Dihydrolipoamide dehydrogenase (E3) catalyzes the reoxidation of dihydrolipoyl moiety of the acyltransferase components of three $\alpha$-keto acid dehydrogenase complexes and of the hydrogen-carrier protein of the glycine cleavage system. His-457 of Pseudomonas putida E3 is suggested to interact with the hydroxyl group of Tyr-18 of the other subunit and with Glu-446, a component in the last helical structure. To examine the importance of the suggested interactions in human E3 function, the corresponding residue of human E3, Asn-473, was substituted to Leu using site-directed mutagenesis. The E3 mutant was expressed in Escherichia coli and highly purified using an affinity column. Its E3 activity was decreased about 37-fold, indicating that Asn-473 residue was important to the efficient catalytic function of human E3. Its slightly altered spectroscopic properties implied that small conformational changes could occur in the E3 mutant.
This paper examined the stability of high-dense dislocation substructures (HDDSs) associated with martensite laths in High Cr steels supposed to be used for FBR, based on a series of dislocation dynamics (DD) simulations. The DD simulations considered interactions of dislocations with impurity atoms and precipitates which substantially stabilize the structure. For simulating the dissociation processes, a point defect model is developed and implemented into a discrete DD code. Wall structure composed of high dense dislocations with and without small precipitates were artificially constructed in a simulation cell, and the stability/instability conditions of the walls were systematically investigated in the light of experimentally observed coarsening behavior of the precipitates, i.e., stress dependency of the coarsening rate and the effect of external stress. The effect of stress-dependent coarsening of the precipitates together with application of external stress on the subsequent behavior of initially stabilized dislocation structures was examined.
The free energy simulation technique is used to evaluate the relative binding affinity of a set of hydroxyethylene isostere inhibitors of the HIV-1 protease. The binding reactions and an alchemical mutation construct the thermodynamic cycle, which reduces the free energy difference of the binding interactions into that of the alchemical processes. In the alchemical process, a methyl group is mutated into a hydrogen atom. Albeit the change is a small perturbation to the inhibitor-protease complex, it results in 25 fold difference in the binding constants. The simulation reproduces the experimentally measured binding affinities within 2% of the free energy difference. The protonation state of the catalytic aspartic acid residues is also investigated through the free energy simulations.
Statistical thinking has a broad definition but focuses on the context of regression modelling in the present study. To foster students' statistical thinking within the context, teaching should no longer be seen as transfer of knowledge from teacher to students but as a process of engaging with learning activities in which they develop ownership of knowledge. This study aims at collaborative learning contexts; students were divided into small groups in order to increase opportunities for peer collaboration. Each group of students was asked to do a regression project after class. Through doing the project, they learnt to organize and connect previously accrued piecemeal statistical knowledge in an integrated manner. They could also clarify misunderstandings and solve problems through verbal exchanges among themselves. They gave a clear and lucid account of the model they had built and showed collaborative interactions when presenting their projects in front of class. A survey was conducted to solicit their feedback on how peer collaboration would facilitate learning of statistics. Almost all students found their interaction with their peers productive; they focused on the development of statistical thinking with concerted effort.
Purpose: In the theory of traditional medicine, Glenditsia spina(GS) can resolve carbuncle, relive swelling, dispel wind and destroy parasites. This study was designed to investigate the effects of GS on gene expression of ovarian tissue in polycystic ovary syndrome(PCOS) rats. Methods: In this experiment, female rats injected with a single dose of 2 mg estradiol valerate(EV) and GS was given for 5 weeks. The genetic profile for the effects on ovarian tissue in PCOS rats was measured using microarray technique, and the functional analysis on these genes was conducted. Results: 985 genes were increased in control and restored to normal level in GS group. (B), 733 genes were decreased in control group and restored to normal level in GS group. (F). Metabolic pathways related in B group genes were Graft-versus-host disease, Allograft rejection, Autoimmune thyroid disease, Cytokine-cytokine receptor interaction, Small cell lung cancer, Type I diabetes mellitus. Metabolic pathways related in F group genes were Antigen processing and present, Adipocytokine signalling pathway, Focal adhesion, ECM-receptor interaction, Pancreatic cancer, Notch signalling pathway, Tight junction. The network of total protein interactions was measured using cytoscape program, and some key molecules, such as c-Fos, c-Myc, ABL1 related in B group, MAPK8, RASA1, CALR related in F group that can be used for elucidation of therapeutical mechanism of medicine in future were identified. Conclusion: These results suggest possibility of GS as anti-cancer and anti-hyperplasia drug in PCOS. In addition, the present author also suggests that related mechanisms are involved in suppression of proto-oncogene such as c-Fos, c-Myc and ABL1, and in regulation of cell cycle such as RASA1.
본 연구에서는 학업 성취도, 과학 수업에 대한 태도, 과학에 대한 태도에 미치는 협동학습과 개별학습의 효과를 학습자의 성취 수준에 따라 비교하였다. 협동학습과 개별학습에서는 학습의 책임을 학습자에게 보다 많이 부여하고, 고급 사고 수준의 활동이 포함된 과제를 학습자 중심으로 해결하도록 하였다. 협동학습 집단은 소집단 토론을 통하여 이러한 과제를 해결하였고, 개별학습 집단은 동일한 활동지를 개별적으로 해결하였다. 전통적인 수업은 교사 설명 위주로 진행하였다. 연구 대상은 남녀 공학 중학교 1학년 학생이었으며, '혼합물의 분리' 단원에 대하여 총 10차시 동안 수업을 실시하였다. 이원 공변량 분석 결과, 협동학습 집단의 학업 성취도 검사 점수가 개별학습 집단이나 전통적 수업 집단에 비하여 유의미하게 높았다. 또한, 과학 수업에 대한 태도 및 과학에 대한 태도도 협동학습 집단이 보다 긍정적이었다. 모든 변인에서 수업 처치와 학습자 성취 수준 사이의 상호작용 효과는 나타나지 않았다. 즉, 협동학습은 학습자의 성취 수준과 무관하게 효과적이었다.
이 연구에서는 과학적 논변활동에서 드러나는 학생들의 인식론적 프레이밍을 확인하고, 소집단 구성원들의 프레이밍 사이의 상호작용이 논변활동에 어떠한 영향을 미치는지를 탐구하였다. 21명의 과학영재 학생들이 세 명 혹은 네 명이 한 집단을 이루어 연구에 참여하였고 학생들이 광합성 속도에 대한 탐구 데이터를 해석하는 논의 과정이 분석되었다. 학생들의 활동은 소집단별로 녹화되었고 학생들의 담화 전사본과 행동이 분석 자료로 활용되었다. 탐구 과정에서 변칙 사례에 반응하는 학생들의 발화 및 행동을 분석하여 그들의 인식론적 프레이밍을 확인하였고 주장에 대한 정당화의 근거와 소집단 논변활동 수준이 분석되었다. 연구 결과, 학생들은 탐구에 대해 '현상 이해'와 '교실 게임'이라는 두 가지 방식의 프레이밍을 보였다. 구성원들이 '현상 이해'의 프레이밍을 보인 경우에는 다른 구성원들에게 데이터의 신뢰성과 타당성 점검을 통해 변칙 사례의 원인을 정당화할 것을 요구하였으며, 이는 높은 수준의 논변활동으로 나타났다. 반대로 구성원들이 '교실 게임'에 해당하는 프레이밍을 보인 경우에는 변칙 사례를 설명할 필요성을 인식하지 않았고, 단순한 경험적 근거만 고려함으로써 낮은 수준의 논변활동을 보였다. 논의 전반에 걸쳐 구성원들의 프레이밍이 일치하지 않는 경우에는 집단 내의 감정적 갈등이 유발되어 깊은 논의가 이루어지지 않았다. 한편, 이러한 논의 과정에서 학생들의 프레이밍 전환이 관찰되었는데, 여기에는 집단의 리더가 큰 영향을 끼친 것으로 나타났다. 본 연구는 과학 교실에서 과학적 논변에 요구되는 생산적인 프레이밍 형성을 위한 기초 정보를 제공할 것으로 기대된다.
이 연구의 목적은 Thinking Science의 모둠 활동에서 나타나는 교사들의 도움 특성과 교사들의 도움 특성에 따른 학생들의 반응 특성을 알아보는 것이었다. 연구를 위해 초등학교 5학년 24명과 중학교 1학년 24명을 선정하여 TS 활동을 처치하였으며, 그 중 학생들의 논의가 비교적 활발했던 활동 4와 6에 대해 초 중학교 각각 2모둠의 언어적 상호작용을 분석하였다. 활동 처치 전에 과학적 사고력 검사를 통해 학생들의 인지수준을 확인하였으며, 이에 따라 이질집단으로 모둠을 구성 하였다. 각 모둠마다 도움 교사를 두었으며 TS 활동의 적용과정에서는 모둠 내 상호작용을 강조하였다. 교사들의 도움은 단순 확인 질문의 활용이 높았으며 메타질문의 활용은 부족하였다. 또한 교사의 개인적인 성향이나 교직 경험, 활동에 대한 숙지 정도에 따라 교사들의 도움 특성이 다르게 나타났다. 단순 확인 질문이 많고 교사의 개입이 많으면 학생들의 논의는 자발적이지 못하고, 메타질문의 활용과 피드백이 고르게 활용된 교사의 경우 학생들의 논의과정 요소의 활용 또한 다양하게 나타났다. 그러므로 과학적 사고력을 향상시키기 위한 TS 활동을 처치할 때 처치의 효과를 높이기 위해서는 TS 프로그램의 내용과 TS에서 요구되는 전략뿐만 아니라, 교사들이 학생들과 상호작용하는데 있어서 열린 교수-학습 분위기 속에서 학생들의 사고를 자극시켜 그들 자신의 아이디어를 제안하게 하고 또한 그들의 의견을 정당화 할 수 있는 기회를 제공할 수 있는 교수 기술을 배울 수 있는 교사 연수를 실시할 필요성이 있음을 시사한다.
본 연구에서는 과학영재 학생들을 대상으로 생물 영역의 동물 발생 실험수업을 통해 나타나는 언어적 상호작용을 분석하여 과학영재성을 보여주는 특성들을 알아보고자 하였다. 연구 대상은 서울대학교 과학영재센터의 중학교 1학년 학생 20명이었으며, Creative Problem Solving (CPS) 모형을 기반으로 개발된 동물 발생 실험 프로그램으로 소집단 토의 중심의 실험 수업을 총 4회 진행하면서 학생들의 언어적 상호작용을 녹음 및 녹화, 관찰하였다. 전사된 내용으로부터 질문-응답 유형과 과학영재의 특성을 추출하여 분석하였다. 질문-응답의 유형 분석에서는 보다 높은 사고력을 요하는 사고형 질문이 기본정보형 질문보다 3~6배나 높게 나타났다. 특히, 탐구실험 I은 다른 세 탐구실험보다 기본정보형 및 사고형 질문이 40% 이상 높게 나타났다. 언어적 상호작용의 분석 결과 창의적 지필검사에서 보기 어려웠던 창의성의 세부 요인을 확인할 수 있었으며, 문제발견력 및 문제해결력, 고도의 언어능력 등을 확인하였고, 정의적 특성인 과제 집착력과 리더십도 드러나는 것을 볼 수 있었다. 본 연구 결과는 지필고사를 대신하는 영재의 판별 및 선발에서 활용할 수 있는 프로그램 개발에 실질적인 도움을 줄 수 있을 것으로 사료된다.
본 연구의 목적은 논변 중심 과학 수업에서 교사의 반응적 발화가 학생들의 논변 활동에 어떠한 영향을 미치는지 수업 사례를 탐색함으로써 반응적 교수 실행 전략을 제안하는 것이다. 논변 수업의 경험이 없는 중학교 교사 2명과 학생 57명이 연구에 참여하였으며 '자극과 반응' 단원을 중심으로 논변 활동 중심의 과학 수업이 이루어졌다. 모든 수업과 인터뷰는 녹화, 녹음 후 전사하여 주요 분석 자료로 활용하였다. 근거 이론에 따라 두 교사의 반응적 발화를 유형화하여 논변의 구조적 대화적 측면으로 범주화하였으며, 이를 토대로 교사의 반응적 교수 실행이 드러난 수업 사례를 중심으로 질적 분석을 수행하였다. 연구 결과 두 교사는 논변의 대화적 측면 보다는 구조적 측면에 주로 반응하였는데, 특히 전체 논의 보다는 소집단 활동에서 이러한 양상이 두드러졌다. 이는 한 명의 교사가 짧은 시간 내에 여러 소집단에 개입해야 하는 교실의 상황과 논변 활동의 구조적 측면에 주된 목표를 둔 교사의 인식이 주요 요인이었다. 논변의 구조적 측면에 대한 교사의 반응적 발화 중 학생의 사고를 탐색하거나 추론을 촉진하는 발화는 학생들이 자신의 사고를 드러내고 수업의 목표를 인식하며 추론의 책임을 갖고 스스로 사고해 나가도록 지원하였다. 논변의 대화적 측면에 대한 교사의 반응적 발화는 주로 5차시 수업의 전체 논의에서 다수 나타났는데 소집단 간 논변의 차이를 명확히 보여준 수업 과제가 대화적 상호작용 촉진의 기회를 제공하였다. 이때 교사의 논변 간 공통점과 차이점을 부각시키는 발화, 소집단의 논변을 학급 전체에 발표하게 하는 발화, 특정한 학생을 지목하여 논변을 검증하고 평가하게 한 발화들은 소집단 간 논변의 차이를 명확히 드러내게 하여 소집단 간 검증과 반박을 이끌었고, 서로 다른 주장을 담은 논변 간의 연결을 지속적으로 지원하며 대화적 상호작용을 촉진하였다. 이러한 결과를 바탕으로 논변 활동에서 교사의 반응적 교수 실행을 위한 교수 전략을 제언하였다.
