Tolerance of Anaerobic Granular Sludge to Oxygen

  • To study the tolerant capacity of anaerobic granular sludge (AGS) to oxygen using semi -dynamic batch experiment, the aerating time, pH of the basal media, reductive inorganic materials, microorganism, and microorganism metabolite were investigated. When the aerating time was higher or lower than 0.5 h, the producing gas activity of sludge was lower than that of the control. The oxygen tolerance of the experimental sludge was the highest at the initial pH 7.2. The producing gas activity of sludge I was higher than that of sludge II. And storage at $4^{\circ}C$ can low the lose of the oxygen tolerance capacity of granular sludge. The producing gas activity of sludge was the highest when KI was added. The growth of aerobic microorganisms and some metabolite could increase the producing gas activity of granular sludge.

  • The effect of upflow velocity on size distribution and activity of granular sludge was studied in laboratory-scale Expended Granular Sludge Bed(EGSB) reactor fed with non-fat dry milk and sucrose as sole carbon and energy source. High upflow velocity advanced size and activity of granular sludge by distribution and floatation of granular sludge. Therefore, the reactor operation of an apt upflow velocity was needed and an apt upflow velocity in this experimental was estimated to 1-10m/hr.

  • The recovery posibility of granular sludge inactivated due to high organic loading at stawrt-up stage of UASB reactors was examined at various storage periods while kept at a constant temperature of $35^{\circ}C$. It was noticed that the inactivated sludge kept without feeding recovered microbial activity much faster than that kept with continuous feeding. The activity of the sludge gradually recovered to the point where the organic removal rate of 0.15g of 0.15g COD/g VSS-day at the inactivated stage had changed to 0.36g COD/g VSS-day after 60 days of storage without feeding, which was similar to the active granular sludge activity of 0.38g COD/g VSS-day. There was no significant different in the characteristics of activity recovery between granular sludge and smashed sludge.

  • The objectives of this study have been carried out to investigate the solubilization of municipal sewage sludge by single and dual frequency ultrasonic pretreatment, and the methane production characteristics of pretreated sewage sludge by specific methanogenic activity test for sewage sludge reduction. The waste activated sludge was collected from thickened tank of Suyoung sewage treatment plant in Busan city, and its concentration was adjusted to 1.0% total solids. Ultrasonic frequency was varied 15, 20, 15+20 kHz, and acoustic density was used a maximum 176W/L. The dual frequency ultrasonic pretreatment was found to be more effective than single frequency ultrasonic in the solubilization rate and methane production. The $SCOD_{Cr}/TCOD_{Cr}$, rate were 15.2%, 13.9%, 17.0% with single frequency of 15 kH2, 20 kHz, dual frequency of 15+20 kHz, respectively. The application of dual frequency ultrasound for sewage sludge pretreatment can be interest for sewage treatment plants having problems in sludge treatment and disposal.

Adaptation of Betula schmidtii Seedling in Coal-mine Field with Different Sewage Sludge Treatment Methods

  • We tested the field adaptation of Betula schmidtii on the abandoned coal-mine soil with sludge amendment methods for promoting physiological activity of B. schmidtii seedlings under several environmental stress. Sewage sludges were amended to coal-mine soil with B. schmidtii seedlings which grown in the mixture of artificial soil and composted sludge soil before transplanting (before-fertilized treatment, BF) and fertilized with composted sludge after transplanting (after-fertilized treatment, AF). The percent of establishment of seedlings for AF (80.7%) was lower than that for BF (92.7%). Nitrate reductase activity and photosynthetic pigment content were higher in AF than in BF, but malondialdehyde (MDA) content and superoxide dismutase (SOD) activity were lower in AF than in BF These results represent that after-fertilized seedlings increase resistance against physiological stress at field condition using nitrogen source of composted sludge. On the contrary, before-fertilized seedlings were susceptible to environmental stress on abandoned coal-mine soil by exhausting of nitrogen source from composted sludge.

  • Chlorine inhibition on the denitrifing activity of activated sludge treating dairy wastewater was investigated in this study. Filamentous bulking was caused artificially by a sudden load of feed and monitored by measuring sludge volume index. In cases of the activated sludge and bulking sludge which were contacted with chlorine as $7.5\;mgCl_2/gVSS/day$ for bulking control, the decreases of specific denitrification of $32.2{\sim}40.4%\;and\;43.5{\sim}46.5%$ were shown in comparison to the control group which was not reacted with chlorine, respectively. In continuous operation, it was observed that the removal efficiency of total nitrogen was more susceptible to chlorine than the removal efficiency of total phosphorus.

  • Granular sludge formation and it's activity change are the most important factors in achieving successful start-up and operation of UASB reactor. Nevertheless, the detailed mechanism is still unknown. On the basic of the experiments in laboratory-scale UASB reactor, the effect of hydrogen partial pressure on sludge granulation was evaluated. Size distribution method and specific metabolic activity of the sludge with the operation time were used as a means for estimating the degree of the sludge granulation. At the constant hydrogen loading, the granulation increased as starvation periods in hydrogen supply increased, resulting in high organic removal efficiency. It was evidient that hydrogen play very important role in granulation and sludge granulation was achieved through mutual symbiosis between hydrogen utilizing bacteria and hydrogen producing bacteria under the hydrogen dificient conditions. Enzymatic Activities in Petroleum Wastewater Purification System by an Activated Sludge Process

  • The enzymology of an activated sludge system for a petroleum wastewater purification process was investigated. Leucine-aminopeptidase (L-AMP), ${\beta}$-glucosidase (${\beta}-GLC$), and lipase (LIP) were selected for the study. It was found that more than 81.7% of enzymatic activity was associated with microbial cells in the activated sludge floc. The metabolic response of a mixed microbial population to increased phenol concentration showed that L-AMP activity increased in the activated sludge, whereas activities of ${\beta}-GLC$ and LIP decreased, due to the inhibitory effect of the phenol which varied from 100 mg/l to 500 mg/l.

Effects on Microbial Activity of Aerobic Granular Sludge (AGS) in High-Salinity Wastewater (고농도 염분함유 폐수가 호기성 그래뉼 슬러지의 미생물 활성도에 미치는 영향)

  • The purpose of this study was to evaluate the effect of high-salinity wastewater on the microbial activity of Aerobic Granule Sludge (AGS). Laboratory-scale experiments were performed using a sequencing batch reactor, and the Chemical Oxygen Demand (COD), nitrogen removal efficiency, sludge precipitability, and microbial activity were evaluated under various salinity injection. The COD removal efficiency was found to decrease gradually to 3.0% salinity injection, and it tended to recover slightly from 4.0%. The specific nitrification rate was 0.043 - 0.139 mg $NH_4{^+}-N/mg$ $MLVSS{\cdot}day$. The specific denitrification rate was 0.069 - 0.108 mg $NO_3{^-}-N/mg$ $MLVSS{\cdot}day$. The sludge volume index ($SVI_{30}$) ultimately decreased to 46 mL/g. The specific oxygen uptake rate decreased from an initial value 120.3 to a final value 70.7 mg $O_2/g$ $MLVSS{\cdot}hr$. Therefore, salinity injection affects the activity of AGS, causing degradation of the COD and nitrogen removal efficiency. It can be used as an indicator to objectively determine the effect of salinity on microbial activity.

Hydrolysis of Paper Mill Sludge Using an Improved Enzyme System

  • The effects of water soluble materials in paper mill sludge on cellulase and $\beta$-glucosidase activities were studied while the optimization of enzyme system for hydrolysis of the paper mill sludge for production of glucose was made. Water soluble materials in the paper mill sludge showed stimulatory effect on carboxymethyl cellulose (CMC) activity, inhibitory effect on filter paper (FP) activity, and no effect on avicelase and $\beta$-glucosidase activities. CMC and ${\beta}$-glucosidase activities at 5 and 10, 5 or 10 and 10, and 10 and 10 U/ml were optimal for hydrolysis of 5, 10, and 20% of the paper mill sludge, respectively.
