Pathogenic and Morphologic Variation of Toxoplasma gondii by X-Ray and Neutron Flax Irradiation (X-Ray 및 Neutron 조사에 의한 Toxoplasma gondii의 병원성 및 형태학적 변화에 대하여)

  • Series of experiments were conducted to determine lethal does of X-ray and Neutron on Toxoplasma gondii. strain RH and IRI. As well morphological changes of Toxoplasma gondii irradiated or not were compared by use of electron microscope. The pathogenicity test of the irradiated and nonirradiated Toxoplasma gondii was made in mice guinea-pigs, rabbits and pigs: The letahl dose of X-ray and Neutron on RH and IRI strain and the growth rate between two strains after irradiation were shown little differences. Morphological changes were not observed until 18th passage was made. After then, the growth rate was decreased apparently, and atrophied forms were frequently observed in electron microscope. Survival time of animals inoculated with irradiated strain was longer than that of animals giving non-irradiated strain, and Toxoplasma gondii were isolated from all the dead animals. But it is of interest that pigs survived after injection of Toxoplasma gondii remained health and much attempts were failed toisolate Toxplasma gondii remained health and much attempts were slaughtered them. Animals were succumbed after injection of Toxoplasma gondii without any relationship with serum titers. (HA antibody).

Effect of Feed Antibiotics on the Performance and Intestinal Microflora of Weanling Pigs in China

  • Ninety-six crossbred (Large White${\times}$Landrace${\times}$Duroc) pigs, weaned at 35 days of age, were assigned to four dietary treatments in order to investigate the effects of oral antibiotics on the performance and the intestinal microflora of weanling pigs. Pigs were fed either a basal diet, without antibiotics, or the basal diet plus either 50 ppm acetylspiramycin, 50 ppm olaquindox, or 100 ppm bacitracin zinc. The pigs were housed eight per pen with three pens per treatment in an environmentally controlled nursery. Ten days after weaning, three pigs from each treatment were slaughtered and intestinal pH, microflora, and volatile fatty acid concentration were determined. At the end of the 4 week trial, the remaining pigs were weighed and feed consumption was measured. Average daily gains for pigs fed acetylspiramycin, olaquindox, bacitracin zinc and the control diet were 0.43, 0.40, 0.37, and 0.34 kg per day (p=0.001), respectively. Antibiotic addition did not modify feed intake, but acetylspiramycin improved feed conversion (p=0.003). In comparison with the control, acetylspiramycin significantly increased Bifidobacteria numbers in the jejunum (p=0.082) and ileum (p=0.014) and decreased total bacterial counts throughout the intestine (p<0.01 except for the ileum where p=0.079). Acetate production was significantly lower in the cecum (p=0.028) and colon (p=0.079) of pigs fed acetylspiramycin. In addition to increasing numbers of Bifidobacteria in the jejunum (p=0.082) and ileum (p=0.014), olaquindox increased Lactobacillus in the jejunum (p=0.004) and decreased E. coli in the colon (p=0.022). Bacitracin zinc increased Lactobacillus numbers in the jejunum (p=0.004) and Bifidobacterium concentrations in the jejunum (p=0.082) and ileum (p=0.014).

Detection of residual antibiotics by TLC and EEC-4 plate method in slaughtered pigs (도축돈에서 TLC와 EEC-4 plate법을 이용한 항생물질 잔류조사)

  • The antibiotic residues of the urine, the liver, the lung, the kidney and the spleen in slaughtered pigs at Kyongbuk province were detected by TLC(505 kit) and EEC-4 plate method. 1. The positive rate of residual sulfamethazine which was detected by 505 kit in the urine (n=200) was 0.0%. 2. The positive rate of residual sulfamethazine which was detected by EEC-4 plate in the urine (n=126), the liver(n=98), the kidney(n=72), the spleen (n=68) and the lung(n=48) were 63%, 49%, 36%, 34% and 24%, respectively. 3. By EEC-4 plate method, the positive detection rates of the urine were 53.0% in BS(pH 6.0), 29.0% in BS(pH 7.2), 11.5% in BS(pH 8.0) and 13.0% in ML(pH 8.0) medium, that of the liver 41.5% in BS(pH 6.0), 22.0% in BS(pH 7.2), 6.5% in BS(pH 8.0) and 5.0%, in ML (pH 8.0) medium, that of the lung 21.0% in BS(pH 6.0), 9.5%, in BS(pH 7.2) and 8.5% in BS(pH 8.0) medium, and that of the kidney 31.5% in BS(pH 6.0), 14.5% in BS(pH 7.2), 20.0% in BS(pH 8.0) and 3.0% in ML(pH 8.0) medium. In the spleen, only in BS(pH 6.0) medium the positive rate was detected as 33.5 %. 4. The positive rates of samples which shown TLC-positive detected by EEC-4 plate method were 53.9% in no band, 77.8% in one band, 80.9% in two bands, 66.7% in three bands, respectively. In conclusion, the EEC-4 plate method could be applied for the detection of residual antibiotics in samples which shown as out of standard Rf values by TLC-method (SOS kit).

  • Objective: This experiment investigated meat color, myoglobin content, enzyme activities, and expression of genes associated with oxidative potential of pigs slaughtered at different growth stages. Methods: Sixty 4-week-old $Duroc{\times}Landrace{\times}Yorkshire$ pigs were assigned to 6 replicate groups, each containing 10 pigs. One pig from each group was sacrificed at day 35, 63, 98, and 161 to isolate longissimus dorsi and triceps muscles. Results: Meat color scores were higher in pigs at 35 d than those at 63 d and 98 d (p<0.05), and those at 98 d were lower than those at 161 d (p<0.05). The total myoglobin was higher on 161 d compared with those at 63 d and 98 d (p<0.05). Increase in the proportions of metmyoglobin and deoxymyoglobin and a decrease in oxymyoglobin were observed between days 35 and 161 (p<0.05). Meat color scores were correlated to the proportion of oxymyoglobin (r = 0.59, p<0.01), and negatively correlated with deoxymyoglobin and metmyoglobin content (r = -0.48 and -0.62, p<0.05). Malate dehydrogenase (MDH) activity at 35 d and 98 d was higher than that at 161 d (p<0.05). The highest lactate dehydrogenase/MDH ratio was achieved at 161 d (p<0.05). Calcineurin mRNA expression decreased at 35 d compared to that at 63 d and 98 d (p<0.05). Myocyte enhancer factor 2 mRNA results indicated a higher expression at 161 d than that at 63 d and 98 d (p<0.05). Conclusion: Porcine meat color, myoglobin content, enzyme activities, and genes associated with oxidative potential varied at different stages.

Characteristics of outbreak for the classical swine fever (CSF) at Incheon metropolitan area in 2002 (인천지역에서 발생한 돼지콜레라의 역학적 특성)

  • The aim of this survey was to investigate the characteristics of outbreak farm determined as the classical swine fever(CSF) at Gangwha-gun and Seo-gu, Incheon metropolitan area from October 7 to November 25 in 2002. Sixty pigs in six different farms were confirmed to the CSF and a total of 9,106 pigs containing 3,194 related epidemiologically was slaughtered to stop spreading of the disease. Clinical signs of pigs diagnosed with the CSF were high fever, anorexia, depression, paralysis of hindlimbs, cyanosis, etc and gross lesions were typically represented with hemorrhage of submandibular and superficial lymph node, infarction of spleen, and petechial (ecchymotic) hemorrhage of kidney and skin. But some outbreak farms had not shown remarkable symptoms, so they were confused with other bacterial diseases. White blood cell (WBC) counts, the classical swine fever virus(CSFV) antigen and antibody enzyme linked immunosorbent assay(ELISA) and reverse transcriptase polymerase chain reaction (RT-PCR) results about six farms indicated that total 60 pigs were infected with the CSFV. Although the origin and infection route of the CSFV were not clear, but the transmissions between farms were mainly through indirect contact such as the movement of farm personal and vehicles from outbreak farm.

Passive Immunization Approach to Reduce Body Fat in Pigs Using Fat-specific Polyclonal Antiserum

  • Plasma membrane proteins from pig adipocytes, brain, heart, kidney, liver and spleen were isolated using a 32% sucrose gradient. An adult male sheep was immunized three times at 3-wk intervals with the purified pig adipocyte plasma membrane (APM) proteins. Blood samples were taken from the immunized sheep 12 d after the third immunization. Antiserum showed strong reactivity with APM proteins determined by ELISA, and the reactivity could be detected at dilutions in excess of 1:128,000. Antiserum showed very low binding affinity with proteins isolated from brain, heart, kidney, liver or spleen. Ninety weanling pigs were allocated randomly to three treatment groups and were injected i.p. with 40 ml of antiserum (n = 30) or 20 ml of lyophilized antiserum (21.5 mg/ml; n = 30). A control group (n = 30) received 40 ml of saline, and all pigs were slaughtered at 24 wk of age. The polyclonal antiserum did not change BW or ADG. Carcass percentage of pigs was numerically increased by the antiserum treatment compared with control. Both antiserum treatments did not significantly (p>0.05) affect body composition, including body fat content, relative to the control group.

Blood haematology, muscle pH and serum cortisol changes in pigs with different levels of drip loss

  • Objective: An experiment was conducted to study the blood haematology, muscle pH, and serum cortisol changes in pigs with different levels of drip loss. Methods: Two groups (low and high) of 20 animals were selected from 100 pigs based on drip loss. All [$Duroc{\times}(Large\;White{\times}Landrace)$] pigs were slaughtered according to standard slaughtering procedures. At exsanguinations, blood samples were taken for the haematological parameters and serum cortisol analysis. The muscle samples were taken from longissimus dorsi muscle to evaluate the muscle pH and drip loss. Results: Haematological parameters of low drip loss group showed higher content of white blood cells and monocytes than high drip loss group (p<0.05). The low drip loss group had higher muscle pH at 45 min (p<0.05) and 24 h (p<0.001) post-mortem than the high drip loss group. However, there was no significant difference in serum cortisol levels (p>0.05). Conclusion: Drip loss is mainly affected by the muscle pH decline after slaughter and also might be affected by white blood cells and monocytes.

A Survey of Puteurella Multocida Isolated from Pigs Affected with Pneumonia in Eastern Kangwon (강원 동해안 지역 돼지 폐렴에서 분리한 puteurella multocida 에 대한 조사)

  • The present study was conducted to investigate the incidence of pasteurella multocida(p. multocida) infection and some properties of the isolated organisms from the swine herds in Eastern Kangwon during the periods from March 1993 to November 1993. The results obtained were summerized as follows: 1. The lungs of 180 slaughtered pigs were sampled and p. multocida was isolated from 38 pigs (21.1%) and cultured positive. 2. The majority of biochemical md cultural properties of the p. multocida isolates were identical to those of the standard strains. 3. We investigated the capsular serogroup and drug susceptibility of 38 Isolates of p. multocida from pigs with pneumonic lesions 4. p. multocida isolateds were typed for capsular serogroupes A by hyaluronidase inhibition of capsule and D by acriflavin auto agglutination. Most isolates(55.3%) were type A, 15.8% were type D, and the remaining 28.9% were untypable 5. In antimicrobial susceptibility test these isolates of p. multocida were susceptible in order of colistin(94.7%), ampicillin(94.7%), cepalothin(92.1%). gentamicin(92.1%), amikacin(89.5%), but the majority of them were resistant in order of neomycin(26.3%), teracycline(23.7%), streptomycin( 15.8%)

Isolation of Staphylococcus aureus and detection of enterotoxin from pigs and cattle carcass by PCR (소와 돼지 도체표면에서 황색포도상구균의 분리 및 장독소 검출)

  • At the present study, it was aimed to explore the states of antimicrobial resistant Staphylococcus aureus isolates from 320 pigs and cattle carcass (160 pigs and 160 cattle) slaughtered in Busan province from March 2008 to November 2009. Among 320 samples, 26 of Staphylococcus aureus were isolated from pigs (10.6%) and cattle (5.6%). In antimicrobial susceptibility test, all of the isolates were demonstrated susceptibility to oxacillin, cefoxitin, cephalothin, vancomycin, rifampin and linezolid. But the isolates were showed resistance other antibiotics in order of penicillin (92.3%), gentamicin (76.9%), tetracycline (69.2%), erythromycin (65.4%), and clindamycin (61.5%). In case of enterotoxin production, 7.7% of 2 strains produced enterotoxin A.

Effects of marbofloxacin injection for granuloma formation in fattening pigs (마보플록삭신 제품별 접종이 비육돈 목심 부위 이상육 발생에 미치는 효과)

  • The abnormal meat could caused by abscess, fibrosis and granuloma in the region of the neck by adverse effect of vaccination or antibiotic injection under swine practice. In this study, the incidence of abnormal meat due to antibiotics injection was investigated using marbofloxacin from different manufacturers. To evaluate the incidence of abnormal meat via antibiotic treatment, a total of 440 pigs with the age of 140~150 days old were selected. Two hundred of pigs were assigned as a control group and 2 mL of saline was inoculated daily for three days. Two hundred forty of pigs were used as treatment group and further divided into 6 groups, consisting of 16% of original marbofloxacin treated, 10% of original marbofloxacin treated and 4 different kinds of generic marbofloxacin treated groups respectively. When test animals were slaughtered, incidence, type and size of induced abnormal meat were recorded. In the result of this study, statistical analysis found significantly lower incidence of abnormal meat formation was in 16% original marbofloxacin treated groups compared to the other tested groups (P<0.05). In the summary, antibiotic treatment could produce abnormal meat formation but the 16% of original marbofloxacin had the most minimal abnormal meat formation effect.
