Production of Cloned Calves by the Transfer of Somatic Cells Derived from Frozen Tissues Using Simple Portable $CO_2$ Incubator

  • The ability of frozen-thawed fetal skin was examined to generate viable cell lines for nuclear transfer. Fetal skin frozen at -20$-20^{\circ}C$, $-30^{\circ}C$ or $-80^{\circ}C$ in the presence of 5% DMSO used as tissue explants to generate somatic cells. The resultant confluent cells were then used as donors for nuclear transfer (NT). Of the bovine NT embryos reconstracted from the somatic cells, 62.3%, 76.6% to 65% showed cleavage 70.5%, 81.9% to 78.5% reached the stage of morula formation and 39.7%, 43.2% or 47.6% reached the blastocyst stage. There was no significant difference in development when the NT embryos were compared with those reconstracted from fresh somatic cell derieved skin tissues (72%, 75.3%, and 45.2%, for cleavage, and development to morula and blastocyst stage, respectively). NT embryos were then placed in a portable $CO_2$ incubator and carried to China from Japan by air. After reaching to farm, two NT embryos were transferred to each of 5 recipients. We obtained 2 NT calves which birth weights is 30kg and 36kg female, and gestation periods is 281 and 284 days, respectively. There were no observation any abnormality from those calves. The results indicated that cell lines derieved from bovine fetal skin cryopreserved by a simple method could be used as donors in nuclear transfer using the portable $CO_2$ incubator.

Isolation from Gloydius blomhoffii siniticus Venom of a Fibrin(ogen)olytic Enzyme Consisting of Two Heterogenous Polypeptides

  • Objective: This study was undertaken to isolate a fibrin(ogen)olytic enzyme from the snake venom of Gloydius blomhoffii siniticus and to investigate the enzymatic characteristics and hemorrhagic activity of the isolated enzyme as a potential pharmacopuncture agent. Methods: The fibrinolytic enzyme was isolated by using chromatography, sodium dodecyl sulfate-polyacrylamide gel electrophoresis, and fibrin plate assay. The characteristics of the enzyme were determined by using fibrin plate assay, protein hydrolysis analysis, and hemorrhage assay. Its amino acid composition was determined. Results: The fibrin(ogen)olytic enzyme with the molecular weight of 27 kDa (FE-27kDa) isolated from G. b. siniticus venom consisted of two heterogenous disulfide bond-linked polypeptides with the molecular weights of 15 kDa and 18 kDa. When more than $20{\mu}g$ of FE-27kDa was applied on the fibrin plate, fibrinolysis zone was formed as indicating its fibrinolytic activity. The fibrinolytic activity was inhibited completely by phenylmethanesulfonylfluoride (PMSF) and ethylenediaminetetraacetic acid (EDTA) and partially by thiothreitol and cysteine. Metal ions such as $Hg^{2+}$ and $Fe^{2+}$ inhibited the fibrinolytic activity completely, but $Mn^{2+}$ did not. FE-27kDa preferentially hydrolyzed ${\alpha}$-chain of fibrinogen and slowly hydrolyzed ${\beta}$-chain, but did not hydrolyze ${\gamma}$-chain. High-molecular-weight polypeptides of gelatin were hydrolyzed partially into polypeptides with molecular weights of more than 45 kDa. A dosage of more than $10{\mu}g$ of FE-27kDa per mouse was required to induce hemorrhage beneath the skin. Conclusion: FE-27kDa was a serine proteinase consisting of two heterogeneous polypeptides, hydrolyzed fibrin, fibrinogen, and gelatin, and caused hemorrhage beneath the skin of mouse. This study suggests that the potential of FE-27kDa as pharmacopuncture agent should be limited due to low fibrinolytic activity and a possible side effect of hemorrhage.

Anti-obesity Effects and Safety of the Mixture of Herbal Extracts in 3T3-L1 Cells and HR-1 Mice Fed a High Fat Diet (3T3-L1 지방전구세포와 고지방 식이로 유도된 비만 HR-1 마우스 피부에 도포한 한약 추출 복합물의 항비만 효과 및 안전성 평가)

  • The aim of this study was to investigate whether a novel formulation of an herbal extracts has an inhibitory effect on obesity. To determine its anti-obesity effects, we performed anti-obesity-related experiments in vitro and in vivo. Thus, our present study was carried out to evaluate the anti-obesity effect of herbal extracts using a high fat diet (HFD)-induced obese mouse model and 3T3-L1 adipose cells. The effects of each herbal extracts on lipid accumulation in 3T3-L1 cells were examined using Oil Red O staining. Results showed that treatment with each herbal extracts at $10{\sim}100{\mu}g/ml$ had no effect on cell morphology and viability. Without evidence of toxicity, herbal extracts treatment decreased lipid accumulation compared with the untreated adipocytes controls as shown by the lower absorbance of Oil Red O stain. Futhermore, compared with control-differentiated mature adipocytes, each herbal extracts significantly inhibited lipid accumulation in mature 3T3-L1 adipocytes. In the HFD-fed obese mice, body weight, liver weight and white adipose tissue weights were significantly reduced by mixture of herbal extracts administration in mouse skin. Futhermore, we found that mixture of herbal extracts administration suppressed serum triglyceride (TG), and total cholesterol (TCHO) in HFD-induced obese mouse model. The mixture of herbal extracts of permeability was estimated by measuring the transepithelial electrical resistance (TEER) value in pig skin. The optimized formulations of herbal extracts (Test 3 formulation) showed skin permeation. However, test 1 formulation containing essential oil as enhancer showed maximum skin permeation. After confirming the enhanced skin permeability, in vivo studies were performed to assess whether skin irritation potential on the basis of a primary irritation index (PII) in rabbit skin. Reactions were scored for erythema/edema reactions at 24 h, 48 h and 72 h post-application. It was concluded that the test 1 formulation was not irritation (PII = 0). The present study suggests that the test 1 formulation might be of therapeutic interest with respect to the treatment of obesity.

Effects of Egg Shell Membrane Hydrolysates on Anti-Inflammatory, Anti-Wrinkle, Anti-Microbial Activity and Moisture-Protection

  • This study was conducted to examine the effects of eggshell membrane hydrolysates (ESMH) on the anti-inflammatory, anti-wrinkle, anti-microbial activity, and moisture-protection for cosmetic use. Whole ESMH (before fractionation), and fraction I (>10 kDa), fraction II (3-10 kDa), and fraction III (<3 kDa) of the hydrolysates were assessed in this experiment. As lipopolysaccharide (LPS) and IFN-${\gamma}$ caused the inflammation on Raw264.7 cell, whole ESMH and fraction I showed to be effective in inhibiting the induction of cell inflammation depending on the concentration, and also showed outstanding effect to suppress the skin inflammation. Fraction I inhibited collagenase and elastase activities to a greater extent than the other fractions, while all fractions had antibiotic effects at concentrations of 10 mg/disc and 20 mg/disc. In addition, it showed the moisture protection effects of skin on the holding amount and losing amount of moisture in upper-inner arm of the human body with a relatively low loss rate in skin, which confirmed that the hydrolyzed fractions of ESM helps to form the superior protective layer of moisture. It was concluded that ESMH fractions with different molecular weights, especially the 10 kDa fraction, have anti-lipopolysaccharide, anti-IFN-${\gamma}$-induced inflammation, anti-collagenase and elastase activities, and thus can be used as a cosmetic agent to protect skin.

Real-Time Human Tracking Using Skin Area and Modified Multi-CAMShift Algorithm (피부색과 변형된 다중 CAMShift 알고리즘을 이용한 실시간 휴먼 트래킹)

  • In this paper, we propose Modified Multi CAMShift Algorithm(Modified Multi Continuously Adaptive Mean Shift Algorithm) that extracts skin color area and tracks several human body parts for real-time human tracking system. Skin color area is extracted by filtering input image in predefined RGB value range. These areas are initial search windows of hands and face for tracking. Gaussian background model prevents search window expending because it restricts skin color area. Also when occluding between these areas, we give more weights in occlusion area and move mass center of target area in color probability distribution image. As result, the proposed algorithm performs better than the original CAMShift approach in multiple object tracking and even when occluding of objects with similar colors.

A Diagnosis Method of Basal Cell Carcinoma by Raman Spectra of Skin Tissue using NMF Algorithm (피부 조직의 라만 스펙트럼에서 NMF 알고리즘을 통한 기저 세포암 진단 방법)

  • Basal cell carcinoma (BCC) is the most common skin cancer and its incidence is increasing rapidly. In this paper, we propose a diagnosis method of basal cell carcinoma by Raman spectra of skin tissue using the NMF(non-negative matrix factorization) algorithm. After preprocessing steps, measured Raman spectra is used classification experiments. The weight and the basis can be obtained in a simple matrix operation and a column vector of the matrix decompsed by the NMF. Linear combination of bases and weights, it is possible to approximate the average of Raman spectra. The classification method is to select the class which to minimize the root mean square of the difference of the linear combination and the objective spectrum. According to the experimental results, the proposed method shows the promising results to diagnosis BCC. In addition, it confirmed that the proposed method compared with the previous research result could be effectively applied in the analysis of the Raman spectra.

Bioactive and Skin-compatible Properties of Silk Sericin (세리신 단백질의 생리활성과 피부친화성 탐색)

  • Silk sericin protein was extracted by treatment with enzyme or NaOH solution from raw silk and culled-cocoon shell. The extracted sericin was characterized and examined the functional properties as well as subjective properties for its use as a soap and a body cleaner. The optimum extraction conditions on specimen are NaOH (0.02 wt.%) or enzyme, Flavourzyme 3% under N2 gas. Molecular weights of sericin was decreased by treatment with enzyme, Actinase, from 10,000-30,000 to 2,700-4,200. Sericin showed important bioactive properties, for instance, lowering effect on blood glucose and alcohol. Subjective test of sericin soap and body cleaner showed superior in washability, foamability, and skin hydration, etc., to commercial soap and body cleaner. Therefore, it is thought that silk sericin can be expected as the source of bioactive and skin-compatible materials.

Skin Color Detection Using Partially Connected Multi-layer Perceptron of Two Color Models (두 칼라 모델의 부분연결 다층 퍼셉트론을 사용한 피부색 검출)

  • Skin color detection is used to classify input pixels into skin and non skin area, and it requires the classifier to have a high classification rate. In previous work, most classifiers used single color model for skin color detection. However the classification rate can be increased by using more than one color model due to the various characteristics of skin color distribution in different color models, and the MLP is also invested as a more efficient classifier with less parameters than other classifiers. But the input dimension and required parameters of MLP will be increased when using two color models in skin color detection, as a result, the increased parameters will cause the huge teaming time in MLP. In this paper, we propose a MLP based classifier with less parameters in two color models. The proposed partially connected MLP based on two color models can reduce the number of weights and improve the classification rate. Because the characteristic of different color model can be learned in different partial networks. As the experimental results, we obtained 91.8% classification rate when testing various images in RGB and CbCr models.

Anti-Obesity Effect of Herbal Mixture Powder and Water Extracts in Mice Fed a High Fat Diet (나복자, 의이인, 백복령, 다엽, 차전자피로 구성된 혼합분말제(HP)와 진피, 산사, 옥미수, 양총피로 구성된 혼합열수추출물(HE)이 고지방식이로 유도된 비만마우스에 미치는 항비만 효과)

  • Objective : This study was conducted to investigate anti-obesity effects of mixed-Powder(Raphani Semen, Coicis Semen, Poria, Plantaginis Testa & Theae folium)(HP) & mixed water extract(Citri Pericarpium, Crataegi Fructus, Maydis Stigma, Allium Skin) (HE) in mice fed a high-fat diet. Method : 50 Female ICR mice were divided into six groups; Normal group (Ve), high-fat diet group (Po), Po+HP(0.6g/kg) group(HP), Po+HE(1g/kg) group(HE), Po+(HP+HE) group(HPE), Po+(HP+HE)*2 group(DHPE). For 8 weeks, these groups were fed their respective diets. Body weight, liver weight and weights of adipose tissues, GOT/GPT index changes & Blood Glucose index changes were measured respectively. Lipid profiles in serum were analyzed by kit of blood. Results : Groups HP, HE, HPE, DHPE had significantly reduced body weight, liver weight, adipose tissue weights and GOT/GPT index, blood glucose index compared to the Po group. Also, serum triglyceride and total cholesterol, low density lipoprotein, were significantly reduced when compared to the Po group. Group G1 had significantly increased high density lipoprotein levels. Conclusion : Mixed powder (HP) & mixed water extract (HE) show anti-obesity effects in mice fed a high-fat diet.

Effects of DHU001, a Mixed Herbal Formula on Acute Inflammation in Mice

  • The effects of DHU001, a mixed herbal formula consisted of 7 types aqueous extracts for treating respiratory disorders were observed on xylene-induced acute inflammation. The xylene was topically applied 60 min after administration of 500, 250 and 125 mg/kg of DHU001, and all animals were sacrificed 2 hrs after xylene application. The changes on ear weights, histolopathological analyses of ear were evaluated and compared to those of indomethacin and dexamethasone(15 mg/kg treated)-Both of drugs are well-known by anti-inflammatory agents. Xylene application resulted in marked increases in induced ear weights as compared with intact control ear. Severe vasodilation, edematous changes of ear skin and increase in the thickness of the ear tissues, neutrophil infiltration as acute inflammation were detected in xylene-treated control ears at histopathological observation. However, these xylene-induced acute inflammatory changes were dose-dependently decreased by oral treatment of DHU001. Therefore, it is concluded that DHU001 has favorable anti-inflammatory effects on xylene-applicated acute ear inflamed mice.
