Levosulpiride, (S)-(-)-5-Aminosulfonyl-N-[(1-ethyl-2-pyrrolidinyl) methyl]-2-methoxybenzamide, enhances the transduction efficiency of PEP-1-ribosomal protein S3 in vitro and in vivo

  • Many proteins with poor transduction efficiency were reported to be delivered to cells by fusion with protein transduction domains (PTDs). In this study, we investigated the effect of levosulpiride on the transduction of PEP-1 ribosomal protein S3 (PEP-1-rpS3), and examined its influence on the stimulation of the therapeutic properties of PEP-1-rpS3. PEP-1-rpS3 transduction into HaCaT human keratinocytes and mouse skin was stimulated by levosulpiride in a manner that did not directly affect the cell viability. Following 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced inflammation in mice, levosulpiride alone was ineffective in reducing TPA-induced edema and in inhibiting the elevated productions of inflammatory mediators and cytokines, such as cyclooxygenase-2, inducible nitric oxide synthase, interleukin-6 and -1${\beta}$, and tumor necrosis factor-${\alpha}$. Anti-inflammatory activity by PEP-1-rpS3 + levosulpiride was significantly more potent than by PEP-1-rpS3 alone. These results suggest that levosulpiride may be useful for enhancing the therapeutic effect of PEP-1-rpS3 against various inflammatory diseases.

Effect of Amniotic Fluid Stem Cells and Amniotic Fluid Cells on the Wound Healing Process in a White Rat Model

  • Background Amniotic-fluid-derived stem cells and amniocytes have recently been determined to have wound healing effects, but their mechanism is not yet clearly understood. In this study, the effects of amniotic fluid stem cells and amniocytes on wound healing were investigated through animal experiments. Methods On the back of Sprague-Dawley rats, four circular full-thickness skin wounds 2 cm in diameter were created. The wounds were classified into the following four types: a control group using Tegaderm disc wound dressings and experimental groups using collagen discs, amniotic fluid stem cell discs, and amniocyte discs. The wounds were assessed through macroscopic histological examination and immunohistochemistry over a period of time. Results The amniotic fluid stem cell and amniocyte groups showed higher wound healing rates compared with the control group; histologically, the inflammatory cell invasion disappeared more quickly in these groups, and there was more significant angiogenesis. In particular, these groups had significant promotion of epithelial cell reproduction, collagen fiber formation, and angiogenesis during the initial 10 days of the wound healing process. The potency of transforming growth factor-${\beta}$ and fibronectin in the experimental group was much greater than that in the control group in the early stage of the wound healing process. In later stages, however, no significant difference was observed. Conclusions The amniotic fluid stem cells and amniocytes were confirmed to have accelerated the inflammatory stage to contribute to an enhanced cure rate and shortened wound healing period. Therefore, they hold promise as wound treatment agents.

임상간호사와 보건간호사가 지각한 가정간호 교육내용의 중요도 및 업무수행능력 (Importance of Education Contents and Competencies on Home Care Nursing of the Hospital and Public Health Nurses in Korea)

  • This study was conducted during the time period of September 1997 to February 1999. in order to provide data concerning important facets of horne care nursing and the degree to which practicing hospital and public health nurses believe themselves to be competent in each area. The study subjects consisted of 610 hospital nurses, 158 public health nurses and 21 other nurses from Seoul and the province of Kyunggi. Korea. Data was collected through self-reporting questionnaires which was used by Kim et al.(1999) to evaluate the status of home health nursing and the varying ideas of self-competence that practicing nurses have. The results of the study were as follows: 1) The mean scores of perceived important components and competencies on home health nursing measured on the Likert 4 point scale were $3.15{\pm}0.36$ for importance. and $2.56{\pm}0.36$ for competency. Of the four categories regarding nursing services, the 'Nursing skill' factor had the highest importance and competency. 2) There were significantly higher scores for hospital nurses' importance components and competencies of home care nursing as compared to those scores for public health nurses. 3) The significant factors of the importance component of home care nursing listed in order of priority were 'general infection control', 'bed sore care skill', 'rehabilitation care', 'keeping maintenance of the client's confidentiality', 'malpractice', 'diabetes patient care', 'ability for problem solving' respectively. In contrast, of low priority in the importance components of home care nursing were 'referring nursing as a job to others'. 'record keeping', 'family dynamics', 'medical equipment', 'economic problems', 'environmental assessment', 'suture removal', 'multidisciplinary coordination' respectively. 4) The high priority of the degree to which practicing nurses believe themselves to be competent in home health nursing were the 'blood sugar test', 'enema skill', 'injection skill', 'skin care', 'bed sore care skill', etc. In contrast, lower competence for home care nursing as perceived by nurses were 'AIDS care', 'family violence care', 'substance abuse care', 'mental health care' respectively.

Advancement and Application of Somatic Cell Nuclear Transfer Technique in Dog

  • The cloning of canids was succeeded in 2005, several years after the birth of Dolly the sheep and also after the cloning of numerous other laboratory and farm animal species. The delay of successful somatic cell nuclear transfer (SCNT)was due to the unique reproductive characteristics of the female dogin comparison to other domestic mammals, such as ovulation of immature canine oocyte and a requirement of 25 days for the completion of meiosis within the oviduct (Holst & Phemister, 1971). When the technology for the recovery of in vivo matured oocyte was established, the application of cloning also became possible and cloned dog offspring were obtained. This report summarizes the progress of technical procedures that are required for cloning canids and the application of this technique. The first cloned dog, Snuppy, was achieved using an in vivo-matured oocyte which was enucleated and transferred with an adult skin cell of male Afghan hound. After establishment of a criterion of well-matured oocyte for the improvement of SCNT efficiency, we obtained three cloned female Afghan hound and a toy poodle cloned from 14 year-old aged Poodle using SCNT through this factor. To date, cloned dogs appeared to be normal and those that have reached puberty have been confirmed to be fertile. Through application of canine SCNT technique, first, we demonstrated that SNCT is useful for conserving the breed of endangered animal from extinction through cloning of endangered gray wolves using inter-species SCNT and keeping the pure pedigree through the cloning of Sapsaree, a Korean natural monument. Secondly, we showed possibility of human disease model cloned dog and transgenic cloned dog production through cloning of red fluorescent protein expressing dog. Finally, SCNT can be used for the propagation of valuable genotypes for making elite seed stock and pet dog. In summary, dog cloning is a reproducible technique that offers the opportunity to preserve valuable genetics and a potential step towards the production of gene targeted transgenic cloned dogs for the study of human diseases.

Childhood Arsenic Exposure and Health Risk Assessment from Rice Cultivated Near the Mining Areas in Korea

  • 우리나라 유아들이 비소에 노출된 경우의 인체위해성에 대한 상관관계 연구가 미비한 실정이다. 본 연구는 우리나라 광신인근 지역에서 생산된 백미의 섭취를 통한 비소의 노출을 유아기(1-6세)와 타 연령군으로 나누어 평가하고 해당 연령군의 인체위해성 평가를 확률적 접근법을 이용해 실시하였다. 우리나라 광산인근 지역에서 생산된 백미 섭취를 통한 D아기의 비소 노출량은 체중을 기준으로 했을 때 타 연령군에 비해 높게 나타났다. 피부암에 대한 초과발암위해의 경우 1-2세 군과 3-6세 군의 50% percentile값이 각각 만명 당 1.76명과 3.16명으로 나타났다. 어린이와 같은 아만성기간 동안의 비소 노출에 대한 비발암 기준 참고치(0.005 mg/lg/day)를 적용한 결과 비발암위해도의 평균값 뿐 아니라 고섭취군을 나타내는 95% percentile 값 또한 규제수치인 1.0 이하로 나타났다.

대식세포 내에서의 홍화자 추출물의 항염증 활성 (Anti-Inflammatory Activity of Carthamus tinctorious Seed Extracts in Raw 264.7 cells)

  • 본 연구에서는 홍화자 추출물이 항염증에 대한 실험연구가 이루어져 있지 않은 것에 착안하여 LPS에 의해 활성화된 대식세포로부터 유도되는 염증반응에 대한 억제효과를 조사하였다. 홍화자 추출물을 이용하여 피부 염증에 대하여 연구를 하였다. 산화질소와 cytokine의 생산은 면역세포의 대표적인 염증인자이다. 세포는 LPS 처리 후 한 시간 뒤에 홍화자 추출물을 처리를 하였다. 세포 독성이 나타나지 않는 농도인 5, 10, 25 및 50 ${\mu}g/ml$를 사용하였다. 홍화자의 에틸아세테이트 분획물은 NO, $PGE_2$, TNF-${\alpha}$, IL-$1{\beta}$, IL-6, iNOS, COX-2의 생성을 저해 시켰다. $PGE_2$는 50 ${\mu}g/ml$의 농도에서 60%에 가까운 저해율을 나타내었다. iNOS와 COX-2 역시 ${\mu}g/ml$의 농도에서 각각 54%, 65%가 저해가 되었다. 게다가 홍화자 에틸아세테이트 분획물은 염증성 사이토카인인 TNF-${\alpha}$, IL-$1{\beta}$, IL-6의 생성을 감소 시켰다. 이러한 결과로 홍화자 추출물은 염증 예방과 치료에 효과적임을 확인 할 수 있었다.

애기뿔소똥구리 유래 CopA3합성 펩타이드의 항염증 효능에 관한 연구 (Study of Anti-inflammatory Effect of CopA3 Peptide Derived from Copris tripartitus)

  • CopA3를 이용하여 피부 염증에 대하여 연구를 하였다. 산화질소와 cytokine의 생산은 면역세포의 대표적인 염증인자이다. 세포는 LPS 처리 후 한 시간 뒤에 CopA3를 처리하였다. 세포 독성이 나타나지 않는 농도인 5, 25, 50, 100 ${\mu}g/ml$를 사용하였다. CopA3는 NO, TNF-${\alpha}$, IL-$1{\beta}$, IL-6, iNOS, COX-2의 생성을 저해 시켰다. iNOS와 COX-2 역시 100 ${\mu}g/ml$의 농도에서 각각 54%, 65%가 저해가 되었다. 게다가 CopA3는 염증성 사이토 카인인 TNF-${\alpha}$, IL-$1{\beta}$, IL-6의 생성을 감소 시켰다. 이러한 결과로 CopA3는 염증 예방과 치료에 효과적임을 확인 할 수 있었다.

저분자 당단백을 함유하는 다마스크 장미추출물의 항노화 효과 (Anti-aging Effects of Rosa damascena Extract Containing Low Molecular Glycoprotein)

  • 본 연구에서는 다마스크 장미 내 고분자 당단백을 bioconversion 기술을 이용하여 저분자화한 다마스크 장미추출물(Rosa damascena extract containing low molecular glycoprotein, RELG)의 항노화 효능을 측정하였다. DPPH 라디칼 소거능을 이용한 RELG의 항산화능 평가($IC_{50}$)는 $22.6{\mu}g/mL$으로 양성대조군인 ascorbic acid $21.1{\mu}g/mL$와 비교하여 비슷한 수준의 항산화능을 나타내었고 피부세포실험에서는 $15{\mu}g/mL$에서 자외선과 $H_2O_2$에 의한 활성산소 생성을 28% 억제하였다. 또한 같은 농도에서 자외선에 의한 콜라겐 분해효소(MMP-1)의 생성을 48% 억제하고 $10{\mu}g/mL$에서 대조군 대비 보습인자인 aquaporin 3 (AQP3)의 발현을 44%, 중성지질 생합성을 10% 촉진하여 항주름 효능과 더불어 보습효능까지 나타냈다. 추가적으로 두피 모유두세포에서는 스트레스로 인한 세포사멸을 $15{\mu}g/mL$에서 10% 억제하고 활성산소의 생성을 90% 감소시켰다. 본 연구 결과, RELG는 효과적인 항노화 화장품 소재임을 확인하였다.

중환자실 환자의 욕창 관련 경계압력 예측요인 (Factors Predicting the Interface Pressure Related to Pressure Injury in Intensive Care Unit Patients)

  • Purpose: Interface pressure is a factor that contributes to the occurrence of pressure injuries. This study aimed to investigate interface pressure at common sites of pressure injury (occipital, gluteal and peritrochanteric areas), to explore the relationships among risk factors, skin condition and interface pressure, and to identify risk factors influencing interface pressure. Methods: A total of 100 patients admitted to the intensive care unit were enrolled at a tertiary teaching hospital in Korea. Interface pressure was recorded by a scanning aid device (PalmQ). Patient data regarding age, pulmonary disease, Braden Scale score, body mass index, serum albumin, hemoglobin, mean blood pressure, body temperature, and oxygen saturation were included as risk factors. Data collected from July to September 2016 were analyzed using binary logistic regression. Results: The mean interface pressure of the occipital, gluteal, and right and left peritrochanteric areas were 37.96 (${\pm}14.90$), 41.15 (${\pm}16.04$), 53.44(${\pm}24.67$), and 54.33 (${\pm}22.80$) mmHg, respectively. Predictive factors for pressure injuries in the occipital area were age ${\geq}70$ years (OR 3.45, 95% confidence interval [CI]: 1.19~9.98), serum albumin deficit (OR 2.88, 95% CI: 1.00~8.26) and body temperature ${\geq}36.5^{\circ}C$ (OR 3.12, 95% CI: 1.17~8.17); age ${\geq}70$ years (OR 2.81, 95% CI: 1.10~7.15) in the right peritrochanteric area; and body temperature ${\geq}36.5^{\circ}C$ (OR 2.86, 95% CI: 1.17~6.98) in the left peritrochanteric area. Conclusion: Our findings suggest that old age, hypoalbuminemia, and high body temperature may be contributory factors to increasing interface pressure; therefore, careful assessment and nursing care of these patients are needed to prevent pressure injury. Further studies are needed to establish cutoff values of interface pressure for patients with pressure ulcers.

두통(頭痛)의 원인에 따른 형상의학적(形象醫學的) 고찰 -동의보감(東醫寶鑑) 두문(頭門)을 중심으로 (Review on the Causes of Headache in Hyungsang Medicine)

  • The followings are concluded from the treatment of headache in Hyungsang medicine, focussed on 11 kinds of headaches in Donguibogam. Headache is classified into overall headache and migraine according to the affected region. The causes are divided into exogenous affection and internal injury; The former brings on headache due to Wind-Cold and headache due to Damp-Heat. The latter, reversal headache, headache due to adverse rising of phlegm, headache due to regurgitation of Gi, headache due to excessive Heat, headache due to excessive Damp, true headache, and alcoholic headache. Headache due to internal injury generally tends to show deficiency syndrome with external affection. Headache due to exogenous affections is common to those who have big head or white skin and to Bangkwang type, and woman. The primary causes are Wind-Cold and Wind-Heat. When the body is observed in the perspective of eight phases, Damp-Heat is to be produced in the front, and Dry-Damp, in the back. Headache due to Damp-Heat is susceptible to Yangmyeong meridian type whose body develops more in the front and to woman. In the perspective of the upper and the lower, Yangdu(that is, head) is related to Eumdu(that is, glans of penis). Headache is also caused by the problems of Eumdu ,such as deficiency of Essence in man, pathologic change of uterus in women, and San syndrome in lower abdomen. In the case of man, headache is frequently severe and difficult to treat because head is a root for man. Disharmony of Gi and blood between the right and the left brings out migraine and headache due to regurgitation of Gi. Migraine is usually accompanied by symptoms of exogenous affection and often afflicts Gi-type, Shin-type, Soyang meridian type, deer type, and Dam-type. Headache due to regurgitation of Gi is brought by Gi deficiency or blood deficiency so that symptoms of exogenous affection do not show. It is mainly common with old people and those who have sunken eyes induced by deficiency of stomach Gi. In the perspective of the upper, the middle, and the lower, the pathologic change of head, chest and abdomen also bring about headache. The pathologic cause of head is Wind-Heat ,which triggers overall headache, migraine, headache due to Wind-Cold, headache due to excessive Heat, The pathogen of chest is phlegm-Fire and brings out headache due to Damp-Heat and headache due to adverse rising of phlegm. The pathologic factor in abdomen is Cold-Damp and produces headache due to adverse rising of phlegm and headache due to excessive Damp. In case of women, headache is generally caused by phlegm-Fire and retention of undigested food.
