Pig slurry is a good soil amendment not only because of its high organic matter content, but also because of its ability to provide various nutrients. The objective of this study was to estimate the influence of raw and processed pig slurry application on pot soil over chemical fertilizer and non-amended control soil. Change in the chemical parameters (pH, organic matter (OM), organic carbon (OC), macro and micronutrients) and microbial mass of the treated soils were monitored over 30 to 90 days. Pot soil was treated with the recommended dose of pig slurry and chemical fertilizer, and was sampled after 30, 60 and 90 days of incubation. The least significanct difference (p<0.05) was observed on Fe, Cu, Zn, available P and K between treatments. All treatments increased N, P and K content and microbial mass of soil over control soil. Interestingly, no significant effects were detected on OM, OC, total bacteria, actinomycetes and fungi mass in soil irrespective of treatments given. However fungal and bacterial counts, as well as available nutrients, were found to be higher in processed slurry (PS)-treated soil compared to other soils. In general a significant correlation existed between the fungal count and OM, OC, Zn, T Kjeldahl N (TKN), available P and K of soil. A strong negative correlation was observed between pH and Fe in soil. This study clearly demonstrated that the use of processed manure as a fertilizer could be a key for sustainable livestock agriculture.
To examine the effects of lipoic acid on metabolic response to a ${\beta}$-agonist, isoproterenol, in broilers, chicks were fed dietary lipoic acid at levels of 0 (control) or 50 mg/kg for 24 d. At 27 d of age, chickens were randomly selected. Isoproterenol dissolved in 0.9% saline was injected into the wing vein at a dosage of 2 mg per kg BW; then, blood samples were taken at 0, 10, 20, 30, 60, 90, 120, and 180 min. Amounts of plasma glucose, NEFA, triglyceride and total cholesterol were determined. Dietary lipoic acid reduced only plasma total cholesterol by 25%. Following isoproterenol injection, plasma glucose in both groups increased for 20 min; then, it returned to its basal concentration. In contrast, the maximal reductions in plasma NEFA and triglyceride in both groups (20 to 30 min) were confirmed by isoproterenol injection. In addition, while glucose returned to the basal level, plasma NEFA in the lipoic acid-treated chickens increased above the basal or control value during the 60 to 180 min post-injection. The present study suggests that the dietary administration of lipoic acid elicits fatty acid mobilization in ${\beta}$-adrenergic response to isoproterenol when the basal level of plasma glucose is maintained.
The effect of active immunization against porcine somatostatin (SRIF-14) on somatostation and somatotropin secretion profile in 18 gilts was investigated. Gilts were assigned to the following treatments: control (sham injection, n = 6); bovine serum albumin (BSA) (injection of BSA with bacterial protein adjuvant, n = 6); SRIF (injection of BSA-SRIF-14 conjugate with bacterial protein adjuvant n = 6). Serum SRIF and pST were assayed from the blood samples taken on day 7 after the last immunization injection. Anti-SRIF antibody titres were assayed in weekly samples two weeks after the initial immunization to one week after the last immunization. Results revealed that the immunization protocol used in the present investigation failed to produce antibodies capable of neutralizing endogenous somatostatin. In addition, the porcine somatotropin assay revealed no significant differences in baseline pST concentration, mean peak amplitude and number of peaks during a 24 h secretory period among SRIF, BSA and control treatment. There were also no differences in SRIF baseline concentration, peak amplitude, and number of peaks during a 24 h secretory period among any of the three treatments. Circulating concentrations of pST and pSRIF were highly correlated (r = -0.09). Furthermore, anti-SRIF antibody titre was not detected in the serum of the gilts actively immunized against SRIF. These data, collectively, suggest that the protocol employed in the present investigation for active immunization against SRIF is not an effective method for changing SRIF and pST secretion profiles of the gilt and thus to enhance performance.
This study was carried out to investigate physicochemical properties of different concentrations (0.1%, 0.3%, and 0.5%) of red ginseng hydrolyzates (RGH)- or red ginseng extract (RGE)-added Asiago cheeses (AC) during ripening at $14^{\circ}C$ for 4 months. The moisture content significantly increased with increasing concentrations of both RGH- and RGE-added AC (p<0.05). While RGHAC and RGEAC were more yellow and darker with increasing concentrations than that of control (p<0.05), the color was not influenced from the hydrolysis. In texture analysis, hardness, cohesiveness, and chewiness of RGHAC and RGEAC significantly decreased compared to the control during the ripening (p<0.05). In sensory analysis, bitterness and ginseng flavor and taste scores increased significantly with increasing the concentrations of RGH and RGE during ripening (p<0.05). In conclusion, the addition of RGH and RGE into cheese slightly influenced the properties of Asiago cheese, and similarities were observed between RGHAC and RGEAC. Thus, the lower concentrations (0.1% to 0.3%) of RGH and RGE added to AC were preferred for color, texture, and sensory during the ripening, therefore, these cheeses would be worth developing commercially.
In vivo antagonistic effect of Lactobacillus helveticus CU 631 and Lactobacillus spp. against typical enteritis causing pathogen Salmonella enteritidis KU 101 have been determined, which showed an increase in survival rate and the decline in viable cell numbers of pathogen in liver and spleen at sacrifice. A signifcant difference in the antagonistic effect against KU 101 were observed, which was species and/or strain dependent of Lactobacillus (p<0.01), the survival rate of the mice in the Salmonella infection by feeding L. helveticus CU 631 has been shown to be 157%, whereas those of L. rhamnosus GG ATCC 53103, L. acidophilus ATCC 4356, L. johnsonii C-4 were 137%, 132%, 119% respectively on the basis of lactobacilli non-associated control KU101 fed mice to be 100%. Viable cells of S. enteritidis KU101 in the liver and in the spleen at sacrifice were decreased in Lactobacillus spp. fed group with no significant difference. The higher level of total secretory IgA concentration in the intestinal fluid of lactobacilli fed mice than control mice have been observed. In vitro antagonistic activity of Lactobacillus spp. against KU101 have been determined, a prominent antagonistic activity of CU 631 against KU 101 were demonstrated.
An experiment was conducted to investigate the effects of equiprotein replacement of dietary fish meal (FM) with duckweed (DW) and soybean meal (SBM) on the performance of broilers. A total of 112 seven-day-old as hatched broilers were fed on 4 different iso-energetic (2,818 kcal/kg) and iso-nitrogeneous (20.2% CP) diets up to 56 days of age. Diet A was control with 12% FM. In diets B (3% DW + 13.5% SBM), C (6% DW + 11.5% SBM) and D (9% DW +10% SBM). All FM protein of control diet was replaced by DW and SBM. The replacement of dietary FM by DW and SBM depressed feed intake, live weight gain and feed conversion efficiency and increased production cost and thus affected profitability. All those growth parameters had a linear declining trend as the proportion of DW in the diet was increased. It may be concluded that complete replacement of dietary FM by DW and SBM should not be recommended for raising broilers.
Weaner heifers were set stocked at 4/ha on a grass-legume pasture in South Sulawesi, Indonesia, and either unsupplemented (Control) or for 338 days given daily supplements of one of, rice bran (RB) supplied at 1 kg/animal/d, molasses/urea (MU) or 0.5 RB + 0.5 MU (MURB) the amounts of which were adjusted to give similar energy intakes. There were 20 animals in each treatment. A drought resulted in low pasture availability for about half the supplementation period. The LWG per animal in the MURB treatment was 85 kg above that of the control and this was significantly greater (p < 0.01) than those for MU (62.0 kg) or RB (56.2 kg) although the economics favoured the gains from RB which returned over three times the cost of the supplement. Costs could be reduced by supplementing only at times of maximum undernutrition, but such a strategy is of doubtful value in this situation as there was no compensatory LWG and a similar rate of economic return was maintained throughout the period. The results suggest that additional benefits from the supplementation may be improved reproductive performance and more efficient use of pasture.
Objective: The objectives of this study were to investigate the direct antioxidative effect of 90 Kda heat shock protein (Hsp90) obtained from duck muscle. Methods: The interaction of Hsp90 with phospholipids and oxidized phospholipids was studied with surface plasmon resonance (SPR), and their further oxidation in the presence of Hsp90 was evaluated with thiobarbituric acid reactive substances (TBARS) assay. The scavenging effect on the 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azinobis (3-ethylbenzthiazoline-6-sulfonic acid (ABTS) was measured, and the electron paramagnetic resonance (EPR) spectroscopy in combination with 5-tert-Butoxycarbonyl-5-methyl-1-pyrroline-N-oxide and 2-phenyl-4,4,5,5,-tetramethylimidazoline-1-oxyl-3-oxide (PTIO) was utilized to determine the abilities of Hsp90 in scavenging hydroxyl and PTIO radicals. Results: SPR showed Hsp90 could bind with both phospholipids and oxidized phospholipids, and prevent their further oxidation by the TBARS assay. The DPPH and ABTS scavenging activity increased with Hsp90 concentration, and could reach 27% and 20% respectively at the protein concentration of 50 μM. The EPR spectra demonstrated Hsp90 could directly scavenge ·OH and PTIO· radicals. Conclusion: This suggests that Hsp90, a natural antioxidant in meat, may play an important role in cellular defense against oxidative stress, and may have potential use in meat products.
The effects of feeding rice bran and beet pulp mixtures on the site and extent of digestion and microbial synthesis in fattening steers were studied. Three Holstein steers fitted with ruminal, duodenal and ileal cannulas were fed four diets in a $4{\times}3$ Youden square design. The four diets consisted of 15% Italian ryegrass hay and 85% concentrate as a control diet which included 72% rolled barley, 20% rice bran plus 40% beet pulp, 30% rice bran plus 30% beet pulp or 40% rice bran plus 20% beet pulp. All diets provided 1.8 times digestible energy required for maintenance. The digestibility of fat in the small intestine (% of flow) showed an increase with rice bran content among the by-product diets. Digestibility of structural carbohydrate both in the rumen and the whole digestive tract decreased linearly with rice bran content. The digestibility of nonstructural carbohydrate was not affected by rice bran content, but that of nonstructural, nonstarch polysaccharides was higher in the rumen and lower in the large intestine for the by-product diets than for the control diet. A rice bran content of more than 30% in the by-product diets severely inhibits ruminal microbial synthesis and digestible energy intake in fattening steers.
The influence of dietary vitamin E supplementation on meat color and ${\alpha}$-tocopherol concentrations in plasma, longissimus thoracis muscle and subcutaneous fat was investigated. Ten Japanese $Black{\times}Holstein$ crossbred steers were placed in three experimental groups receiving different supplements of vitamin E. Four steers (control) were fed no supplemental vitamin E. Two groups of three steers each, were supplemented with 500 mg dl-${\alpha}$-tocopherol acetate per animal daily for 175 days and 1,000 mg for 100 days, respectively, before slaughter. The ${\alpha}$-tocopherol concentration in plasma increased, as vitamin E were fed, and were related to the length of time and the amount of supplement. The ${\alpha}$-tocopherol concentrations in the muscle and the fat from the two supplemental groups did not differ significantly and were three or more times greater than those in the control group. Vitamin E supplementation did not affect the quantity of marbling of beef. Supplemental vitamin E stabilized the color of displayed beef around wavelengths of 500 nm and 640 nm. The two methods of vitamin E supplementation had similar effects on meat color. The effect of supplemental vitamin E on the color of beef with marbling was observed 2-3 days after slaughter and was followed for another two weeks.
