Monthly Occurrence of Vectors and Reservoir Rodents of Scrub Typhus in an Endemic Area of Jeollanam-do, Korea

  • Monthly surveys were conducted to investigate the occurrence of chigger mites and seroprevalence of scrub typhus among small mammals in Jeollanam-do, the southwestern part of Korea, from November 2006 through October 2007. Fifty-eight small mammals, including 57 Apodemus agrarius (98.3%) and 1 Crocidura lasiura (1.7%), were captured, and a total of 4,675 chigger mites representing 4 genera and 8 species were collected from them. The chigger infestation rate among small mammals was 69.0%. The most predominant species in A. agrarius was Leptotrombidium scutellare (54.0%), followed by Leptotrombidium pallidum (39.4%), Leptotrombidium orientale (4.4%), Leptotrombidium palpale (1.1%), Neotrombicula tamiyai (0.6%), Eushoengastia koreaensis (0.3%), Neotrombicula gardellai (0.3%), and Cheladonta ikaoensis (<0.1%). The chigger index of A. agrarius was the highest in October (740.0), followed by November (242.0), September (134.6), March (98.3), February (38.2), January (35.3), December (34.5), April (30.8), and May (1.7). The average antibody positive rate of scrub typhus in wild rodents was 50.0%. The seropositive rates were high in October (100.0%) and November (83.3%), whereas those in other months were relatively low (28.6-57.1%). The chigger index of L. scutellare rapidly increased in September to form an acuminate peak in October, followed by a gradual decline. These results suggest that the outbreak of scrub typhus in the southwestern part of Korean peninsula is mostly due to L. scutellare.

Effects of $\omega$6 and $\omega$3 Fatty Acid Diets on the Fatty Acid Composition of the Mesenteric and Subcutaneous Fat of Lactating Rats

  • Long chain polyunsaturated fatty acids (LCPUFA) are important components of brain phospholipds and play important role (s) in brain function. In rats, the maximum brain growth occurs during the period of lactation even though it happens during the third trimester of gestation in human. Since milk contained docosahexaenoic acid (DHA) even through the maternal diet had no DHA and/or a very small amount of its precursor, $\alpha$-linolenic acid ($\alpha$-LnA), an emphasis was given to maternal adipose tissue as a reservoir of this fatty acid. We, therefore, investigated the mesenteric and subcutaneous adipose tissues for their fatty acid composition in dams reared with different fat diets. Diets containing various amounts of $\omega$6 and $\omega$3 fatty acids were given to adult female rats (200-250g) throughout the pregnancy and lactation periods. Diets were composed of 10% (wt/wt) corn oil (CO), soybean oil (SO), perilla seed oil (PO) containing about 60% $\alpha$-LnA, or fish oil (FO) rich in eicosapentaenoic acid (EPA) and DHA. The fatty acid ompositions of mesenteric and subcutaneous fat were measured and evaluated at Day-2 and Day-15 after parturition. In general, major characteristics of dietary fatty acid composition was reflected on the fatty acid composition of adipose tissues. Dietary fatty acid composition was reflected more on mesenteric fat as compared to subcutaneous fat. Mesenteric fat was found to contain less arachidonic acid (AA) and mesenteric fats of CO, SO and PO groups contained less DHA than did the subcutaneous fat. The P/M/S ratios of adipose tissues were similar between experimental groups while dietary P/M/S ratios differed significantly. It was noticeable that a small proportion of DHA was found in the adipose tissues of animals of CO, SO and PO groups (Day-2) and in SO and PO groups (Day-15), the groups which do not contain DHA in their diets. The percentage of DHA in mesenteric fat o CO, SO and PO groups decreased as lactation continues, while the proportion of DHA in FO group increased. Adipose tissues of FO group had higher DHA/EPA ratio as compared to the diet. Considering the fact that the body contains a large amount of adipose tissues, our present finding suggests that the adipose tissue can serve as a reservoir of DHA for pregnant and lactating rats.

Sampling Effects on Fishing Gears in the Hoengseong Resevoir (포획도구에 따른 횡성호내 어류채집 효과분석)

  • Fish community in the Hoengseong Reservoir was investigated from April to October 2005. The total number of fish caught from the period was 8,626 fish representing 22 species and 8 families, using tyke net and trammel net. Besides biomass was 122.9 kg. In the fyke net, 8,168 individuals caught during the period represented 21 species belonged to 8 families, its biomass was 90.6 kg. On the other hand, 458 individuals caught in the trammel net during the period represented 16species belonged to 6 families, its biomass was 26.3 kg. The results showed that the trammel net had lower number of species, individuals, and less biomass compared to those of the tyke net. Number of species confirmed was increased according as mesh size of the net was smaller. Besides, the fyke net with small mesh size was able to collect not only small-sized fish but also large-sized fish so that it was suitable to study qualitative and quantitative analysis of fish fauna in detail. Whereas, the trammel net with proper mesh size was effective to determine composition of fish species in the studied area.

Application of DNA Analysis for Identification of Prey Items on Zooplankton: Selective Treatment Method (기수역 요각류 위내용물 유전자 분석: 소화기관 내외부 유전자의 선택적 처리방법)

  • Understanding the selective feeding behavior of zooplankton on phytoplankton is essential for evaluating the nutrient cycle and energy flow in the food web. Although many studies have been conducted regarding the feeding behaviors of zooplankton through gut content analyses, there are limitations in the visual identification of digested contents using a microscope. DNA techniques have been applied to overcome these limitations since they can detect and amplify small amounts of prey DNA remaining in the gut contents. We designed a method to extract prey DNA from the gut contents of the whole body of the copepod specimen and tested the resolution of DNA identification for the prey phytoplankton. The common brackish species, Sinocalanus tenellus, were collected from Saemangeum Reservoir in different sites and seasons, and gut content DNA was extracted using 2.5% bleach treatment for 2 min for removal of potential contamination sources existing in preserved specimens without dissolution of the body. The sequences of the extracted gut contents were confirmed using BLASTn suite based on the NCBI database. The phytoplankton species detected in the gut showed temporal and spatial differences. Although DNA analysis of small copepod gut contents has been suggested as an effective method to examine the dynamics of primary prey sources at the genus or species level, uncertainties such as misidentification and limitations in the detailed information of the composition still exist.

Evaluation of the Water Quality Changes in Agricultural Reservoir Covered with Floating Photovoltaic Solar-Tracking Systems (수상 회전식 태양광 발전시설 설치에 따른 농업용 저수지의 수질변화 평가)

  • To evaluate the water quality changes in agricultural reservoir covered with floating photovoltaic solar-tracking systems, the water quality variations with time and depth were monitored on both six sites for light blocking zones and four sites for light penetration zones after the installation of floating photovoltaic solar-tracking systems in Geumgwang reservoir at Anseong-si, Kyeonggi province. For one year with 16 monitoring events, water quality parameters [i.e., water temperature, pH, dissolved oxygen (DO), chlorophyll-a (Chl-a), and blue-green algae (BGA)] were monitored at depths of 0.3 m, 1 m, 3 m, and 5 m, while chemical oxygen demand (COD), total nitrogen (TN), and total phosphorus (TP) were monitored at depths of 0.3 m. Statistically, the difference in all water quality parameters was not significantly different (p > 0.05) at the level of significance of 0.05. Based on these results, the water quality data from light blocking zones (site 1~6) and light penetration zones (site 7~10) were clustered, and were compared with time and depth. As a result, the difference in water temperature, pH, DO, COD, TN, TP, Chl-a, and BGA between light blocking zones and light penetration zones was not significant (p > 0.05) with different time and depth. For Chl-a and BGA, some data from light blocking zones greater than light penetration zones were temporary observed due to the severe drought, low water storage rate, and over growth of periphyton. However, this temporal phenomenon did not impact the water quality. Considering the small water surface area (${\leq}0.5%$) covered by floating photovoltaic solar-tracking systems, the mixing effect of whole Geumgwang reservoir caused by Ekman current and continuous discharge were more dominant than the effect of reduced solar irradiance. Further study is warranted to monitor the changes in water quality and aquatic ecosystems with greater water surface area covered by floating photovoltaic solar-tracking systems for a long time.

  • Sustainable energy generation is becoming extremely imperative due to the expected limitations in current energy resources and to reduce pollution. Especially, because of its considerable energy potential, ocean wave energy has been investigated with regard to power generation. To develop large high power wave generator system, it is important to make a small scale proto type and to test that. Thus the objective of this research is to examine the characteristics of a mechanically excited generator system having small power capacity experimentally. The water reservoir (4 m length, 1.5 m width and 1.8 m depth) having a wave maker to make arbitrary height and period of the water wave was made. The proto type consists of three main parts; a buoy, rack-pinion base one-way mechanism, and a wave generator(Fig.1). The water wave is going up and down and the hexahedron buoy is following the wave. The rack gear attached to the buoy is also going up and down to roll the pinion connected to an electric generator then it produces electricity. The experiments were performed with several conditions of water waves, and the power outputs over 30 W could be measured for some conditions. In future works, to achieve higher performance for the proto type, the effects of primary parameters (buoy shape and mass, etc.) on the system efficiency will be identified.

Numerical Modeling of Free Surface at the Fish Migration Spillways (어류 이동용 여수로의 자유수면 수치해석)

  • A level-set finite-element method is employed to investigate the free surface flow of two conceptual fish passage designs. While one design has a right-angled entrance from the reservoir to the fish passage chute, the other has a curved-shaped entrance. The numerical results are validated with hydraulic experiments through comparison of the free surface location and the pressure distribution in the spillway. It is observed that the right-angled design yields a curved free surface and pressure distribution in the vicinity of the entrance due to large strains, whereas the curved-shaped design yields a smooth flow transition with small strains. The subatmospheirc pressure distributions near the spillway ogee crest for both designs exhibit similar flow patterns. It is concluded that the curved-shaped design is more favorable for fish passage because of the feature of a smooth flow transition with small strains.

Use of the cast net for monitoring fish status in reservoirs distributed in the Korean peninsula

  • Reservoirs consist of two different environments, the littoral and the pelagic zone, and different fishing gear is commonly used in each zone-gill nets in the pelagic zone and electrofishing in the littoral zone. However, an active fishing gear, the cast net, is normally used instead of electrofishing for scientific studies in South Korea. In order to estimate cast net effectiveness for determining fish status in reservoirs, the study was conducted at 15 reservoirs with two different fishing gears: a cast net in the littoral zone and gill nets in the pelagic zone. When combining catches of both gears, species richness increased substantially compared to using one gear only. There was a size difference in fish caught by each net, and small fish were predominantly caught with the cast net due to its small mesh size (7 mm). The combined length of six species, used for length-weight relationship analysis, collected with the cast net was smaller than that collected with gill nets (independent t-test, P < 0.05). In this study, cast net sampling provided sufficient data for the littoral zone, but not enough to identify the overall fish assemblage in studied reservoirs. Utilization of only one gear can therefore lead to substantial underestimation of fish status, and a combination of both gears is recommended for determining more reliable estimates of fish status in reservoirs.

History and Current Situation of River Management using Physical Habitat Models in the U.S. and Japan

  • History of Instream Flow Incremental Methodology (IFIM) Following the large reservoir and water development era of the mid-twentieth century in North America, resource agencies became concerned over the loss of many miles of riverine fish and wildlife resources in the arid western United States. Consequently, several western states began issuing rules for protecting existing stream resources from future depletions caused by accelerated water development. Many assessment methods appeared during the 1960's and early 1970's. These techniques were based on hydrologic analysis of the water supply and hydraulic considerations of critical stream channel segments, coupled with empirical observations of habitat quality and an understanding of riverine fish ecology. Following enactment of the National Environmental Policy Act (NEPA) of 1970, attention was shifted from minimum flows to the evaluation of alternative designs and operations of federally funded water projects. Methods capable of quantifying the effect of incremental changes in stream flow to evaluate a series of possible alternative development schemes were needed. This need led to the development of habitat versus discharge functions developed from life stage-specific relations for selected species, that is, fish passage, spawning, and rearing habitat versus flow for trout or salmon. During the late 1970's and early 1980's, an era of small hydropower development began. Hundreds of proposed hydropower sites in the Pacific Northwest and New England regions of the United States came under intensive examination by state and federal fishery management interests. During this transition period from evaluating large federal reservoirs to evaluating license applications for small hydropower, the Instream Flow Incremental Methodology (IFIM) was developed under the guidance of the U.S. Fish and Wildlife Service (USFWS).

The Study on the Evaluation of Environment Function at Small Stream - In the Case of Hongdong Stream in Hongsung-gun - (농촌지역 소하천의 환경성 평가 연구 - 홍성군 홍동천을 사례로 -)

  • This study was conducted to understand the environmental and ecological function of habitat through evaluation of water environment, soil environment, vegetation characteristics, macro-invertebrate characteristics, and visual habits environment evaluation (SVAP) in Hongdong stream located in Hongseong-Gun, Chungnam Province, and hereafter to utilize the results for the habits reconstruction and improvement project. As the results of water quality analysis, BOD, COD, T-P was almost below the standard quality from upper stream to down stream. The construction of small reservoir, wetland and water purification facility, and the management of non-point pollution are proposed to improve these problems. The soil texture was sandy soil, which is unfitted with vegetation development. The construction of shallows and bogs, and induction of soil sedimentation and biotope formation are proposed to improve these problems. In the plant flora, total 90 kinds were observed with 81 species and nine varieties in total 36 families and 85 genera, and the naturalization rate was higher at down area than upper area. As the results of macro-invertebrate fauna survey, total 26 species and 297 individuals in 20 families and 22 genera were collected. Peltodytes sinensis, Chironomidae sp., and Culicidae sp., which are observed at polluted environment, were collected as dominant species. An appropriate vegetation management party idea is necessary, and it is done an idea in consideration of the soil and a physical characteristic. Visual habits environment evaluation (SVAP) result was mostly determined with below normal (Fair) grade. Pollution source interception, purification pond establishment, and various bog establishment are proposed to improve these problems. With the above results, the ecological environment was determined with bad condition, and the improvement of biotope was urgently needed through sustainable monitoring and management of streams habitat in rural area.
