Characteristics of Wet Distillers Grains on In vitro Ruminal Fermentation and Its Effects on Performance and Carcass Characteristics of Finishing Hanwoo Steers

  • Two experiments were conducted to determine the nutrient composition, in vitro ruminal ammonia concentrations and pH of wet distillers grains (WDG, produced from tapioca 70% and rice 30%) and to evaluate dietary effects of fermented total mixed ration (TMR) using WDG on the performance, blood metabolites and carcass characteristics of Hanwoo steers from mid fattening to slaughter. In Exp. I, average dry matter (DM), crude protein, ether extract, crude fiber, ash, neutral detergent fiber, acid detergent fiber, and nitrogen free extract of seven WDG samples from an ethanol plant with different sampling dates were 19.9%, 24.8%, 3.8%, 21.8%, 8.87%, 60.3%, 34.5%, and 40.7% (DM basis), respectively. For in vitro ammonia concentrations and pH, each sample was assigned to 7 incubation times (0, 4, 8, 12, 24, 48, and 72 h). Linear increase was observed between 12 and 48 h for ammonia concentrations, but final ammonia concentrations (72 h) were not significantly different among WDG samples and fermentation patterns of WDG samples showed similar tendency. In vitro pH varied among treatments from 0 to 24 h, but were not different statistically after 48 h. In Exp. II, 45 Hanwoo steers of 23 months ($641{\pm}123kg$) from mid fattening period to slaughter (248 days) were randomly divided into three groups of 15 pens each (five repetitions/each treatment) and assigned to one of three dietary treatments; i) Control (TMR), ii) WDG 15 (TMR containing 15% of WDG, as fed basis) and iii) WDG 28 (TMR containing 28% of WDG, as fed basis). The body weight (BW), ADG, and feed conversion ratio (FCR) of control and WDG 15 and 28 during 248 days were 760.8, 740.1, and 765.5 kg, and 0.50, 0.50, and 0.52 kg/d, and 18.6, 17.6, and 17.1, respectively. The dry matter intake (DMI) (kg/d) of control (9.11) was higher (p<0.05) than WDG treatments (WDG 15%, 8.57; 28%, 8.70). Nevertheless, DMI did not affect BW, ADG, and FCR of Hanwoo finishing steers. Blood metabolites were in normal ranges and were not different among treatments except the albumin concentration. In carcass characteristics, WDG 15 (30%) showed higher frequency of A-carcass yield grade than WDG 28 (15%) and control (7%), and WDG 28 (61%) showed higher frequency of $1^{{+}{+}}$ and $1^+$-carcass quality grade than WDG 15 (40%) and control (60%). In conclusion, using WDG up to 28% in TMR did not show any negative effect on the performance and blood metabolites, and improved carcass quality of Hanwoo steers. Therefore, WDG can be a useful feed ingredient for Hanwoo steers in mid-fattening period to slaughter.

Blood haematology, muscle pH and serum cortisol changes in pigs with different levels of drip loss

  • Objective: An experiment was conducted to study the blood haematology, muscle pH, and serum cortisol changes in pigs with different levels of drip loss. Methods: Two groups (low and high) of 20 animals were selected from 100 pigs based on drip loss. All [$Duroc{\times}(Large\;White{\times}Landrace)$] pigs were slaughtered according to standard slaughtering procedures. At exsanguinations, blood samples were taken for the haematological parameters and serum cortisol analysis. The muscle samples were taken from longissimus dorsi muscle to evaluate the muscle pH and drip loss. Results: Haematological parameters of low drip loss group showed higher content of white blood cells and monocytes than high drip loss group (p<0.05). The low drip loss group had higher muscle pH at 45 min (p<0.05) and 24 h (p<0.001) post-mortem than the high drip loss group. However, there was no significant difference in serum cortisol levels (p>0.05). Conclusion: Drip loss is mainly affected by the muscle pH decline after slaughter and also might be affected by white blood cells and monocytes.

Investigation of blood biomarkers related to meat quality and quantity in Hanwoo steers

  • Objective: This study was conducted to investigate the correlation between blood components and carcass traits, and to find the biomarkers related to meat quality and quantity in Hanwoo steers. Methods: One hundred twenty-six Hanwoo steers (8 to 9 months of age, body weight of $185.6{\pm}24.38kg$) were used to find the correlation between blood compositions and carcass traits. The steers were fed concentrate and rice straw (30 steers) or total mixed rations (96 steers) during the whole experimental period. Blood samples were collected from the jugular vein at the growing (8 to 12 months), fattening (13 to 23 months) and finishing phases (24 to 30 months). Steers were slaughtered at 30 to 31 months of age (body weight of $701.6{\pm}60.45kg$) and the carcass traits were evaluated. Blood metabolites and hormones were analyzed and the correlation coefficients and regression equations with carcass traits were determined. Results: Average concentrations of retinol, leptin and insulin were 1.10 IU, 30.34 ng, and 35.35 ng per mL of blood plasma, respectively. Retinol has negative correlations (p<0.01) with insulin and leptin. Blood insulin and total protein decreased with the age of steers, but retinol, aspartic acid transaminase (AST), glucose, cholesterol and triglyceride increased. In the finishing phase, significant (p<0.01) negative correlations occurred between blood retinol content and marbling score, and between blood AST content and longissimus muscle area of 13th rib, and the following regression equations were obtained: Marbling score $(1-9)=-0.009{\times}retinol(IU/100mL)+9.125$ ($R^2=0.643$), Longissimus muscle area $(cm^2)=-0.250{\times}AST(U/L)+112.498$ ($R^2=0.450$). Conclusion: It is possible to make highly marbled beef by controlling the blood retinol content during the fattening and finishing phases of Hanwoo steers. Accordingly, blood retinol and AST could be biomarkers for determining beef quality and quantity, respectively, prior to slaughter.

The Effects of Pre-slaughter Stress and Season on the Activity of Plasma Creatine Kinase and Mutton Quality from Different Sheep Breeds Slaughtered at a Smallholder Abattoir

  • The objective of the current study was to determine the effect of pre-slaughter stress, season and breed on the activity of plasma creatine kinase (CK) and the quality of mutton. One hundred and seventy-three (173) castrated sheep from Dormer (DM), South African Mutton Merino (SAMM), Dorper (DP) and Blackhead Persian (BP) sheep breeds were used in the study. The animals were grouped according to age-groups as follows: Group 1 (6 to 8 months), Group 2 (9 to 12 months) and Group 3 (13 to 16 months). Blood samples were collected during exsanguinations using disposable vacutainer tubes for CK analysis. Representative samples of the Muscularis longissimuss thoracis et. lumborum (LTL) were collected from 84 castrated sheep, of different breeds (28 per breed) 24 h after slaughter. The following physico-chemical characteristics of mutton were determined; meat pH ($pH_{24}$), color ($L^*$, $a^*$ and $b^*$), thawing and cooking losses and Warner Braztler Shear Force (WBSF). The activity of plasma CK was significantly higher (p<0.001) in summer ($1,026.3{\pm}105.06$) and lower in winter ($723.3{\pm}77.75$). There were higher values for $L^*$ ($33.7{\pm}0.94$), $b^*$ ($11.5{\pm}0.48$) and WBSF ($29.5{\pm}1.46$) in summer season than in winter season; $L^*$ ($29.4{\pm}0.64$), $b^*$ ($10.2{\pm}0.33$) and WBSF ($21.2{\pm}0.99$). The activity of plasma CK was influenced by the type of breed with Dormer having the highest (p>0.001) levels ($1,358.6{\pm}191.08$) of CK. South African Mutton Merino had higher values for $pH_{24}$ ($5.9{\pm}0.06$), $L^*$ ($34.2{\pm}0.97$), $b^*$ ($12.2{\pm}0.50$) and WBSF ($26.8{\pm}1.51$) and Blackhead Persian had higher values ($35.5{\pm}2.17$) for cooking loss (CL%) than the other breeds. Computed Principal Component Analyses (PCA) on the activity of plasma CK and physico-chemical characteristics of mutton revealed no correlations between these variables. However, positive correlations were observed between $pH_{24}$, $L^*$, $a^*$, $b^*$, CL% and WBSF. Relationships between pre-slaughter stress, CK activity and physico-chemical characteristics of mutton were also observed. It was therefore concluded that although mutton quality and creatine kinase were not related, pre-slaughter stress, season and breed affected the activity of creatine kinase and mutton quality.

Effect of stress during slaughter on carcass characteristics and meat quality in tropical beef cattle

  • Objective: This study aimed to determine the effects of stress during slaughter of beef cattle on physiological parameters, carcass, and meat quality at a Federal Inspection Type slaughterhouse located in the southeast of Mexico. Methods: A total of 448 carcasses of male Zebu×European steers with an average age of 36 months were included. Carcass assessment of presence of bruises and bruise characteristics was carried out on each half-carcass. Blood variable indicators of stress (packed cell volume, neutrophil to lymphocyte ratio, glucose, cortisol concentration) and meat quality parameters (pH, color, shear force, drip loss) were evaluated. Results: Of the 448 carcasses evaluated, 81% of the carcasses showed at least one bruise; one bruise was detected in 36.6% and two bruises in 27.0% of animals. Of the 775 bruises found, 69.2% of the bruises were grade 1 in region 3. Of the 448 carcasses studied, 69.6% showed hyperglycemia (6.91 mmol/L); 44.3% and 22.7% showed high (74.7 ng/mL) and extremely high (108.8 ng/mL) cortisol levels, respectively, indicative of inadequate handling of animals during preslaughter and slaughter. Of the carcasses evaluated, 90.4% had a pH ≥5.8 with an average of pH 6.3. In both pH groups, meat samples showed L values >37.0 (81.6%) and a shear force >54.3 N; meat pH≥5.8 group showed a drip loss of 2.5%. These findings were indicative of dark, firm, and dry (DFD) meat. According to principal component analysis, grades 1 and 2 bruises in region 3 and grade 1 bruises in region 5 were highly associated with cortisol, drip loss, and color parameters b and h and were negatively associated with L, a, and C. Conclusion: The bruises probably caused by stress-inducing situations triggered DFD meat. Appropriate changes in handling routines in operating conditions should be made to minimize stress to animals during the slaughter process to improve animal welfare and meat quality.

Net micromineral requirements for maintenance and growth of ewe lambs at the latter fattening period

  • Objective: The objective of this study was to investigate the net micromineral (Cu, Fe, Mn, Zn) requirements for maintenance and growth of Dorper×Jinzhong crossbred ewe lambs at their latter fattening period. Methods: Thirty 1/2 Dorper × 1/2 Jinzhong crossed F1 ewe lambs (35±0.5 kg of body weight [BW]) were used and divided into five groups in a randomized design for a comparative slaughter trial. At the beginning of the experiment, six lambs were randomly selected and slaughtered at 35 kg BW to determine their initial body composition. When their BWs reached 43 kg, another six lambs fed ad libitum were slaughtered to serve as an intermediate slaughter group. The retained eighteen lambs were randomly distributed into three groups and offered one type of feed at 100%, 65%, and 40% of the ad libitum intake. When the lambs fed ad libitum reached a BW of 50 kg, the three groups were slaughtered. The body composition (muscle, fat, bone, blood with viscera, skin, and wool) were weighted, ground, mixed, and subsampled for mineral content analysis. Results: The net maintenance requirements of Cu, Fe, Mn, and Zn were 0.017, 0.160, 0.004, and 0.067 mg/kg BW/d, respectively, and the net growth requirements per 100 grams of average daily gain ranged from 0.48 to 0.51 mg of Cu, 2.63 to 2.17 mg of Fe, 0.12 to 0.15 mg of Mn, and 2.07 to 2.00 mg of Zn, respectively, for Dorper × Jinzhong crossed ewes from 35 to 50 kg BW. Conclusion: Our results suggest that the micromineral requirements for both maintenance and growth of Dorper × Jinzhong crossbred ewe lambs were quite different from the recommendations of NRC (2007), except for Zn.

Effects of different stocking density in lairage of fattening pigs in high temperatures

  • Lairages serve several functions, such as providing post-mortal inspections and providing a reservoir of animals to ensure the slaughter line runs efficiently. High stress lairage conditions can contribute to the accumulation of transport stress in pigs, causing poor pork quality and still stressed pigs at slaughter. The objective of this study was to investigate meat quality, blood profile and behavior changes according to lairage stocking density in in high temperature. Density treatments were as follows: LD, low density (lower than 0.5 m2/100 kg); ND, normal density (0.5 m2/100 kg to 0.83 m2/100 kg); HD, high density (higher than 0.83 m2/100 kg). Air temperature treatment was as follows: HT, high temperature (higher than 24℃). Pigs stocked with LD showed lower pH, WHC (water holding capacity), and higher DL (drip loss) and CL (cooking loss) than those stocked with HD. Pigs stocked with LD showed lower cortisol level than those stocked with HD. Therefore, Pigs exposed to high stock density (lower than 0.5 m2/100 kg) in high air temperature during pre-slaughter caused acute stress and lead to PSE (pale, soft, exudative) pork incidence. Based on obtained results, stocking of too high (lower than 0.5 m2/100 kg) density is generally not good for meat quality and animal welfare at high temperatures.

천연물질을 이용한 반추동물 항생제 대체용 미량광물질 및 천연유화제의 이용 (Supplmentation of the Mineral and Surfactant for Replacement of Antibiotics for Ruminant Aninmal)

  • The present study investigated the effect of inclusion of chromium propionate (Cr-P) and lysophopholipid (LPL) in diet on blood parameters and meat quality of Hanwoo steer. Feeding trial was performed from late fattening period to slaughter and blood parameters (insulin, blood glucose and non-esterified fatty acid (NEFA) concentration) and meat quality were examined. Total 4 experimental groups including control (no addition), T1 (Cr-P 0.2%), T2 (LPL 0.2%) and T3 (Cr-P 0.1% + LPL 0.1%) were employed. For blood parameters, insulin concentration in T1 and T3 showed an elevating patterns from $3.13{\mu}U/mL$ to $3.35{\mu}U/mL$ (T1) and from $4.38{\mu}U/mL$ to $5.23{\mu}U/mL$ (T3). The changes of NEFA in all groups were detected as a decreasing patterns according to days of feeding. However, significant difference was not found. In growth performance, T2 showed greater daily gain and T1 showed greater carcass yield compared to others. However, there were no significance in difference. In meat quality, T1 showed greater yield and intra-muscular fat levels and lower sharing force compared to others. However, significant differences were not detected.

젖소에서의 간질환 진단을 위한 혈액화학치의 조사 (Blood Chemical Investigations on Liver Diseases of Holstein Cattle)

  • The incidences of liver abscess, fatty liver and fascioliasis were 11.9%, 23.2% and 36.0%, respectively in 211 Holstein cattle observed at the slaughter house. Blood chemical values of them were measured. The results were as follows : 1. Liver abscess group was significantly higher than normal group in the levels of SDH, AST, TP, globulin and fibrinogen, but it was significantly lower than normal group in the levels of albumin and A/G ratio (p<0.01). 2. Fatty liver group was significantly higher than normal group in the levels of SDH, AST, OCT, TP and globulin, but it was significantly lower than normal group in the levels of albumin and A/G ratio (p<0.01). 3. Fasciolisais group was significantly higher than normal group in the levels of AST. TP and globulin (p<0.01), but it was significantly lower than normal group in the levels of albumin and A/G ratio (p<0.01).
