Isolation of a Pseudomonas sp. Strain Exhibiting Unusual Behavior of Poly(3-hydroxyalkanoates) Biosynthesis and Characterization of Synthesized Polyesters

  • A Pseudomonas sp. strain that is capable of utilizing dicarboxylic acids as a sole carbon source was isolated from activated sludge by using the enrichment culture technique. This organism accumulated polyhydroxyalkanoates (PHAs) with an unusual pattern of monomer units that depends on the carbon sources used. Polyhydroxybutyrate (PHB) homopolyester was synthesized from glucose or small $C_{-even}$ alkanoic acids, such as butyric acid and hexanoic acid. Accumulation of PHB homopolyester was also observed in the cells grown on $C_{-odd}$ dicarboxylic acids, such as heptanedioic acid and nonanedioic acid as the sole carbon sources. In contrast, a copolyester consisting of 6 mol% 3-hydroxybutyrate (3HB) and 94 mol% 3-hydroxyvalerate (3HV) was produced with a PHA content of as much as 36% of the cellular dry matter. This strain produced PHAs consisting both of the short-chain-length (SCL) and the medium-chain-length (MCL) 3-hydroxyacid units when heptanoic acid to undecanoic acid were fed as the sole carbon sources. Most interestingly, polyester consisting of significant amount of relevant fractions, 3HB, 3HV, and 3-hydroxyheptanoate (3HHp), was accumulated from heptanoic acid. According to solvent fractionation experiments, the polymer produced from heptanoic acid was a blend of poly(3HHp) and of a copolyester of 3HB, 3HV, and 3HHp units. The hexane soluble fractions contained only 3HHp units while the hexane-insoluble fractions contained 3HB and 3HV units with a small amount of 3HHp unit. The copolyester was an elastomer with unusual mechanical properties. The maximum elongation ratio of the copolyester was 460% with an ultimate strength of 10 MPa, which was very different from those of poly(3HB-co-3HV) copolyesters having similar compositions produced from other microorganisms.

Adsorption and Activity Changes of Nitrofen by Clay Minerals (점토광물(粘土鑛物)에 의한 Nitrofen의 흡착(吸着) 및 활성(活性) 변화(變化))

  • The nitrofen adsorption on several clay minerals was determined by sludge method to know the effect of clay minerals on the nitrofen activity. The bioassay was conducted with wheat seedlings to study the influence of the adsorbed nitrofen on the nitrofen activity. It is apparant that a four hours shaking was enough to reach the equilibrium concentration. The more the amount of sample, the more nitrofen was adsorbed by clay minerals, whereas the more nitrofen adsorption per unit gram of sample was observed at a 200 mg addition than a 400 mg in the same nitrofen solution. A little amount of nitrofen was adsorbed on Ca-zeolite or Ca-kaolinte, and much more nitrofen was adsorbed on Na-montmorillonite than the other clay minerals in the experiment. Little effect of pH on the adsorption would be attributed to physical adsorption between nitrofen molecule and clay surface. Na-and Ca-montmorillonite were the most effective in reducing the phytotoxicity of nitrofen to the growth of wheat seedlings among clay minerals which nitrofen was added to the growth media.

  • This paper was investigated the research regarding the effects of several factors such as initial ammonium nitrogen concentration, aeration rate. biomass amount and C/N ratio on nitrification process using synthetic wastewater and activated sludge obtained from wastewater treatment facility. As a result, in high ammonium nitrogen concentration above 100 mg/L, the pH of wastewater was dropped to pH 6.8. The increases of initial ammonium nitrogen concentration, aeration rate and initial biomass amount were linearly enhanced the removal rate of ammonium nitrogen. In the condition of C/N ratio of 0 to 3, high ammonium nitrogen removal rate was obtained.

Influence of hydraulic retention time on biogas production during leachate treatment

  • The main objective of this study is to investigate the influence of hydraulic retention time (HRT) on biogas production during leachate treatment using an anaerobic reactor type Upflow Anaerobic Sludge Blanket. For this purpose, four HRTs ranging from 12 to 48 h were experienced. The obtained results showed that higher amount of biogas could be produced during longer HRTs. However, HRTs longer than 48 h could not affect clearly the biogas generation and considered as unnecessary given the small additional amount of biogas produced during the degradation process. A volume of $0.434L/L_{leachate}/d$ was achieved during the treatment with a HRT of 48 h. The higher biogas production, the smaller chemical oxygen demand (COD) values achieved. Besides, COD removal and biogas production positively correlate, showing that the active biomass has degraded effectively the organic load to produce biogas. Moreover, all the analyzed physicochemical parameters have experienced a decrease after 48 h except for the pH, which increased to approximately neutral value. More precisely, a decrease of 93.8%, 89.7%, 95%, 70%, 77%, and 84.4% was recorded for COD, electrical conductivity, total suspended solid, turbidity, $NH_4{^+}-N$, and $NO_3{^-}-N$, respectively.

The effect ot mixing ratio of four rvastc mater~als ou the composnlg phascs in melled slag was investigated. Thc mixed wastes were meltcd to slag by heat under a constant basicity at 1370C. The shg consisted of p -CIS and C,AS(2CaO - A I P , . SiO,). The ratio of two phases was varied with mixing ~atioo f the waste materials. In order Lo increasc the amount ot j -C2S phase, the coal fly ash content should be reduced, while amount of the chemical glass be increased. The coal fly ash contcnt was the most imporlant factor in controlling phases of thc melted-slag.

Effects of Food Waste Mixed Organic Fertilizer Treatment on Growth and Yield of Capsicum annuum

  • The global population is increasing every year, and the amount of food waste is also increasing. Direct landfilling of food waste has been prohibited since 2005, and in accordance with the London Convention in 2013, the discharge of livestock manure, sewage sludge, and food waste into the sea is prohibited. In the case of incineration to treat the discharged food waste, the heat point is lowered due to the moisture in the food waste itself, so fuel must be added. Therefore, this study was conducted to get basic data for setting the limit of application by investigating the growth and yield of crops after treating food waste dry powder mixed fertilizer (MF) on red pepper. In the experiment, continuous cultivation was carried out for two years in 2021 (1st year) and 2022 (2nd year). The treatment groups were set as Not Treatment (NT), Chemical Fertilizer (CF), Mixed Fertilizer (MF), Mixed Fertilizer×2 (MF×2). After harvest, crop growth and yield were investigated. As a result of the 1st years of growth survey, CF, MF, MF×2 show significant difference in shoot length compared to NT. About fresh weight and dry weight, CF show significant difference compared to NT. The 2nd years of growth survey, the shoot and root length, fresh weight did not show significant difference with NT. In case of dry weight, MF is significant increased compared to NT. As a result of the yield survey of the 1st year, all treatment groups did not show a significance in yield compared to the NT. In case of 2nd year, all treatment groups show significantly increased value compared to NT. The yield of MF was highest among the treatment groups. In the future, it is thought that it is necessary to quantitatively evaluate the effect of food waste dry powder mixed fertilizer through additional experiments and continuous cultivation, and to establish an appropriate amount of use and establishment of a manual based on this.

Denitrification Characteristics and Mircoorganism Composition of Acclimatec Denitrifier Consortium

  • The effect of the COD/N ratio on denitrification characteristics was evaluated for the development of a denitrification process. Activated sludge, acclimated to an anoxic condition, was used as the denitrifier consortium (mixture of denitrifying organisms) for enhanced nitrogen removal in a recirculating aquarium system. Synthetic wastewater containing nitrate was used as the influent solution and glucose was used as the carbon source for denitrification. The COD/N ratio varied within a range of 1.5-7.2. The denitrification efficiency was higher than 97% even at a COD/N ratio of 1.5. Under a theoretical COD/N ratio of 3.0, nitrite was detected, however, the amount was less than 1% of the total influent nitrogen. The number of both nitrate-reducing bacteria and denitrifying bacteria reached $3.5{\times}10^5/ml$ with a COD/N ratio of 1.5 after 45 days of operation.

A Study on the Application of Pre-Chemical Treatment on the Decentralized Domestic Wastewater Reclamation System (도시의 분산형 생활오수 재생시스템에 화학적 전처리공정도입에 관한 연구)

  • The purpose of this study was to investigate applicability of pretreatment on the existing biological treatment for domestic wastewater reclamation. From Jar Tests, it was found that optimum dosage of coagulant was PAC 0.5mg/L and $FeCl_3$ 180mg/L for urban sewage. In this study, PAC 0.5mg/L was selected considering sludge production and the amount of coagulant required. In a continuous experiment performed with combining chemical coagulation and biological treatment, a considerable removal efficency was obtained in term of BOD, SS, T-N, T-P and ABS. When the raw sewage was supplied into the pre-chamical treatment facility, the removal of BOD and SS was 48.3% and 81.1%. However T-N removal was very low which means T-N consists of $NH_3-N$ mostly. T-P was almost completely recluced by the chemical addition. The effluent BOD & SS was 57~76 and 21~43mg/L, which could reduce the size of biological treatment facility. From the cost estimation pre-chemical treatment could save around half of the area required for biological treatment with post ceagulation.

The Antimicrobial Effect of Structure Modified Apatite (구조개질 Apatite의 항균효과)

  • The hydroxyapatite(HAp) as a carrier the for ion exchange agent of $Ag^+$ions was prepared in semiconductor fabrication, The Ca/P molar ratio of the HAp was 1.65. The HAp is molded in shape of the antimcrobial ball and then sintered at 100$0^{\circ}C$ Ag-containing HAp(HAp-Ag) was prepared by incorporating $Ag^+$/ions in HAp crystals through an ion-exchange reaction in solutions containing 0.01M $AgNO_3$. The antimicrobial effect of HAp-Ag for bacteria such as Escherichial coli and Staphylococcus aureus has been Investigated. The concentrations of silver in the antimicrobial ball was determined by inductively coupled plasma and the amount of $Ag^+$ions was 9.0$\mu\textrm{g}$/g. The HAp-Ag exhibited excellent antimicrobial effect for bacteria such as E. coli and S. aureus. The bactericidal activity was considered to be caused by direct contact of bacteria with $Ag^+$ions in the HAp crystals. The HAp would likely to be possible as a carrier of antimicrobial metal ions such as Ag, Cu, and Zn by recycling of waste sludge in the semiconductor fabrication process.

Current Status on the Domestic Recycling of Magnesium (국내(國內) 마그네슘 리싸이클링 현황(現況))

  • Magnesium has been used as parts of vehicles, case materials of notebook PC and mobile phone, and its demand has been increasing recently. So until now, there has little magnesium scraps from the end of life vehicles or electronic parts, and most scraps has been generated from magnesium processing lines such as melting, die casting and machining. It is to review the present status of magnesium recycling. Here, domestic demand of magnesium, recycling amount and technologies used in domestic recycling companies were surveyed in recent years. In 2010, 8,840 tons of magnesium scraps were processed and used as raw materials for die casting products. The recycling ratio was estimated as 32.5%.
