the likelihood of a healthier life span.

Optimization of Extraction Conditions for Cabbage (홍월적 양배추의 추출조건 최적화)

  • This study was conducted to monitor the extraction yields and functional properties from cabbage by a response surface methodology. The extract yield was maximized as 44.47$\%$ under the temperature of 79.86$^{\circ}C$, ethanol concentration of 56.84$\%$ and solvent to sample ratio 25.58 mL/g . The maximum value of electron donating ability was 85.46$\%$ at 46.38$^{\circ}C$,57.06$\%$ of ethanol concentration and 27.71 mL/g of solvent to sample ratio. The maximum value of tyrosinase inhibitory effect was 69.37$\%$ at 37.5$^{\circ}C$,47.71$\%$ of ethanol concentration and 16.03 mL/g of solvent to sample ratio. The maximum value of SOD-like activity was 48.36$\%$ in 66.12$^{\circ}C$, 70.35$\%$ of ethanol concentration and 29.13 mL/g of solvent to sample ratio. Estimated conditions for the maximized extraction including yield, electron donating ability and SOD-like activity were 20 $\∼$ 30 mL/g in ratio of solvent to sample, 25$\∼$85$\%$ in ethanol concentration, and 40$\∼$90$^{\circ}C$ in extraction temperature.

Biological Activities of Maca (Lepidium meyenii) Extracts (마카 추출액의 생리활성 효과)

  • This study was conducted to determine the optimal extraction conditions for maca by comparing the yields, total polyphenol contents, superoxide dismutase (SOD)-like activity and the nitrite scavenging ability. The proximate composition analysis showed 6.57% moisture, 12.83% crude protein, 1.05% crude fat, 4.80% ash and 74.75% carbohydrate. Maca was extracted with 7 different solvents (water, methanol, ethanol, acetone, ethyl acetate, chloroform and hexane) and the extracts were tested for biological activities. The extraction yields of water, methanol and ethanol extracts were 46.2%, 21.4% and 16.8%, respectively. Acetone, ethyl acetate, chloroform and hexane exhibited very low extraction yield, ranging from 0.2 to 1.0%. Total polyphenol contents and the nitrite scavenging ability were the highest in water extract. Electron donating ability and the SOD-like activity were the highest in methanol extract. When water extract was drawn out at different extraction temperatures (30, 70, $100^{\circ}C$) and time (1, 3, 5 hr), the improved biological activities (total polyphenol contents, electron donating ability, SOD-like activity and nitrite scavenging ability) were found in extracts treated at $100^{\circ}C$ for 3 or 5 hrs.

The Detection of Superoxide Dismutase Activity and Isozyme Pattern of Panax ginseng C.A. Meyer Leaves (인삼엽에서 Superoxide Dismutase Activity 측정 및 Isozyme Pattern 검정)

  • We studied a assay method on the measurement of superoxide dismutase (SOD Superoxide : superoxide oxidoreductase, EC. 1. 15. 1. 1) activity with photoreduced flavin and nitroblue tetrazolium (NBT) as superoxide (${O_2}^{-}$) source and detector, respectively. The $\Delta$E (1000 ng SOD$.$$min.)^{-1}$ of photoreduced flavin-NBT system was 0.08, whereas that of xanthine-xanthine-cytochrome system used broadly in experiments was 0.014. Therefore, the new method was regarded more simple and utilizable than xanthine-xanthine cytochrome system method. In the present paper, we also carried out to investigate the SOD activity and isozyme pattern for the parpose of study of leaf-burning disease in ginseng (Panax ginseng C.A. Meyer) leaves.
